1.Knock-down ATG5 gene inhibits autophagyand enhances celastrol-induced apoptosis in human lung cancer cell H1299

Objective To establish the lung cancer cell strain with low ATG5 expression and to detect the effect of celastrol on lung cancer cell apoptosis after downregulation of autophagy.Methods H1299 was infected by lentivirus-mediated ATG5 shRNA.RT-qPCR and Western blot assays were applied to confirm the effect of ATG5 knock down.Autophagy was measured by Western blot and RFP-LC3 transfection.Cell apoptosis of ATG5 normal expression group and of ATG5 low expression group of H1299 cells was detected by FACS.Finally, Western blot was used to detect the expression of apoptosis-related proteins Bcl-2, Bax and cleaved caspase-3.Results The expression of ATG mRNA and protein significantly decreased after ATG5 knockdown in H1299 cells (P<0.05).The autophagy marker of LC3-Ⅱ level was downregulated and P62 expression was upregulated after inhibition of ATG5, and the RFP-LC3 puncta reduced significantly after ATG5 knockdown (P<0.05).Compared with control group,the apoptosis rate in ATG5 downregulation group increased significantly after celastrol treatment (P<0.01).Pro-apoptotic proteins of Bax and cleaved caspase-3 levels were upregulated and anti-apoptotic protein of Bcl-2 level decreased after ATG5 inhibition (P<0.05).ConclusionsThe effect of celastrol-induced apoptosis of lung cancer cells was enhanced after downregulation of autophagy, demonstrating inhibition autophay may be a new target of lung cancer treatment.

Intracerebral Hemorrhage (ICH) is a common cerebrovascular disease with high disability rate and high mortality rate.A large number of clinical and experimental studies have shown that the abnormal metabolism of iron in the brain tissue around the hematoma after ICH is an important cause of secondary brain damage such as brain edema and neuronal apoptosis.It is an important factor affecting the outcome of patients.This article reviews the abnormal metabolism of iron and its significance after ICH.
4.Study of positive results of electrocardiogram treadmill exercise test for patients with chest pain

Objective To correctly analyze the positive results of electrocardiogram treadmill exercise test (TET) and improve the level of diagnosis of CHD Methods Two hundred and fifty eight patients with chest pain underwent TET and CAG; the case history and different clinical parameters were collected (1) Clinical data of the true positive group and the pseudo positive group were compared and underwent a statistic analysis (2) Clinical data of the female positive group were compared and underwent a statistic analysis Results The number of females in the pseudo positive group was larger than in the true positive group;The number of patients with typical chest pain, hyperlipidemic history, history of smoking, family history of CHD was smaller than in the true positive group;In the pseudo positive group,the changes of ST segment were mainly in Ⅱ, Ⅲ and aVF The number of extent of changes of more than 0 2 mV and with angina induced during exertion in pseudo positive group was smaller than in true positive group In females, under 60 years, the number of ST segment changes of resting ECG and left ventricular hypertrophy was larger in pseudo positive group than in true positive group The difference was significant Conclusion The factors including sex, typical chest pain,risk factors of CHD, the location and extent of ECG changes and angina induced following exertion should be combined to analyze the positive results of TET
5.Inhibitory Effect of Chinese Herbal Compound Dongju on Tongue Carcinogenesis in Rats

Objective To examine the inhibitory effect of Chinese herbal Compound Dongju on 4-nitroquinoline 1-oxide(4NQO)-induced tongue carcinogenesis in rats. Methods A total of 85 Wistar rats were divided into four groups: model group(n=35),administration group(n=35),normal control group(n=5) and herb control group(n=10).4NQO dissolved in drinking water was administered orally to induce tongue carcinogenesis in rats.The herb compound was injected into the stomach of the rats during oral carcinogenesis.Rats were sacrificed at 9,13,20,24 and 32 weeks from the beginning of the experiment.The tongues of the rats were dissected for gross and histological assessment,and SP immunohistochemical method was also employed to detect the expression of cyclin D1 in the specimens. Objective To study the relationship between the single nucleotide polymorphisms (SNP)of X-ray repair cross-complementing group-3(XRCC3),human 8-hydroxyguanine glycosylase-1 (HOGG1)gene of repair DNA and the radiosensitivity in esophageal squamous cell carcinoma(ESCC).Methotis Genome DNA was exracted from whole blood cells of ESCC patients.XRCC3 and HOGG1 genotypes were determined by polymerase chain reaction-restriction fragment length polymorphism method.The relationship was analyzed between SNP of XRCC3 Thr242 Met,HOGG1 Ser326 Cys and the response to radiotherapy in ESCC.Results Totally 199 ESCC patients were treated with radical radiotherapy.The response rate w88 81.4%.The response rate in patients with the XRCC3 Thr/Met genotype was significant higher than that in patients with Thr/Thr genotype(91.5%vs 77.1%,X2=5.67,P=0.017),with the same in HOGGlSet/Ser,Ser/Cys and Cys/Cys genotype(74.1%,84.2%,83.9%,X2=2.64,P=0.268).The esophageal radioactivity injury rate in patients with the XRCC3 Thr/Met genotype was also significant higher than that in patients with Thr/Thr genotype(35.6%vs 20.0%,X2=5.44,P= 0.020),with the same in HOGG1 Ser/Ser,Ser/Cys and Cys/Cys genotype(24.1%,24.6%,25.8%,X2=0.03,P=0.984).In Iogistic multivariate analysis,the SNP of XRCC3 was the independent influencing factor closely correlating with the mdiosensitivity of esophageal cancer(X2=16.12,4.43,P=0.009.0.035).No significant difierence was observed between the SNP in HOGG1 Ser326 Cys and the response rate of radiotherapy or the rate of esophageal radioactivity injury(X2=3.74,0.58,P=0.053,0.445).Conclusions SNP of XRCC3 Thr/Met is associated with the radiosensitivity in ESCC patients,which suggests that the XRCC3 Thr/Met SNP may be a predictor for ESCC patients who likely to response to radiotherapy.
8.How to deal with cerebral palsy in 21st century--a new epoch in clinic treatment.

The aims of this paper were to define (1) criteria of cerebral palsy; (2) classification of cerebral palsy; (3) etiology, neuroimaging, and epidemiology of cerebral palsy; (4) different kinds of treatments of cerebral palsy. Data were drawn from an international survey of PUBMED (1994-2014) and CNKI (1994-2014). An expert panel used a consensus building technique. The10-point Jadad scale was used to assess the quality of the trials based on the following items, including allocation sequence generation, randomization concealment, methods of blinding, and descriptions of withdrawals and dropouts. Our clinical experience was also summarized. Below is a summary. (1) Further work is warranted to reach agreement for the classification of cerebral palsy. (2) A worldwide prevalence of 1.5-4.0 per 1 000 live births, with an average lifetime cost of 1 million dollars per person in the United States, while it is 1.8-6.0 per 1000 live births in China. (3) Comparison of clinical efficacy of different treatments. In this review, the current advances in different kind of treatments of brain injury are discussed with specific relevance to cerebral palsy.

9.Diagnostic Value of Breast Galactography in Nipple Discharge Diseases

Objective To explore the value of galactography in diagnosis of nipple discharge diseases. Methods Galactographies in 101 cases with nipple discharge confirmed by operation and pathology were analyzed restrospectively. Results There were breast duct ectasia in 23 cases, breast cystic hyperplasia in 12 cases , ductal inflammation in 8 cases, intraductal papilloma in 46 cases, ductal carcinoma in 9 cases, ductal tuberculosis in 1 case and normal duct in 2 cases. Conclusion The galactography is beneficial in diagnosis of the pathogenesis of galactorrhea and is of great value in differential diagnosis between benign and malignant lesions.
