Acta Pharmaceutica Sinica 2001;36(4):296-299
AIM To study the electrochemical behavior of omeprazole (OPZ) and a new method for the determination of the compound was established by adsorptive stripping voltammetry. METHODS Linear-sweep polarography and cyclic voltammetry were used. RESULTS In a supporting electrolyte containing 0.1 mol*L-1 NH3-NH4Cl, a reduction peak of OPZ was observed by linear-sweep voltammetry at Hg electrode. The peak showed a potential of -1.04 V (vs Ag/AgCl at 100 mV*s-1) and adsorptive characteristics. The adsorbed species is most probably the neutral molecular of OPZ. The saturated adsorption of OPZ at Hg electrode is 9.55×10-11 mol*cm-2 and every OPZ molecule occupies an area of 1.74 nm2. On the surface of the hanging mercury drop electrode, the adsorption of OPZ obeys Frumkin adsorption isotherm. The adsorption coefficient β is 1.32×106, the Gibbs energy of adsorption ΔGθ is -34.93 kJ*mol-1, the number of electrons transferred n is 4, the kinetics parameter αnα is 0.78, the rate constant of the surface electrode reaction ks is 0.18 s-1, the diffusion coefficient D of OPZ in solution is 1.03×10-5 cm2*s-1. The detection limit of the adsorptive stripping voltammetry of OPZ is 2.0×10-9 mol*L-1 under optimized condition. CONCLUSION This indicates an irreversible process with adsorption.

Objective To test the hypothesis that isoflurane-preconditioning (ISO-P) provides added protection of myocardium with hypothermia against ischemia-reperfusion injury through KATP channels. Methods Thirty-two SD rats of both sexes weighing 230-270 g were studied. The animals were anesthetized with intraperitoneal ketamine 100 mg?kg-1 and heparinized. Chest was opened and heart was immediately removed and perfused in a Langendorff apparatus with oxygenated (95% O2, 5% CO2) Krebs-Hensleit buffer (KHB) for 50 min, followed by 150 min of hypothermic-ischemia (perfusion was suspended and heart was immersed in 17℃ KHB) and 60 min of normothermic reperfusion (heart was again reperfused with 37.7℃ KHB and immersed in 37.7℃ KHB) (I/R). The animals were randomly divided into 4 groups of 8 animals:(l) control group; (2) ISO-P group; (3) ISO-P + GLB group and (4) ISO-P + 5-HD group. The control group received no pretreatment before I/R. In ISO-P group the heart was perfused with KHB gassed with 1% isoflurane for 15 min followed by 15 min wash-out before I/R. In group 3 and 4 the heart was perfused with KHB containing glibenclamide 20?mol?L-1 (group 3) or 5-hydroxydecanoate 150?mol ?L-1 (group 4) in addition to 1% isoflurane before I/R. Cardiac function was measured with a fluid filled latex balloon inserted in left ventricle (LVSP, LVDP, LVEDP, dp/dtmax, dp/dtmin). During hypothermic ischemia left ventricle pressure (LVP),S-time (the interval between the beginning of ischemia to the time point when LVP increased by 1 mm Hg from the baseline pressure) and C-time (the interval between the beginning of ischemia to the time point when LVP was lmmHg below the plateau pressure) were measured. Other criteria included coronary flow (CF),infarct size and creatine kinase. Results (l) S-time and C-time were negatively correlated with infarct size.(2)In ISO-P group (group 2) S-time and C-time were significantly longer during ischemia; LVDP and LVDP x HR were significantly higher during normothermic reperfusion and infarct size was significantly smaller than those in control group (groupl) (P

Medical Journal of Chinese People's Liberation Army 2001;0(11):-
Objective To compare the effect of pressure regulated biphasic positive airway pressure ventilation(BiPAP) and airway pressure release ventilation(APRV) on cardiopulmonary function of pigs with or without acute lung injury. Methods after anaesthesia, 18 healthy pigs were given BiPAP or APRV ventilation through tracheotomy. Inspiration pressure and expiration pressure in BiPAP equal to APRV. Inspiration time and expiration time is 1.5/2.5s in BiPAP and 4.0/2.0s, 4.5/1.5s, 5.0/1.0s, 5.5/0.5s in APRV respectively. Respiration dynamics, hemodynamics, blood gases and oxygen metabolize scales were measured during two mechanical ventilatory modalities pigs acquired from Swam-gans catheter in internal jugular vein and from catheter in femoral artery. Then oleic-induced acute lung injury model was made, modalities pigs were distributed to BiPAP group (n=9) and APRV group (n=8) randomly. Forenamed scales were repeat measured. Results BiPAP has not difference with APRV on respiration dynamics, hemodynamics, blood gases and oxygen metabolize during normal condition, BiPAP has higher cardiac output (CO) and lower oxygen extraction ratio (O2ER) compare with APRV in ALI pigs. If expiration time is short to 0.5s in APRV, it would represent intrinsic peak end-expiratory (PEEPi), heart rate (HR), mean pulmonary arterial pressure (MPAP) and systemic vascular resistance (SVR) increased and CO decreased. Conclusion Both BiPAP and APRV are beneficial in ALI, but BiPAP has fewer side-effect compared with APRV in early ALI.
