1.Pharmacokinetic Analysis for Lactosaminated Recombination Human Growth Hormone and Recombination Human Growth Hormone by Radioimmunoassay

OBJECTIVE:To study the pharmacokinetics of L-rhGH and rhGH in mice.METHODS:The concertrations of L-rhGH and rhGH in plasma and liver were measured by radioimmunoassay.RESULTS:After intravenous injection,there were significant differences between L-rhGH and rhGH in pharmacokinetic parameters.The Cmax of L-rhGH in the liver was higher than that of rhGH but the T1/2 of L-rhGH was only 17.9% of T1/2 of rhGH;the Cmax of L-rhGH in plasma was lower than that of rhGH and the T1/2 was shorter.CONCLUSION:L-rhGH reveals the pharmacokinetic characteristics which fits in with the physiology of human body.

As a main cellular organelle for bioenergy production , the mitochondrion plays a pivotal role in aerobic respiration , substance metabolism , oxidative stress , apoptosis and calcium homeostasis .Increasingly studies have shown a close relationship between mitochondrial dysfunction and cancer .Mitochondrial metabolic disturbance , reactive oxygen species ( ROS ) increase, mitochondrial gene mutation , calcium overload and abnormal apoptosis can influence tumorigenesis , growth, invasiveness and metastasis of multiple tumors .We aimed to summarize the mechanisms and influences of mitochondrial dysfunction on cancer .
3.Anticoagulation of Warfarin Therapy in Patients After Mechanical Valve Implantation

OBJECTIVE:To discuss the influencing factors of anticoagulation with warfarin in patients who had undergone artificial mechanical heart valve replacement and the relation between dosage regimens and international normalized ratio(INR).METHODS:123 patients who had undergone artificial valve replacement patients were followed-up.The patients were treated with warfarin therapy for anticoagulation after surgery,and given a regular follow-up.Patients' age,gender,and INR value etc were recorded and the adverse events including bleeding,embolism and death were observed.RESULTS:No death case was reported;femoral vein thrombosis occurred in 1 case;gingival bleeding occurred in 11 cases and urinary tract bleeding was noted in 3 case.The incidence of anticoagulation-associated ADR was close to that reported in overseas literature.CONCLUSION:According to INR,individual dose of warfarin should be administered on day 5 after artificial valve replacement.Maintaning INR at 1.5~2.0 can achieve certain anticoagulation efficacy and is relatively safe.
4.Carbamazepine-induced Adverse Reactions:Clinical Characteristics and Related Factors

OBJECTIVE:To study the clinical characteristics and related of the adverse drug reactions(ADR) induced by carbamazepine,and propose preventive solutions.METHODS: 102 cases of carbamazepine-induced ADR,reported in medical journals of China from January 2001 to December 2005,were collected and analyzed.RESULTS:Skin and its appendages received most impairment from carbamazepine-induced adverse reactions(representing 35.3%),followed by general impairment of the body and impairment of the blood & hematopoietic system.Combined use of drugs is liable to induce adverse reactions,indicated by the fact that 16.7% of the sufferers received treatment by combined use of multiple drugs.Most of the ADR occurred 1h after administration.CONCLUSION:Individual differences of patients and combined use of drugs are among the factors that result in carbamazepine-induced adverse reactions.
5.Analysis of the Clinical Characteristics and Related Factors in Occurrence of ADRs of Chinese Prepared Medicines

OBJECTIVE:To analyze the ADRs caused by Chinese prepared medicines.METHODS:638ADRs were col?lected from medical journals of2001~2002in consideration of sex,age,administration route,the time of the occurrence and clinical manifestations.RESULTS:There were638ADR cases involving76kinds of Chinese prepared medicines reported in256articles.Of them,shuanghuanglian injection was the most common preparation(87cases).The commonest clinical manifesta?tions were allergic reactions(157cases),followed with the liver and kidney injury.The ages of patients were mostly above60years,the time of the occurrence was mostly within30minutes after administration and about92%of the patients recovered within24hours.CONCLUSION:The occurrence of ADRs of Chinese prepared medicines is related to the route of administra?tion,dosage form of drug,quality of drug,patient’s age and constitution.

7.The effects of conditioned medium from keloid fibroblasts under hypoxia on angiogenesis.

OBJECTIVETo observe the effects of conditioned medium from keloid fibroblasts under hypoxia on angiogenesis, and to investigate the role of hypoxic microenvironment in invasive growth of keloid.
METHODSPrimary keloid fibroblasts and human umbilical endothelial cells (HUVEC) were cultured as conventional method. Keloid fibroblasts were cultured either in a hypoxic incubator (2% O2) for 48 h or in a normoxic incubator (20% O2) as control. Then those cell culture mediums were collected and mixed with endothelial cell medium by the proportion of 1:1 as conditioned medium. The mRNA and secreted protein of pro-angiogenic factors such as vascular endothelial growth factor (VEGF), angiopoietin-1 (Ang-1) and periostin of keloid fibroblasts under hypoxia were detected by real time PCR and ELISA. The proliferation, migration and invasion, tube formation of HUVEC cultured with conditioned medium were evaluated by CCK-8 assay, Transwell assay and matrigel tube formation assay, respectively.
RESULTSHypoxia increased the expression of VEGF, Ang-1 and periostin in both mRNA (increased by 75%, 43% and 118% respectively, P < 0.05) and secreted protein (increased by 30.2%, 14.2% and 19.5% respectively, P < 0.05) levels; the proliferations of HUVEC in hypoxic conditioned medium in 1, 2 and 3 d were 0.67 +/- 0.07, 0.84 +/- 0.09 and 1.08 +/- 0.10 respectively, which were higher compared to those in control group (0.52 +/- 0.08, 0.72 +/- 0.10 and 0.91 + 0.14, P < 0.05); the numbers of migration, invasion and tube formation of HUVEC were (73.2 +/- 8.9), (56.3 +/- 12.5), (9.66 +/- 1.96) cells/HP, which were higher compared to those in control group [(59.0 +/- 8.0), 35.5 +/- 8.5), (6.5 +/- 1.87) cells/HP, P < 0.05].
CONCLUSIONSHypoxia increases the expression of pro-angiogenic factors of keloid fibroblasts, and its conditioned medium under hypoxia could promote angiogenesis. The results suggest hypoxic microenvironment may play a significant role in the invasive growth of keloid by inducing angiogenesis.

8.Literature Analysis of Compatibility Taboos of Polyene Phosphatidyl Choline Injection

Objective: To analyze the compatibility taboo of polyene phosphatidyl choline injection with several kinds of commonly used clinical drugs.Methods: The medical literatures on the compatibility taboos of polyene phosphatidyl choline injection with several commonly used drugs were retrieved and statistically analyzed.Results: After the compatibility of polyene phosphatidyl choline injection with vitamin C, raceanisodamine injection, doxofylline and sodium chloride injection,ambroxol hydrochloride injection,ondansetron hydrochloride injection and the other commonly used drugs, physical and chemical reactions happened in varying degrees.Conclusion: Polyene phosphatidyl choline injection has compatibility taboos with a variety of commonly used drugs.
9.Clinical value of ultrasonography in the diagnosis of carotid plaque neovascularization

Objective To evaluate the clinical value of ultrasonography in the diagnosis of carotid plaque neovascularization .Methods 32 patients with carotid artery plaques diagnosed by conventional ultrasonography were enrolled.The patients were followed up for contrast-enhanced ultrasonography (CEUS) and ultrasonography micro-vascular imaging(SMI),and the results of the two-dimensional detection of carotid plaque and plaque neovasculariza-tion were compared .Results A total of 63 plaques were detected by SMI , including 39 hypoechoic plaques , 19 episodes of echo patches ,58 of plaques ,and 17 hypoechoic plaques .The diagnostic rate of neovascularization was 10.34%.Among the 113 carotid plaques,48 carotid plaques were detected by SMI ,46 carotid plaques were detected by CEUS.The distribution of neovascularization was similar to that of CEUS ,and the neovascularization of SMI and CEUS was better(Kappa=0.669,P<0.001).Conclusion For the diagnosis of carotid plaques ,ultrasonography can accurately detect the number , location and neovascularization of plaques , and the accuracy of carotid plaques is effective.It provides a radiographic basis for the diagnosis and early intervention of patients ,which has great signifi-cance to the prevention of stroke and cardiovascular and cerebrovascular adverse events .It is worthy of popularizing and application .
10.The histological study of the capsules formed around the mammary prosthesis.

OBJECTIVETo observe and analyze the pathohistological characteristics of capsules which formed around the mammary prosthesis with different contents. And to provide the selective basis for ideal and safe prosthesis in clinical practice.
METHODS20 specimen of the capsules were taken from 20 cases who receive the operation of prothesis removal for different reasons. HE, Masson and Mallory staining were used to analyse the tissue structure and characteristics under the light microscope.
RESULTSThe common structure including the collagen fibers accumulation, inflammatory cells infiltration and the capillary hyperplasia were found in all specimen. A layer of squamous epithelium-like cell was detected in some specimen. The specific characteristics were also found in different capsules formed around different prosthesis. In the capsules around vegetable oil prosthesis, there was excessive collagen fiber accumulation, and the capsules were much thicker. In the PVP (polyvinylpyrolidone) prosthesis capsules, there was severe inflammatory cell infiltration, and the number of eosinophilic granulocyte increased obviously. In the silicone gel and saline prosthesis capsule, the collagen fibers were well-arranged and the inflammatory cells were much less. Synovial metaplasia was detected in two cases.
CONCLUSION1. The capsules form around the prosthesis in all cases after mammary augmentation. 2. There will be synovial metaplasis in some cases, for vegetable oil prosthesis, the collagen over-accumulated which lead the capsules become thicker and harder. So it is not a kind of ideal mammary prothesis. 4. The severe infiltration of the inflammatory cells especially the large quantity of eosinophilic granulocyte indicate the possibility of the delayed hypersensitive reaction mediated by eosinophilic granulocyte. Cautious attitude should be taken during application.
