1.Relationship between benign prostatic hyperplasia and blood pressure variability in elderly patients with H type hypertension

Objective To investigate the correlation between benign prostatic hyperplasia (BPH) and blood pressure variability (BPV) in the elderly patients with H type hypertension.Methods 561 elderly patients with H type hypertension from 2010 to 2012 were enrolled in this study,including 436 BPH patients and 125 non-BPH (NBPH) patients.All patients underwent ambulatory blood pressure monitoring (ABPM) and ultrasound examination of prostate.The 24-hour systolic blood pressure variation (24 h SBPV),24 hour diastolic blood pressure variation (24 h DBPV),daytime systolic blood pressure variation (dSBPV),daytime diastolic blood pressure variation (dDBPV),nocturnal systolic and diastolic blood pressure variations were recorded and compared.The differences in blood pressure variation between the two groups were compared.Correlation between BPV and prostatic volume (PV) was analyzed.Results There were no statistical differences in variabilities of 24 h SBPV,nSBPV and dSBPV between BPH group and NBPH group(all P>0.05).24 h DBPV,nDBPV and dDBPV were significantly higher in BPH group than inNBPH group [(18.5±3.3) mmHg vs.(14.1±3.7) mmHg,(17.1±3.4) mmHg vs.(14.9 ±3.4) mmHg,(16.6±4.1) mmHg vs.(12.8±5.1) mmHg,respectively,all P<0.05].There were correlations of 24 h DBPV,nDBPV and dDBPV with PV (r =0.137,0.032,0.079,respectively,all P<0.05).Conclusions The diastolic blood pressure variability may associate with the degree of prostatic hyperplasia in the elderly patients with H type hypertension.
2.Age distribution of prostate specific antigen and its correlation with blood glucose and lipid in the male elderly

ObjectiveTo investigate the age distribution of serum prostate specific antigen (PSA) and explore its correlation with fasting blood glucose(FBG)and lipid in the male elderly.MethodsThe levels of FBG,triglyceride (TG),low-density lipoprotein (LDL-C),high density lipoprotein (HDL-C) and PSA were reviewed in 2903 elderly men (aged ≥ 60 years) in our hospital.The subjects were classified into 6 groups by age,and PSA levels were compared among groups.The correlations of FBG,TG,LDL-C,HDL-C with PSA were analyzed using multiple linear regression analysis.Results The level of PSA had the trend to increase with age.Positive association of TG,LDL with serum PSA level,but negative correlation between HDL and serum PSA level were noted in the group aged 60-64 years (r=0.10,0.15,0.12,P<0.05).In the group aged 66-69 years,there was negative relationship between FBG and serum PSA(r= 0.10,P<0.05).No relationship could be found in other groups (P>0.05).ConclusionsPSA level may increase with age,but without relation with FBG,TG,LDL-C and HDL-C.
3.Mass Culture and Purification of Vero-based Influenza Virus

Objective:Establish mass-scale purification technology of cell-derived influenza virus. Methods: A microcarrier-based process was used to produce human influenza virus in serum free-adapted Vero cells. The virus was purified in a sequence of downstream processing steps including inactivation, clarification, anion exchange chromatography, affinity chromatography and size-exclusion chromatography. Results: The recovery of HA reached 102%. 95.3% total protein, including 99.77% host cell protein, and 99.99% host cell DNA were removed during downstream processing. Conclusion: Providing a high effective purification technology for cell-derived influenza virus.
4.Research progress of new isothermal amplification technology in rapid diagnosis of Plasmodium and other parasites

Abstract: At present, nucleic acid detection technology based on the PCR principle is commonly used to detect malaria parasites, the existing Plasmodium detection methods mainly include microscopy, antigen immunoassay, and nucleic acid detection,but due to the long detection time, high personnel and equipment requirements, and other shortcomings, its popularization, and application at the grassroots level are limited. What challenges previous Plasmodium detection methods are the lack of experienced professionals and advanced equipment at the grassroots as well as the requirement of rapid detection of large samples under extreme conditions. The isothermal amplification technology developed in recent years has potential application prospects due to its simplicity, rapidity, high sensitivity, and high specificity. This article attempts to review the principles, characteristics, and prospects of various isothermal amplification technologies, and on this basis, focuses on the introduction of recombinase polymerase amplification (RPA) and recombinase⁃aided isothermal amplification (RAA) assay technologies and proposes the use of such recombinant enzyme amplification technologies to achieve rapid and accurate diagnosis of common Plasmodium species possibility and imagination.
5.Correlation between renal artery resistance index and serum creatinine level early after renal transplantation.

OBJECTIVETo investigate the relationship between the resistance index (RI) of the renal artery and serum creatinine (Cr) level in patients early (within one month) after renal transplantation.
METHODSA total of 123 patients receiving renal transplantation underwent examinations by color Doppler ultrasound for measurement of the RI of the renal artery within one month after the operation. According to the results of RI measurement, the patients were divided into RI≥0.75 and RI<0.75 groups for analyzing the correlation between RI and serum Cr level measured at the same time points.
RESULTSThe RI and Cr levels in patients with RI≥0.75 showed a significant positive correlation (P<0.05), whereas they showed an inverse correlation in patients with RI<0.75 (P<0.05). The patients with RI≥0.75 had significantly lower RI of the renal artery and Cr level than those with RI≥0.75.
CONCLUSIONRI is significantly correlated to Cr, and may serve as an indicator for predicting renal graft function after transplantation.

6.Advances of research on autophagy regulation in leukemia cells.

Autophagy is a conservative self-degradation system in eukaryotic cells, which involves in multiple physiologic and pathologic processes. Autophagosome is a typical characteristics of autophagic process, and its formation and degradation are the key points to control autophagy. Due to its dual characteristics to promote survival and death, to some extent, autophagy determines cell fate for survival or die. Autophagy plays important roles in cancer development, metastasis and drug-resistance. Thus targeting autophagy may provide novel strategies for treating cancer and overcoming drug resistance. With the advances of study on autophagy regulation in leukemia cells, the novel therapeutic targets and strategies to cure leukemia will be developed. This review focuses autophagy characteristics and regulation, autophagy and tumor, autophagy and leukemias as well as autophagy regulation in leukemia cells.

7.Protein synthesis and characterization in isolated mitochondria from rat brain.

AIMTo establish a system of mitochondrial translation in vitro of rat brain and identify it's production of protein by molecular weight.
METHODSMitochondria isolated from hemisphere of rat brain by differential centrifugations. The optimization of mitochondrial translation in vitro by 3H-Lencine incorporation was explored. 35S-methionine labeled products of mitochondrial protein synthesis were identified by SDS-PAGE and fluorography.
RESULTSIsolated mitochondria had a highly activity oxidative phosphorylation and respiratory control ratio (RCR) was between 3.5 and 5.5. The activity of 3H-Lencine incorporation in isolated mitochondria in vitro increased with time of incubation in 60 min and maintained a steady level. The maximal activity of 3H-Lencine incorporation per milligram mitochondria protein occured at 1 mg mitochondria/ml of incubation mix.The major auto radiographic bands could be observed at 86, 68, 56, 43, 33, 29, 25 and 18(kD) molecular weight separated on SDS-PAGE.
CONCLUSIONThe translation system of rat brain mitochondria in vitro is faithful and high activity, can be used to study mtDNA expression and regulation in mammalian brain at the level of translation.

8.Monocyte human leukocyte antigen-DR expression in elderly patients with sepsis.

OBJECTIVETo investigate the changes of human leukocyte antigen DR (HLA-DR) expression in the monocytes of elderly patients with sepsis.
METHODSA total of 213 elderly patients were divided into 4 groups according to the criteria defined by the American College of Chest Physicians/ Society of Critical Care Medicine (ACCP/SCCM), namely the non-sepsis group (group A), sepsis group (group B), severe sepsis group (group C) and septic shock group (group D). Another 60 healthy subjects were recruited as the control group. HLA-DR expression in the monocytes from each subject was determined by flow cytometry, and the HLA-DR levels were compared between the groups.
RESULTSCompared with that of the healthy control, HLA-DR expression was significantly increased in group A and lowered in group D. No significant differences were found between the control group and groups B and C. HLA-DR expression was significantly different between groups A, B, C, and D (P<0.01), increasing in the order of group D (32.74-/+13.4)%, group C (61.9-/+14.29)%, group B (69.99-/+12.8)%, and group A (85.06-/+15.37)%.
CONCLUSIONSAlthough with multiple organ diseases and repeated or long-term hospitalization, the non-septic elderly patients do not show lowered expression of HLA-DR. In the elderly patients with sepsis and severe sepsis, HLA-DR expression is not a sensitive index, but obviously lowered HLA-DR expression may serve as a specific indicator in elderly patients with septic shock.

9.Effect of Chinese medicine for resolving dampness on activated and functional T lymphocyte subsets in chronic hepatitis B patients with dampness syndrome.

OBJECTIVETo explore the relation between the effect of Huashi Decoction (HD) and levels of activated and functional T lymphocyte subsets in chronic hepatitis B (CHB) patients of dampness syndrome (DS).
METHODSSeventy CHB patients were nonrandomly assigned to two groups, the treated group (n = 45) treated by HD and the control group (n = 30) by nucleotide analogies. The clinical efficay was observed and levels of activated and functional T lymphocyte subsets were detected before and after treatment.
RESULTSThe peripheral blood levels of CD8+ CD28+ and CD4+ CD28+ T cells were significantly lower in CHB patients than those in the healthy subjects (P < 0.01), which were higher in the treated group after treatment compared with those before treatment (P < 0.05), while the CD8+ CD38+ T cell level was significantly higher in CHB patients than in the healthy subjects (P < 0.05), which was lower in the treated and the control groups after treatment than those before treatment respectively (P < 0.05). No significant difference was found in the markedly effective rate and total effective rate between the two groups (P > 0.05). The levels of CD8+ CD28+ and CD4+ CD28+ T cells increased and the CD8+ CD3+ and CD4+ CD25+ T cell level decreased obviously after treatment in those who acquired marked effectiveness (P < 0.05), while they changed insignificantly in those who did not acquire marked effectiveness (P > 0.05).
CONCLUSIONHD is effective in regulating activated and functional T lymphocyte subsets in CHB patients with DS, which might be its mechanism of inhibiting virus replication and eliminating virus.

10.Toll-like receptor 4: the potential therapeutic target for neuropathic pain.

Activation of microglia plays a vital role in the initiation and maintenance of specific neuropathic pain states. By activating microglia in central nervous system, Toll-like receptor 4 (TLR4) can promote the release of proinflammatory cytokines and neuroactive compounds, participate in the initiation and maintenance of neuropathic pain, and trigger the opiate side effects. Therefore, TLR4 may be a potential therapeutic target for neuropathic pain. Inhibition of TLR4 has shown some biological effects in neuropathic pain models and ibudilast (the TLR4 pathway-inhibiting agent) has been approved for for phase 2 clinical trials. This article briefly reviews the structure, function, and mechanism of TLR4 as well as the development of TLR4-targeted drugs.
