1.Acupuncture at Shangjuxu (ST37) to Inhibit Distention of Colon Induced Discharge Reaction of LC: an Experimental Study.

OBJECTIVETo study possible mechanisms of Shangjuxu (ST37) and the large intestine.
METHODSTotally 40 SD rats were selected. The distension of end colon was used as injured afferent stimulus. Activities of locus coeruleus (LC) neurons were recorded by extracellular microelectrode technique. Shangjuxu (ST37) and Hegu (L14) were needled to observe general features of discharge reactions, distention of colon induced discharge reactions of LC, and its effects on distention of colon induced discharge reactions of LC.
RESULTSDistention of colon could induce incrased discharge of LC neurons by 127.33% ± 45.48%. But needling at Shangjuxu (ST37) and Hegu (L14) could inhibit this injured response by 38.24% ± 7.69% and 21.29% ± 13.16% respectively (all P < 0.01).
CONCLUSIONSNeedling at Shangjuxu (ST37) and afferent signals of colon distension converged and interacted with each other. Needling at Shangjuxu (ST37) could significantly inhibit colon distension induced discharge of LC neurons, which might be one of mechanisms for Shangjuxu (ST37) and the large intestine relationship.

2.Imageologic study and clinical treatment of pelvic fractures combined with artery injuries

Objective To explore the imageologic characteristics of pelvic fractures with artery injuries and the treatment methods for embolization of arteries.Methods From January 1999 to June 2005,60 cases(42 males and 18 females)aged 21-52 years(average 34.5 years)with pelvic fractures and unsteady blood dynamics were admitted into our hospital.There were 32 cases with traffic injury,13 with crushing injury,nine with fall injury and six with other injuries.The mean injury severity score was 39?16(16-66).All cases were hypetensive with systolic blood pressure less than 90 mm Hg on the arrival.Routine X-ray examination of dorsaventral,debouch and porch of pelvis was performed.The aver- age amount of blood transfusion was 2 886 ml.All cases underwent iliac artery angiography and pelvic ar- teriography.Results X-ray examination of pelvic fractures showed posterior pelvic fracture in 25 ca- ses,with 64 branches of blood vessels injured;anterior pelvic fracture in 13,with 17 branches of blood vessels injured;acetabular fracture in six,with 12 branches of blood vessels injured;and combined pel- vic fracture in 16,with 36 branches of blood vessels injured.Three cases died,with mortality rate of 5%.One case with common arterial thrombosis was treated with artificial blood vessel transplantation, four cases with external iliac artery injuries including one with artery rupture were treated with prosthesis, and among the three cases with external iliac artery thrombosis,one was treated with dislodgment of thrombosis,one treated with recanalization of thrombolysis and one did not give any treatment.Fifty cases with injury and bleeding of internal iliac artery and its branches were treated with arterial embolization. Five cases showed no obvious injury.Conclusions The types of artery injuries can be predicted through X-ray of pelvic fracture.Posterior pelvic fracture may easily cause injury to superior gluteal arter- ies,iliac lumber arteries,and lateral sacral arteries.While anterior pelvic fracture will cause injury to obturator arteries.Superior gluteal artery is susceptible to injury.Embolization of injured arteries and an- astomosis are preferred treatment for pelvic arterial disruptions.
3.Contents of secondary metabolites and anti-bacterial activity of compound Caulis Sargentodoxae decoction.

OBJECTIVETo compare the contents of secondary metabolites and the anti-bacterial activity of seven different compound Caulis Sargentodoxae decoctions.
METHODSKirby-Bauer disk agar diffusion method was used for measuring anti-bacterial activity. The contents of secondary metabolites were determined by spectrometry. The correlation and path analysis were used to identify the goal compound.
RESULTThe secondary metabolites contents and anti-bacterial activity of seven different compound Caulis Sargentodoxae decoctions varied significantly. The total contents of six secondary metabolites of Shushao Caulis Sargentodoxae decoction was the highest, while that of Caihu Caulis Sargentodoxae decoction was the second highest and that of Yiyi Caulis Sargentodoxae decoction was the lowest. The anti-bacterial activity of Shushao Caulis Sargentodoxae decoction was the highest and that of Cangbai Caulis Sargentodoxae decoction was the lowest. The correlation and path analysis showed that the total tannin and total chlorogenic acid might be the major factors related to the anti-bacterial activity of compound Caulis Sargentodoxae decoctions.
CONCLUSIONThe total contents of six secondary metabolites and the anti-bacterial activity of Shushao Caulis Sargentodoxae decoction are the highest among seven different decoctions and there is a significant correlation of the anti-bacterial activity with the contents of total tannin and total chlorogenic acid.

5.Anticancer effects of various fractions extracted from Dioscorea bulbifera on mice bearing HepA.

OBJECTIVETo investigate the anti-cancer activities and the possible mechanism of Chinese herb Dioscrea bulbifera.
METHODThe herb was extracted sequentially with petroleum ether, ethanol and water. The anticancer screen were carried out in vivo with HepA in mice.
RESULTThe inhibitory effects on the formation of ascites volume and HepA cell viability in ascites were found in those extracted fractions except water fraction, the petroleum ether fraction being the strongest. Life span of mice bearing HepA ascites was prolonged after exposed to 100 mg x kg(-1) petroleum ether fraction and shortened after exposed to water fraction significantly. Besides, abnormal microstructure on HepA cells surface was found and it was supposed to be potential effect against viability of HepA which was convinced with the regeneration of HepA cells from ascites in mice exposed to petroleum ether fraction.
CONCLUSIONAnticancer active compounds are mainly extracted by petroleum ether from hydrophobic constituents of Dioscrea bulbifera and the anticancer effects were related to direct toxicity on tumor cell.

6.Correlation of anti-bacterial activity with secondary metabolites content in Sargentodoxa Cuneata tables.

OBJECTIVETo explore the correlation between the content of the secondary metabolites and the anti-bacterial activity of Sargentodoxa cuneata tables.
METHODSThe content of secondary metabolies was determined by spectrometers. Staphylococcus aureus and Bacillus subtilis were used as the experimental strains. The minimal inhibition concentration (MIC) was calculated using agar plate dilution method on Kirby-Bauer disk. The minimal bacterialcidal concentration (MBC) was calculated using tube dilution method. Two-way analysis and Turkey's test was employed in the statistical analysis.
RESULTThe anti-bacterial activities on Staphylococcus aureus and Bacillus subtilis of the first batch of S. cuneata tables produced in Anhui was the highest [the diameter of anti-bacterial zone, MIC, MBC was (9.67 +/-0.29) mm, 12.5 mg/ml, 31.25 mg/ml and (10.17 +/-0.58) mm, 12.5 mg/ml, 31.25 mg/ml, respectively]. Those of the second batch of S. cuneata tables produced in Jiangsu was the lowest [the diameter of anti-bacterial zone, MIC, MBC was (7.67 +/-0.29)mm, 25 mg/ml, 125 mg/ml and (8.17 +/-0.29)mm, 25 mg/ml, 125 mg/ml, respectively]. There was significant difference in the antibacterial activity on Staphylococcus aureus between different producing areas of S. cuneata tables and batches (P<0.01), while there was also significant difference in the antibacterial activity on Bacillus subtilis between different producing areas (P<0.01) and no significant difference between different batches (P>0.05). The contents of 6 secondary metabolites of S. cuneata tables produced in different areas and in different batches were different. The total content of 6 secondary metabolites of the first batch of S. cuneata tables produced in Anhui was the highest with mean 3.61%. Those of the second batch produced in Jiangsu were the lowest with mean 1.90 %. There was significant difference in the content of total alkaloid and dissociated emodin between different producing areas (P<0.01) and no difference between different batches (P>0.05). For the content of total chlorogenic acid, flavonoids, total tannin and total saponin, there was significant difference between different producing areas and batches (P<0.01). The correlation analysis showed that the main secondary metabolites influencing the anti-bacterial activity were total saponin, total tannin, dissociated emodin and total chlorogenic acid.
CONCLUSIONThere is difference among the content of the secondary metabolites and anti-bacterial activity of S. cuneata tables produced in different areas and different batches. There is significant correlation of the anti-bacterial activity with the content of total saponin, total tannin, dissociated emodin and total chlorogenic acid in S. cuneata tables.

7.A correlation study between ITGA6 gene, chromosome 8q24, MSMB genes and prostate cancer

Objective To explore the correlation between ITGA6 gene (rs12621278, G), MSMB gene (rs10993994, T), chromosome 8q24 (rs10086908, T) and prostate cancer (PCa) in Beijing residents, and to explore the correlation between genotype and phenotype in PCa patients. Methods PCa patient phenotypes were collected including clinical, genetic, dietary habits, hobbies and blood samples. ITGA6 gene (rs12621278, G), chromosome 8q24 (rs10086908, T) and MSMB gene (rs10993994, T) compared the allele distribution between 112 PCa and 91 healthy control age matched patients. The genotype and phenotype analysis was conducted in the 2 groups. Results Between the case and control groups, only rs10993994, T of MSMB gene (case 56.4%,control 46.2%) was significantly different (P=0.001; OR=1.97, 95%CI:1.28-3.04). The rs10086908, T of 8q24 (case 83.5%, control 79.2%) and rs12621278, G of ITGA6 gene (case 27.2%, control 27.0%) were not significantly associated with PCa in the study sample (P>0.05). Quantitative trait analysis showed that the disease duration of ITGA6 risk genotypes (G/G,1.40±0.55 years) in PCa patients was significantly shorter (P=0.003) than the other genotype carriers (A/G, 4.38±3.10 years; A/A, 2.37±1.84 years). Conclusion The genetic variation in MSMB is possibly associated with PCa susceptibility, suggesting that MSMB genes might be associated with PCa in a Chinese population.
8.Study on the risk of age-related diabetes mellitus among 8280 cases with metabolic syndrome patients and normal persons in Beijing

Objective To investigate the impact of age on patients with metabolic syndrome (MS) and normal persons. Methods Data was gathered from 8280 persons including 4873 males and 3407 females who were randomly selected. All subjects were devided into normal group and MS group. According to the interval of ten years, the subjects were devided into seven age groups, to calculate the difference of impaired fasting glycaemia (IFG) between patients with diabetes mellitus (DM) and normal people, as well as the related portions. Results (1) The risk of IFG and DM appeared to be different among age groups among the target subjects as well as in the normal and the MS groups (P<0.05). (2) Among the whole subjects, the overall prevalence of IFG was increasing with age. The prevalence of DM had an increasing trend with age augment in 20-79 years group, whereas a decreasing trend appeared in people over 80 years of age. (3) For normal persons, the prevalence of IFG and DM were all increasing with age augment in 20-79 years group, and then decreasing with age augment in the over-80-years group. (4)For MS patients, the prevalence of IFG had an increasing trend with age augment in 20-69 years group, whereas a decreasing trend appeared in people over 70 years of age. There was no tendency of variation with age augment in DM.Conclusions (1) For normal persons, high prevalence rates of IFG and DM were correlated to age augment, especially in senior persons. (2) For MS patients, high prevalence of IFG was also correlated to age augment, but no association between prevalence of DM and age augment was seen. (3)Age from 70 to 79 years appeared to be in high risk with MS.
9.Randomized and controlled clinical study of modified prescriptions of Simiao Pill in the treatment of acute gouty arthritis.

OBJECTIVETo investigate the compatibility of a modified prescription of Simiao Pill in the treatment of acute gouty arthritis and to verify the clinical efficacy and safety of the drug through a clinical trial.
METHODSA randomized and controlled clinical trial was designed based on clinical epidemiological principles. A total of 107 patients with acute gouty arthritis were enrolled and randomly assigned to four groups. The first group (Group I) included 27 patients taking gout prescription I; the second group (Group II) included 27 patients taking gout prescription II; the third group (Group III) included 28 patients taking gout prescription III; and the fourth group (control group) included 25 patients taking indomethacin and Benzobromarone as a control group. The duration of the treatment in all 4 groups was two weeks. After the treatment, the index of blood uric acid, blood leukocyte count, score of clinical symptoms, etc. were observed and measured.
RESULTSThe total clinical effective rate of the three different modified prescriptions of the Simiao Pill was above 96%, significantly superior to that of the control group (68%, P<0.05). In terms of the improvement of main symptoms, the scores of four symptoms in all TCM treatment and control groups decreased after treatment, with statistically significant differences (P<0.05). Moreover, the scores markedly fell more so in the three Chinese herb groups than in the control group, and especially in Group III (P<0.05). There was a statistically significant difference in blood uric acid values before and after the treatment in the same group but no significant inter-group difference was seen.
CONCLUSIONThe modified prescriptions, based on the clinical research, clinical experience and traditional Chinese medicine theory, did show a better effect than Western medicine in this clinical study. Moreover, the prescriptions were precise, with the herbs inexpensive and readily available. The patients had good compliance with less adverse reactions noted. The modified prescription has a favorable prospect for future development and is worthy of further blind trials with larger samples.

10.Relationship between interleukin-6 gene-572C/G polymorphism and chronic periodontitis.

OBJECTIVETo investigate the relationship between interleukin-6 (IL-6) gene -572C/G polymorphism and the risk of chronic periodontitis disease.
METHODSIL-6-572C/G genotype polymorphism was examined in 93 patients with chronic periodontitis and 96 control subjects by PCR restriction fragment length polymorphism (PCR-RFLP) method.
RESULTSIL-6-572GG genotype and G allele were more frequent in chronic periodontitis patients than that in control subjects (P<0.05). -572CC, CG and GG genotype frequency were 52.7 % , 40.9 %, 6.4 % in chronic periodontitis patients and 67.7 %, 31.3%, 1.0 % in control subjects, respectively. -572CG + GG genotype and G allele were more frequent in patients than that in control subjects (P<0.05). Compared with CC genotype,the odds ratio for chronic periodontitis was 1.88 (95% CI: 1.04-3.40, P<0.05) for CG + GG genotype.
CONCLUSIONSThe frequency of -572CG + GG genotype was higher in patients with chronic periodontitis compared with the control subjects. IL-6-572C/G polymorphism may be a genetic susceptibility factor for chronic periodontitis in Chinese Hans population.
