2.Changes of the anti-oxidation capability of rat model of Parkinson disease after acupuncture at subthalamus

BACKGROUND: Acupuncture has been confirmed to be effective in treating Parkinson disease both clinically and experimentally, but there is still lack of systematical and complete therapeutic principle and law to be followed in clinic.OBJECTIVE: To observe the influence on glutathione (GSH), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), malondialdehyde(MDA)and nitric oxide synthase (NOS) in corpus striatum of the damaged side in rat models of Parkinson disease by acupuncture at subthalamus.DESIGN: A randomized controlled animal experiment.SETTING: The Second Affiliated Hospital of Heilongjiang University of Traditional Chinese Medicine.MATERIALS: The experiment was performed in the Experimental Animal Center of Heilongjiang University of Traditional Chinese Medicine from January to June in 2003. Eighteen healthy adult Wistar rats of pure breed were randomly divided into 3 groups with 6 rats in each:electroacupuncture group, model group and normal saline group.METHODS: ① Model group: The rats were made into models of Parkinson disease with damaged corpus striatum by injecting 6-hydroxydopamine (6-OHDA, 12 μg), which was dissolved in 5 μL normal saline containing 2 g/L ascorbic acid, slowly into caudal putamen. without intervention. ②Normal saline group: 6-OHDA was replaced by normal saline of the same dosage, and other treatments were the same as those in the model group. ③ Electroacupuncture group: The model preperation was the same as that in the model group. The needles were embedded in subthalamus at 6 weeks postoperatively, and then electroacupuncture (in frequency of 100 Hz, and the intensity about 1 mA) was given one week later for 2 weeks, once a day,15 minutes for each time.MAIN OUTCOME MEASURES: All the rats were killed by cutting heads to remove brain under anesthesia at 2 weeks after electroacupuncture, and the activities of GSH, GSH-Px, SOD and NOS and the concentration of MDA were detected with chromatometry.RESULTS: All the 18 rats were involved in the analysis of results. ① MDA concentration was higher in the model group and electroacupuncture group than in the normal saline group [(5.53±0.71),(4.10±0.27), (5.53±0.71) μmol/L, P < 0.01], but lower in the electroacupuncture group than in the model group (P < 0.05). ② The activities of GSH and GSH-Px were both lower model group and electroacupuncture group than in the normal saline group (P < 0.01), but lower in the electroacupuncture group than in the model group (P < 0.05).③ The activity of SOD was higher in the model group and electroacupuncture group than in the normal saline group [(113.36±2.17),(73.85±5.17), (42.34±1.83) μkat/g, P < 0.01], but lower in the electroacupuncture group than in the model group (P < 0.01).CONCLUSION: The anti-oxidation capability of the rat models of Parkinson disease has been recovered and the nerve injury has been reduced after acupuncture, indicating that it is available and effective to treat Parkinson disease by acupuncture at subthalamus.
3.Clinical application of serological detection in primary hepatic car-cinoma and metastatic hepatic carcinoma patients

Objective:To investigate the clinical value of serological test indicators in the identification of primary and metastatic liver cancers. Methods:We detected the serum levels of ALT, AST, ALP, TBIL, DBIL, GGT, CHE, 5'-NT, CA199, CEA, and AFP in primary hepatic carcinoma (PHC;120 cases), metastatic hepatic carcinoma (MHC;135 cases), and no liver metastatic control (135 cas-es) groups. The methods used were variance analysis and Scheffe test. ROC curve analysis was used to determine the diagnostic value of CA199 , CEA, and AFP in PHC and MHC. Results:The difference between the serum levels of AST, ALP, GGT, 5'-NT, AFP, and CEA of the PHC and MHC groups was statistically significant (P<0.05). The AFP, CEA, and CA199 areas under the ROC curve of the PHC and MHC groups revealed that AFP diagnosis of primary liver cancer had certain accuracy, whereas CEA and CA199 have some diagnostic value in differentiating primary and metastatic liver cancers. Conclusion: The detection of serum levels of AST, GGT, 5'-NT, AFP, CA199, and CEA in malignant tumor was a preliminary diagnosis of liver metastasis and can provide evidence for the dif-ferential diagnosis of PHC and MHC.

7.Expression of translationally controlled tumor protein in squamous cell carcinoma tissue and cell lines A431 and SCL-1

Objective To detect the expression of translationally controlled tumor protein (TCTP) in squamous cell carcinoma (SCC) tissue and cell lines A431 and SCL-1,and to evaluate the effect of TCTP on apoptosis in and proliferation of SCC cells.Methods An immunohistochemical method was used to measure the expression of TCTP in tissue specimens from 65 patients with SCC.Western blot was performed to detect the expression of TCTP in A431 and SCL-1 cells.Three small interference RNAs (siRNAs) targeting the TPT1 gene were designed,synthesized,and transfected into A431 cells.Then,reverse transcription (RT)-PCR and Western blot were conducted to measure the expression of TPT1 mRNA and TCTP,respectively,methyl thiazolyl tetrazolium (MTT) assay and flow cytometry were carried out to detect cell proliferation and apoptosis,respectively.Results TCTP was overexpressed in SCC tissue specimens,and the expression level was positively correlated with the histologic grading of SCC (P < 0.05).Western blot showed that TCTP was expressed in both A431 and SCL-1 cells,and the expression was relatively high in A431 cells.The transfection efficiency of siRNAs varied from 90% to 95%.A decrease in the expression of TPT1 mRNA and TCTP was induced by the siRNAs in A375 cells (all P < 0.05).Conclusion The downregulation of TCTP expression may increase the apoptosis in and suppress the proliferation of A431 cells.
9.Correlation among troponin,CK-MB and acute myocardial infarction range in aged patients

Objective:To explore the correlation among cardiac troponin I (cTnI),cardiac troponin T (cTnT),crea-tine kinase isoenzyme (CK-MB)levels and range of myocardial infarction in aged patients with acute myocardial in-farction (AMI).Methods:Clinical data of 80 aged AMI patients hospitalized in our internal medicine department from Sep 2012 to Sep 2013 were retrospectively analyzed.Correlation among levels of cTnI,cTnT,CK-MB and range of myocardial infarction were analyzed before and one week after treatment.Results:Compared with before treatment,there were significant reductions in levels of cTnI [(4.63±0.21)μg/L vs.(0.15±0.03)μg/L],cTnT [(0.71±0.05)μg/L vs.(0.17±0.01)μg/L],CK-MB [(40.21±2.13)U/L vs.(23.32±2.11)U/L]and myo-cardial infarction area [(35.25±4.65)mm2 vs.(23.17±3.76)mm2 ]after treatment,P <0.01 all;Pearson cor-relation analysis indicated that cTnI,cTnT and CK-MB were all positively correlated with myocardial infarction area (r =0.99,0.98,0.95,P <0.01 all).Conclusion:Cardiac troponin I,T and creatine kinase isoenzyme are closely related to myocardial infarction range.Combination of these three indexes can be used as important basis for judging myocardial infarction range in patients with myocardial infarction.
