1.Effect of Antisense Oligodeoxynudeotides Targeting Nuclear Factor ?B on Expression of Caspase-3 in Glomerulosclerosis

Objective To investigate the effect of antisense oligodeoxynudeotides(ODN)targeting nuclear factor ?B(NF-?B)on the expressions of Caspase-3 and NF-?B in glomerulosclerosis.Methods Male SD rats were divided into 3 groups:sham operation(group A,n=6),glomerulosclerosis(group B,n=6),intervention(group C,n=9).Group C was divided into 3 groups:sense ODN(group C1,n=3),non-sense ODN(group C2,n=3),antisense ODN(group C3,n=3).Glomerulosclerosis models were made for SD rats by unilateral nephrectomy and being injected with adriamycin into caudal vein.After 8 weeks,various ODN applied to corresponding group for intervention.At the end of the 9th week,kidneys were taken out from all rats for the measurement of expressions of NF-?B p65 and Caspase-3 by immunohistochemistry staining.Results After intervention,on the d7,the rats urine protein,glomerular sclerosis index(GI)and expressions of Caspase-3,NF-?B p65 in group C3 were significantly lower than those in group B,C1,C2(Pa0.05).Conclusion Antisense ODN targeting NF-?B can inhibit the activities of NF-?B,Caspase-3 in rats kidneys and retard glomerulosclerosis and glomerular intrinsic cell apoptosis.

3.Clinical observation on 45 cases of gout treated with Jiangzhuo Huoxue Yishen Decoction.

OBJECTIVETo observe the curative effect of the self-formulated Jiangzhuo Huoxue Yishen Decoction (JHYD) combined with Western medicine in treatment of gout.
METHODSA total of 90 gout patients were randomly assigned to two groups, the control group (45 cases) administrated with Finbid orally in the acute stage and allopurinol in the remission period; and the treatment group (45 cases) treated with the same treatment plus oral administration of JHYD. The therapeutic course for both was 4 weeks. The total curative effect, the effect on main symptoms and on different syndrome types, changes of blood uric acid, adverse reactions and recurrence of gout were observed and compared between the two groups.
RESULTSThe cured and markedly effective rate and the total effective rate in the treatment group were all obviously higher than those in the control group (P<0.05). And the effect was better in the treatment group in decreasing blood uric acid, improving symptoms of main joints and stabilizing the curative effect. The curative effect of the combined therapy on different syndrome types was basically consistent, and it showed few adverse reactions.
CONCLUSIONThe self-formulated JHYD showed synergistic effect with Western medicine in treatment of gout, which could decrease blood uric acid level and improve clinical symptoms.

5.Effect of Yijing Recipe on apoptosis of testis spermatogenic cells and the expression of Bcl-2 and Bax protein in infertility rats.

OBJECTIVETo observe the effect of Yijing Recipe (YR) on the apoptosis of testis spermatogenic cells and the protein expression of Bcl-2/Bax in rats with adenine induced infertility.
METHODSTotally 75 Wistar rats were randomly divided into 5 groups, i.e., the blank control group, the model group, the high dose YR group, the middle dose YR group, and the low dose YR group, 15 in each group. Except those in the blank control group, rats in the rest groups were intragastrically administered with adenine for 10 successive days. From the 11th day, rats in the blank control group and the model group were fed with equal volume of normal saline. Rats in the YR groups were intragastrically administered with YR at different doses (3.38 g/100 g; 1.69 g/100 g; 0.85 g/100 g), once daily for 20 consecutive days. All rats were killed by the end of the experiment and their testes extracted. The apoptosis of spermatogenic cells and the expression of Bcl-2/Bax proteins were detected by terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick end labeling (TUNEL) and SABC method.
RESULTSCompared with the blank control group, the Bcl-2 protein expression decreased, the Bax protein expression increased, and the apoptosis index increased in the model group, showing statistical difference (P <0.01). Compared with the model group, the Bcl-2 protein expression increased in the three YR treated groups (P <0.01, P <0.05). The Bax protein expression level decreased in the high and middle dose YR groups (P <0. 01, P <0. 05). The apoptosis index decreased in the middle dose YR group (P <0.01).
CONCLUSIONYR could inhibit the apoptosis of spermatogenic cells through regulating the expression of Bcl-2 and Bax protein in the testis.

6.Guiding role of three-column theory in manipulative reduction, small splint fixation and early rehabilitative exercises for Colles fracture.

OBJECTIVETo explore guiding role of three-column theory in manipulative reduction, small splint fixation and early rehabilitative exercises of Colles fracture.
METHODSFrom August 2011 to February 2012,47 patients with Colles fractures were treated by manipulative reduction small splinting fixation and early rehabilitative exercises under the guidance of three-column theory, including 21 males and 26 females aged from 40 to 76 years old with an average of (65.5 +/- 2.3). According to AO fracture classification, 27 patients were type A (including 18 cases with type A2 and 9 cases with type A3) and 20 patients were type C (including 10 cases with type C1, 6 cases with type C2 and 4 cases with type C3). Pain and recovery time of swelling, postoperative complications were observed and recorded, Gartland-Werley scoring system were applied for evaluate functional recovery and biomechanical analysis of wrist joint at 12 weeks after operation.
RESULTSAll patients were followed up for 3 to 9 months with average of 5 months. Pain relief time ranged from 5 to 15 d with average of (7.6 +/- 2.2) d,recovery time of swelling of opisthenar was for 6 to 13 d with an average of (8.9 +/- 1.9) d. Two patients occurred tension vesicle within 3 days after operation, but no other complications occurred. According to Gartland-Werley scoring system, 25 cases got excellent results, 18 cases good and 4 cases moderate at 12 weeks after operation.
CONCLUSIONUnder the guidance of three-column theory, treating Colles fracture by manipulative reduction, small splinting fixation and early rehabilitative exercises can reduce pain and swelling time, promote union of fracture, effectively rehabilitate wrist function, improve clinical efficacy, and fit for concept of biomechanics.

The ancient discussion and modern clinical application regarding Yingxiang (LI 20) and Neiyingxiang (EX-HN 9) in acupuncture verses are explored. Acupuncture verses are the summary of clinical experiences of acupuncture scholars in the past dynasties, of which the records on application of Yingxiang (LI 20) and Neiyingxiang (EX-HN 9) mainly focused on nasal, facial and head-eye diseases, which is characterized with overall summarization, brilliant exposition, simple language, and distinctive characteristics. Nowadays, based on the ancient usage, the clinical application about these two acupoints is continuously developing. If the ancient acupuncture verses could be understood thoroughly, the clinical practice could be better guided and its effects could be improved.

8.Intervention of Antisense Oligonucleotide Targeting Nuclear-?B on Inflammation of Cell in Glomerulosclerosis

Objective To investigate the correlation between nuclear factor(NF)-?B and inflammation of cell in adriamycin-induced nephritic rats,and the intervention of antisense oligonucleotide targeting NF-?B to adriamycin-induced nephritic rats by measuring the expression of NF-?B p65 in renal cortex and the secretions of interleukin-6(IL-6),tumor necrosis factor-?(TNF-?)in serum.Methods Thirty male SD rats were divided into 5 groups(6 rats in each group):group A(sham operation),group B(glomerulosclerosis),group C(sense oligonucleotide),group D(non-sense oligonucleotide),group E(antisense oligonucleotide).Glomerulosclerosis models were made for SD rats by unilateral nephrectomy and being injected with adriamycin into caudal vein.In the 8th week,various medicine applys to correspon-ding group for intervention.At the 4th day and 8th day after intervention,the excretion amounts of 24 hours urine protein were determined.Hematoxylin-Eosin(HE) staining was used to observe the renal pathological changes.Using image analysis system to calculate glomerulosclerosis index(GI).The expression of NF-?B p65 in renal cortex was measured by immunohistochemistry staining,the secretions of IL-6,TNF-? in serum were performed by means of enzyme-linked immunosorbnent assay,respectively.Results At different time points,the expressions of active-NF-?B p65,IL-6,TNF-? in group E were significantly lower than those in group B.The intervention effects of antisense oligonucleotides at the 8th day were stronger than that at the 4th day.Conclusions NF-?B is significantly correlated with glomerulosclerosis and inflammations of glomerular intrinsic cells.Using NF-?B antisense oligonucleotide to be injected into renal artery can block directly the translation of NF-?B gene,delay inflammations of glomerular intrinsic cells and the progressions of glomerulosclerosis.
9.Effect of citric acid stimulation on salivary alpha-amylase, total protein, salivary flow rate and pH value in Pi deficiency children.

OBJECTIVETo compare the effect of citric acid stimulation on salivary alpha-amylase (sAA), total protein (TP), salivary flow rate, and pH value between Pi deficiency (PD) children and healthy children, thereby providing evidence for Pi controlling saliva theory.
METHODSTwenty PD children were recruited, and 29 healthy children were also recruited at the same time. Saliva samples from all subjects were collected before and after citric acid stimulation. The sAA activity and amount, TP contents, salivary flow rate, and pH value were determined and compared.
RESULTS(1) Citric acid stimulation was able to significantly increase salivary flow rate, pH value, sAA activities, sAA specific activity and sAA amount (including glycosylated and non-glycosylated sAA amount) in healthy children (P<0.05), while it could markedly increase salivary flow rate, pH value, and glycosylated sAA levels in PD children (P<0.05); (2) Although there was no statistical difference in determined salivary indices between the two groups (P>0.05), salivary indices except salivary flow rate and glycosylated sAA levels decreased more in PD children. There was statistical difference in sAA activity ratio, sAA specific activity ratio, and the ratio of glycosylated sAA levels between PD children and healthy children (P<0.05).
CONCLUSIONPD children had decreased response to citric acid stimulation.

10.Studies on Preparation of Herba EpimediiTotal Flavonoids Phyt osomes and Their Pharmaceutics

Objective: To optimize preparation techniques for Herba E pimedii flavonoid phytosomes (EFP) and explore their suitable pharmaceutics. Methods: To optimize the preparation conditions by means of unifo rm design and step regression, prepare Herba Epimedii total flavonoid phytosomes by means of solvent evaporation and investigate the accumulative dissolution of different ratios of EFP-PVP precipitates by means of dissolution release. Resu lt: The optimized preparation conditions are as follows: solvent-tetrahydrofura n, lecithin to PVP-2.5 times, temperature-40 ℃ and reaction-3 hours. Oil/wa ter apparent partition coefficient of icariin was enhanced more than 4 times by phospholipid. The accumulative dissolution of Herba Epimedii flavonoids of EFP- PVP precipitate was significantly higher than that of its physical mixture and H erba Epimedii extract tablet. Conclusion: Phospholipid can ef fectively enhance the oil/water apparent partition coefficient of icariin, and P VP can improve the dissolution of Herba Epimedii phytocomes, but the pharmacokin etics needs further study.
