1.Effect of proliferation and invasiveness by turmeric volatile oil on neuroblastoma cell line SH-SY5Y

Objectives To investigate the effect of proliferation and invasiveness by turmeri cvolatile oil on human neuroblastoma cell line SH-SY5Y. Methods Cells were incubated with different concentrations of TVO in vitro. Then cell survival rate was measured by MTT assay. The effect of 160 mg/L TVO on cell migration was assessed by cell scuffing test. Invasive ability of cell was detected by Transwell test. Apoptosis of cells was detected observed by flow cytometry assay. Results Survival rate of SH-SY5Y cells decreased and apoptisis rate was abated with elevated TVO concentration and prolonged cultivation time. Level of cell migration was lower than that in control group after being cultured with 160 mg/L TVO solution for 12 , 24 and 48h. With the in-crease of TVO concentration , the invasion ability of cells gradually decreased , and the invasive force and cis-platin had no obvious difference when the concentration of drug reached 160 mg/L. Conclusion The prolifera-tion of cells can be inhibited by inhibiting the proliferation and invasiveness ability with TVO.
2.Studies on effect of microemulsion in enhancing solubility of baicalin and puerarin.

OBJECTIVETo study the effect of O/W Microemulsion in enhancing solubility of baicalin and puerarin.
METHODThe Pseudo-ternary phase diagram was employed to selected the formulations of O/W microemulsion. The partical size of microemulsion, and solubility of baicalin and puerarin in water, oil phase, surfactant, microemulsion and alcohol were determined, respectively.
RESULT11 types of microemulsion formulations have the effect in enhancing the solubility of baicalin and puerarin; the solubility of Baicalin in microemulsion is higher as 5-15 times than it in water, the solubility of Puerarin in microemulsion is higher as 3 - 4 times than it in water.
CONCLUSIONMicroemulsion is good in enhancing solubility of effective compounds extracted from Chinese medical herbs.

3.Effect of turmeric volatile oil on proliferation and apoptosis of human skin SCC A431 cells.

To investigate the effect of turmeric volatile oil (TVO) on the apoptosis and proliferation of human skin SCC A431 cells, A431 cells were incubated with different concentrations (5-80 mg•L⁻¹) of TVO in vitro.The proliferation and cell cycle were assessed by CCK8 assay. The change of morphology was observed with inverted microscope. Apoptosis was evaluated with AO/EB double staining and flow cytometry (FCM); cell cycle was analyzed with FCM .Western blot method was used to detect caspase-3 and caspase-9 protein expression. Results indicated that TVO has significant inhibitory effects on the growth of A431 cells in a dose dependent relationship, the difference between groups has statistically significant (P<0.05). TVO group compared with control group, concentrations in cells shrivel and broken phenomenon, cell apoptosis rate increased, and a dose dependent and increased the expression of caspase-3 and caspase-9. The experiment results suggested that TVO could restrain skin squamous carcinoma A431 cells proliferation, and induce its apoptosis. The mechanism may be related to increase the expression of caspase-3 and caspase-9.
