1.Simultaneous Determination of Three Components of Saridon Tablet in Human Plasma by RP-HPLC

OBJECTIVE:To establish a RP-HPLC method for simultaneous determination of caffine,acetaminophen and isopropylphenazone in human plasma after oral dose of saridon.METHODS:Hypersil Hypurity TM Advance was used as chromatographic column and the mobile phase was a mixture of0.01mol/L phosphoric acid-methanol-tetrahydrofuran(86∶12∶2).The detection wavelength was273nm.RESULTS:The calibration curves were linear in the ranges of0.39~100.0?g/ml for caffine,1.96~500.0?g/ml for acetaminophen,and1.17~300.0?g/ml for isopropylphenazone,respectively.The accuracy of all was higher than95.0%.The inter-day and intra-day RSD was less than10.0%.CONCLUSION:The present method is sensitive,accurate,convenient,and highly specific,which can meet the requirement in plasma determination of three components in Saridon tablet.

Objective To study the effects of complex polysaccharides immune milk (CPIM) on cellular and humoral immune functions in mice. Method Sixty ICR mice were divided into five experimental groups and a control group. The mice in experimental groups were given i.p. Ganoderma lucidum polysaccharide (GLP) immune milk,Lycium barbarum polysaccharides (LBP) immune milk and different doses (low,medium and high) CPIM,and the control group was given equivalently skimmed and sterilized goat's milk respectively for 15 d. 3.Stabilization of unstable pelvic fractures with a novel internal-external fixator.

An internal-external fixator for stabilization in three cases with unstable pelvic fractures were reported. Compared with external fixation, the internal-external fixator, which is placed in the subcutaneous layer, decreased the risk of pin-track infection, pin site pain, and bowel obstruction; meanwhile, it had the advantage of external fixation: it was easy to apply, controlled damage, and resulted in minimal injury.

4.Diagnostic value of 3D-indirect magnetic resonance arthrography in detecting meniscal tears of the knees.

OBJECTIVETo explore the diagnostic value of 3D-indirectmagnetic resonance (MR) arthrography in detecting meniscustears of the knees.
METHODSForm January 2013 to January 2014, routine plain MR of the knees followed by the 3D-indirect MR arthrography was performed in 42 patients with suspected meniscal tears clinically. There were 31 males and 11 females, with an average age of 38.4 years old (ranged, 21 to 67 years old). The duration of the course ranged from 2 h to 15 d. The clinical symptom was knee pain. All the patients got subsequently arthroscopic examination or operation. The sensitivity and specificity of routine plain MR and 3D-indirect MR arthrography were compared based on the results of arthroscopic examination or operation.
RESULTSThe signal intensity in the area of meniscal tears on image of 3D-indirect MR arthrography was obviously higher than that of routine plain MR. The sensitivity of 3D-indirect MR arthrography was 85.79% (87/102), while the routine plain MR was 52.94% (54/102), and the specificity improved from 67.78%(61/90) to 86.67%(78/90). The difference was statistically significant (χ2 = 25.90, P < 0.01; χ2 = 9.13, P < 0.01).
CONCLUSIONIn comparison with the routine plain MR findings, 3D-indirect MR arthrography demonstrates meniscus tears of the knee with the better sensitivity and specificity.

5.The effect of emodin on the contraction of isolated jejunum smooth muscle of rats.

OBJECTIVETo investigate the effect of emodin on the contraction of jejunum smooth muscle and its underlying mechanisms.
METHODSRats were randomly divided into 7 groups (n = 6): control group, emodin group (1, 5, 10, 20 micromol/L), propranolol (PRO) plus emodin group, glibenclamide (GLI) plus emodin group, NG-Nitro-L-arginine Methyl Ester (L-NAME) plus emodin group, calcium free control group and calcium free emodin group. The rats were sacrificed by cervical dislocation and the small intestine was isolated. The jejunum segment specimens were mounted on an Organ Bath System with a tension transducer. The effect of emodin on contraction of jejunum smooth muscle was measured by BL-420E+ biological signal processing system and the amplitude (AM), tension (TE) and frequency (FR) of contraction were determined.
RESULTS(1) Emodin inhibited the tension and amplitude of jejunum smooth muscle contraction in a dose-dependent manner (P < 0.05, P < 0.01) while the frequency was not obviously influenced. (2) PRO (P < 0.05) or GLI (P < 0.01) partly abolished the inhibitory effect of emodin on jejunum smooth muscle. (3) L-NAME had no obvious effect on the inhibitory effect of emodin. (4) Emodin attenuated the contraction of jejunum smooth muscle induced by calcium chloride application into calcium free K-H solution (P < 0.01).
CONCLUSIONEmodin obviously inhibits the amplitude and tension, while has no influence on the frequency of jejunum smooth muscle contraction in rats. Activation of beta adrenergic receptor, open of ATP sensitive potassium channels, and inhibition of the extracellular calcium influx through calcium channels of smooth muscle cell membrane might be involved in the process.

6.The effect of chronic intermittent hypoxia to hypothalamus-pituitary-adrenal axis and growth hormone level in rats during sleep.

AIMTo research the effect of intermittent hypoxia during sleep on hypothalamus-pituitary-adrenal (HPA) axis and growth hormone (GH) level.
METHODSRats were respectively exposed to intermittent hypoxia, room air and continuous hypoxia, after 1 day, 3 days, 7 days and 30 days, mRNA levels of corticotropin-releasing hormone (CRH) and growth hormone releasing hormone (GHRH) in hypothalamus of rats were detected using RT-PCR, and the levels of CRH, GHRH, corticotropin(ACTH), cortex ketone, and growth hormone in plasma were measured.
RESULTSAfter 30 days, the CRH mRNA levels in rats hypothalamus which exposed to intermittent hypoxia were increased significantly than those exposed to continuous hypoxia as well as normal control but GHRH decreased, there was no difference between continuous hypoxia and normal control. After 1 day, 3 days, and 7 days, there was no difference between continuous hypoxia and intermittent hypoxia. After 30 days, the plasmic level of CRH,ACTH and cortex ketone increased, GHRH decreased and GH had no obvious change.
CONCLUSIONThe rats' HPA axis level increases and GHRH restrained with chronic intermittent hypoxia during sleep, feedback regulation disorders.

7.The value of diffusion tensor imaging and fiber tractography in cervical spondylotic myelopathy.

OBJECTIVETo study the diagnostic value of diffusion tensor imaging (DTI) in cervical spondylotic myelopathy.
METHODSTwenty healthy volunteers and fifty patients with cervical spondylotic myelopathy underwent DTI in the Affiliated Hospital of Medical College of Ningbo University from January 2014 to April 2015. Healthy volunteers served as controls. Fifty patients were divided into three groups (group A , B, C) according to cervical MRI scan standard. Group A (17 cases) had only the dura mater spinalis compressed; Group B (23 cases) showed the cervical spinal cord compressed, but no high signal in it; Group C (10 cases) had the cervical spinal cord compressed with high signal in the same level. The average apparent diffusion coefficients(ADC) and fractional anisotropy (FA)values in these examinee were analyzed and all subjects were performed fiber tracking.
RESULTSThere was no statistically significant differences in ADC and FA values in C2/C3, C3/C4, C4/C5, C5/C6, C6/C7 of control group (P>0.05). The average ADC and FA values in control group were (0.875 +/- 0.096) x10(3) mm2/s and 0.720 +/- 0.051, respectively; compared with group A,there was no statistically significant difference; compared with group B and C, there was significant difference; comparison among group A, B, C, there was significant differences.
CONCLUSIONDTI can early and accurately quantify the changes of microstructure in cervical spondylotic myelopathy. Fiber tracking can show the damage range of spinal cord lesions.

8.Granulocyte colony-stimulating factor gene polymorphism in patients with systemic lupus erythematosus in Northern Chinese

Objective To analyze the polymorphism(s) or mutation(s) of the granulocyte colony-stim- ulating factor gene (G-CSF,Csf3) and its possible association with the susceptibility to systemic lupus erythe- matosus (SLE).Methods Polymorphism screening was carried out by the polymerase chain reaction-single strand conformation polymorphism method (PCR-SSCP),using DNA from 204 Northern Chinese patients with SLE,459 ethnically matched healthy control,78 patients with other autoimmune diseases.Results It was i- dentified that 4 polymorphisms at position 1869,1205,1931,and 394.The nucleotide sequences of the sample that showed different SSCP patterns and different distribution between SLE and healthy controls were deter- mined by direct sequencing.Only the distribution of the fifth extron of Csf3 (1931) between SLE and controls was different.The frequency of A/G genotype (60.5%) and A allele (53.2%) at position 1931 in patients with SLE was significantly higher than that in healthy controls (51.6% and 45.2%,respectively),whereas,the fre- quency of G/G genotype (16.2%) and G allele (46.8%) in patients with SLE was lower than that in healthy controls (28.9% and 54.8% respectively).Conclusion The results suggest that Csf3 (1931) is significantly associated with the susceptibility to SLE in Northern Chinese patients .Further population and functional stud- ies will be followed to establish Csf3 (1931) as one of the susceptible genes for SLE.
9.Meta?analysis on the prognosis of preoperative glucose pretreatment in patients with digestive tract malignant tumor

Objective The clinical benefits of carbohydrate intake 2-3 h before surgery in patients with digestive tract malignant tumor without diabetes mellitus have been confirmed by foreign experts,the"High level evidence" for perioperative nutrition support were also written in China"Guidelines for perioperative nutritional support in adults".However,there are few hospitals in China that prefer preoperative glucose pretreatment,taking into account the differences between domestic and foreign data.In this paper,the clinical randomly controlled experimental data about the effect of preoperative glucose pretreatment on the prognosis of patients with malignant tumor of digestive tract was analyzed by Meta analysis,in order to evaluate the efficacy and safety of preoperative glucose pretreatment in Chinese patients,and to achieve the clinical significance.Methods Retrieving the database from establishment to March 2017,the related literatures about preoperative glucose pretreatment and prognosis of patients with digestive tract malignant tumor were collected.Revman5.3 software was used to perform Meta analysis of 10 randomized controlled trials(606 patients)who met the inclusion criteria.Results Compared with the traditional fasting and non drinking group,the preoperative subjective discomfort of the preoperative group was significantly reduced(SMD = -1.29,95%CI = -2.00--0.59,P = 0.000 3), postoperative insulin resistance was significantly reduced(SMD=-1.92,95%CI=-2.68--1.16,P<0.01), postoperative blood glucose concentration,insulin concentration increased,and no vomiting and aspiration occur in any patient as shown by the literature.Conclusion In the patients with malignant tumor of digestive tract, perioperative nutrition support and pretreatment show significant clinical benefits,preoperative subjective comfort improves,the risk of aspiration does not increase significantly,postoperative insulin resistance decreases.It is proved that preoperative carbohydrate pretreatment is effective and safe in the operation of domestic digestive tract cancer.
10.Expression of insulin-like growth factor 1 and its receptors in colorectal cancer and their significances

Objective To investigate the expression of insulin-like growth factor 1 (IGF-1), insulin growth factor receptor 1(IGF-1R)and insulin growth factor receptor 2(IGF-2R)in colorectal cancer, and their relationship with the relevant clinicopathological factors. Methods Immunohistochemical SP method was used to detect the expression of IGF-1,IGF-1R and IGF-2R in 154 cases of colorectal cancer tissues,58 cases of benign disease tissues (colorectal adenoma, polyps) and 90 normal tissues. Results The positive rate of IGF-1 and IGF-1R expression in colorectal cancer tissues [93.5%(144/154), 70.1%(108/154)] was higher than that in benign diseases [51.7%(30/58), 51.7%(30/58)] and adjacent normal tissues [18.9%(17/90), 35.6%(32/90)] (P=0.001). The positive expression rate of IGF-1 and IGF-1R in colorectal cancer tissue, benign disease tissue and adjacent normal tissues decreased gradually, and there was significant difference between any two groups (P<0.05). The positive expression rate of IGF-2R had no significant difference between any two groups (P>0.05). IGF-2R was significantly different between any two groups (P<0.05). The expression of IGF-1R and IGF-2R in colorectal cancer tissues were not significantly correlated with gender, location, tumor size, family history, depth of tumor invasion and local lymph node metastasis (all P>0.05).IGF-1 was positively correlated with the body mass index(r=0.169,P=0.036).IGF-2R was negatively correlated with age (r=-0.196, P=0.015), and positively correlated with TNM staging in patients with colorectal cancer (r=0.227, P=0.005). The expression of IGF-1 was positively correlated with IGF-1R (r=0.281, P=0.000 1). There was no significant correlation between IGF-1 and IGF-2R in cancer tissues (P>0.05). Conclusion IGF-1 and IGF-1R may promote the occurrence of colorectal cancer, and IGF-2R may be associated with the progress of colorectal cancer,and obesity is a risk factor for incidence of colorectal cancer.
