1.Isolation and purification of basement membrane zone mono-specific autoantibodies by spot affinity purified method

Objective:To investigate the value of spot affinity purified method in the isolation and purification of mono specific basement membrane zone autoantibodies(BMZ Ab).Methods:Utilizing human dermoepidermal antigen transmitted on NC membrane,We purified 3 cases with mono specific BMZ Ab in sera of SABD patients which was oppositive by immunoblotting.Results:The BMZ Ab purified by this method could specifically combine its target antigen.Conclusion:Spot affinity purified method could be widely used in the isolation and purification of autoantibodies produced by mixed micro antigen.
2.The relationship of inhibin-B in serum and endometrium

Objective To detect the frequency and tissue distribution pattern of inhibin subunits in normal endometrium and periph-eral blood, and investigate the precise action in endometrium. Methods Normal cndometrial tissues were obtained from 16 women in the proliferative phase with salpingo-infertility, 11 women in the early secretory phase and 17 women in the late secretory phase with cervical cancer from Jan to Dee 2006. Expression of INH subunits were detected by immunohistochcmistry. INH-B in the blood samples were detec-ted. None of patients took any steroid drugs before 6 months of giving the samples. Results Immunohistochemistry showed that INH sub-units expressed in normal endometrial tissues. The expression intensity of INH subunits gradually increased in different menstrual phases. The level of inhibin-B in proliferative phase was higher than that in late secretory phase. Conclusions The relationship of inhibin-B level in serum and expression of inhibin subunits in endometrium showed that inhibin could be involved in autocrine/paracrine signaling and contrib-ute to several aspects of endometrial maturation.
3.Related investigation and thinking on the course of Laboratory diagnostics in general medical train-ing from the Navy physicians

Objective To investigate and analyze the application and effect of the course of Labo-ratory diagnostics in general medical training from the Navy physicians. Methods 37 students attending the general medical training in our university in September 2015 were selected as the subject. At the end of the course, the “Questionnaire Survey on Teaching Feedback” was designed and conducted among them to understand the students'!opinions on the arrangement, teaching content and teaching methods of the experi-mental diagnostics. A total of 37 questionnaires were issued and retrieved effectively and the survey data were collected and summarized. Results Among 37 questionnaires, 100%of trainees (n=37) thought it was necessary to set up the Laboratory diagnostics course in the general medical training, 72.9%(n=27) suggested 3 to 4 teaching hours, and on the demand side of teaching content settings, 91.9%(n=34) chose analyses of blood, urine, stool routine tests and then tests of liver and kidney function. The proportions of students to recommend theory teaching, case discussion and practice operation were 24.3% (n=9), 51.4% (n=19) and 48.6% (n=18) respectively. Conclusions The setup of the Laboratory diagnostics course plays an active role in the general medical training from navy physicians. In the future, according to the demand of the general medical training, teachers should optimize the teaching contents and learning hours, improve the teaching methods and enhance the training quality.

5.Sensitivity of Plasmodium vivax to Chloroquine in Laza City,Myanmar

Fourty-eight patients infected by Plasmodium vivax were randomly divided into chloroquine regimen groups A and B.Each case in group A received a total dosage of 1 200 mg(base) over a 3-day period(600 mg on the first day then 300 mg daily).Cases in group B each received total dosage of 1 500 mg(base) in 3 days(750 mg on the first day then 375 mg daily).Body temperature and blood smears were examined on days 0, 1, 2, 3, 7, 14, 21 and 28, respectively.All patients were negative for asexual parasites within 3 days.The cure rate on day 28 was 100%.Results showed that P.vivax is sensitive to chloroquine in Laza, Myanmar, and chloroquine can still be used for the imported vivax malaria cases at the China-Myanmar border.
6.Local recurrence after complete resection of primary retroperitoneal liposarcoma

Objective To investigate factors impacting on local recurrence in cases of primary retroperitoneal liposarcoma (RPLS) after complete enbloc resection.Methods The clinical data of 47 cases of primary retroperitoneal liposarcoma were retrospectively analyzed.Results 3-year local recurrence rate was 62%.Single and multiple regression analysis showed preoperative needle biopsy,histological subtype,tumor size and capsular invasion were all risk factors related to postoperative local recurrence in primary retroperitoneal liposarcoma undergoing complete resection (P < 0.05).Conclusion Preoperative aspiration biopsy,tumor poor differentiation,large tumor size tumor extracapsular spread all lead to high local recurrence rate after complete resection of primary retroperitoneal liposarcoma.
7.Separation, Identification and Inhibitory Effect on HepG2 Cells of Flavonoids from Humulus Scandens

Objective:To extract, separate and identify the flavonoids in Humulus Scandens and detect their cytotoxic effect on cells HepG2. Methods: After Humulus scandens bing was extracted by alcohol, luteolin-7-O-β-D-glucoside ( LGL) and cosmoiin ( AGL) were obtained by using column chromatography and identified by spectral analysis. The inhibitory effects of LGL and AGL on HepG2 cells were detected by a typan blue dye method. Results:The main flavonoids in Humulus Scandens were LGL and AGL. When the concentration of LGL and AGL was increased from 6. 25 μmol·L-1 to 100. 0 μmol·L-1 , the inhibition rate of LGL was increased from 28. 8% to 95. 2% and that of AGL was increased from 3. 6% to 21. 3%. Conclusion: LGL has notable inhibitory effect on HepG2 cells in a dose-dependent manner, while the inhibitory effect of AGL is not obvious.

Aims To extract,separate and identify the flavonoid constituents in Humulus Scandens and to ex-plore the relationship of monomers and alveolar fluid clearance (AFC)in mice in vivo.Methods Humulus scandens were extracted with alcohol and then isolated by the technology of Column and the structures were i-dentified by spectrometry.In vivo AFC was measured using bovine serum albumin protein assays affected by luteolin-7-O-β-D-glucoside (LGL ) and cosmsiin (AGL).Results The main constituents of flavanones
in Humulus scandens were LGL and AGL.Both of them could improve the AFC.Conclusion The AFCs of LGL and AGL,compared to the blank control group, increased which explains the effect of flavonoid constit-uents on removing edema and promoting water absorp-tion.
9.Detection of TNF-related apoptosis inducing ligand gene expression by real-time fluorescent quantitative method

Objective To establish a real time fluorescent quantitative revers transcripatase PCR(FQ-RT-PCR) method to detect the expression level of TNF-related apoptosis inducing ligand (TRAIL) mRNA in peripheral blood mononuclear ceils (PBMC) and determine its expression level in healthy donors, HBV-caused cirrhosis patients and PBC ones. Methods Specific primers and Taqman-MGB probe were designed and β-actin was used as endogenous control. The amplified fragment was obtained by RT-PCR. The quantitative template was constructed and then the fluorescent intensity was documented on the ABI Prism7000 analyzer. The standard curve was established, according to which, the TRAIl. mRNA levels in 30 healthy individuals, 30 patients with primary biliary cirrhosis (PBC) and 25 ones with HBV-caused cirrhosis were calculated automatically by software after the values of cycle threshold (Ct) were detected continuously during amplification. Results The linear detection range of the assay for TRAIL gene was 103 - 109 copies/ ug RNA ( r=-0.997). The coefficients of variation of both intra-and inter-assay reproducibility for high concentration samples were 5.6% and 6. 3% , respectively, and those for low concentration samples were12.5% and 14. 6%. The TRAIL mRNA expression level in PBC patients was [ (3.3±2.5)×105copies/ugRNA] significantly higher than that of healthy control [ (0.5±0.2)×105 copies/ug RNA ] (t=5.994,P <0.01). TRAIl. mRNA level of HBV-caused cirrhosis patients[ (2.1±0.9)×105 copies/ug RNA] wasalso significantly elevated (t=8.536, P<0.01). However, the difference between these two diseased groups had no significance. Conclusion We have successfully set up a FQ-RT-PCR method for detecting TRAIL gene expression and found that its expression levels of peripheral blood mononuelear cells in PBC and HBV caused cirrhosis patients are elevated, which provides a new insight into mechanism study of liver injury caused by cirrhosis.
10.Evaluation of the Enzyme-linked Immunosorbant Assay in Detecting Circumsporozoite Protein of Anopheline Vectors in Yunnan

Objective To detect circumsporozoite protein (CSP) in anopheline vectors from south Yunnan and to evaluate ELISA in the detection. \ Methods\ Salivary glands of the anopheline mosquitoes were taken for finding sporozoites by microscopy and part of the glands was used for detecting CSP by ELISA. An. minimus was experimentally infected by blood from vivax malaria patient (with Plasmodium vivax) and examined for sporozoites and CSP. Eight species of anopheline mosquitoes were caught in the field and examined. Monoclonal antibodies to P.falciparum (Pf2A10) and P.vivax (Pv210, Pv247) were used in ELISA for detecting CSP. \ Results\ Sporozoites were found in the salivary glands of 27 out of 36 An. minimus experimentally infected (75\^0%), 29 were ELISA CSP positives (80\^6%), and 26 of the 27 mosquitoes showed Pv210 CSP positive. Among \{1 010\} parous anopheline mosquitoes from the field, 7 were found sporozoite positive (0\^69%), 8 were ELISA CSP positive (0\^79%), and 6 of the 7 mosquitoes showed CSP positive. Of \{4 675\} wild mosquitoes in 8 anopheline species with different ages, 11 were found CSP positive (0\^24%) including An.minimus, An.sinensis and An.maculatus with a positive rate of 0\^20%, 0\^24% and 0\^39% respectively.Among the 11 mosquitoes, 9 were Pv210 positive and 2 were Pf2A10 positive. Conclusion CSP detection by ELISA is a useful method to monitor the malaria transmission capacity of anopheline vectors.
