1.The investigation of reference intervals of thyroid hormone within 18-60 years old healthy people in Haikou area

Objective To establish the reference intervals for thyroid-stimulating hormone(TSH) ,triiodothyronine(T3 ) ,thy-roxine(T4 ) ,free triiodothyronine(FT3 ) and free thyroxine(FT4 ) in healthy people aged between 18 and 60 years old in Haikou are-a .Methods According to the recommendations of clinical and laboratory standards institute(CLSI)C28-A3 document ,401 persons aged between 18 and 60 years old were recruited from Haikou(275 male ,126 female) .The makers were analyzed by gender .Estab-lished the reference intervals of TSH ,T3 ,T4 ,FT3 and FT4 in Haikou area .Results Reference intervals based on gender were estab-lished as TSH(0 .58-4 .60 mIU/L) ,T3 (1 .11-2 .31 nmol/L for male ,0 .97-2 .13 nmol/L for female) ,T4 (72 .24-152 .22 nmol/L ) ,FT3 (4 .17-6 .33 pmol/L for male ,3 .95-5 .47 pmol/L for female) ,FT4 (13 .16-21 .29 pmol/L for male ,12 .13-19 .61 pmol/L for female) .Conclusion This study has established the reference intervals of TSH ,T3 ,T4 ,FT3 and FT4 in healthy people aged between 18 and 60 years old in Haikou area .
2.Separation, Identification and Inhibitory Effect on HepG2 Cells of Flavonoids from Humulus Scandens

Objective:To extract, separate and identify the flavonoids in Humulus Scandens and detect their cytotoxic effect on cells HepG2. Methods: After Humulus scandens bing was extracted by alcohol, luteolin-7-O-β-D-glucoside ( LGL) and cosmoiin ( AGL) were obtained by using column chromatography and identified by spectral analysis. The inhibitory effects of LGL and AGL on HepG2 cells were detected by a typan blue dye method. Results:The main flavonoids in Humulus Scandens were LGL and AGL. When the concentration of LGL and AGL was increased from 6. 25 μmol·L-1 to 100. 0 μmol·L-1 , the inhibition rate of LGL was increased from 28. 8% to 95. 2% and that of AGL was increased from 3. 6% to 21. 3%. Conclusion: LGL has notable inhibitory effect on HepG2 cells in a dose-dependent manner, while the inhibitory effect of AGL is not obvious.
3.Separation and identification of flavonoid constituent in Humulus Scandens and their effects on alveolar fluid clearance in mice

Aims To extract,separate and identify the flavonoid constituents in Humulus Scandens and to ex-plore the relationship of monomers and alveolar fluid clearance (AFC)in mice in vivo.Methods Humulus scandens were extracted with alcohol and then isolated by the technology of Column and the structures were i-dentified by spectrometry.In vivo AFC was measured using bovine serum albumin protein assays affected by luteolin-7-O-β-D-glucoside (LGL ) and cosmsiin (AGL).Results The main constituents of flavanones
in Humulus scandens were LGL and AGL.Both of them could improve the AFC.Conclusion The AFCs of LGL and AGL,compared to the blank control group, increased which explains the effect of flavonoid constit-uents on removing edema and promoting water absorp-tion.
4.Detection of TNF-related apoptosis inducing ligand gene expression by real-time fluorescent quantitative method

Objective To establish a real time fluorescent quantitative revers transcripatase PCR(FQ-RT-PCR) method to detect the expression level of TNF-related apoptosis inducing ligand (TRAIL) mRNA in peripheral blood mononuclear ceils (PBMC) and determine its expression level in healthy donors, HBV-caused cirrhosis patients and PBC ones. Methods Specific primers and Taqman-MGB probe were designed and β-actin was used as endogenous control. The amplified fragment was obtained by RT-PCR. The quantitative template was constructed and then the fluorescent intensity was documented on the ABI Prism7000 analyzer. The standard curve was established, according to which, the TRAIl. mRNA levels in 30 healthy individuals, 30 patients with primary biliary cirrhosis (PBC) and 25 ones with HBV-caused cirrhosis were calculated automatically by software after the values of cycle threshold (Ct) were detected continuously during amplification. Results The linear detection range of the assay for TRAIL gene was 103 - 109 copies/ ug RNA ( r=-0.997). The coefficients of variation of both intra-and inter-assay reproducibility for high concentration samples were 5.6% and 6. 3% , respectively, and those for low concentration samples were12.5% and 14. 6%. The TRAIL mRNA expression level in PBC patients was [ (3.3±2.5)×105copies/ugRNA] significantly higher than that of healthy control [ (0.5±0.2)×105 copies/ug RNA ] (t=5.994,P <0.01). TRAIl. mRNA level of HBV-caused cirrhosis patients[ (2.1±0.9)×105 copies/ug RNA] wasalso significantly elevated (t=8.536, P<0.01). However, the difference between these two diseased groups had no significance. Conclusion We have successfully set up a FQ-RT-PCR method for detecting TRAIL gene expression and found that its expression levels of peripheral blood mononuelear cells in PBC and HBV caused cirrhosis patients are elevated, which provides a new insight into mechanism study of liver injury caused by cirrhosis.

6.Analyses of genetic model of psoriasis vulgaris.

OBJECTIVETo explore the possible genetic model of psoriasis vulgaris.
METHODSThe complex segregation analysis and heritability calculation were performed with the aid of Penrose method, Falconer regression method and SAGE-REGTL program.
RESULTSIt was found that in 1043 patients with psoriasis vulgaris, 305 patients (29.24%) have the family history of psoriasis, and 738 patients have not the family history. A ratio of s/q approached 1/(square root of q) with Penrose method, and the heritability values of psoriasis in the first-degree and second-degree relatives were 67.04%, 46.6% respectively. By complex segregation analysis, Mendelian, non-major-gene model and environment model were rejected for psoriasis.
CONCLUSIONThe results suggest that psoriasis follows a pattern of polygenetic or multifactorial inheritance rather than single-gene inheritance.

7.Effect of Morindae Officinalis Radix Processed with Different Proportions of Glycyrrhizae Radix et Rhizoma on Improvement of Renal Function and HPG Axis of Kidney Yang Deficiency Model Rats Induced by Adenine

ObjectiveTo explain the scientific connotation of Morindae Officinalis Radix (MOR) processed by Glycyrrhizae Radix et Rhizoma (Gly) by comparing the effect of raw products of MOR and processed products of MOR with different proportions of Gly (GMOs) on the improvement of renal function and hypothalamic-pituitary-gonadal (HPG) axis, the protein expression of Wnt/β-catenin and transforming growth factor-β1 (TGF-β1)/Smad signal pathways in kidney Yang deficiency model rats induced by adenine. MethodGMOs were prepared according to method under MOR in 2020 edition of Chinese Pharmacopoeia. Rat model of kidney Yang deficiency was established by intragastrical administration of adenine, levels of follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2) and testosterone (T) were measured by enzyme-linked immunosorbent assay (ELISA). Levels of urea nitrogen (BUN) and serum creatinine (SCr) were measured by spectrophotometry, hematoxylin-eosin (HE) staining was used to evaluate the pathological changes of kidney, testis and epididymis. Immunohistochemistry (IHC) was used to analyze the protein expression of E-cadherin, α-smooth muscle actin (α-SMA), Wnt2b, β-catenin, Smad1 and Smad4. ResultMOR processed with 100∶6 and 100∶12 proportions of Gly (short for GMO/100∶6 and GMO/100∶12) had the most obvious improvement on the body posture of kidney Yang deficiency model rats. GMO/100∶12 had the best effect on reducing the levels of BUN, SCr, FSH, LH and the ratio of E2/T. GMO/100∶6 and GMO/100∶12 had the best effect on regulating the protein expression of E-cadherin, α-SMA, Wnt2b, β-catenin, Smad1 and Smad4. ConclusionGMO/100∶6 and GMO/100∶12 have the a good effect on the improvement of renal function and HPG axis in kidney Yang deficiency model rats induced by adenine, which is related with the fact that they can regulate Wnt/β-catenin pathway in renal and testicular tissue and TGF-β1/Smads pathway in testicular tissue.
8.Comparison of Effect of Euodiae Fructus Processed with Different Proportions of Glycyrrhizae Radix et Rhizoma on Oxidative Stress and Autophagy in Mouse Liver Under Toxic Dose

ObjectiveTo investigate the principle and scientific connotation of Euodiae Fructus(EF) processed with Glycyrrhizae Radix et Rhizoma(Gly) by comparing the effects of unprocessed products of EF(UEF) and processed products of EF with the different proportions of Gly(GEFs) at toxic doses on oxidative stress and autophagy in the liver of mice. MethodSeventy mice were randomly divided into 7 groups, namely the control group, the UEF group, the group of the processed products of EF without Gly(PEF) and 4 groups of GEFs(the mass ratios of EF to Gly were 100∶3, 100∶6, 100∶12 and 100∶24, respectively, hereinafter referred to as the processed products of EF with the mass ratios of 100∶3, 100∶6, 100∶12 and 100∶24 of Gly). The mice were given purified water, the decoction of UEF, PEF and GEFs by gavage at a dose of 30 g·kg-1. PEF and GEFs were prepared according to the method under EF in the 2020 edition of Chinese Pharmacopoeia. Levels of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were determined by ultraviolet-visible spectrophotometry, hematoxylin-eosin(HE) staining was used to evaluate the pathological changes of liver tissue, the level of reactive oxygen species(ROS) was detected by fluorescence method, the mRNA expression of heme oxygenase-1(HO-1), quinone oxidoreductase-1(NQO1), glutathione-S-transferase 1(GSTA1), Kelch-like epichlorohydrin-associated protein 1(Keap1) and p62 were measured by Real-time fluorescence quantitative polymerase chain reaction(Real-time PCR), western blot was used to detect the protein expression of phosphorylated mammal target of rapamycin(p-mTOR), phosphorylated ribosomal p70 S6 protein kinase(p-p70S6K), p62, microtubule-associated protein 1 light chain 3Ⅰ(LC3Ⅰ) and LC3Ⅱ. ResultCompared with the control group, after 7 d of administration, the increase in body mass of mice in the UEF group began to slow down and the difference gradually increased, and the liver body index significantly increased(P<0.01), pathomorphological observation showed that the structure of hepatic lobules was disordered, and local hepatic sinuses were narrowed or disappeared, and there were inflammatory infiltration and local bleeding, the levels of ALT and AST in serum and ROS in liver tissue were significantly increased(P<0.01), and the expressions of Keap1, HO-1, NQO1, GSTA1, p62 mRNA and p-mTOR, p-p70S6K, p62 protein in liver tissue were significantly decreased(P<0.01), and LC3Ⅱ/LC3Ⅰ was significantly increased(P<0.01). Compared with the UEF group, the body mass of mice increased, and the liver body index, the levels of ALT and AST in serum, and the level of ROS in liver tissue all decreased in the groups of PEF and GEFs. Among these groups, only the liver lobules in GEF(100∶6) group were intact, and the size of liver sinuses was close to that in the control group. The mRNA expressions of Keap1, HO-1, NQO1, GSTA1 and p62 in liver tissue showed an overall upward trend in the groups of PEF and GEFs. Among these groups, only the ones of the above mRNA in the GEF(100∶6) group had a significant increase(P<0.05, P<0.01). The protein expressions of p-mTOR, p-p70S6K, p62 and LC3Ⅱ/LC3Ⅰ had a callback in the groups of PEF and GEFs, of which the protein expressions of p-mTOR, p-p70S6K and LC3Ⅱ/LC3Ⅰ in the GEF(100∶6) group and the expression of p62 protein in the GEF(100∶24) group had the largest callback. Except for p-mTOR protein, other protein expressions were statistically significant(P<0.05, P<0.01). ConclusionThe hepatotoxicity of EF is closely related to its ability to induce oxidative stress, which leads to pathological autophagy and hepatocyte damage. This ability can be reduced by the processing with different proportions of Gly, especially the ratio of 100∶6.
9.The structure and function analysis of bacterial community during aerobic composting of chicken manure.

In order to determine the changes of bacterial community structure and function in the early, middle and late stage of aerobic composting of chicken manure, high-throughput sequencing and bioinformatics methods were used to determine and analyze the 16S rRNA sequence of samples at different stages of composting. Wayne analysis showed that most of the bacterial OTUs in the three composting stages were the same, and only about 10% of the operational taxonomic units (OTUs) showed stage specificity. The diversity indexes including Ace, Chao1 and Simpson showed a trend of increasing at first, followed by decreasing. However, there was no significant difference among different composting stages (P < 0.05). The dominant bacteria groups in three composting stages were analyzed at the phylum and genus levels. The dominant bacteria phyla at three composting stages were the same, but the abundances were different. LEfSe (line discriminant analysis (LDA) effect size) method was used to analyze the bacterial biological markers with statistical differences among three stages of composting. From the phylum to genus level, there were 49 markers with significant differences among different groups. The markers included 12 species, 13 genera, 12 families, 8 orders, 1 boundary, and 1 phylum. The most biomarkers were detected at early stage while the least biomarkers were detected at late stage. The microbial diversity was analyzed at the functional pathway level. The function diversity was the highest in the early stage of composting. Following the composting, the microbial function was enriched relatively while the diversity decreased. This study provides theoretical support and technical guidance for the regulation of livestock manure aerobic composting process.

10. Clinical features, diagnosis and treatment of silent sinus syndrome

Objective:
To explore the clinical features, diagnostic methods and therapeutic strategy of silent sinus syndrome (SSS).
Methods:
A retrospective study was made on eight SSS patients treated during 2013-2016 in Longgang ENT Hospital and Otorhinolaryngology Hospital of the First Affiliated Hospital of SUN Yat-sen University. The following clinical data, including demographic data, symptoms, history of trauma and surgery, signs, imaging examination, endoscopic surgery and postoperative outcomes, were analyzed to summarize the diagnosis and treatment experiences.
Results:
Eight SSS patients showed the following clinical features: the proportions of both sexes and sinus sides were 4 to 4; seven cases (7/8) were adult, with an average of (48.1±11.8)y; seven cases (7/8) had long history of trauma or surgery, with an average of (17.9±10.5)y; seven cases (7/8) cannot recall the exact course of SSS; six cases (6/8) had no nasal symptoms; eight cases (8/8) had unilateral ocular discomforts; eight cases (8/8) had signs of unilateral enophthalmos (2-5 mm), accompanied with hypoglobus; and by CT and MRI scanning, eight cases (8/8) showed the unilateral maxillary sinus outlet obstruction, sinus full opacification, sinus wall bony rarefaction, sinus wall contraction, sinus volume loss, and the ipsilateral orbital floor bowing descent and orbital volume increase. After treated by endoscopic sinus surgery and followed-up for one year, four cases were cured, and the other four improved; no intra- or post-operative complications occurred; and no individual need a two-stage orbital plastic reconstruction.
Conclusions
SSS often develops in the unilateral maxillary sinus of adult patient with long history of trauma or surgery, but the nasal symptoms and signs are silent. Diagnostic for SSS depends on characteristic ocular sign and sinus CT imaging. Endoscopic sinus surgery helps to improve ocular and nasal signs and promote orbital self-reconstruction.
