1.Experimental studies on hematopoiesis reconstitution by using of transplantation of gene-modified bone marrow cells

Objective To explore the biological features of hematopoiesis reconstitution by using genetic marking.Methods NeoR gene was transduced into bone marrow(BM)cells of mouse mediated by liposome.Then these cells were transplanted into lethally irradiated recipients.The hematopoiesis reconstitution was observed and the marker gene in spleen and BM cells of recipients after hematopoiesis reconstitution was examined.Results The transplanted recipients remained alive and healthy.But the irradiated mice with no transplantation died from BM aplasia soon.Meanwhile,the cells from spleen and BM of transplanted mice could be alive in G418 system,and contained the DNA fragments of NeoR gene by PCR.Conclusion Gene-modified BM cells could be used to reconstitute hematopoiesis successfully and express the foreign gene to some extent stably.
2.Functional expression of CD95/Fas antigen and Bcl-2 on cord blood hematopoietic progenitor cells.

The cell-surface expression and functional status of the CD95/Fas antigen on primitive hematopoietic progenitors isolated from human cord blood (CB) were studied. The CD34+ cells freshly isolated from CB displayed low CD95 expression. The combinations of cytokines such as SCF + FL could up-regulate the expression of CD95 in vitro culture and tumor necrosis factor-alpha (TNF-alpha) and interon-gamma (IFN-gamma) further increased the CD95 expression induced by positive cytokines. The functional status of CD95-mediated apoptosis were analyzed by incubation of CD34+ CB cells in the presence of anti-CD95 monoclonal antibodies (McAbs). The effects of anti-CD95 McAbs were measured by viable cell counting, flow cytometry, LTIC and CFU-C assays. A decrease of viable cells, CFU-C and LTIC numbers were observed in the presence of anti-CD95 McAbs and TNF-alpha or IFN-gamma. However, growth factor deprivation or the early-acting cytokine such as SCF and FL cross-linking to CD95 caused low apoptosis of CD34+ cells. The correlation of increased intracytoplasmic levels of bcl-2 and the presence of CD95 on fresh CB CD34+ cells suggested that bcl-2 might be involved in protecting against CD95-mediated apoptosis of CB CD34+ cells.

3.Description and mechanism of cognitive impairment post-HSCT

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is a highly successful treatment for hematological malignancies,severe congenital immunodeficiencies and some other diseases.Approximate 50 % of the disease-free survival patients suffered from long-lasting severe cognitive impairment after allo-HSCT.Pathological evidences showed parenchymal lymphocytic inflammation,microglia activation,and mild cerebral angiitis-like changes in allogeneic transplanted animals and patients' autopsy.Pay much importance attention to the pathological changes of central nervous system and its impact on cognitive function and take suitable measures would help the patients to achieve a state of complete physical mental and social well-being.

5.Estrogen receptor ERα36 gene silencing impacts the expression of growth-associated protein in PC12 cells

Objective To investigate the effects of ERα36 (a novel subtype of estrogen receptor alpha) on growth and proliferation in PC12 cells via examining the expression of growth-associated protein in differentiated PC12 cells after ERα36 gene silencing.Methods Transfection of ERα36-shRNA plasmid into PC12 cells was performed to establish the ERoα36 gene silencing cells model (PC12-36L1,PC12-36L2).Immunocytofluorescence was used to examine the expression of ERα36,and Western blot was used to analyze the expression of PCNA,cyclinD1 and MAPK in the PC12 cells.Results ① ERα36 was expressed in both cell types(PC12-36C1,PC12-36L1 and PC12-36L2).Compared with PC12-36C1,PC12-36L2 cells(OD value were respectively 0.95±0.05,0.78±0.10),PC12-36L1 cells significantly decreased expression of ERα36(OD value 0.47±0.12,P<0.01).② Compared with PC12 and PC12-36C1 cells,PC12-36L1 cells were significantly higher expression of PCNA,CyclinD1 and p-MAPK(P<0.01)(OD value of PCNA,CyclinD1 and p-MAPK:PC12 cells were respectively 1.00±0.05,1.00± ±0.11,1.00±0.05,PC12-36C1 cells were respectively 1.09±0.15,0.92±0.23,1.12± 0.08,PC12-36L1 cells were respectively 1.74±0.12,2.20±0.25,1.77±0.06).Conclusion ERα36 gene silencing can promote the growth and proliferation in PC12 cells.It suggests that the lower expression of ERα36 may be related to the diseases in nervous system such as brain tumor.

Objective In order to study the effect of the Fas system on normal peripheral blood lymphocytes(PBL)activated by allospecific antigen.Methods The isolated and purified lymphocytes werc subjected to one-way mixed lymphocyte culture.Fas antigen exprexsion on the surface of T lymphocytes freshly isolated and activated was detected dynamically by using flow-cytometry(FCM).The influence of the Fas system activated or not on the rate of lymphocytic apoptosis after mixed culture was investigated by using DNA electrophoresis and TdT-mediated dUTP nick end labeling(TUNEL).Results The Fas expression on the surface of activated T lymphocytes was increased and the rate of apoptosis of the grow with addition of anti-Fas monoclonal antibody was higher than that of the group without addition of antibody(P<0.05).Conclusion It was possible to dispose of the lymphocytes activated by specific antigen in vitro by anti-Fas monoclonal antibody.
9.Efficacy and safety of polyethersulfone highflux dialyzer for maintenance hemodialysis patients

Objective To investigate the efficacy and safety of polyethersulfone highlux dialyzer for maintenance hemodialysis patients. Methods Thirty-six maintenance hemodialysis patients were randomizedresigned into two groups,polyethersulfone 14HF(PES-14HF)group and polyethersulfone 150DS(PES-150DS)group,based on a random number table. The patients from two groups received different dialyzer hemodialysis for over 3.5 hours/each time respectively. Changes of serum creatinine,urea,β2-microglobulin,hemoglobin and albumin levels were measured for determination of the efficacy and safety evaluation. Results In the comparison between before or after hemodialysis,the level of serum creatinine,urea,β2-microglobulin levels decreased significantly to(333.8 ± 89. 5)μmol/L,(7. 0 ± 1.9)mmol/L,(22. 9 ± 1.7)mmol/L from(990. 2 ± 191.2)μmol/L,(24. 7 ± 4. 1)mmol/L,(13.6 ± 3.3)mmol/L respectively in the PES1 4 HF group(P < 0.01);the level of serum creatinine,urea,β2-microglobulin levels decreased significantly to(395.5 ± 86.1)μmol/L,(8. 1± 2. 8)mnol/L,(18.0 ± 3.0)mmol/L from(1059. 5 ± 179. 4)μmol/L,(25.3 ± 4. 8)mmol/L,(22. 3 ± 2. 9)mmol/L respectively in the PES-150DS group(P < 0. 01). We found no significant differences in each measured index between two types of hemodialysis(P > 0. 05 respectively),while the level of β2 -microglobulin levels decreased more significantly in the PES14HF group(42. 81 ± 12. 48)mg/L than PES-150DS group(24. 21 ± 13. 24)mg/L(P =0. 017). Conclusions The efficacy and safety of the PES-14HF hollow fiber membrane hemodialyzer is equivalent to that of the PES-150DS hemodialyzer in hemodialysis for uremic patients.
