Chinese Journal of Microbiology and Immunology 2011;31(4):299-304
Objective To investigate the effects of platelet-derived growth factor(POGF),basic fibroblast growth factor(bFGF)and epidermal growth factor(EGF)on the proliferation, migration and phenotypie modulation of airway smooth muscle cells(ASMCs)in vitro. Methods Cultured human ASMCs were stimulated with PDGF.bFGF,EGF or DMEM(control).Cell pmliferation was detected by BrdU incorporation and cell count.Cell migration was measured using Boyden's chamber.Reverse transcription-poly-merase chain reaction(RT-PCR)and Western blot analysis were used for the detection of sm-α-actin and sm-MHC mRNA and protein expression in ASMCs.Resuits The mitotic index(%BrdU-positive per total number of cells)of ASMCs stimulated with PDGF.bFGF and EGF were significantly higher than those in control(P<0.001 or P<0.05).And the cell number of ASMCs stimulated with PDGF,bFGF and EGFwere also significantly higher than those in control(P<0.05).The number of migrating cells treated with PDGF,bFGF and EGF were higher than that in eontrol(P<0.05).The expressions of sm-α-actin mRNA treated with bFGF and EGF were significantly lower than those in control.but it was higher than control when treated with PDGF.The expressions of sm-MHC mRNA treated with bFGF and EGF were significantly lower than those in control, but it was higher than control when treated with PDGF(P<0.01 orP<0.05).Theexpressions of sm-α-actin protein treated with bFGF and EGF were significantly lower than those in control, but it was higher than control when treated with PDGF(P<0.05).The expressions of sm-MHC protein treated with bFGF and EGF were significantly lower than those in control.but it was higher than control when treated with PDGF(P<0.05).Conclusion PDGF,bFGF and EGF can directly induce ASMCs,proliferation and migration in vitro;and meanwhile, the expressions of cells, contractive phenotype increased treated with PDGF and decreased treated with bFGF and EGF.
2.Chronic inflammatory cytokines: Associations with type 2 diabetes and obesity
Academic Journal of Second Military Medical University 2000;0(11):-
Role of chronic inflammation in type 2 diabetes and obesity has been recognized in the past few years. Examples of potent inflammatory cytokines are C-reactive protein (CRP),tumor necrosis factor-?(TNF-?) ,plasminogen activator inhibitor-1 (PAI-1) and interleukin-6 (IL-6). Inflammatory processes may have important roles in the development of type 2 diabetes, obesity and insulin resistance. This article reviewed the relationship between some inflammatory cytokines and diabetes, providing references for clinical diagnosis and treatment.

Objective To investigate the use of antimicrobial drugs in our hospital and provide guide for clinical use of antimicrobial drugs. Methods Tabulate and analyze the usage of antimicrobial drugs according to the discharged patients' medical records from Jan 2004 to Dec 2006.Then use statistical methods for retrospective survey. Results Among 4003 medical records,2756 of them used antibiotics, with the percentage being 68.85%.1103 recodes used combined medication, which accounted for 40.02% of the total records.Type I incision occupied 90.86% in surgical departments. The percentage of the inpatients who used antimicrobial drugs after operation for about five to seven days was 78.85%.For selecting drugs, the majority were empirical antibiotics treatment, which achieved 93.72%.The ratio of irrational use of drugs was 25.90%.Conclusion The clinical use of antimicrobial drugs in our hospital is dramatically lower than the average level of the whole nation.However,non-standardized administrations of medication,super-standardization,super-range use of drugs are still the problems needed to solve.

Objective To investigate the characteristics in vitro and hemostatic function of O -carboxymethyl chitosan multi-hemostatic sponge. Methods Freeze-drying technology was applied on preparation of O -carboxymethyl chitosan hemostatic sponge. The physicochemical properties of the self-produced multi-hemostatic sponge were observed for its appearance, porosity rate, water absorption, and density. The hemostatic effect was compared between the self-produced hemostatic sponge and commercially available absorbable gelatin sponge in rabbit models with ear venous and arterial injuries. Results The self-produced hemostatic sponge was off-white and full of pores with good tenacity and even net texture. Its porosity ratio, water absorption rate, and density were 67. 23%,38. 77%,and 0. 043 4 g·(cm3)-1,respectively. The bleeding time volume were significantly lower from the self-produced sponge than that from the commercially available gelatin sponge. No secondary re-bleeding was observed. Conclusion The self-produced O -carboxymethyl chitosan multi-hemostatic sponge displays stable physicochemical characteristics and reliable hemostatic effect.

Objective To observe the therapeutic effect and effects on coagulation indexes ofXuefu Zhuyu Capsules combined with chemotherapy in treating patients with mid-advanced small cell lung cancer (SCLC).Methods Totally 100 patients with mid-advanced SCLC were randomly divided into observation group and control group with 50 cases in each group. The observation group was given EP chemotherapy combined withXuefu Zhuyu Capsules, while the control group was given EP chemotherapy for 4 cycles. The coagulation indexes of two groups before and after treatment were observed, and the clinical efficacy and TCM syndrome efficacy were evaluated. Results After treatment, the tumor volume and TCM syndrome scores of two groups were lower than those before treatment (P<0.01), and the observation group was lower than the control group (P<0.01). The effective rate of the observation group was 100%, higher than 88% of the control group, the difference was statistically significant (P<0.05). The improvement rate of TCM syndrome score was 98% in the observation group, and was 84.0% in the control group. The observation group was superior to the control group (P<0.05). In observation group, D-dimer, platelet count and fibrinogen after treatment were lower than those before treatment (P<0.05,P<0.01), and were lower than those in control group (P<0.01). There was no significant difference of all kinds of adverse reactions between the two groups (P>0.05).Conclusion EP chemotherapy combined with Xuefu Zhuyu Capsules can improve hypercoagulable state in patients with SCLC, improve the effect of chemotherapy, and do not increase the adverse reactions.

Objective:To investigate the dynamic change of tetramethylpyrazine (TMP) in processing of Chuangxiongchatiao Granules. Methods: To explore the influences of the different extraction, concentration and drying method on the transferring rate of TMP by HPLC. Results: Transferring rate of TMP is higher by alcohol than by water. The loss of TMP in concentrating process is lower by rotating distillation than by direct heating concentration and by reduced pressure concentration. The transferring rate of TMP is higher by reduced pressure desiccation than by ordinary pressure. Conclusion: The better extrating way of Chuangxiong is by alcohol in the processing of Chuangxiongchatiao Granules; the concentrating and drying process should be done at the low temperature and by rapid dry process.
