1.THE ANALYSIS OF GENITAL TRACT MYCOPLASMA INFECTION AND TUBAL OBSTRUC-TION IN 60 CASES OF INFERTILITY PATIENTS

Objective To investigate the relationship between women's reproductive tract mycoplasma infec-tion and tubal obstruction effusion .Methods A retrospective analysis of 60 cases infertility infertility clinic patients and healthy control were detected in our hospital from Junuary 2013 to December 2014.The cervical secretion sam-ples were used for detection of Mycoplasma ( leucorrhea , Ureaplasma urealyticum ( Uu ) and Mycoplasma hominis ( Mh) lipiodol angiography and fallopian tube , and two groups of patients with mycoplasma infection and tubal cir-cumstance tube obstruction were compared .Results The two groups infertile patients with Cervical Mycoplasma ex-amination with Uu positive were 41 cases, accounting for 68.3%of the patients in the control group .Uu was positive in 16 cases, accounting for 26.6%; Infertile patients with Uu +Mh positive were 15 cases, accounting for 25%, while the control group were no cases were positive for Uu +Mh;Infertility group patients detected negative for Myco-plasma in 4 cases, accounting for 6.7%, while the control group were negative for the detection of Mycoplasma in 44 cases, accounting for 73.3%.The results showed that infertility group and Uu in patients with Mh infection was high-er than that of the control group, and the difference is statistically significant (p<0.05).The infertile patients with fallopian tube patency were 12 cases, accounting for 20%; The control group of tubal patency were 33 cases, ac-counting for 55%, and it was accounted for 19.3% of 21 cases of tubal infertility group , obstruction.The control group of 13 cases of tubal obstruction patients , accounting for 21.7%;27 cases had complete obstruction of fallopian tube infertility patients group , accounting for 45.0%, and the control group had complete tubal obstruction in 14 ca-ses, accounting for 23.3%.The results showed that infertile patients with fallopian tube obstruction was significantly higher than that in the control group , and the difference was statistically significant (p<0.05).Conclusion Infer-tility women with genital tract mycoplasma infection occurrence are closely related .We should make a comprehensive inspection to discover occult infection of Mycoplasma timely and treat actively in women with infertility treatment .
2.Comparison analysis of cellular morphology and immune phenotypic in Uygur and Han patients with myelodysplastic syndrome

Objective To investigate the cell morphology and immune phenotypic characteristics in Uygur and Han patients with myelodysplastic syndrome (MDS) in Xinjiang Uygur Autonomous Region.Methods Bone marrow smears of 67 cases diagnosed as MDS were systematicly observed and recorded,flow cytometry (FCM) was used to test the immunophenotyping.Results There were abnormal hematopoiesis in different extent in granulocyte series,erythron series and megakaryocytic series of all patients.Occurence of pathological hematopoietic performance in Uygur patients with MDS was similar to that in Han,granulocyte series [52 cases (77.6 %)] >megakaryocytic series [44 cases (65.7 %)] > erythrocyte series [36 cases (53.7 %)],the differences in two groups were not significant (x2 values 1.02,0.30,0.02,respectively,all P > 0.05).The changes type of pathological hematopoietic performance in two groups were similar,single garden nuclear megakaryocyte [36 cases (53.7 %)],false Pelger nuclear anomaly granulocyte [36 cases (53.7 %)],erythroid gigantic young cell change [33 cases (49.3 %)],granulocyte cell particles reduced or absent [27 cases (40.3 %)] etc.The result of FCM showed that along with the progressive change of RA/RAS to RAEB/RAEB-t,the expression ratios of mature CD15 were decreased,while the expression ratios of early stage CD117 and CD34 were raised gradually (x2 values 6.23,12.06,8.95,7.37,8.95,8.08,respectively,all P < 0.05),the differences in two groups were not significant (x2 values 0.715,0.024,0.146,respectively,all P > 0.05).Meanwhile,Uygur patients with MDS has great expression of CD56 and Han had HLA-DR,there were significantly differences in two groups (x2 values 3.91 and 3.90,respectively,both P < 0.05).Conclusion Occurence of pathological hematopoietic performance in Uygur patients with MDS are similar to that in Han.Most MDSs has two or more levels of pathological hematopoietic performance.The differences of immunophenotype characteristics in two groups are partially significant and test of immunophenotype are helpful in the diagnosis,classification,prognosis of MDS.
3.The prognostic value of dysplasia features in myelodysplastic syndromes

Objective To analyze the characteristics of dysplasia in patients with myelodysplastic syndromes (MDS),and identify the risk factors for evolution of MDS to acute leukemia.Methods According to the WHO criterion 2008 of MDS,98 cases were included.As the characteristics of dysplasia,cell morphology,cytogenetics and flow cytometry were analyzed in these patients.Results To the last follow-up,27 of 98 patients transformed to acute myeloid leukemia,including M2 (15 cases),M4 (10 cases),M6 (2 cases).Patients with abnormal Pelger-Hu(e)t had significantly higher risk for evolution of MDS to leukemia,when compared to those without Pelger-Hu(e)t (23/67 vs 4/36,x2 =4.87,P =0.03).Auer corpuscle and small nuclear pathological were also the risk factors of developing to leukemia (19/37 vs 8/61,x2 =16.87,P =0.000; 21/57vs 6/46,x2 =9.14,P =0.003,respectively).37 patients died,the median suvival time was 26 months (95 % CI 13-38) for patients with abnorrnal Pelger-Hu(e)t,19 months (95 % CI 11-26) for patients with Auer corpuscle,13 months (95 % CI 6-19) for patients with small nuclear pathology,respectively (x2 =11.05,13.04,21.05,P =0.001,0.000,0.000).Conclusion Abnormal Pelger-Hu(e)t,Auer corpuscle and small nuclear pathology are indentified as the risk factors for evolution of MDS to leukemia,and they are the infavorable predictors of survival.
4.Clinical study of 36 cases with severe pre-eclampsia complicated with ascites

Objective To analyse clinical characteristics and preventive methods of pregnancy-induced hypertensive severe pre-eclampsia with ascites.Methods 184 cases of patients with severe pre-eclampsia complicated with ascite were divided into two groups and the clinical characteristics were compared(severe pre-eclampsia complicated with ascite group,n=36;Severe pre-eclampsia group,n=148 cases).Results The average time of the termination of pregnancy was (34±4) weeks in severe pre-eclampsia complicated with ascites group,and (36±3) weeks in severe pre-eclampsia group (t=2.71,P<0.01).There were 6 cases of intrauterine stillborn (16.67%),14 cases of neonatal mortality (38.89%);22 cases of premature labor (61.11%) in severe pre-eclampsia complicated with aacites group;There were 4 cases of intrauterine stillborn (2.7%);16 cases of neonatal mortality (10.81%);and 35 cases of premature labor (23.65%) in severe pre-eclampsia group (X~2=10.99,16.73,19.01,P<0.001).There were differences in the total protein,albumin,white/ball ratio,lactic acid dehydregenase,serum creatinine,24-hour urinary protein between the two groups [total protein:(51.68±6.08) g/L vs (59.34±8.28)g/L,albumin:(24.32±4.06) g/L vs (32.16±5.63) g/L,white/ball ratio:(0.92±0.26) vs (1.16±0.26),lactic acid dehydrogenase:(495.87±312.56) U/L vs (323.81±185.00) U /L,serum creatinine:(131.62±95.34) μmol/L vs (91.52±86.83) μmol/L,24 h urinary protein(4.21±3.51)g/24 h vs (2.38±2.57)g/24h,t=6.17,8.77,5.50,4.79,2.72,3.98,P<0.05 or <0.01].Conclusion Tbe condition of severe pre-eclampsia with ascites is extremely serious,so pregnancy should be timely terminated.The aacites can be dissipated after delivery.Early,systematic and regular prenatal care can prevent severe pre-eclampsia and complications in time.
5.Application of Ureteroscopy in the Diagnosis and Treatment of Ureteral Injury Caused by Laparoscopic Hysterectomy

Objective To explore the value of ureteroscopy in the diagnosis and treatment of ureteral injury caused by laparoscopic hysterectomy . Methods Suspect ureteral injury occurred in 23 cases out of 578 laparoscopic hysterectomy from January 2010 to December 2015 in our hospital.Among them, 12 cases were testified by ureteroscopy .A double-J catheter was placed in for 3 months. Results All the 12 cases were followed up for 3-26 months (average, 19 months).The double-J catheter was removed at 3 months after surgery .The continuity of the affected side urinary tract was restored in 10 cases and hydronephrosis was not found by B ultrasound .Two cases were found ureteral obstruction and were given ureterovesical reimplantation at 3 months after surgery . Conclusions The ureteroscopy should be carried out as early as possible for patients with suspect ureteral injury after laparoscopic hysterectomy .Double-J catheter placement can avoid parts of patients converting to open surgery .
6.Effect of Penehyclidine Hydrochloride on Prevention of Catheter Related Bladder Discomfort after General Anesthesia

Objective:To compare the efficacy of penehyclidine hydrochloride and atropine in the prevention of catheter related bladder discomfort(CRBD) in patients undergoing gynecology laparoscopic surgery.Method;Eighty ASA classⅠorⅡpatients undergoing elective gynecology laparoscopic surgery were randomly assigned into 2 groups of P and A with 40 cases each.Fifteen minutes before the anesthesia induction,penehyclidine hydrochloride 0.5 mg and atropine 0.25 mg were administered in Group P and Group A,respectively.After the induction of anesthesia,patients were catherized with a 14 Fr Foley's catheter.HR,bladder discomfort,Ramsay Sedative Score(RSS) and side reactions were recorded at 0 h,0.5 h,1 h and 6 h after the end of surgery in the post-anaesthesia care unit(PACU).The severity of bladder discomfort was graded as mild,moderate and severe.Result:The total incidence rate of CRBD in Group P was apparently lower than that in Group A(P0.05).The incidence rate of CRBD of Group P was apparently lower than that in Group A at 6 h after the end of surgery(P0.05).Conclusion:The application of enehyclidine hydrochloride before anesthesia induction provides a better prophylactic effect of CRBD than that of atropine but with a higher incidence rate of dry mouth.
7.The efficacy of injecting insulin in family diabetes mellitus in Xiaofeng town community in Huzhou area and the analysis of the problems of injection technique

Objective To investigate the efficacy of injecting insulin in family diabetes mellitus in Xiaofeng town community in Huzhou area and the problems of injection technique.Methods From February 2015 to December 2016, 94 patients with diabetes were injected with insulin in Xiaofeng town community in Huzhou area.From February 2015 to 3016, the fasting blood glucose(FPG), 2 h postprandial blood glucose 2hPG) and glycosylated hemoglobin(HbA1c), and to investigate the problems in the process of injecting insulin by questionnaire.Results After 3 months of injection, FPG, 2hPG and HbA1c were decreased in all patients(P<0.05).71 cases(75.53) were regularly rotated in the injection of insulin and 82 cases were alcoholic(81.9%), the use of 5 mm needle 86 cases(91.49%)[our hospital only 5 mm and 8 mm Needle, jurisdiction of the surrounding hospital needle is 6 mm.(76.20%), 12 cases(12.77%) of insulin needle, and 2 to 3 days of insulin needle(72.60%) were injected in the subcutaneous 10s(71.28%) after injection, 16 cases(17.2%) were replaced, 10 cases(48.94%) were replaced on the 10th day, and 11 cases(11.70%) were replaced when the insulin was replaced.9 cases(9.57%) were replaced after the pain.Conclusion Diabetic patients have definite therapeutic effect on insulin injection in family, but there are some defects in injection technique and injection hygiene.Therefore, health care workers need to strengthen their knowledge of injection of insulin injections in families.
8.Analyses for the ?/? T Cell Receptor Gene Rearrangement and CDR3 Repertoire in Active Pulmonary Tuberculosis Patients

Objective:To establish a method of multi-PCR to amplify the complete DNA sequence (CDS) of TCR ? and ? chain of the antigen-specific T lymphocytes in local pathologic specimen of active pulmonary tuberculosis patients, and to analyze ?/? T cell receptor gene rearrangement and CDR3 repertoire.Methods:The lymphocytes in bronchoalveolar lavage (BAL) of active pulmonary tuberculosis patients were separated. Following total RNA extraction, cDNA synthesis, Multi-PCR, recombinant clones construction, and sequencing, the CDS of TCR ? and ? chains from these lymphocytes were analyzed by using software of DNAstar and internet TCR resources.Results:24 of ? chain CDS and 13 of ? chain CDS from 3 samples of BAL were obtained. As for TCR ? chain, AV1S2 (54%), AV12S3 (41%), and AV12S2(5%) appeared frequently. BV2(38%), BV29S1(46%), BV14(3%), and BV4S2(3%) in TCR ? chain appeared more often. There were CDR3 diversities between samples and even in the same sample by amino acid sequence analysis, but there were a few identical or similar amino acid sequences. There was the same amino acid sequence of SVGTGTLHQETQY in CDR3 region of ? chain of BAL sample No.1 and No.2; The sequence of AVRDWAGNMLT appeared in two ? chains of BAL sample No.2 and No.3; Moreover, the sequence of AV…DNN…RLM appeared in ? chains of BAL sample No.2 and No.3.Conclusion:A method of Multi-PCR is used to amplify TCR ? and ? chain CDS of tuberculosis patients. There are characteristic T cell clones to proliferate,with TCR ? and ? chain repertiore skewing in local infective focus. The sequences of CDR3 in different TCR clones are mostly different but there are a few identical or similar sequences in the same patient or even between different patients. The identical amino acid sequences of CDR3 are possibly specific for recognizing MTB polypeptide.

Objective To investigate the effect of dexamethasone on proliferation of human periodontal ligament fibroblasts(hPDLF) cultivated in vitro,and search for optimal culture condition for hPDLF in vitro,and provide basis for further study on regeneration of periodontinum.Methods The hPDLF cultivated in vitro were divided into 5 groups and cultivated in 5 different culture media separately which all included 10% fetal bovine serium: ①control group(no DEX);②5 mg?L-1DEX;③10 mg?L-1 DEX;④20 mg?L-1 DEX;⑤50 mg?L-1 DEX.The proliferation of hPDLF was assyed by MTT method.The phase contrast microscope was used to observe the morphological changes of the cells.Results The PDLF appeared aggregation and cells on bottom of culture bottle showed squamae-shape after inoculated on plate with 24 hole for 3 d.There were more layers of smaller hPDLF with blunt prominency came forth after cultivated in DEX culture media.MTT method showed that DEX promoted the proliferation of hPDLF obviously compared with control group(P

Objective To investigate the effect of insulin-like factor-I(IGF-Ⅰ)on the proliferation activity of human periodontal ligamental fibroblasts(PDLF).Methods The PDLF transfected with IGF-Ⅰ and PDLF untransfected with IGF-Ⅰ prepared in our lab were digested by 0.25% trypsin and prepared into cell suspension.The two sorts of cells were counted after being vaccinated,then the growth curves of the two cells were mapped and the growth speeds were compared.The PDLF transfected with IGF-Ⅰ and PDLF untransfected with IGF-Ⅰ were digested by 0.25% trypsin and then cultivated.The proliferation activities of the PDLF transfected with IGF-Ⅰ and PDLF untransfected with IGF-Ⅰ were observed using MTT methods.The activity of alkaline phosphatase(ALP) was determined by using ALP kit.Results The growth curves of the two cells showed that the growth speed of the PDLF transfected with IGF-Ⅰ was significantly faster than that of the PDLF untransfected with IGF-Ⅰ(P
