1.Effects of pramipexole on parkinson's disease and serum CRP, Cystatin C, BDNF and oxidative stress

Objective To study curative efficacy of Pramipexole in treatment of Parkinson's Disease and its effects on Serum C-reactionprotein (CRP), cystatin C, brain derived neurotrophic factor(BDNF) and oxidative stress.Methods 100 patients of Parkinson's Disease who received therapy from February 2014 to April 2016 in our hospital were selected as research objects were randomLy divided into control group and observation group, 50 patients in each group.The control group was treated with Dobasidate tablets;The observation group was given pramipexole tablets on the basis of the control group.Then the clinical effects and adverse reactions were compared.CRP, Cystatin C, BDNF and oxidative stress were measured. Results After treatment, the total effective rate of the observation group [(90.00% (45/50)] was significantly higher than the control group[74.00%(37/50)] (P<0.05);The symptoms of palpitation, depression, nausea, anorexia, diarrhea and vomiting were lower in observation group than in observation group, and the differences of depression, anorexia and vomiting were statistically significant(P<0.05);The BDNF index of the observation group (13.38 ±1.93)ng/L was higher than that of the control group (12.03 ±1.83) ng/L(P<0.05);CRP (3.53 ±0.97) mg/L and Cystatin C index (0.98 ±0.12) mg/L were lower than the control group(4.62 ±1.15)、(1.06 ±0.14) mg/L (P<0.05).After treatment, the glutathione peroxidate (GPX),superroxide dismutase(SOD),glutathione(GSH)and catalase(CAT) of oxidative stress in observation group were higher than those in control group,the difference was statistically significant (P<0.05).Conclusion Pramipexole in the treatment of Parkinson's disease is obvious, can effectively improve the serum CRP, Cystatin C and BDNF index, high security, it is worth promoting the application.
2.Effects of sodium salicylate on the expression of HSP27 protein during oxidative stress in tissue-cultured human lens epithelial cells.

The effects of sodium salicylate on the expression of heat shock protein 27 (HSP27) during oxidative stress in tissue-cultured human lens epithelial cells were investigated. Cultured human lens epithelial cells (HLB-3) were divided into 3 groups: control group (group A), oxidation injury group (group B) and sodium salicylate group (group C). Apoptosis of human lens epithelial cells cultured in vitro was induced in the presence of 150 micromol/L H2O2. Cells viability and the expression of HSP27 were analyzed. Viability of the cells was measured by methyl thiazole tetrazolium (MTF) chromatometry. The expression of HSP27 in HLB-3 cells was detected by using immunohistochemistry and image analysis system. Sodium salicylate could induce the expression of HSP27, and the cells viability in group C was significantly higher than in group B (0.2667+/-0.01414 vs 0.2150+/-0.01080, P=0.012<0.05). The average gray value of HSP27 in group B was less than that in group C (P=0.000<0.05). The increased expression of HSP27 by sodium salicylate might play an important role in the protection of hydrogen peroxide-induced injury of human lens epithelial cells, suggesting that sodium salicylate could suppress, at least in part, the apoptosis of human lens epithelial cells.
3.On clinical effects of uterosacral ligament amputation in combination with lesion electrofulguration under laparoscope for endometriosis-related dysmenorrhea

Objective To evaluate the efficacy of uterosacral ligament amputation in combination with lesion electrofulguration under laparoscope in the treatment of endometriosis-related dysmenorrhea. Methods A total of 86 patients with endometriosis-related dysmenorrhea were treated by uterosacral ligament amputation combined with electrofulguration of lesions under laparoscope from March 2000 to August 2004 in this hospital. Among the 86 patients, enucleation of ovarian endometriotic cysts and electrofulguration of pelvic endometriotic lesions were also simultaneously conducted in 37 patients, and electrofulguration of all visible pelvic endometriotic lesions was also performed in the rest of 49 patients. The operation was accomplished with the aid of the placement of an infrared ureteral detector in 3 patients because of the thickening of the uterosacral ligament and local adhesion. Results The 86 patients were followed for 6~48 months (median, 23 months). Dysmenorrheal completely disappeared in 36 patients, partially subsided in 29, and remained unchanged in 21, the effective rate being 75.6% (65/86). Conclusions Uterosacral ligament amputation combined with electrofulguration of lesions under laparoscope is effective for the treatment of endometriosis-related dysmenorrhea.
4.ESI fragmentation studies of six unusual nucleotide sugars.

Unusual dTDP-sugars are key intermediate in many pathogenic bacteria. In this study, negative-ion electrospray tandem mass spectrometry (ESI-MS-MS) with collision-induced dissociation (CID) was used to study the fragmentation characteristics of six unusual nucleotide diphosphate sugars. The results indicated the major fragment of the six unusual nucleoside sugars observed in the ESI-MS-MS spectra resulted from cleavage of diphosphate moiety and their characteristic fragment ions at m/z 401, 383, and 321, correspond to [TDP-H] together with fragment ions resulting from the loss of water and phosphate moiety, respectively. Furthermore, 4-position substituted change of unusual sugar rings affected the stability of two important characteristic fragment ions of [glycosyl-1"-PO3](-) and [glycosyl-1"-P2O6](-).

6.Clinical observation of integrated traditional Chinese and western medicine in the treatment of intractable hiccup caused by cerebrovascular disease Medicine

Objective To explore the clinical effect of integrated traditional Chinese and western medicine in the treatment of intractable hiccup caused by cerebrovascular disease.Methods A total of 74 patients with intractable hiccup caused by cerebrovascular disease were selected.The patients were randomly divided into observation group and control group according to random number table method.28 patients in the control group were given simple western medicine treatment.46 patients in the observation group were treated with integrated traditional Chinese and western medicine.The total effective rate and symptom improvement were observed after treatment.Results The total effective rate of observation group was 94.48%,that was 78.57% in the control group,the difference was statistically significant(x2=2.0,P<0.05).Conclusion Combined traditional Chinese and western medicine can significantly reduce intractable hiccup caused by cerebrovascular disease,improve total efficacy,improve the quality of life.
7.Analysis of chromosome karyotype characteristics in patients with chronic myeloid leukemia

Objective To explore the characteristics of chromosome karyotypes in patients with chronic myeloid leukemia (CML),and to provide help to individualized treatment.Methods The date of chromosome karyotypes of 313 patients and FISH of 45 of these patients with CML excluding Ph chromosome negative (Ph-) after treatment were collected from January 2014 to June 2015.Karyotypes were detected by R-banding.Results In the 313 cases,307 cases (98.08 %) were Ph chromosome positive (Ph+) and 6 cases (1.92 %) were Ph-.In the Ph+ patients,288 cases (93.81%) were classical Ph+,and 19 cases (6.19 %) were variant rearrangements.There were 48 cases (15.34 %) with additional chromosome changes in all patients,including 41 cases (13.10 %) with classical Ph+ and 7 cases (2.24 %) with variant rearrangements.The most common additional chromosome changes were in the following order:+der(22) Ph (35.42 %),+8 (33.33 %) and +21 (12.50 %).The most frequent pattern of combination was +der(22) combined with +8 (16.67 %),followed by +8 combined with +21 (10.42 %).The proportion of pure Ph+ patients in chronic phase was higher than that of advanced phase,but proportion of classical Ph+ patients with additional chromosome changes in chronic phase was lower than that in advanced phase (x2 =1 11.55,P < 0.01).The proportions of chronic phase and advanced phase patients with simple variant rearrangements were not different from those with complex variant rearrangements (P =0.582).The results of FISH in 45 cases were all positive,including 5 cases with 2 GIR1Y.Conclusion Karyotype analysis can reveal the instability of genetic and the characteristics of disease progression by identifying the evolution of Ph,which provides the basis for clinical doctors to choose suitable treatment.
8.Effects of dexmedetomidine on plasma IL-8,IL-10 levels and lung AQP1 expression in patients undergoing one lung ventilation

Objective To investigate the effects of dexmedetomidine on plasma IL-8,IL-10 level and lung AQP1 expression in patients undergoing one lung ventilation(OLV).Methods Forty patients,ASA Ⅰ or Ⅱ,scheduled for lung cancer radical surgery were randomly divided into two groups:control group (group C)and dexmedetomidine group (group D).In group D,dexmedetomi-dine 1 μg/kg (initial dose)was given over more than 10 min before anaesthesia induction,followed by continuous infusion at 0.5 μg·kg-1 ·h-1 until 30 min before the end of operation.In group C,the e-qual volume of normal saline was infused.Blood samples were collected at immediately before OLV (T1 ),30 (T2 ),60 (T3 ),120 (T4 )min of OLV,30 min after lung inflate (T5 )and 2 h after opera-tion (T6 )for determination of plasma IL-8 and IL-10 levels.Lung tissues were obtained at T1 for de-termination of AQP1 expression.Results Compared with the value at T1 ,plasma IL-8 levels in group C at T3-T6 and in group D at T3-T5 were increased,plasma IL-10 levels were increased in both groups at T2-T5 (P <0.05).Compared with group C,plasma IL-8 levels at T3-T6 were decreased and plasma IL-10 levels at T2-T5 were increased in group D (P <0.05).Compared with the value at immediately before OLV,the expression of AQP1 was decreased at the time of lobectomy of lungs in group C (P<0.05).Compared with group C,the expression of AQP1 at the time of lobectomy of lungs in group D was increased (P <0.05).Conclusion Dexmedetomidine 1 μg/kg followed by continuous infusion at 0.5 μg·kg-1 ·h-1 can reduce inflammatory response,up-regulate the expression of AQP1 in pa-tients undergoing OLV.
9.Relationship of the expression level of monocyte chemotactic protein-1 and nuclear factor-κB with atrial fibrosis of atrial fibrillation patients with rheumatic heart disease

Objective To explore the relationship of the expression level of monocyte Chemotactic protein-1 and nuclear factor κB(NF κB)with atrial fibrosis of atrial fibrillation patients with rheumatic heart disease.Methods About 200 mg right atrial tissue were taken from 26 patients with rheumatic heart disease undergoing valve replacement surgery and divided into sinus rhythm (SR) group (n=12) and atrial fibrillation (AF) group (n=14).Masson staining and atrial myocardial fibrosis markers were used to determine the level of fibrosis.The mRNA levels of cytokines in atrial tissue were measured by reverse transcription polymerase chain reaction (RT-PCR)The protein level of MCP-1 and phosphorylation of NFκB in atrial tissue were detected by Western blotting.Results Compared with SR group,the AF group showed that collagen volume fraction (AF:0.42 ± 0.03;SR:0.13 ± 0.02),the mRNA levels of myocardial fibrosis markers such as transforming growth factor (TGF)-β1,type Ⅰ collagen and type Ⅲ collagen,and cytokines such as IL-1β and TNF-α,and the protein level of MCP-1 (AF:0.170±0.003;SR:0.040±0.005) and level of phosphorylation of NF-κB (AF:0.35 ± 0.02;SR:0.12 ± 0.03) were significantly increased (all P<0.05).In addition,the expression levels of cytokines,the protein expression level of MCP1 and the phosphorylation level of NF-κB were positively correlated with collagen volume fraction of the right atrial myocardial tissue (all P<0.05).Conclusions Activation of NF-κB may induce the expression of MCP-1 in the myocardial tissue of patients with rheumatic heart disease,and sequentially stimulate the secretion of cytokines and extracellular matrix deposition,and finally participate in the occurrence and persistence of atrial fibrillation.
10.Biological characteristics of mouse erythroleukemia cell FBL-3

Objective To investigate the biological characteristics of mouse erythrolenkemia cell FBL-3.Methods The morphological feature, growth characters, clone formation and immunochemistry of mouse erythroleukemia cell FBL-3 were examined by light microscope. The cell cycle distribution and expression of MHC were detected by flow cytometry. Drug sensitivity was measured by MTT assay. Tumorigenicity was evaluated after intravenous injection FBL-3 cells into C57BL/6 mice. Results FBL-3 cells were smaller,fusiform or polygon, adherence. Doubling time was 24.12 h. The clone formation rates were (35.23±1.44) %and (60.27±5.56) % at 14 th and 21 th day, respectively. The reactions for PAS and chloracetic acid were positive, while the POX, NAP and butanoic acid reactions were negative. The cell cycle distribution was as follow: G0/G1 phase (50.9±2.5) %, S phase (36.3±1.4) %, G2/M phase (13.8±0.8) %. The IC50 of FBL-3 cells to Ara-C, VDS, DDP, MMC and MTX were (0.49±0.04), (0.87±0. 09), (3.77±0.32), (1.66±0.16) μmol/L and (2.77±0.24) nmol/L, respectively. Chromosome number was at 34 to 41. MHC of FBL-3 cell was H-2b. Sexual gene Sry was positive. All C57BL/6 mice were morbidity with erythroblastic leukemia when FBL-3 cells had been intravenously inoculated. There was a linear relationship between the survival time and the number cell injected. The main targets for the leukemic FBL-3 cells were liver, spleen, kindey and lung. Conclusion FBL-3 cell has typical features of mouse leukemia cell, was easily cultured in vitro and tumorigenesised in C57BL/6 mice. FBL-3 cell could be as a satisfactory tool for the research of leukemia.
