1.Evaluation of application of enzyme immunoassay workstation in detection of antibodies against Streptococcus pneumoniae

Objective To apply automated enzyme immunoassay workstation instead of manual operation to facilitate detection of Streptococcus pneumoniae(S.pneumoniae) IgG antibodies, so as to improve the detection efficiency.Methods Procedures of the assay were set up on the automated workstation according to the World Health Organization(WHO) Pneumococcal Capsular Polysaccharide Types-pecific IgG Antibody Detection Manual Pn PS ELISA. The number of working station panels was maximized to preset all reagent consumables and more samples to be tested, thereby improving the detection flux and reducing the number of manual interventions. The serum PnG was repeatedly detected to verify the test precision. The WHO S.pneumoniae international calibration serum panel 12/278 was detected to verify the accuracy of the test. To compare the consistency of workstations and manual operation, 16 same serum samples were tested by manual operation and on the workstation meanwhile. Detections of four calibration serum samples were performed on the workstation by double-(WHO standard method) and single-column methods, and the consistency of the results was compared.Results With the layout of 21 plate positions, 24 serum samples could be examined in one program run, requiring only 2 manual interventions. The CV of 32 detection results of serum PnG was lower than 15%. The assay results of 75% of the calibration serum panel in 14 out of 23 serotypes exhibited a percent error of 40% or less compared to the assigned values, and the total accuracy was 80%.The correlation coefficient(Pc) of agreement between workstation and manual test results was 0. 996 7, and the correlation coefficient of agreement between workstation single-and double-column test results was 0. 999 8, both of which showed high agreement.Conclusion The automated enzyme immunoassay workstation has shown high precision, accuracy and large detection throughput, which is labor-saving, and can be applied to the detection of S.pneumoniae antibodies. The results of single-and double-column assay provide experimental basis for single-column detection in place of double-column detection.
2.Study on influencing factors and clinical characteristic of High Intensity Focused Ultrasound ablation analgesic in middle-late stage pancreatic carcinoma

Objective To study the influencing factors and clinical characteristic of High Intensity Focused Ultrasound(HIFU) abhtion analgesic in middle-late stage pancreatic carcinoma.Methods Seventyone cases pancreatic cancer patients underwent HIFU treatment were grouped according to the pain efficacy of related influencing factors and the data was analyzed by single factor correlation analysis.The relationship between influencing factors and analgesic effect were analyzed through the pain outcome score.Results Pain score decreased ≥50% in advanced pancreatic cancer patients occurred in <60 years old,Body mass index (BMI) < 22 kg/m2,tumor location in the pancreatic head and neck,tumor size of 2-5 cm,and the average power of HIFU treatment > 400 W,Ductal adenocarcinoma of the tumor histological type were related to the degree of pain rehef,and the difference was statistically significant (P < 0.05).And fewer complications occurred after HIFU ablation.Conclusion Analysis of the relationship between the different factors and the analgesic effect in advanced pancreatic carcinoma can be used to predict the efficacy of HIFU treatment.

Transcatheter arterial chemoembolization (TACE) is the first choice of patients with hepatocellular carcinoma (HCC) missed opportunity of exairesis.Angiogenesis is one of the key factors that affect prospective efficacy.Counter-measures against angiogenesis mainly contain complete embolization of tumor blood supply and integration with angiogenesis inhibitors.The angiogenesis after TACE and the corresponding countermeasures were reviewed in this article.
4.Dynamic changes in ATP-binding cassette transporter A1 in peripheral blood monocytes of patients with type 2 diabetes mellitus

Objective To investigate the expression level of monocytes ATP-binding cassette transporter A1(ABCA1) stimulated by oxidized low-density lipoprotein (ox-LDL) in patients with coronary heart disease (CHD) for exploring the mechanism of ABCA1 in the pathogenesis of atherosclerosis (AS). Methods Human monocytes of patients in CHD group (n=42) and normal control group (n=40) were isolated by density gradient centrifugation with separating medium (1.077). ABCA1 was labeled by immune reaction PE fluorescence, then the expression of ABCA1 before and after being incubated with the ox-LDL (30mg/L) for 24 hours was investigated by flow cytometer, and the changes in ABCA1 expression were observed. The differences between the two groups and the correlation between ABCA1 expression and blood lipids were analyzed. Results The high-density lipoprotein cholesterol (HDL-C) and apolipoprotein A1 (apoA1) levels in CHD group (1.15?1.40 and 1.19?0.17, respectively) were significantly lower than that in control group (1.36?0.40 and 1.34?0.22, respectively, P0.05). Conclusions ABCA1 expression in the patients with CHD may be inhibited by high concentration of ox-LDL. The lowered expression of ABCA1 and the abnormal lipids spectrum may decrease the reverse cholesterol transport (RCT), and contribute to the development of AS and CHD.
6.Analysis of somatic mutations in membrane proteins of tumor cells

Objective To screen specific mutations on extracellular regions of membrane proteins ( extracellular membrane protein mutations ) in tumor cells and provide the reference information for target searching in cancer precision medicine .Methods Somatic mutations on extracellular regions of membrane proteins of 7042 tumor samples were collected to screen all specific extracellular membrane protein mutations, and the overall distribution of these mutations were obtained by statistical analysis.Genes, gene site and cancer types occured high frequency of extracellular membrane protein mutations were identified.Results 97193 specific extracellular membrane protein mutations were obtained from 4938362 somatic mutations in 7042 tumor samples (30 cancer types), the statistical analysis showed that 4347 genes and 65532 sites were involved in these specific mutations.The study further analyzed five genes (MUC16、LRP1B、CSMD3、RYR2、USH2A), one site (17:37868208) and six cancer types (including colorectal cancer, melanoma, uterine cancer, brain lower grade glioma, lung adenocarcinoma and stomach adenocarcinoma) which occured high frequency of extracellular membrane protein mutations.Conclusion An information library of specific mutations on extracellular regions of membrane proteins was established and the distribution of these specific mutations was obtained which can provide reference information for target detection in targeted cancer therapy and immunological therapy.
7.Advances in the Silent Stroke

Wtth the development of imaging technique,particularly the use of magnetic resonance diffusion weighted imaging,it can easily detect silent stroke lesiom.Both the prevalence and incidence of asyrrotomatic stroke were higher than those of symptomatic stroke.Age,hypertemion and atrial fibrillation are its accepted risk factors.Although silent stroke lacks clinically overt stroke-like symptoms by definition,it often has mild physical and cognitive impairment with careful examination.Furthermore,asymptomatic stroke is also correlated with the occurrence of subsequent symptomatic stroke.decreased cognitive function and dementia.Therefore,clinicians should pay more attention to it,positively control its risk factors and improve its prognosis.
8.Influence of baicalin combined with NB-UVB on melanin synthesis and tyrosinase activity in melanocytes

Objective To observe the synthesis of melanin and tyrosinase activity of the cultured normal human melanocytes after treated with baicalin and NB-UVB, and to provide theoretical evidence for study on the new therapy method of vitiligo.Methods The normal human menlanocytes cultured in vitro were divided into control group,different concentrations of baicalin groups,different doses of NB-UVB group and baicalin combined NB-UVB group.The proliferation rate of melanocytes was measured by MTT assay,the level of melanin was tested by NaOH pyrolysis,and the tyrosinase activity was measured by dopa oxidization.Results Compared with control group,the proliferation rates in 10-4 mol·L-1 baicalin group and 30 mJ·cm-2 NB-UVB group had no significant difference(P>0.05 ). Compared with control group, the levels of melanin and tyrosinase activities in baicalin group,NB-UVB group and combined group were increased (P<0.05 or P<0.01);compared with baicalin group and NB-UVB group,the level of melanin and tyrosinase activity in combined group were increased(P<0.01). Conclusion Baicalin combined with NB-UVB has better promotion effect on melanin synthesis than used alone.

Objective To study the anti,radiation effect of beta,carotene(?,C). Methods The different doses of ?,C (30,60 and 90 mg/kg bw) were orally administered separately to three groups of mice, and a control group was also arranged. The four groups of mice were exposed to 60Co,? rays after 30 days, then all groups were continuously orally administered, the survival rates, survival time, number of white blood cells(WBC), micronucleus frequency(MF), spermatocyte chromosome abberation, immune function, superoxide dismutase(SOD) activity were measured. Results The results showed that compared with control group, groups treated with?,C expressed lower range WBC decrease, stronger immune function, higher SOD activity, less MF, longer survival time and improved survival rates. Conclusion ?,C had radioprotective effect.

Salmonella is an important zoonosis.China is the largest pork consumer.Contaminated pork is the main source of Salmonella disease at home and abroad.There are many swine Salmonella vaccine in use in Europe.Now the current swine Salmonella vaccine is a live attenuated vaccine,Salmonella can reduce colonization in pigs,and can induce immune response efficiently after the two immunizations.The control effect is poor,at this stage of swine vaccine cross protection research,type of Salmonella and vaccine antigen are not the same.This paper reviews the research progress of swine Salmonella vaccine.
