3.Study on preventive and therapeutic effects of astragali radix on denervated tibial muscle atrophy in rats.
China Journal of Chinese Materia Medica 2014;39(6):1083-1087
OBJECTIVETo study the effect of Astragali Radix on the denervated tibial muscle atrophy in rats, and discuss its mechanism.
METHODTotally 60 SPF-grade Sprague-Dawley rats were selected in the common peroneal nerve crush model, and then randomly divided into 6 groups: Astragali Radix high-dose, medium-dose, low-dose groups, the Mecobalamin group, the model group, and the sham operation group. They were administered with drugs after the operation. At 18 d, the pathological section staining and morphological analysis were performed. The wet-weight ratio and section area of tibial muscles were also measured. The real-time fluorescence quantification was adopted to detect the differential expression between Angptl4 and PI3K genes.
RESULT(1) Wet-weight ratio: The wet-weight ratio in Astragali Radix high-dose, medium-dose groups was much higher than that in the model group (P < 0.05 or P < 0.01). (2) Section area: The sham operation group was higher, with regular morphology; Whereas the model group showed significant decrease, with chaotic structure and obvious connective tissue proliferation; Astragali Radix groups and the mecobalamin group showed relatively small section areas, with chaotic structure and unobvious connective tissue proliferation. Compared with the model group, Astragali Radix groups showed significant increase (P < 0.01). (3) Motor end plate: The sham operation group was in uniform brownish black color and oval or round shape; Astragali Radix medium-dose and high-dose group and the mecobalamin group showed rough line edges; Astragali Radix medium-dose and low-dose groups and the model group showed decline in the number, with irregular morphology, rough line edges and a light color. (4) Angptl4 and PI3K: Compared with the model group, the Astragali Radix high-dose group showed significant increase (P < 0.05).
CONCLUSIONAstragali Radix has a significant effect in preventing and treating denervated tibial muscle atrophy. It may delay the muscle atrophy by increasing Angptl4 and PI3K gene expressions.

Chinese Journal of Obstetrics and Gynecology 2009;44(11):846-850
Objective To investigate the effect of vagina reconstruction using tissue-engineering biological material (acellular dermal matrix) in an animal model. Methods Vagina excision and vagina reconstruction with tissue-engineering biological material were performed in 12 Chinese experimental miniature pigs. The control group was matched with two of normal vagina specimens resected. At week 1,2,4,6,8,12 after surgery, the animals were sacrificed, respectively, and the neovaginas were prepared for immunohistochemical and Van Gieson (VG) staining to evaluate the status of various layer growth of vagina.Epithelial broad spectrum of monoclonal antibodies of AE1/AE3 and α-actin were used to test the existence of epithelial and smooth muscle tissue by immunohistochemical staining. The ultrastructure of neovagina was studied by transmission electron microscope at week 1 and 12 after surgery.Contractile function of isolated smooth muscle of neovagina was evaluated by chemical and electronic stimulation after 12 weeks' reconstruction. Results (1) Epithelization of 2/3 neovaginal mucosa was observed within 1 week.Only 1-2 layer epitheliums were observed under the light microscopy and epithelial cells with characteristics of loose and disarrangement were shown with the electron microscopy.Within 4-6 weeks, epithelization in mucosa of neovaginal canal was intensified to 4-5 layers. After 12 weeks, the differences between the neovagina and the native vagina were harldy noted either in the gross or microscopically.(2) After 4 weeks,a few smooth muscle cells were observed with VG and immunohistochemical staining, and homogeneous muscle bundle was formed.(3) After 12 weeks, similar contractile responses between neovagina and native vagina were observed when KCI and electrical stimulation with different frequency and voltage were given [(2.96±0.29)g vs.(3.14±0.30)g,(3.43±0.34)g vs.(4.65±0.73)g,(4.92±0.38)g vs.(4.89±0.44)g].Conclusion The tissue-engineering biological material might be an ideal graft used in the reconstruction of vagina.

Objective To study the CT symptom and diagnosis value of pancreatic trauma. Methods Retro-spectively analyzed the CT and clinical data of 11 pancreatic trauma. Results Bleeding, edema in pancreas parenehy-ma, uprightness low density lines appear along long axis of pancreas when enhancement were performed ,which werethe direct symptom of pancreatic trauma and rupture; hematoeele, hydrops around pancreas, front fascia of left kidney broaden, pancreatic pseudoeyst pneumatosis in bile duet, which were the indirect symptom of pancreatic trauma. Con-clusion CT is the first choice in the diagnosis of pancreatic trauma. Master the CT symptom and make right diag-nosis, which were important to clinical operation.

Chinese Pharmacological Bulletin 2010;26(4):421-424
Nowadays, cerebrovascular disease is one of the main causes of human death.Activation and aggregation of the platelet play core roles in the development of cerebrovascular diseases.A lot of clinical applications have showed that there are great interindividual differences in the response to antiplatelet drugs, and many patients have developed drug resistances.Pharmacogenetic studies have revealed that genetics is one of the key factors resulting in the interindividual differences of the response to antiplatelet drugs. In this overview, the newest advancement in pharmacogenetics is summarized with respect to the antiplatelet drugs.
