1.Synthesis of 10-Position Schiff Base Derivatives of Emodin and Preliminary Investigation of Anti-tumor Ac-tivity in vitro

Objective: To synthesize 10-position schiff base derivatives of emodin and study the anti-tumor activity. Methods:The 10-position schiff base derivatives of emodin were synthesized through emodin reacting with ammonia derivatives in absolute alcohol with acetic acid as the catalytic agent. Their structures were characterized by 1 H-NMR and ESI-MS, and the anti-tumor activity on k562 cells was determined by MTT method. Results:Three 10-position schiff base derivatives were synthesized and the structures were confirmed, and the IC50 on k562 cells was 4. 5, 3. 8 and 2. 0 μmol·L-1 , respectively. Conclusion:The 10-position schiff base de-rivatives of emodin can improve the anti-tumor activity and druggability of the parent compound, which are worthy of further study.
2.Effect of whole course health education on heart rehabilitation of old patients with percutaneous coronary intervention

Objective To investigate the effect of the whole course health education on heart rehabilitation of old patients with percutaneous coronary intervention(PCI).Methods One hundred and twentyone old patients with coronary heart disease who all underwent complete PCI in Tangshan Gongren Hospital from January 2008 to December 2009 were selected as our subjects.All subjects were randomly divided into the intervention group (60 cases) and the control group (61 cases).All patients were treated by PCI ± standard drug therapy.Patients in control group were given the conventional health education,while in intervention group were given the extending health education plus the conventional health education.Questionnaire was used to collect the coronary heart disease knowledge,recovery and secondary prevention behavior.The levels of blood pressure,density lipoprotein cholesterol,fasting blood glucose (FB) were measured at 6,12,24 months at discharge hospital.Meanwhile the reaching standard rate of coronary heart disease risk factors control was also used to evaluate the effect of education.Results The scores of knowledge of coronary heart disease rehabilitation of intervention group at 6,12 and 24 months after discharge were (79.69 ± 4.66),(83.26± 8.38),(81.81 ± 9.21) respectively,higher than that of control group ((72.36 ± 8.16),(69.96 ± 10.69),(70.00 ± 11.32) respectively),and there were significant differences between groups(t =3.26,5.39 and 4.41respectively,P < 0.05).The scores of the second level prevention act of intervention group were (79.00±5.65),(79.88 ± 6.34) and (79.13 ± 11.21) respectively,higher than that of control group((70.72± 7.59),(68.84 ± 5.36) and (66.52 ± 9.15) respectively),and there was significant difference between groups (t =3.75,5.99 and 3.63 respectively; P < 0.05).The reach standard rate of low density lipoprotein cholesterol(LDL-C),exercise of intervention group at 6 months were 60.0% (36/60),65.0% (39/60),higher than that of the control group(37.7% (23/61),32.8% (20.61)),and there was significant difference between groups(x2 =6.045,10.871,P <0.05),while there was no significant difference in terms of the reach standard rate of blood pressure,FB and quit smoking(P > 0.05).The reach standard rate of blood pressure,LDL-C,FB,quit smoking,exercise of intervention group at 12 months were 75.0% (45/60),76.6% (46/60),96.6%(58/60),81.7 % (49/60),75.0% (45/60),higher than that of control group (49.2 % (30/61),32.8 %(20/61),80.3% (49/61),57.4% (35/61),31.1% (19/61),and there was significant difference between groups(x2 =8.021,28.438,8.431,10.435 and 23.128 respectively,P < 0.05).The reach standard rate of blood pressure,LDL-C,FB,quit smoking,exercise of intervention group at 12 months were (75.0% (45/60),81.7% (49/60),95.0% (57/60),86.7% (52/60),81.7% (49/60)),higher than that of control group (44.3% (27/61),31.1% (19/61),75.4% (46/61),47.5% (29/61),34.4% (21/61)),and there was significant difference between groups (x2 =11.710,35.810,8.112,25.450 and 25.650 respectively,P<0.05).Conclusion Whole course health education is proved to reach the purpose of long-term heart rehabilitation,and decrease the risk of the risk factors for PCI.Meanwhile it is a safe,effective,compliant heart rehabilitation model after PCI in old patients.
3.STUDY ON ANGIOTENSIN CONVERTING ENZYME INHIBITORY PEPTIDES FROM DIGESTION OF SOY PROTEIN IN VITRO

Objective: To investigate if angiotensin converting enzyme inhilitory(ACEI) peptides would be produced from SPI digested by a batch digestion system using enzymes similar to digestive enzymes in humans.Method: Simulate the conditions of human gastrointestinal digestion in a model digestion system in vitro and produce soy peptides from SPI digested using pepsin and pancreatin.In addition to monitoring ACEI activity in the total soy protein digest,the possibility of generating soy peptide fractions with more potent activity than the unfractionated digest was investigated by measuring activity of fractions obtained after ultrafitration,anion exchange,and RP-HPLC.Results: The generation of ACEI activity in SPI was determined after sequential digestion with pepsin and pancreatin.The inhibitory activity was highest within the first 20 min at pepsin digestion and decreased upon subsequent digestion with pancreatin.An IC50 value of 0.28?0.06 mg/ml was determined after 180 min of digestion,while no ACEI activity was measured for the undigested SPI at 0.73 mg/ml.Chromatographic fractionation of the SPI digest resulted in IC50 values of active fractions ranging from 0.13?0.03 to 0.93?0.08 mg/ml.Conclusion: Many different peptides with ACEI activities were produced after pepsin-pancreatin digestion of SPI in vitro and lead to the speculation that physiological gastrointestinal digestion could also yield ACE inhibitory peptides from SPI.
4.Diagnostic value of MRI for spontaneous intracranial hypotension syndrome

Objective To analyze the magnetic resonance imaging ( MRI ) appearances of spontaneous intracranial hypotension syndrome ( SIHS) ,in order to improve the understanding of this disease .Methods The clini-cal data of 7 patients with SIHS confirmed by lumbar puncture were retrospectively analyzed .MRI was performed in all cases,in which there were 6 cases with contrast-enhanced MRI.Results All of them showed diffusely thickening of dural.6 cases with brain contrast-enhanced MRI demonstrated linear ,non-nodular and diffusely dural enhancement . The dilation of sinus and great vein were observed in 5 cases,subdural hydroma in 2 cases,brain sagging in 3 cases, pituitary gland plumpness in 2 cases,subdural hematoma in 1 case and 1 case with linear spinal dural enhancement . Conclusion The MRI manifestations of SIHS are characteristic ,and it will be helpful for the diagnosis by combining clinical data.
5.Evenly-designed and optimised technological parameters for Maxingshigan Decoction by semi-bionic extraction

Objective:Optimising the technological parameters for the extraction of Maxingshigan Decoction (Herba Ephedrae, Semen Pruni Armeniacae, Gypsum Fibrosum, Radix Glycyrrhizae). Methods:Using the even design and TCL, ephedrine, glycyrrhetinic acid, hydrocyanic acid, calciumion and extract weight were taken as the indexes. Results:Optimal parameters: the pH values of the water for the 1st, 2nd and 3rd decoctions are 2.0 ,6.5 and 9.0,respectively; and the time of 3 decoctions are 2.0,1.0and 0.5h, respectively. Conclusion:Optimal parameters for Maximpshipan Decoction are feasible in extraction.
6.Comparison of genomic DNA content of liver and spleen tissue for different age of elderly

Objective To investigate the change of genomic DNA of liver and spleen tissue for different age of the elderly,and provide the experimental data for aging-related research. Methods 35 livers and 33 spleens of autopsied samples preserved in refrigerator at-80 ℃ were divided into 3 groups according to age:age 65y to 79y,age 80y to 89y,age≥90y. The content of DNA in liver and spleen was determined by ultraviolet absorbent method. Results Compaired with age 80y to 89y (0. 310 ± 0. 286)mg/mL,the content of DNA in liver was significant higher at age 65y to 79y (1.464 ±0.488)mg/mL and age ≥90y(1.147 ±0.333)mg/mL(P<0.05);Compared with age 80y to 89y(0. 938 ± 0. 589)mg/mL,the content of DNA in spleen was significant higher at age 65y to 79y(1. 723 ± 0. 726)mg/mL and age≥90y(1. 688 ± 0. 963)mg/mL(P<0. 05). The content of DNA was significant lower in liver (0. 856 ± 0. 658)mg/mL than that in spleen (1. 414 ± 0. 852)mg/mL. Conclusion The content of DNA in human liver and spleen tissue may be decrease along with aging. The content of DNA in the group at age≥90y may be increase. There were some differences between different viscera tissue in content of DNA.
8.Diagnostic value of anti-carbamylate protein antibod for rheumatoid arthritis: a meta analysis

Objective We aimed to evaluate the diagnostic value of anti-carbamylate protein antibody (anti-CarP antibody) for rheumatoid arthritis (RA).Methods All domestic and foreign database were searched by computerized and manual methods,including PubMed database,Embase database,Cochrane library.All studies about anti-CarP antibody on the diagnostic value of RA were included.The deadline of the search was March 15,2018.Studies that did not meet the population and threshold to select criteria requirements were excluded.Results Manager 5.2,Meta-disc 1.4 and Stata12.0 software were used for literature quality evaluation and statistical analysis.Results A total of 8 papers were included.The heterogeneity test showed that there was heterogeneity caused by nonthreslold effects among the studies,and random effects model was used for Meta analysis.The sensitivity,specificity,positive and negative likelihood ratios of antiCarP antibody for the diagnosis of RA were [45%,95%CI(43%,48%)]、[97%,95%CI(95%,98%)]、[12.65,95% CI (8.87,18.05)]、[0.56,95% CI (0.53,0.6)].DOR was [23.97,95% CI (16.33,35.17)] respectively,and the AUC was 0.777 5.Conclusion Anti-CarP antibody is highly specific for the diagnosis of RA,but its sensitivity is poor.
9.Effects of inflammatory microenvironment mediated by macrophage on the proliferation and osteogenic differentiation of periodontal ligament cells

Objective:
The present study investigated the effects of the inflammatory microenvironment mediated by macrophages on the proliferation and osteogenic differentiation of periodontal ligament cells (PDLCs).
Methods:
Conditioned medium containing inflammatory factors was collected following macrophage activation with 1 μg/mL lipopolysaccharide (LPS). PDLCs were isolated from healthy teeth and cultured in conditioned medium (LPS-CM group) or normal medium (control group), and the proliferation of PDLCs was detected using the MTT assay. The cells were cocultured with an osteogenic inducer for 3 d and 7 d, and the alkaline phosphatase (ALP) activity of PDLCs was detected using an ALP kit. The mRNA expression levels of runt-related transcription factor 2 (RUNX2), osteocalcin (OCN), and collagen I (COL-I) were detected using real-time PCR, and the protein levels of RUNX2, OCN, and COL-I were detected using Western blotting. Mineralization nodules were observed using Alizarin red staining after osteoinduction for 14 d.
Results:
The OD value of PDLCs in the LPS-CM group was lower than that in the control group (P < 0.05). The mRNA levels of RUNX2, OCN, and COL-I in the LPS-CM group were lower than those in the control group (P < 0.05). In addition to the OCN 3 d group (t = 2.75, P = 0.056), the protein expression of RUNX2, OCN, and COL-I in the LPS-CM group was lower than that in the control group (P < 0.05). However, the ALP activity of the LPS-CM group was higher than that of the control group, which was 1.58-fold greater (t = 5.91, P = 0.030) at 3 d and 1.29-fold greater (t = 6.01, P = 0.046) at 7 d. The number of calcified nodules in the LPS-CM group was significantly less than that in the control group (t = 8.63, P = 0.048).
Conclusion
The inflammatory microenvironment mediated by macrophages may inhibit the proliferation and osteogenic differentiation of PDLCs.
10.Metabonomic study on protective effect of ethanol extracts of drynariae rhizoma on osteoporosis in rats urine by using UPLC-MS/MS.

This paper was designed to study metabonomic characters of the osteoporosis induced by high dose of hydrocortisone and the protective effects of Drynariae Rhizoma, which can replenish the kidney and strengthen the bones. A urinary metabonomics method based on ultra-performance liquid chromatography coupled with mass spectrometry (UPLC-MS/MS) was developed. Clear separation of healthy control group, model group and treatment group was achieved by using the principal components analysis (PCA) and 9 significantly changed metabolites were identified as potential biomarkers of osteoporosis. Compared with the health control group, the model group rats showed lower levels of creatinine, citric acid, azelaic acid, hippurate, tryptophan and indoxyl sulfate together with higher levels of phenylalanine, cresol sulfate and phenaceturic acid. These changes in urinary metabolites suggest that the disorders of amino acid metabolism, energy metabolism, gut microflora and anti-oxidative damage are related to osteoporosis induced by high dose of hydrocortisone and the potential effect of Drynariae Rhizoma on all the four metabolic pathways.
