Oyster protein was extracted from fresh oyster using salting-out method. It was determined by Kjeldahl procedure that the protein content was about 38. 6% and free amino-ni-trogen content of total nitrogen was about 21. 6%. Hydrolytic characteristic of oyster protein was analyzed by hydrolytic enzymes. SDS-PAGE results showed there were 4 types of protein zones. And the average molecular weight was 16172, 17633, 43227 and 57909, respectively.

The study is to establish an HPLC method using fluorescence detector for the determination of doxazosin enantiomers and investigate their chiral inversion in vitro and in vivo. Ultron ES-OVM was taken as the chiral chromatographic column, and the column temperature was 30 degrees C. Isocratic elution using a mobile phase of phosphate buffer-acetonitrile (85 : 15, v/v) at a flow rate of 0.8 mL x min(-1) was done. The fluorescence detection was set at lambda(Ex) = 255 nm and lambda(Em) = 385 nm. Prazosin was used as the internal standard. (-) Doxazosin or (+) doxazosin added into rat plasma in vitro was determined after incubating in 37 degrees C water bath for 2, 5 and 10 days. (-) Doxazosin or (+) doxazosin was administered orally to the rats for one months. Plasma samples were taken at 8 h after the last administration. A good linear relationship was achieved when the concentration of doxazosin enantiomers was within the range of 4 - 2 000 ng x mL(-1). The average recovery for (-) doxazosin was 99.5% with RSD 3.6%, and for (+) doxazosin was 99.3% with RSD 4.3%. Chiral inversion was observed neither in vitro nor in vivo studies. The method is selective, accurate and reproducible, which is suitable for the detection of doxazosin enantiomers in rat plasma. The in vitro and in vivo studies indicate that chiral inversion occurs uneasily between (-) doxazosin and (+) doxazosin in the rat.

Objective To study the effects of flavone compound α-NF on CYP1B1 gene in transplanted nude mice tumor with human paclitaxel resistance ovarian cancer cell. Methods Using 24 nude models of orthotopic transplantation of human ovarian cancer, they were divided randomly into four groups: control group, α-NF group, paclitaxel group and combined group. RT-PCR and Western blotting method were used to detect CYP1B1 mRNA and protein expression. Results There was no significant difference in CYP1B1 mRNA expression between α-NF group, combined group and control group(P >0.05). Compared with control group, CYP1B1 protein expression was distinctly lower in α-NF and combined group (P<0.01). Conclusion α-NF has no effect on CYPIB1 gene in mRNA level, whereas α-NF down-regnlate, CYP1B1 expression of transplanted A2780TS nude mice tumor in protein level, thus α-NF has the effect on reversing paclitaxel resistance of A2780TS cell.

Objective To test the reliability and validity of the self-assessed questionnaire of the elderly health care needs.Methods Based on the Zuluaga-Raysmith model, the initial questionnaire was formulated by literature reviewing and interviews.Through Delphi experts consultation, a questionnaire containing 12 factors, 61 items was constructed.A total of 750 elderly residents were investigated to test the reliability and validity of the questionnaire.Results The effective rate of questionnaires was 88.67％ (665/750).The total Cronbach α was 0.871.The test-retest reliability of all the items were all greater than 0.75.The item-level content validity index were all greater than 0.80.The scale-level content validity index was 0.95.14 factors were extracted by exploratory factor analysis, explaining 66.653％ of the total variance.Conclusions The elderly health care needs self-assessed questionnaire is comprehensive with good stability, which can be used in a multi-center, large sample research.The research may help to get a deep understanding of health care needs of the elderly as well as influencing factors.

Objective: To investigate the effects of fosinppril on myocardial cell apoptosis and apoptosis-associated gene expression in chronic heart failure (CHF) rats.
Methods: CHF model was established by partially banding of abdominal aorta superior to renal artery. The experimental rats were randomly divided into 3 groups: Sham operation group and CHF group, the rats in both groups received stomach normal saline; Fosinopril group, the rats received stomach fosinopril 10 mg/kg?d.n=10 in each group and all animals were treated for 8 weeks. LVEDP, ±dp/dtmax and LVMI were examined, left ventricular myocardial cell apoptotic index (AI) was measured by TUNEL method, Bcl-2 and Bax protein levels were detected by immunohistochemistry and caspase-3 protein expression was assessed by Western blotting.
Results: Compared with Sham operation group, CHF group had increased LVEDP, LVMI, AI, elevated protein expressions of Bax and Caspase-3,P<0.01; while decreased ±dp/dtmax, reduced protein expression of Bcl-2 and the ratio of Bcl-2/Bax,P<0.01. Compared with CHF group, Fosinopril group presented decreased LVEDP, LVMI, AI, reduced protein expressions of Bax and Caspase-3,P<0.01; while increased ±dp/dtmax, elevated protein expression of Bcl-2 and the ratio of Bcl-2/Bax, P<0.01.
Conclusion: Fosinopril may inhibit myocardial cell apoptosis which occurred during CHF, by up-regulating Bcl-2 expression and down-regulating expressions of Bax and Caspase-3 in CHF rats, therefore improve the cardiac function.

Objective To observe the effect of Chaihushugansan combined with fluoxetine on interleukin -6 ( IL-6 ) , interleukin -1β( IL-1β) , tumor necrosis factor ( TNF-α) in the treatment of patients with postpartum depression.Methods 83 patients from November 2013 to July 2015 in Jiaxing Maternity and Child Health Care Hospital were randomly divided into 41 patients of observation group and 42 patients of control group.The control group were treated with fluoxetine treatment, the obsercation group received Chaihushugansan on the basis of control group.The Hamilton depression scale (HAMD) and Hamilton anxiety scale (HAMA),IL-6, IL-1β, TNF-α, high-sensitivity C-reactive protein (hs-CRP) and adverse reactions were followed up and recorded.Results The HAMD and HAMA score in observation group post-treatment were (8.31 ±2.05,9.03 ±2.08)points, which were lower than (13.96 ±2.16, 13.61 ±2.14) points in control group (P<0.05).The serum IL-6 and IL-1βlevels in observation group post-treatment were (6.59 ±3.20,5.01 ±2.83)pg/mL, which were lower than (10.64 ±3.86,9.31 ±3.42)pg/mL in control group (P<0.05).The serum TNF-αand hs-CRP levels in observation group post-treatment were ( 9.16 ±2.01 ) pg/mL, ( 3.62 ±1.06 ) mg/L, which were lower than ( 12.30 ±2.37 ) pg/mL, (5.29 ±1.14)mg/L in control group (P<0.05).The total adverse reaction rate in observation group was 7.32%, significantly lower than 23.81% in control group ( P <0.05 ) .Conclusion Chaihushugansan combined with fluoxetine has a good therapeutic effect in the treatment of postpartum depression, could significantly reduce the IL-6, IL-1β, TNF-α levels, with fewer side effects, it is better than fluoxetine alone.

OBJECTIVE To evaluate risk factors of the death among lung cancer patients with nosocomial pneumonia.METHODS A total of 312 lung cancer patients with nosocomial pneumonia in our hospital were retrospectively analyzed.RESULTS Eighty-seven patients died and remaining 225 patients were cured finally.Risk factors by multivariate analysis were more than 62 years old(OR 1.75,95%CI 1.06-2.98),metastatic disease(OR 9.24,95%CI 5.21-16.17),bacteria/fungi infection(OR 2.31,95%CI 1.15-4.67),and lower serum albumin concentration(OR 1.87,95%CI 1.08-3.13).CONCLUSIONS Older,metastatic diseases,double infections and lower serum albumin concentration are the risk factors correlated with the mortality among lung cancer patients with nosocomial pneumonia.

Objective To compare the effects of combination of maternal hydration and hands and knees posture with simple hands and knees posture on correction of breech pregnancy. Methods One hundred patients who were diagnosed breech pregnancy from November 2015 to November 2016 were assigned to experimental group and control group with 50 cases each according to individual willingness. The patients in experimental group received the combination of maternal hydration and hands and knees posture;while the patients in control group received simple hands and knees posture. Results Amniotic fluid index was (18.94±2.44)cm in experimental group and (15.97±2.33)cm in control group, the difference had statistic significance (t=6.239, P<0.05). The effective transfer rate and successfully used time was 92%(46/50) , (2.10 ± 0.75) weeks in experimental group and 64%(32/50), (4.27 ± 0.98) weeks in control group, the difference had statistic significance (χ2=11.422, P<0.05; t=-12.463, P<0.05). For primipara and multipara, the effective transfer rate was 90.91%(30/33), 16/17 in experimental group, and 68.75%(22/32), 10/18 in control group, the difference had statistic significance (χ2=4.986, 6.806, all P<0.05). The rate of vaginal and cesarean section deliveries was 86%(43/50), 14%(7/50) in experimental group, and 56%(28/50), 44%(22/50) in control group, the difference had statistic significance(χ2=10.928, P=0.001). Patients who had fetal movement or who didn′t have time to do correction or missed was 17.10%(46/269), 1.49%(4/269) in experimental group, and 12.55% (22/255), 7.06% (18/255) in control group. Conclusions @Therapic effects of combination of maternal hydration and hands and knees posture is much better than that of the simple hands and knees posture therapy on correction of breech pregnancy, which is worthy of popularization and application in clinic.

Objective To explore the feasibility of hemochrome identification by micro spectrophotometer. Methods We use dip lotion by Centrifugation, reaction with modiifed takayama reagent,measured absorption spectra with a micro spectrophotometer to determine whether samples contain blood. On the basis of optimized parameters, we use diluted human hemoglobin to measure sensitivity of the method; using suspected blood / blood spots, and different storage time and matrix of blood spots to measure speciifcity and samples of adaptability. Result With micro spectroscopic method of hemochromogen,only blood (spot) has speciifc spectral consisting of three absorption peaks at 415,525 and 555nm and no cross suspected common blood / spot.Reaction 2 min, sample volume2.5ul, 1000-fold diluted human blood stably obtains primordial spectrum. By prolonging the immersion time, set the vehicle control, 10 years old blood gauze. Blood stains in colored cloth can be effective detective.. The height of absorption peaks and blood content were significantly corelated, and the change in absorbance at 525 and 555nm consistent trend (y=0.5232x+0.0274, R2=0.9971). Conclusion Micro spectroscopic method of hemochromogen has high sensitivity and speciifcity, quick and easy, can be incorporated into the DNA testing process. There is a good prospect in the actual seizure case. Instead of the traditional crystallization method for teaching, training requirements more in line with the skills and awareness for current students.
