1.Effects of PNS on the Apoptosis and Expression of C-myc of Vascular Smooth muscle Cells

Objective To observe the effects of panax notoginseng saponins on the apoptosis and expression of C-myc of vascular VSMC in vitro.Methods VSMC derived from rabbit aorta were cultured in vitro and different concentrations of PNS were added in experimental groups.VSMC apoptosis was evaluated with cell ultrastructure under electron microscopy and flow cytometry.Expression of C-myc was observing by immunocytochemical method.Results The ratio of apoptosis and the expression of C-myc in PNS groups were high than control group.Conclusions PNS can increase the ratio of VSMC apoptosis which may be related the expression promoting of C-myc.
2.The effects of valsartan on the proliferation,migration,p-ERK1/2 and p-P38 MAPK expression of vascular smooth muscle cells

Objective To investigate the effect of valsartan on the proliferation and migration of rat aortic smooth muscle cells and the expression of p-ERK1/2 ,p-P38 promoted by angiotensinⅡ (AngⅡ ) .Methods VSMCs from the rat thoracic aorta were cultured by attachment-block culture .VSMCs from3-5 passages were used .The proliferation of VSMCs was tested by MTT meth-od .Pipettetip wounding injury was used to access the migration of VSMCs .The expression of ERK1/2 ,p-ERK1/2 ,P38 and p-P38 of VSMCs was detected by Western Blot .Results (1) AngⅡ could significantly promote the proliferation of VSMCs .The prolifer-ation of VSMCs promoted by AngⅡ could be inhibited by Valsartan and PD98059 .Valsartan inhibited the proliferation of VSMCs in a concentration-dependence .SB23015 could significantly promote the proliferation of VSMCs .(2 ) The migration activity of VSMCs in the Ang Ⅱ was increased significantly .Valsartan and PD98059 inhibited Ang Ⅱ-induced migration of rat VSMCs and SB23015 was on the contrary .(3) Valsartan and PD98059 could inhibit AngⅡ-induced VSMCs intracellular p-ERK1/2 protein ex-pression ,and this role was enhanced by SB23015 .Valsartan and SB23015 could inhibit Ang Ⅱ-induced VSMCs intracellular p-P38 protein expression ,and PD98059 had no significant effect .(4)VSMCs had no significant effect on cell proliferation ,migration and p-ERK1/2 ,p-P38 protein expression under valsartan alone .Conclusion (1)Valsartan could inhibit Ang Ⅱ-induced proliferation and migration of VSMCs in a concentration-dependence .(2)The effects that valsartan inhibits the proliferation and migration of Ang Ⅱ-induced VSMCs might be related to its inhibiting Ang Ⅱ-induced p-ERK1/2 protein expression .(3)p-P38 might play a negative reg-ulatory role on the AngⅡ-induced p-ERK1/2 activation .

4.Study on therapeutic effect of cerebrospinal fluid replacement combined with intrathecal injection on tuberculous meningitis

Objective To explore the therapeutic effect of cerebrospinal fluid (CSF) replacement combined with intrathecal injection on tuberculous meningitis. Methods Fifty-five patients with tuberculous meningitis were randomly divided into treatment group and control group. The patients in the treatment group were treated with CSF replacement combined with intrathecal injection on the basis of routine anti-tuberculosis therapy. The curative effects between these two groups were analyzed after 8 weeks. Results The total effective rate was 95.7% in the treatment group,compared with that 61.9% in the control group (P < 0.01); the healing rate in the treatment group was 56.2% vs 28.6% in the control group. In addition,the decay rate for WBC and CSF pressure in the treatment group are more rapid than those in the control group(P < 0.05). Conclusion CSF replacement combined with intrathecal injection has better curative effect in patients with tuberculous meningitis.
5.Effects of long-and mid-term indwelling of AO clavicular hook plate on the shoulder function following acromioclavicular dislocation surgery

Objective To investigate the effects of long-and mid-term indwelling AO clavicular hook plate (CHP) on the shoulder function following surgery for acromioclavicular dislocation(from grade Ⅲ to grade Ⅴ by Rockwood's classification).Methods A retrospective study was done of 95 patients with acromioclavicular dislocation who had been fixed with AO CHP from December 2002 to May 2011.They were 80 men and 15 women,aged from 20 to 63 years (average,45.0 years).By the Rockwood's classification,there were 40 cases of type Ⅲ,35 ones of type Ⅳ and 20 ones of type Ⅴ.Time from injury to surgery ranged from 2 to 7 days (average,4.6 days).The CHP was indwelling in 70 patients but removed in 25 patients from 12 to 23 mouths post-surgery (average,16.5 months).The University of California (UCLA) shoulder scoring system was used to compare the shoulder functions between those with an indwelling CHP and those without,as well as between the healthy and affected shoulders among those with an indwelling plate.Results The follow-up time for all the 95 cases ranged from 7.5 months to 9 years,with an average of 38 months.Of those with an indwelling CHP,subacromial bone erosion occurred in 45,internal implant failure in 3,impingement in 13,screw loosening in 2 and frozen shoulder in 5,giving a complication rate of 97.1% (68/70).Of those without an indwelling CHP,subacromial bone erosion occurred in 3 and no redislocation was found in this group,giving a complication rate of 12.0% (3/25).The UCLA score for the affected shoulder among those with an indwelling CHP(27.0 ± 11.0 points) was lower than that for the healthy one (34.0 ± 1.0 points) and lower than that for the affected shoulder among those without an indwelling CHP(32.0 ± 6.5 points).Conclusion Long-and mid-term indwelling AO CHP can have a significant adverse impact on shoulder function,leading to a very high rate of postoperative complications.
6.Quality standard for Anshen Wendan Pill

AIM:To establish a quality standard for Anshen Wendan Pill(Rhizoma Pinelliae Praeparata,Pericarpium Citri reticulatae,Poria,Radix et Rhizoma Ginseng,etc.). METHODS:Pericarpium Citri Reticulatae,Fructus Schisandrae Chinensis,Radix et Rhizoma Ginseng,and Radix et Rhizoma Glycyrrhizae were identified by TLC,and the contents of schisandrin and hesperidin were determined by HPLC. The content of cinnabaris was determined by titration. RESULTS:Pericarpium Citri reticulatae,Fructus Schisandrae Chinensis,Radix et Rhizoma Ginseng,and Radix et Rhizoma Glycyrrhizae could be identified by TLC. The calibration curve of schisandrin was linear in the range of 0.017-0.52 ?g with the correlation coefficient of 1.000 0(n=5).The average recovery of schisandrin was 99.83%(RSD=0.86%,n=6). The calibration curve of hesperidin was linear in the range 0.506 -2.53 ?g,with the correlation coefficient of 1.000 0(n=5).The average recovery hesperidin was 100.43%(RSD=1.39%,n=6). The average recovery of cinnabaris was 98.30%(RSD=1.06%,n=6). CONCLUSION :The methods are simple,accurate and reproducible. They are suitable for the quality control of Anshen Wendan Pill.

8.Application of ~(18)F-FDG hybrid PET/CT imaging in clinical staging of non-small cell lung cancer

Objective To investigate the application of ~(18)F-fluorodeoxyglucose hybrid PET/CT (~(18)F-FDG hPET/CT) imaging in clinical staging of non-small cell lung cancer (NSCLC). Methods Seventy two consecutive patients with NSCLC undergoing ~(18)F-FDG hPET/CT before radiotherapy were analyzed. The results were compared with previous CT scan and conventional clinical staging. The patients were followed-up for at least 6 months. Results Among the 72 patients, the staging grade was changed in 27 patients due to the result of ~(18)F-FDG hPET/CT in whom with 20 patients having their grade raised and 7 lowered. Radical treatment was changed to palliative treatment in view of a raise of staging grade. Because of distant metastasis was detected by ~(18)F-FDG hPET/CT imaging, 14 patients received palliative treatment. Compared with CT, 47 more lymph nodes and 26 distant metastases were found with ~(18)F-FDG hPET/CT imaging, and the patients in question received radiotherapy and palliative treatment accordingly. Conclusion ~(18)F-FDG hPET/CT imaging, by changing the clinical staging in 37.5% (27/72) NSCLC patients, has impact on treatment strategy and treatment planning of radiotherapy in NSCLC patients. Patients were frequently spared unnecessary treatment, and management was more appropriately targeted.
9.Preliminary results of osteotomy with short fusion and dual growing rod technique for congenital scoliosis

Objective To evaluate the clinical outcomes of the hybrid technique of posterior osteotomy with short segmental fusion and dual growing rod technique for severe rigid congenital scoliosis.Methods Seven patients (2males,5 females) undergoing this hybrid technique for severe rigid congenital scoliosis in our hospital from 2006 to 2011 were retrospectively studied.The average age was 5.9 years (range,2-10).The Risser sign of all the patients was 0.The follow-up time was 59.4 months (range,36-83 months).The patients' charts were reviewed.The analysis included age at initial surgery and the latest follow-up,number and frequency of lengthening,and complications.Radiographic evaluation included measured changes in scoliosis Cobb angle,thoracic kyphosis,lumbar lordosis,trunk shift,length of T1-S1 and instrumentation.Results All patients were treated with posterior osteotomy with short segmental fusion and dual growing rod technique.There were 48 total surgeries,41 of which were lengthening procedures,for 7 patients.The average lengthening was 5.9 per patient.The mean scoliosis improved from 81.4° to 40.1 ° after initial surgery and was 41.1 ° at the final follow-up.The average T1-S1 length was of 1.12 cm per year.The Campbell's space available for lung ratio increased from 0.87 to 0.97.Conclusion Osteotomy with short fusion could help to improve the correction of the growing rod and eliminate the large asymmetric growth potential around the apex,with little influence to the length of the spine.Dual growing rod technique could maintain correction achieved at initial surgery while allowing spinal growth to continue.However,this technique is relatively more aggressive and technique demanding.And the patients need numbers of surgeries.
10.The Expression of MCP-1 in the Diabetes Mellitus Skin Defect Healing

Objective To study the expression of MCP-1 in the diabetes mellitus skin defect healing, then discuss its effect. Methods The diabetes mellitus models were established, and the expression of MCP-1 was detected by immunohistochemical staining and Elisa. Results The immunohistochemistry stain showed that the expression of MCP-1 in DM group was higher than that in control group at all time. And the serum value of MCP-1 in DM group had statistical significance ( <0.05) at 3 days or 1 week, but that in 2 weeks and 4 weeks was no statistical significance ( <0.05) . Conclusion The MCP-1 might play a role in the healing of diabetes mellitus skin defect through mediate abnormal inflammatory response.
