3.Advances in studies on the application of platelet-rich plasma in the field of orthopedicsand the underlying mechanisms

Platelet-rich plasma(PRP) is a highly-concentrated plasma enriched in platelets and various growth factors. These growth factors will be released abundantly by platelets when PRP is activated by thrombin, which plays a key role in accelerating wound healing, bone repair, and meanwhile, in controlling of inflammatory process. Biocompatibility and versatility of PRP have been confirmed in clinical studies as well as its excellent effects on tissue reparation. These have lead to the extensive use of PRP in many medical fields as a therapeutic alternative. Mechanism of PRP in bone and tissue repair the application of PRP in the field of orthopedics is reviewed in this article. In addition, attempt is made to discuss about some problems still existing in the clinical application of PRP.
4.Osthole alleviates cerebral ischemia-reperfusion inj ury by suppressing mitochondrial mediating apoptosis

ABSTRACT:Objective To observe the influence of Osthole on cerebral ischemia-reperfusion injury and its acting mechanism.Methods We randomly divided 50 SD rats into sham (Sham)group,ischemia-reperfusion injury (IRI)group and Osthole (25,50,and 100 mg/kg)(Osthole)groups (n=10).The neurological symptoms, extent of cerebral infarction and cerebral water content were evaluated by physical approach.The activities of ROS and ATP were examined by the kit;the mitochondrial membrane potential (MMP ) was measured by flow cytometry.The expressions of Bax,Bcl-2,Caspase3,clevage-Caspase3,Caspase9,clevage-Caspase9,cytoplasmic AIF and cytochrome C were measured by Western blotting.Results Compared with those in sham group,the extent of cerebral infarction and cerebral water content,changes of neurological symptoms,activities of ROS,and expressions of clevage-Caspase3 ,clevage-Caspase9 ,Bax,cytoplasmic AIF and cytochrome C in IRI group were significantly increased (P<0.05).However,the activities of ATP,expressions of Caspase3,Caspase9,Bcl-2 and MMP,and ATP activity were decreased (P<0.05).Compared with those in IRI group,the extent of cerebral infarction and cerebral water content,changes of neurological symptoms,activities of ROS,and expressions of clevage-Caspase3 ,clevage-Caspase9 ,Bax,cytoplasmic AIF and cytochrome C in Osthole group were significantly decreased (P<0.05).But the activities of ATP,expressions of Caspase3,Caspase9,Bcl-2 and MMP,and ATP activities were increased (P < 0.05 ). Conclusion Osthole pretreatment can attenuate cerebral ischemia-reperfusion injury,which is associated with suppressing mitochondria-mediated apoptosis.
5.Pathogenesis and new therapeutic targets of liver fibrosis

In recent years,the basic research on liver fibrosis has been progressed rapidly.This article briefly reviews the cellular and molecular mechanisms of liver fibrosis,including the origin of myofibroblasts,immune regulation,autophagy,and epigenetic regulation,and introduces several new therapeutic targets.More and more evidences show that successful removal of causes is the most important antifibrotic therapy.At present,although the antifibrotic drugs acting on different targets have been emerging,most of them are still in the early stage of research and development,and well-designed clinical trials are needed to confirm their clinical efficacy.
6.Effect of Electroacupuncture on Hand Dysfunction after Stroke

Objective To observe the effect of electroacupuncture on hand dysfunction after stroke. Methods Forty hemiplegic patients after stroke were selected from a multi-center large-scale trail, belonged to rehabilitation group (n=20) and electroacupuncture group (n=20), who accepted routine rehabilitation and additional electroacupuncture, respectively, for six weeks. They were assessed with Brunnstrom Grade, Fugl-Meyer Assessment (FMA), Wolf Motor Function Test (WMFT) and modified Barthel index (MBI) before treatment, two, four and six weeks during treatment, and twelve weeks after treatment. Results There were differences between two groups in scores of FMA, WMFT and total time to complete WMFT twelve weeks after treatment (t>1.900, P<0.05). There was no difference between two groups in scores of MBI and Brunnstrom Grade (t=0.532, Z<1.79, P>0.05), but electroacupuncture group seemed to improve more. Conclusion Elec-troacupuncture may improve the muscle strength for patients in flaccid paralysis, and promote the recovery of hand function for patients af-ter stroke.

OBJECTIVE:To establish a RP-HPLC method to determine the content of ofloxacin in compound ofloxacin eye drops METHODS:Using Shim-pack ODS as fixed phase,a mixture to citric acid(0 05mol/L)-acetonitrile-ammonium acetate(0 5mol/L)-0 3% phosphoric acid(78∶22∶1∶3)as mobile phase,and detecting wavelength:293nm,flow rate:1 0ml/min RESULTS:The linear range of ofloxacin was 8 12?g~40 6?g(r=0 9 996,n=5) The average recovery was 99 29% and RSD(n=6)was 0 57% CONCLUSION:The method is rapid,simple and accurate
8.Relationship between cerebral oxygen saturation and postoperative cognitive dysfunction in elderly patients

Objective To investigate the relationship of intraoperative cerebral oxygen saturation(rSO_2 )monitored with near-infrared cerebral oximeter (INNOS 5100) and postoperative cognitive dysfunction in patientsoperated upon under isoflurane or sevoflurane anesthesia and to determine the critical rSO_2 value below whichpostoperative cognitive dysfunction may occur.Methods Sixty ASA Ⅰ- Ⅱ patients of both sexes aged 62-80 yr,weighing58-77 kg schedules for elective abdominal surgery or surgery on the low limb were divided into threegroups according to their levels of education: group Ⅰthe illierate and uneducated (n = 20); group Ⅱ primaryschool education (6yr education (n = 20). Each group was furtherdivided into isoflurane and sevoflurane subgroups (n = 10 in each subgroup). The patients were unpremedicated.Anesthesia was induced with intravenous atropine 0. 3mg, propofol 1 .0-1. 5 mg?kg~(-1), fentanyl 2-3?g?kg~(-1) andvecuronium o. 1-0.2 mg?kg~(-1) and maintained with isoflurane or sevoflurane inhalation (0.9-1. 1 MAC )supplemented with intermittent i.v. boluses of fentanyl and recorded before anesthesia (baseline), after O_2inhalation(T_1), after induction of anesthesia(T_2), after skin incision (T_3), during operation (T_4)and at the end ofsurgery (T_5). 9.Effect of Chuan Shen Tong Injection in the Treatment of Prostatic Hyperplasia: An Observation of 2090 Cases

Objective: To make further evaluation on the efficacy and safety of Chuan Shen Tong Injection (CSTI) on benign prostatic hyperplasia with blood-stasis syndrome. Methods: A self-contrast between pre-and post-treatment and multicenter trial was carried out in 2090 subjects treated with CSTI over 4 weeks. Results: The markedly effective rate of CSTI in controlling symptoms is 60.77 %and the total effective rate is up to 91.67 %. Conclusion: Chuan Shen Tong Injection is effective and safe in treating benign prostatic hyperplasia with blood-stasis syndrome.

Objective: To observe the clinical efficacy of Tuina (Chinese therapeutic massage) plus Shen Ling Bai Zhu San for pediatric diarrhea due to spleen deficiency.Methods: A total of 105 children with diarrhea due to spleen deficiency were selected and randomized into two groups by the random number table method. Fifty-two cases in the control group were treated with oral Shen Ling Bai Zhu San, and 53 cases in the observation group were treated with additional Tuina treatment. The clinical efficacy was evaluated after treatment, and the changes of traditional Chinese medicine (TCM) symptom scores were observed. Results: During the trial, there were three dropouts in the observation group, and two in the control group. After the treatment, the total effective rate was 100.0% in the observation group, and 90.0% in the control group. The difference between the two groups was statistically significant (P<0.05). The scores of primary and secondary symptoms, and the total scores of TCM symptoms in both groups were significantly lower than those before treatment (P<0.05). And the scores of primary and secondary symptoms, and the total score of TCM symptoms in the observation group after treatment were lower than those in the control group (P<0.05). There were no obvious adverse reactions in either group during the treatment. Conclusion: Tuina plus Shen Ling Bai Zhu San has a better therapeutic effect in treating pediatric diarrhea due to spleen deficiency than Shen Ling Bai Zhu San alone.
