The ecological balance of gastrointestinal microbiota plays an important role in digestion, absorption, metabolism, immunity and protection against pathogens. Functional disturbance of gastrointestinal microecology is closely related to gastrointestinal diseases. Recent studies have shown that gastrointestinal microecology was involved in the pathogenesis process of gastric cancer. This article reviewed the progress in study on relationship between gastrointestinal microecology and gastric cancer.
2.Human cervical cancer oncogene and tumor
Journal of International Oncology 2009;36(12):883-885
Human cervical cancer oncogene(HCCR) is newly identified in cervical cancer tissues, and expresses in most human tumors. Resarches show that HCCR is a candidate marker for human hepatocelular carcinoma and breast cancer. Moreover, the expression of HCCR is regulated by mitogen-activated protein kinase (MAPK)and phosphatidylinositol 3-kinase signal pathway. The proliferation, apoptosis, migration and invasion of tumor cells could be inhibited by siRNA of HCCR.

Results In Group A,B and C,the expression of DLL4 protein were (144.99 ±10.12 ),(1 15.42 ±10.39 ),(90.08 ±7.29 ),and there were significantly diffiences among three groups (F=314.220,P<0.05);The expression of DLL4 protein atⅠ~Ⅱstage were significantly lower than that inⅢ~Ⅳ stage (P<0.05).Conclusion DLL4 protein may play an important role in the pathogenisis of endometriosis.
4.Functional mechanism of the mutual regulation between long non-coding RNAs and other epigenetic mechanisms in cancer
Journal of International Oncology 2015;42(9):671-674
Abnormal expressions of long non-coding RNAs (LncRNAs) are associated with a variety of tumors.Recent studies show that lncRNA can regulate other epigenetic processes such as DNA methylation, histone modifications, chromatin remodeling and miRNA through various ways;on the other side, other epigenetic processes can influence the expression of lncRNA.The mutual regulation of them paly important roles in the occurrence and development of malignant tumors. The better understanding of the crosstalk between lncRNA and epigenetic modulation will contribute to clearly clarify the mechanism of tumor, which provides a theoretical basis for the anti-cancer therapies targeting lncRNAs.

Objective To investigate the protective effects of pretreatment with low-dose ketamine on intestinal mueosa against ischemia-reperfasion injury (I/R) and the underlying mechanism. Methods Forty-eight male SD rats weighing 230-270 g were randomly divided into 6 groups (n = 8 each) : group Ⅰ sham operation (S) ;group Ⅱ ketamine + sham operation (KS);group Ⅲ l/R;group Ⅳ ketamine + I/R (K);groupⅤ ketamine + ZnPPⅨ + I/R (KZ) and group Ⅵ ZnPPⅨ + I/R. Ketaminc 10 mg/kg and/or ZnPPⅨ 5 mg/kg were injected intraperitoneally 30 min before I/R in group Ⅳ, Ⅴ and Ⅵ. Intestinal I/R was produced by clamping of superior mesentery artery for 1 h followed by 6 h reperfusion. The animals were then killed and a segment of small intestine of 2 cm in length at about 10 cm from ileocecal valve was removed for determination of MDA content and SOD activity, HO-1 and iNOS expression and histological examination. The degree of intestinal tissue damage was graded using Chiu's scoring system (0 = normal, 5 = severe damage-exfoliation of villi, disruption of proper lamina, bleeding and ulceration). Results Intestinal MDA content was significantly increased, SOD activity was decreased and HO-1 and iNOS expression was significantly up-regulated by I/R (group Ⅲ) as compared with group Ⅰ and Ⅱ . Ketamine pretreatment significantly attenuated I/R-induced increase in MDA content and decrease in SOD activity and the protective effects of ketamine against I/R were counteracted by concomitant asministration of ZnPPⅨ. Microscopic examination showed that the pathological changes induced by I/R were significantly attenuated by ketamine and the protective effects of ketamine pretreatment against I/R were reversed by ZnPPⅨ in group Ⅴ. Conclusion Low-dose ketamine pretreatment can significantly ameliorate intestinal ischemia-reperfusion injury and up-regulation of intestinal HO-1 expression and down-regulation of iNOS are involved in the mechanism.

Objective To seek an effective and safe therapy for acute variceal hemorrhage of patients with Child class C cirrhosis. Methods One hundred and nine Child class C cirrhotic patients with an acute hemorrhage from esophageal varices were reviewed. Sixty - one patients received medical therapy;29 patients were underwent sur- gical operations and 19 patients received TIPSS. Results The surgical group had a higher mortality rate during hos- pitalization(65. 5 % ) than either of the other groups(36. 1 % and 26. 3 % respectively ). The rebleeding rates during hospitalization were 24. 1 %, 36. 1 % and 15. 8 % respectively in the surgical group, medical group and TIPSS group. Rebleeding rate during following- up period were 33. 0%, 36. 1 % and 21 .4% respectively. The rebleeding rate was significantly higher in the medical group than that in either of the other groups during the two phases. Kaplan - Meier survival analysis didn' t show significant difference among the three groups. Conclusions The results showed that TIPSS is a good alternative to surgical or medical therapy for patients with Child class C cirrhosis and acute vericeal bleeding.

Objective It has been unclear how osteopontin, one of metastasis-associated genes, could be regulated in HepG2 cells. The aim of this study was to investigate the effect of protein kinase B(Akt) on the expression of osteopontin in HepG2 cells, and to explore the relationship between Akt and osteopontin expression. Methods HepG2 cells were transfected with constitutively active Akt and dominant negative Akt by lipofectmine, and transfectants were confirmed using Western blot for Akt. Osteopontin expression was detected by Northern blot and Western blot. Results HepG2 cells were successfully transfected with Akt genes including constitutively active Akt and dominant negative Akt, and overexpression of exozegenes Akt could be detected in HepG2 cells by Western blot. Using Northern blot and Western blot, we found that Akt gene regulated osteopontin expression in RNA level and protein level. In serum-free condition, HepG2 cells constitutively expressed low levels of osteopontin. Transfection of constitutively active Akt increased osteopontin mRNA and protein expression. Transfection of dominant negative Akt decreased osteopontin expression. Conclusions The fact that osteopontin gene expression can be regulated by Akt indicates that osteopontin synthesis can be blocked by inactivation of Akt gene and that metastasis of liver cancer might be inhibited by this intervention.
