1.Investigations of glucocorticoid receptor mutant without ligand binding domain

Objective:To construct expression vector of human glucocorticoid receptor mutant.Methods:cDNA sequence of human glucocorticoid receptor mutant having no LBD was amplificated through RT-nested PCR,PCR product was directionally cloned to pEGFP-N2 plasmid vector,and recombinant plasmids were screened through sequencing.Expressions of recombinant plasmids in human macrophage cell line U937 were observed through fluorescence microscope.Transcriptional activation activity of variant was evaluated through methods of relative activity of luciferase.Results:The 1 601 bp length cDNA was obtained,and result of sequencing confirmed the cloned cDNA sequence is completely coincidence with that of glucocorticoid receptor in Genebank.Expression products of recombinant plasmid mainly located in nucleus.Transcriptional activation activity incresed after recombinant plasmid transfected.Conclusion:Expression vector of glucocorticoid receptor mutant was constructed successfully,which has transcriptional activation activity independent of glucocorticoid.
2.Factors Influencing the Release in Vitro of Atenolol Nasal Gel

OBJECTIVE:To investigate the factors influencing the drug release of atenolol nasal gel for references of optimization of its formulation.METHODS:The effects of carbopol 941(0.8%,1.0% and 1.2%)which used as the gel stroma,the amount of surface acting agent Tween-80(0,6% and 8%),and absorption enhancer(DM-?-CD vs.?-CD)on the in vitro release of atenolol nasal gel were investigated.RESULTS:All the above-mentioned 3 factors had marked effects on the in vitro release of atenolol nasal gel.The in vitro release of atenolol nasal gel was markedly enhanced by 0.8% carbopol 941,6% Tween-80 and 10% DM-?-CD absorption enhancer.CONCLUSION:The results in the study serve as references for the optimization of the formulation of atenolol nasal gel.

The paper researches the co-authorship situation in the field of foreign medical ethics through the social network analysis method,taking 9 foreign core journals of medical ethics included in the Journal Citation Report as the data source,it extracts and sorts out the information through BICOMB and UCINET software to generate the co-occurrence matrix of the author and draw the map,reveals the overall structural features and co-authorship situation of the co-authorship network in the field of foreign medical ethics.
4.Germicidal Efficacy and Corrosiveness of San Xiao Pyrogeny Inactivator:An Experimental Observation

OBJECTIVE To determine the germicidal efficacy and corrosiveness of San Xiao pyrogen inactivator.METHODS Its germicidal efficacy was measured with carrier test.RESULTS The killing rate of Bacillus subtilis var.niger spores exposed to its solution containing available chlorine 750mg/L for 30 min attained 100%.Its solution containing available chlorine 500 mg/L with a 15 min contact time could destroy HBsAg antigenicity in suspension.Its germicidal efficacy decreased in presence of organic substance.Its water solution containing available chlorine 1000mg/L was not corrosive to stainless steel,moderately corrosive to carbon steel and severely corrosive to copper and aluminum.CONCLUSIONS San Xiao pyrogen inactivator is a kind of original disinfectant which has better germicidal efficacy and uses safely.
5.Simultaneous Determination of Six Flavonoid Ingredients in Shuxuening Injection by Quantitative Analysis of Multi-components by Single-marker

Objective:To simultaneous determine six flavonoids in Shuxuening injection by quantitative analysis of multi-compo-nents by single marker ( QAMS) and examine the durability and system suitability of the analysis. Methods:Shuxuening injection was the study object and rutin was used as the internal material, the relative correction factor ( f) between rutin and narcissus glycosides, quercetin-3-O-β-D-glucosyl (1-2)-α-L- rhamnoside, kaempferol-3-O-β-D-glucose group (1-2)-α-L-rhamnopyranoside, quercetin-3-O-[2-O-(1-O)-p-coumaric acid-β-D-glucose-yl]-α-L-rhamnoside, kaempferol-3-O-[2-O-(6-O)-on coumaroyl-β-D-glucose-based]-α-L-rhamnosides was respectively established. The result differences between the internal standard method and external standard method were compared, and the internal standard method was re-validated. Results: The contents of the six flavonoids in Shuxuening injection were calculated by the correction factors. Compared with those of the external standard method, the differences in the contents of the method were small( RSD<5%) . Conclusion:QAMS can be used as a simple and accurate quantitative quality evaluation meth-od for the determination of a variety of ingredients in Shuxuening injection, which provides reference for the subsequent study on quality standard.

7.Aquaporin-1 and Aquaporin-9 Expressed by Reactive Astrocytes after Traumatic Brain Injury in Rat

Objective To investigate the expression changes of aquaporin-1 (AQP1) and aquaporin-9 (AQP9) in rats after traumatic brain injury, and explore the roles of AQP1 and AQP9 in the development of brain edema.Methods In an impact-acceleration head injury model of rat, brain water content was measured by wet-dry weight method at 1 h, 6 h, 24 h, 48 h and 168 h after brain trauma. The expressions of AQP1 and AQP9 in brains were investigated by immunohistochemistry and Western blot.Results Brain water content increased in the injuried brain tissues after 1 h post-injury, reached peak at 24 h and returned at 168 h. Both expressions of AQP1 and AQP9 were induced in reactive astrocytes adjacent to injury sit at 6 h and 24 h after brain trauma. Staining of AQP1 in the endothelial cells was not present in normal rat brains but appeared strong after brain trauma.Conclusion A remarkable induction expression of AQP1 or AQP9 after brain trauma may participate in the development of brain edema.
8.The effect of a program of physical therapy on function of patients with total hip replacement

To study the effect of a program of physical therapy,40 subjects with total hip replacement for femoral neck fracture were randomized into rehabilitation and control groups.The program included ROM exercises,low resistance exercises of hip abductors and ADL exercises.Muscle strength of hip abductors were measured by Kendall's Muscle Testing and Function,hip ROM,functional ability and pain were evaluated by Harris Hip score on the lst and 3rd month after surgery.Statistical analysis with t test was used for comparing Harris Hip scores.The result shows that muscle strength and hip function were improved significantly in rehabilitation group (p<0.05).That means the program is effective in long term after hip replacement.
9.Ultrastructural characterization of macular pucker

Objective To study the ultrastructure of macular pucker (MP) from the patients with rhegmatogenous retinal detachment (RRD) and the mechanism associated with its development. Methods Twenty specimens of MP surgically removed by vitrectomy from 13 patients were dissected into two layers in each of them.The ultrastructure of two layers,i,e,near the vitreous and near the retina,was studied with electron microscopy. Results Seven sections of the near vitreous ones appeared prodominant collagen deposits and a few of epithelial-like cells,and pigment particles might be present in the cytoplasm.While cells with foot processes were found in 13 membrane sections near the retina and increasing number of various types of cells rich in collagen around were observed including fibroblast-like cells and glial cells. Conclusion The findings suggest that the MP after surgery of retinal detachment may possess a characteristic lamination,and posterior hyaloid cortex was involved in the developmetn of MP.The adhesion between posterior hyaloid cortex and macular area might be a key factor for forming MP.
10.Clinical efficacy of atorvastatin in patients with lacunar infarctions and influence on cerebrovascular reactivity

Objective To investigate the clinical efficacy of atorvastatin in patients with lacunar infarctions (LI) and influence on cerebrovascular reactivity(CVR). Methods 60 patients with LI were randomly divided into observation group and control group. The patients of control group were given aspirin treatment, patients of observation group were given atorvastatin combination with aspirin therapy. The breath-holding index(BHI) before and after treatment was calculated. After continuous treatment for 3 months the clinical efficacy was evaluated. Results The total effective rate of the observation group was 90. 0% ,it was significantly higher than that of the control group(73.3% ),compared with the control group the difference was significant( x2 = 10. 428 ,P < 0. 05). The bilateral BHI of observation group significantly increased, compared with per-treatment and control group the difference was significant ( t =9. 455,6. 135 ,all P < 0. 05 ) ,but the bilateral BHI of control group had no significant difference compared with pertreatment ( P > 0. 05 ). Conclusion Atorvastatin could effectively improve CVR in patients with LI. Therefore, atorvastatin had great significance for the prevention of stroke.
