1.Investigations of glucocorticoid receptor mutant without ligand binding domain

Objective:To construct expression vector of human glucocorticoid receptor mutant.Methods:cDNA sequence of human glucocorticoid receptor mutant having no LBD was amplificated through RT-nested PCR,PCR product was directionally cloned to pEGFP-N2 plasmid vector,and recombinant plasmids were screened through sequencing.Expressions of recombinant plasmids in human macrophage cell line U937 were observed through fluorescence microscope.Transcriptional activation activity of variant was evaluated through methods of relative activity of luciferase.Results:The 1 601 bp length cDNA was obtained,and result of sequencing confirmed the cloned cDNA sequence is completely coincidence with that of glucocorticoid receptor in Genebank.Expression products of recombinant plasmid mainly located in nucleus.Transcriptional activation activity incresed after recombinant plasmid transfected.Conclusion:Expression vector of glucocorticoid receptor mutant was constructed successfully,which has transcriptional activation activity independent of glucocorticoid.

The paper researches the co-authorship situation in the field of foreign medical ethics through the social network analysis method,taking 9 foreign core journals of medical ethics included in the Journal Citation Report as the data source,it extracts and sorts out the information through BICOMB and UCINET software to generate the co-occurrence matrix of the author and draw the map,reveals the overall structural features and co-authorship situation of the co-authorship network in the field of foreign medical ethics.
3.Factors Influencing the Release in Vitro of Atenolol Nasal Gel

OBJECTIVE:To investigate the factors influencing the drug release of atenolol nasal gel for references of optimization of its formulation.METHODS:The effects of carbopol 941(0.8%,1.0% and 1.2%)which used as the gel stroma,the amount of surface acting agent Tween-80(0,6% and 8%),and absorption enhancer(DM-?-CD vs.?-CD)on the in vitro release of atenolol nasal gel were investigated.RESULTS:All the above-mentioned 3 factors had marked effects on the in vitro release of atenolol nasal gel.The in vitro release of atenolol nasal gel was markedly enhanced by 0.8% carbopol 941,6% Tween-80 and 10% DM-?-CD absorption enhancer.CONCLUSION:The results in the study serve as references for the optimization of the formulation of atenolol nasal gel.
4.Germicidal Efficacy and Corrosiveness of San Xiao Pyrogeny Inactivator:An Experimental Observation

OBJECTIVE To determine the germicidal efficacy and corrosiveness of San Xiao pyrogen inactivator.METHODS Its germicidal efficacy was measured with carrier test.RESULTS The killing rate of Bacillus subtilis var.niger spores exposed to its solution containing available chlorine 750mg/L for 30 min attained 100%.Its solution containing available chlorine 500 mg/L with a 15 min contact time could destroy HBsAg antigenicity in suspension.Its germicidal efficacy decreased in presence of organic substance.Its water solution containing available chlorine 1000mg/L was not corrosive to stainless steel,moderately corrosive to carbon steel and severely corrosive to copper and aluminum.CONCLUSIONS San Xiao pyrogen inactivator is a kind of original disinfectant which has better germicidal efficacy and uses safely.
5.Total knee arthroplasty-associated factors in 39 cases

Data of 39 patients (51 knees) undergoing total knee arthroplasty (TKA) in the Department of Orthopaedics,Fourth Affiliated Hospital of Guangxi Medical University between October 2001 and March 2008,were analyzed retrospectively,including 6 males (6 knees) and 33 females (45 knees) aged 59-72 years. Among the 39 cases,there were 35 cases of osteoarthritis,2 of rheumatoid arthritis,1 of osteoarthritis and pigmented villonodular synovitis,and 1 of advanced tuberculosis. Different degrees of pain,swelling,genu valgum,genu varumand and functional impairment of knee joints were found in all cases. The prosthesis adopted included 3 knees of DePuy,10 of Stryker,3 of Link,5 of Zimmer,7 of Taiwan Unite,and 23 of Pulus (made in China). Among all the cases,47 knees were treated by posterior stabilized prosthesis,and 4 knees were treated with posterior cruciate ligament-retaining TKA. Careful medical examination and evaluation were performed to control blood sugar level before operation; the operation duration was shortened as possible. Reasonable proposals were formulated to prevent the postoperative infection and deep venous thrombosis. Clinical data were analyzed using the HSS score standard. All patients were followed up for 6-57 months. No deep infection,prosthetic loosening or death was found. Postoperatively,2 patients (2 knees) developed cutaneous necrosis at incision margin,of which 1 patient (1 knee) was cured by changing dressings,and 1 patient (1 knee) was healed after incision of skin and relaxation suture. The range-of-motion of 51 knees all exceeded 90?. The motion of knee joint were increased from 65?(32?-95?) in average preoperatively to 96?(90?-110?) after the operation. The preoperative pain of all patients disappeared instantly after operation,beside one case (2 knees) and the other had a little pain. The Harris knee joint scores were increased from 39 points (25-53 points) in average preoperatively to 91 points (82-97 points) after the operation at the final follow-up; 25 cases (34 knees) were excellent,12 cases (15 knees) were good,2 cases (2 knees) were fair and the excellent and good rate was 96%. Although TKA has a satisfactory surgical effect,associated factors should be paid more attention.
6.Effect of LEEP knife combined with biopsy in patients with small invasive cervical cancer

Objective To explore the effect of the application of cervical loop electric knife (LEEP) in the patients with cervical micro invasive cancer.Methods Clinical data of 66 patients with minimal invasive carcinoma diagnosed by LEEP cone resection in our hospital from June 2013 to June were retrospectively analyzed. Preoperative and postoperative patients with LEEP were compared with 66 patients who underwent biopsy with biopsy, the diagnosis of micro invasive carcinoma was compared with LEEP cone resection.Results The results of colposcopy biopsy were detected in 2 cases of LSIL, 60 cases of HSIL and 1 cases of AIS, 3 cases were diagnosed as cervical microinvasive carcinoma or suspicious cervical microinvasive carcinoma, the sensitivity was 4.54%, the misdiagnosis rate was 95.46%; LEEP conization were detected in 1 cases of LSIL, 44 cases of HSIL and 1 cases of AIS, 20 cases negative margin. There was no significant difference in the thickness and area of the resected tissue between the three groups (P > 0.05). The depth of the resected tissue was significantly higher than that in the HSIL and AIS group, the HSIL and AIS groups were significantly higher than the LSIL group, the difference was statistically significant (P < 0.05). Interstitial infiltration depth is less than or equal to 1mm group, the 1 depth of stromal invasion than 3mm group and 3 < rate were 15.00%, 19.51% and 20.00% of the remaining lesion depth of stromal invasion than the patients in the 5mm group after operation, there was no statistically significant difference (P > 0.05). Cut edge positive group, positive margin group and interstitial fiber positive margin group of patients with postoperative residual disease rates were 25.00%, 15.38% and 23.80%, the difference was not statistically significant (P > 0.05).Conclusion The rate of missed diagnosis of small invasive cervical cancer is higher, and the diagnosis rate can be improved by the combination of LEEP and cone resection.
7.Effect of Ruanjian Xiaoying Decoction on the Relative Expression of Cytokines and Apoptosis-regulated Proteins Fas/Fasl in Experimental Autoimmune Thyroiditis Rats

Objective To evaluate the therapeutic effect of Xiaoying Ruanjian Decoction on experimental autoimmune thyroiditis (EAT) from the levels of autoantibodies, interleukin-1, tumor necrosis factor-? and apoptosis-regulated proteins Fas/Fasl of EAT rats, and investigate the mechanism from the relation between cytokines and apoptosis-regulated proteins. Methods Sixty SD female rats of SPF level were divided into normal group and model group. An animal model of EAT in rats was developed by injecting thyroglobulin mixed with Freund’s adjuvant immunization and drinking periodate water. The rats modeled were divided into Chinese medicine group, western medicine group and model group. The levels of autoantibodies, interleukin-1, tumor necrosis factor-? were detected with enzyme-linked immunosorbent assay. The level change of Fas/Fasl in the thyroid of rat was observed with immunohistochemical staining. Results Ruanjian Xiaoying Decoction significantly reduced the levels of autoantibody, interleukin-1, tumor necrosis factor-? and lowered the expression of Fas and Fasl in thyroid cells. Conclusion Ruanjian Xiaoying Decoction alleviates the levels of apoptosis-regulated proteins Fas/Fasl by decreasing the relative expression of antibodies and cytokines in serum levels, which plays a role in reducing apoptosis of thyroid cells.

9.Dedifferentiated liposarcoma in proximal tibia accompanied by angiosarcoma: One case report

One case with dedifferentiated liposarcoma in proximal tibia accompanied by angiosarcoma was analyzed. The dedifferentiated liposarcoma was further analyzed by imaging, clinical and pathological features. One 51-year-old male patient was suspected to bone tumor due to space occupying lesion for 6 days in left proximal tibia and admitted in Changhai Hospital of Second Military Medical University of Chinese PLA in May 2007. The clinical features, including onset features, physical examination, blood routine, imaging alterations, pathological characteristics, operation approach, and follow-up period were retrospectively analyzed. The tumor was treated by curettage and inactivation, iliac grafting, bone cement filling, tibial plateau T type plate and screw internal fixation and tumor tissue biopsy. However, tumor relapsed 40 days postoperatively, and the patient was treated by amputation surgery. Two weeks after surgery, the patient developed emptysis and died due to multiple organ failure. Dedifferentiated liposarcoma accompanied by angiosarcoma displays no noticeable characteristics in imaging, clinical manifestation, and laboratory examination, which make it difficult to diagnose it. This tumor is highly malignant, and has short disease course, and poor prognosis.
10.INVESTIGATION OF HEALTH CONDITIONS OF THE MEDICAL STAFFS IN A GENERAL HOSPITAL

Objective To investigate the health status of the medical staffs in our hospital , find risk factors and make corresponding strategies to improve health of status .Methods Results of physical examination on 1203 medical staffs were analyzed by SAS 9.2.Results Total residual test ratio is 9.3%, and the residual test ratio of male was significantly higher than female (χ2=38.399,p <.0001).The prevalence ratio of all subject is 60.9%, and there was no significant difference between male and female , but the higher age category, the higher prevalence ratio (χ2=211.4037,p <.0001).The top 5 diseases our staffs suffering are waist and cervical vertebra disease , thyroid nodule, hyperlipidemia, breast hyperplasia and fatty liver, in which the prevalence ratio of thyroid nodule and hyperlipidemia was different between male and female .Conclusion The health status of our medical staffs should be improved by paying high attention to medical examination and health management .
