1.Levels of plasma VEGF,TPS and TuM2-PK before and after interventional therapy in lung cancer

Objective To explore the expression and significance of vascular endothelial growth factor (VEGF),tissue polypeptide specificity antigen (TPS) and tumor type M2 pyruvate kinase (TuM2-PK) before and after interventional therapy in lung cancer (LC).Methods Sixty cases of LC treated by interventional treatment in our hospital from January 2013 to January 2015 were analyzed.The levels of VEGF,TPS and TuM2-PK on preoperative 1 d,postoperative 1,3,7,14,28 d were detected by enzyme-linked immunosorbent assay(ELISA).The change of VEGF,TPS and TuM2-PK levels in the patients with different treatment outcomes was observed.Contemporaneous 40 individuals undergoing healthy physical examination served as the healthy control group.Results The levels of VEGF,TPS,TuM2-PK on preoperative 1d,postoperative 3,7,14,28 d had statistical differences between the observation group and control group (P<0.05,P<0.01).Compared with pre-treatment,the levels of VEGF,TPS and TuM2-PK in partial remission patients were significantly improved,which in the patients with stable and progressive condition,the levels of VEGF,TPS and TuM2-PK were significantly increased (P<0.05).Conclusion The levels of VEGF,TPS and TuM2-PK are significantly elevated before and after interventional treatment in LC patients,moreover the worse the therapeutic efficacy,the higher the levels of VEGF,TPS and TuM2-PK,which can be used as the detection indexes of LC interventional treatment.
2.Prokaryotic expression and purification of human renin and mature renin

Objective To express and purify the human renin(REN)and mature renin(REN-340)in prokaryotic cells,and detect the concentration. Methods According to the human REN gene sequence(5972)registered in NCBI,REN and REN-340 gene fragments were artificially synthesized and cloned into vector p ET-28a(+)after E.coli codon optimization,and the recombinant expression plasmids 6 × His-REN-p ET and 6 × His-REN-340-p ET were constructed. The recombinant plasmids were transformed into competent E.coli BL21(DE3)and induced at different induction temperatures(20,25,30and 37 ℃)and different final concentrations(0. 1,0. 2,0. 4 and 0. 5 mmol/L)of IPTG. Recombinant REN and REN-340were induced under the optimal induction conditions,and the concentrations were detected by ELISA after nickel ion affinity chromatography with Ni-NTA. Results The recombinant expression plasmids 6 × His-REN-p ET and 6 × His-REN-340-p ET were constructed correctly as identified by restriction analysis and sequencing. The optimum induction temperature of recombinant REN and REN-340 was 37 ℃ and 20 ℃,and the best final concentration of IPTG was 0. 1 and 0. 2 mmol/L,respectively. The concentrations of purified recombinant REN and REN-340 were 9 148. 21 and 15 115. 31 m IU/m L,respectively. Conclusion The prepared recombinant REN and REN-340 proteins have high concentration with short production cycle,which are expected to be used as candidate quality control products and standard substances for external quality assessment(EQA).

The surgical intervention and criteria of severe acute pancreatitis has significantly changed in the last several years with the advent of enhanced imaging techniques and minimally invasive surgery.Many kinds of minimally invasive technologies which take laparoscopy as a representative are constantly applied to the treatment of severe acute pancreatitis and make its mortality and postoperative complication rate decrease obviously.However,there are many different ways using laparoscopic techniques to treat severe acute pancreatitis.In this review,the author will discuss the current status of the different approaches.

6.Observations on the Treatment of 32 Cases of Insomnia by Manipulations of Digital-pressing Acupoints and Tuina along Meridians

Five kinds of manipulations of plucking, circular rubbing, pecking, pinching and patting were performed on the channels of Governor Vessel and bilateral Bladder Meridian. According to syndrome differentiation, all the 32 cases were divided into 5 syndromes of the stagnated liver-qi causing fire syndrome, the phlegm-heat attacking upper parts syndrome, the hyperactivity of fire due to yindeficiency syndrome, the deficiency of both heart and spleen syndrome and the timidity due to insufficiency of qi and deficiency of blood of the heart syndrome and were then treated by different manipulations. After 2 courses of treatment, 25 cases were cured, 7 cases got marked effectiveness,no case failed and the effective rate was 100%.

At the end of last century,the snlall-molecule selective kinase inhibitor,imatinib mesylate (IM),Was successfully exploited.This resulted in a revolutionary step in the treatment of chronic myeloid leukemia(CML).However,along with the expansion of its clinical application and the process,drug resistance Was commonly observed in patients, especially during accelerated or blast phases, and has become a significant therapeutic problem in clinical application.The problem has prompted the appearance of novel small molecule tyrosine kinase inhibitors,and its development is reviewed.
8.Ultrasonic Detection of Mesenteric Lymph Nodes in Children with Intermittent Abdominal Pain

Objective To examine the clinical application of ultrasonography to detection of mesenteric lymph nodes in chil‐dren with intermittent abdominal pain.Methods A total of 196 children who underwent abdominal ultrasonography for differ‐ent reasons were divided into the intermittent abdominal pain group and non‐abdominal pain group.The location ,size and num‐ber of mesenteric lymph nodes were recorded.Results Statistical difference in the long‐axis diameter(P=0.005)and ratio of short‐to‐long‐axis diameter was found among patients with different ages in non‐abdominal pain group(P= 0.015) ,while no significant difference was seen in short‐axis diameter(P=0.773).No significant difference was observed in the diameter of each axis between different genders in non‐abdominal pain group.There was a statistical difference between abdominal pain group and non‐abdominal pain group in the incidence of lymph nodes with short‐axis diameter of 6 mm and larger(P=0.002)and long‐axis diameter of 14 mm and larger(P=0.007).Conclusion Mesenteric lymph node with short‐axis diameter larger than 6 mm should be considered enlarged in children ,but should not be diagnosed with mesenteric lymphadenitis.It’s common to find en‐larged mesenteric lymph nodes in children without abdominal pain.Further investigations with a larger number of patients are required to confirm these findings .
9.Sensitivity of gastric cancer stem cells to 5-fluorouracil

BACKGROUND:5-Fluorouracil is a common chemotherapy drug for gastric cancer, but it is more likely to develop drug resistance in clinical treatment. Studies have shown that tumor stem cels are lowly sensitive to chemotherapeutic drugs, which may be an important cause of chemotherapy resistance. OBJECTIVE:To analyze the sensitivity of gastric cancer stem cels to 5-fluorouracil in vitro, and to understand the mechanism of drug resistance associated with gastric cancer. METHODS:Based on the sorting strategy, human gastric cancer cel clones were isolated from AGS cel lines. CD44 and thymidylate synthetase expression in different clones was detected using immunocytochemistry analysis. Clone formation assay was used to evaluate self-renewal capacity of different clones. Cel counting kit-8 was used to determine the clonal growth inhibition rate of AGS under treatment with different concentrations of 5-fluorouracil. RESULTS AND CONCLUSION:The AGS cels that were inoculated with low density and cultured could differentiate into 32 forms, including the ful clone (16%, 5/32), the second clone (66%, 21/32) and the accessory clone (19%, 6/32). Among them, the ful clones highly expressed CD44 and thymidylate synthetase, and could generate a great amount of passage 2 clones after inoculation; the second clones showed a weak expression of CD44 and thymidylate synthetase, and could generate a smal number of passage 2 clones after inoculation; the accessory clones showed a weak or no expression of CD44 and thymidylate synthetase, and there was no passage 2 clone after inoculation. Under the effect of different concentrations of 5-fluorouracil, the growth inhibition rates of the secondary clones and AGS cels were both higher than that of the ful clones (bothP < 0.05). These findings indicate that gastric cancer stem cels have a relatively lower sensitivity to 5-fluorouracilin vitro, which is speculated to be an important mechanism of drug resistance.

Objective To explore the application of RNA interference(RNAi) in colorectal cancer gene therapy.Methods The related literatures in recent years were reviewed.Results RNAi causes a high effective and distinctive degradation of mRNA homologous in sequence to the dsRNA.This new technology has been successfully applied to research the genesis and the growth of colorectal cancer.Conclusion RNAi has been a new focus in gene therapy for colorectal cancer.
