Objective To detect and localize the expression of macrophage colony stimulating factor (M CSF) in human luteinized granulosa cells and investigate the effects of follicle stimulating hormone (FSH) on M CSF production by human luteinized granulosa cells in vitro Methods Human luteinized granulosa cells were isolated from follicular fluid of superovulated infertile patients undergoing intracytoplasmatic sperm injection Some of the luteinized granulosa cells were used for detecting M CSF by immunocytochemical staining (ABC method) Most of them were cultured with HAM′s F 10 medium alone or plus various concentrations of FSH (2, 5, 15, 25 IU/ml) The media were collected after 72 hours and their M CSF contents were measuered by a solid phase enzyme linked immunosorbant assay Results About 80% of the luteinized granulosa cells were positively stained for M CSF antibody The immunoreactive signals of M CSF were specially located in the cytoplasma of the luteinized granulosa cells No immunoreactivity of M CSF was observed in vaginal squamous epithelial cells through contaminations during transvaginal oocyte retrieval The concentration of M CSF in cultured luteinized granulosa cells medium without FSH stimulation was low However, addition of 2, 5, 15, 25 IU/ml concentrations of FSH caused significant dose dependent increases of M CSF production after 72 hours culture by luteinized granulosa cells ( P
2.Advance in Research on Brain Protection Mechanisms of Fastigial Nucleus Stimulation (review)

The research on fastigial nucleus could be traced back to 19th century. However the great achievements on cerebrum overshadowed the importance of cerebellum and its nucleus. In recent 30 years, with the finding of brain protection effect of fastigial nucleus stimulation, much attention has been given to this field. Now its widely accepted that electrical stimulation of fastigial nucleus has brain protection effect and concrete mechanisms can be concluded as: improving the electrical instability around the infarct zone, reducing peri-infarction depolarizing wave and cortical spreading depression; suppressing microvessels inflammatory reaction; inhibiting cell apoptosis; reducing the neurons damage and facilitates neovascularization and so on. These new discoveries was reviewed and further expectation on this field was put forward by the authors.
3.Mental Health and Achievement Motivation of Undergraduates

Objective: To study the relationship between mental health and achievement motivation of undergraduates. Method: 532 undergraduates completed SCL-90 and Academic Achievement Motive Scale. Results:17.29% subjects had mental problems. Undergraduates had higher score in self-oriented motive than society-oriented motive, while the score of SCL-90 correlated positively with score in society-oriented motive. In students with poor academic achievement, the score of SCL-90 negatively correlated with score in self-oriented motive. Conclusion:Society-oriented motive had negative influence on mental health of undergraduates and in those with poor academic achievement, poor mental health positively correlated with lower self-oriented motive.
4.Fast-track surgcry for cirrhotic patients with portal hypertension: a comparative study

ObjectiveTo study the effect of a fast-track surgery (FTS) program on the clinical outcomes in cirrhotic patients with portal hypertension.Methods42 cirrhotic patients with portal hypertension were randomly allocated into the FTS group (n=21) and the conventional therapy control group (n=21).The postoperative time to first defecation,hospital stay,hospitalization cost,and postoperative complications were compared between the two groups.ResultsCompared with the control group,the postoperative time to first defecation was significantly shorter in the FTS group (P=0.0287).Furthermore,the postoperative hospital stay was significantly shorter in the FTS group than the control group (P=0.002).Additionally,hospitalization cost was significantly lower in the FTS group than the control group (P<0.001).The postoperative complication was also significantly different between the two groups (7.15 % vs 21.5 %,P=0.001).ConclusionA FTS program contributed to better postoperative rehabilitation in cirrhotic patients with portal hypertension.
5.Analysis on HLA B27 results detected by real-time fluorescence PCR method and flow cytometry

Objective To compare the clinical application value of the real-time fluorescence PCR method and flow cytometry method for detecting human leukocyte antigen B27(HLA-B27).Methods Blood HLA-B27 level in 225 patients with suspected ankylosing spondylitis was detected by using real-time fluorescence PCR method and flow cytometry method.The detection results were compared and analyzed between the two methods.Results The results of 95.1% sample were identical detected by two methods without statistically significant difference(P>0.05).Taken the results of flow cytometry as reference, the sensitivity of real-time fluorescence PCR for detecting HLA-B27 was 94%, the specificity was 96%.Gene sequencing was performed if results of a sample detected by two methods were different, which was identical with the result detected by real-time fluorescence PCR.Conclusion Both methods for detecting HLA-B27 all have high sensitivity and specificity.Real-time fluorescence PCR method is more superior to the flow cytometry method in the results accuracy.
6.Establishment of MALDI TOF-MS Technique Platform for Detecting Cytochrome P4 5 0 Gene Polymorphism

Objective To establish the MALDI-TOF-MS technique platform for detecting Cytochrome P450 gene polymor-phismfrom of patients.Methods Collected 53 EDTA anticoagulation peripheral blood samples from October 2013 to June 2014 in Peking Union Medical College Hospital.The whole genomic DNA was extracted from patients’peripheral blood. Used MALDI-TOF-MS to identify the SNP polymorphism of CYP2C9*2(rs1799853),CYP2C9*3(rs1057910),CYP2C19*2(rs4244285),CYP2C19*3(rs4986893),CYP2C19*4(rs28399504),CYP2C19*5(rs56337013)and CYP2C19*17 (rs12248560).To verify the above SNP polymorphism by Sanger sequencing method.Results The MALDI-TOF-MS could perform 53 samples on two cytochrome gene and 7 SNP locus simultaneously.In all the 53 patients,25 AG,6 AA and 22 GG genotypes were identified in CYP2C19*2(rs4244285),the allele frequency of A genotype was 34.9%.4 AG and 49 GG genotypes were identified in CYP2C19*3 (rs4986893),the allele frequency of A genotype was 3.8%.5 CA and 48 AA gen-otypes were identified in CYP2C9*3 (rs1057910),the allele frequency of C genotype was 4.7%.No mutation loci were i-dentified in CYP2C9*2 (rs1799853),CYP2C19*4 (rs28399504),CYP2C19*5 (rs56337013)and CYP2C19*17 (rs12248560).All the identification data were confirmed by Sanger sequencing.The coincidence rate was 100%.Conclusion The MALDI-TOF-MS technique platform for the cytochrome enzyme SNP was established.This platform has high throughput and accurate characteristics.It has important clinical application value for the treatment of personalized medicate.
7.Study on relationship between the beginning plan and the anatomy and dosimetry change during intensity modulated radiotherapy for esophageal carcinoma

Objective To study the dynamic changes of anatomy and the dosimeter distribution those changes influenced. Methods Initially simplified intensity modulated radiation therapy (sIMRT)were performed to twenty-nine patients with phase Ⅲ - Ⅳa esophageal carcinoma from January 2007 to March 2009. The target volumes and involving organs were contoured on the primary spiral CT pictures.After sIMRT planning being finished, secondary CT scan was acquired to rectify the treatment center. For eleven patients at every other week and eighteen patients at the fourth week, spiral CT images were acquired according to the same treatment center, and thereafter fused with the first CT images. Firstly, the law of change and the best time of replanning were searched:the changed gross tumor volume (GTV), gross node volume (GTVnd), plan target volume (PTV) and normal organs (lung, spinal cord, heart and outline) on the fusion interface were modified by a single physician, the changes for each structure throughout treatment were measured by system software. Secondly, dose distributions were computed and evaluated for replanning CT using the same beams arrangement as the initial plan. Cumulative dose was estimated using weighted average and compared with the original plan. Results For eleven patients, The law of change:the volume of outlines and GTV gradually decreased, and the change come to peak in the fourth week. The conformal index for PTV gradually decreased, whereas the heterogeneous index gradually increased. For twenty-nine patients on the fourth week, the dose of GTV were more than 60 Gy. The dose of PTV-D95 and CTV-D99 decreased ( t = 1.49, P = 0. 147 and t = 2. 07, P = 0. 048 respectively). The dose of CTV-D99 in two patients deceased to 54 Gy or less. The cord-Dmax and lung V30 increased significantly ( t = - 2. 42, P = 0. 022 and t = -2. 26,P =0. 032). Conclusions During the course of sIMRT for esophageal cancer, the volume of GTV decreased and the change come to peak in the fourth week. It is the best time for evaluating the change of dose of target volume using CT-CT fusion. For some patients, revise of the treatment plan is needed to ensure adequate target volume dosage and safety of normal tissues.
8.Effects of As2 O3 in combination with cinobufacini on proliferation and apoptosis of the K562 cells

Objective To investigate the effect of As2 O3 in combination with cinobufacini on proliferation and apoptosis of the K562 cells and provide theoretical basis for clinical application.Methods Cell proliferation was assayed by analyzing the growth and viability of the cells.Apoptosis was assayed by performing cell morphology,Annexin-V/PI staining,DNA-PI staining,and DNA gel electrophoresis.Results After exposure to As2O3 and cinobufacini,the growth of K562 cells was inhibited and the viability of K562 cells was decreased. After treated with 1.0μmol/L As2O3,0.125μg/ml cinobufacini,0.25μg/ml cinobufacini,1.0μmol/L As2O3 + 0.125 μg/ml cinobufacini,1.0μmol/L As2O3 + 0.25μg/ml cinobufacini for 24 and 48 hours,the proliferation inhibition rate were(24 ± 1.3)%,(21 ± 1.5)%,(38 ± 3.1)%,(57 ±2.7)%,(66 ±3.3)% and(49 ±2.9)%,(48 ±2.7)%,(61 ±2.1)%,(77 ±3.8)%,(82 ±4.2)%,the apoptosis rate of K562 cells were(4.8 ± 0.5)%,(5.6 ± 0.7)%,(9.8 ± 0.6)%,(11.9 ± 1.2)%,(15.2±1.5)% and(11.0 ±0.9)%,(12.9 ±1.1)%,(18.4 ±1.5)%,(21.0 ±2.0)%,(28.0 ±1.9)%.The percentage of apoptotic cells was a time- and dose-dependent manner.Typical DNA ladder was shown by DNA gel electrophoresis.Conclusions As2O3 combined with cinobufacini inhibited the proliferation of K562 cells and induced apoptosis of the K562 cells,the combination of the two drugs had better effect.
9.The change of T helper 17 cells in the peripheral blood monouclear cells in patients with liver cystic echinococcosis

Objective To investigate the change of T helper 17 cells (Th17) in the peripheral blood mononuclear cell (PBMC) in patients with liver cystic echinococcosis. Methods Fifty-six subjects were divided into three groups: healthy controls (HD, n = 20), patients with cystic echinococcosis (CE, n= 18) and patients with cystic echinococcosis combined with bile fistula (BF,n= 18). The frequency of Th17 cells in CD4+ T lymphocytes was detected by flow cytometry. Th17-related cytokines including interleukin (IL)-17 and IL-23 were measured by enzyme-linked immunosorbent assay (ELISA). The data were analyzed by t test and Pearson correlation analysis.Results The frequency of Th17 in the peripheral blood was significantly lower in CE group compared to BF group and HD group [(0. 23±0. 11)% vs (0. 76±0.43)% vs (0.52±0.50)%; t=2. 225 and4. 077 respectively, both P<0.05), while there was no statistical difference between BF group and HD group (t=1. 931, P>0.05). The levels IL-17 and IL-23 were (12.1±3.7) ng/L and (84.4±46.0) ng/L respectively in CE group, which were lower than those in BF group [(15.5±4.1) ng/L and (138.6±37. 9) ng/L, respectively; t=2. 515 and 3. 649 respectively, both P<0.05] and those in HD group [(14.8±4.4) ng/L and (138.1±48. 7) ng/L, respectively; t=2. 401 and 3. 706 respectively,both P <0.05], while there was no statistical difference between BF group and HD group (t=0. 534,P >0.05). Serum concentrations of IL-17 were all positively correlated with the concentrations of IL23 in these three groups (r=0. 657, P<0.05). Conclusion The frequeny of Th17 cells in PBMC and the serum concentrations of IL-17, IL-23 are significantly reduced in patients with cystic echinococcosis.

The role of serum and glucocorticoid-induced kinase 1 (SGK1) pathway in the connective tissue growth factor (CTGF) expression was investigated in cultured human mesangial cells (HMCs) under high glucose. By using RT-PCR and Western blot, the effect of SGK1 on the CTGF expression in HMCs under high glucose was examined. Overexpression of active SGK1 in HMCs transfected with pIRES2-EGFP-S422D hSGK1 (SD) could increase the expression of phosphorylated SGK1 and CTGF as compared with HMCs groups transfected with pIRES2-EGFP (FP) under high glucose or normal glucose. Overexpression of inactive SGK1 in HMCs transfected with pIRES2-EGFP-K127N hSGK1 (KN) could decrease phosphorylated SGK1 and CTGF expression as compared with HMCs groups transfected with FP under high glucose. In conclusion, these results suggest that high glucose-induced CTGF expression is mediated through the active SGK1 in HMCs.
