1.Protective Effect of Nimesulide on Experimental Gastric Ulcer in Rats

OBJECTIVE: To study the protective effects of nimesulide, a COX- 2 selective inhibitor on experimental gastric ulcer in rats. METHODS: The gastric ulcer model was established in rats by intragastric administration them with indometacin, 5 min later, which were assigned to receive nimesulide at different dosages ( 200, 100, and 50mg? kg- 1) , 6h later, all the rats were sacrificed. The primary outcome measures were the area of ulcer, levels of SOD, MDA, and GSH in sera and gastric mucosa. RESULTS: As compared with model group, in nimesulide ( at different dosage) groups, the area of ulcer was markedly decreased, level of MDA was decreased while activities of SOD and GSH were increased ( P

6.The clinical study on western combined with traditional Chinese medicine in treatment of lumbar disc herniation

Objective To investigate the curative effect of western combined with traditional Chinese medicine in treatment of acute lumbar disc herniation.Methods 200 patients with lumbar disc herniation were randomly divided into western combined with traditional Chinese therapy group(group A) and western medicine group(group B) ,each group 100 cases.B group was conducted with the general anti - inflammatory, dehydration, nerve pain and nutritional therapy,and A group was given the traditional Chinese medicine treatment on the base of B group.After 3 weeks treatment, the curative effect, VAS score and hospital stay were observed.Results A total effective rate 90.0% in group A was higher than B group 73.0% ( x2 = 3.987 ,P < 0.05); The scores of VAS after treatment 4 weeks[(1.1 ±1.23、0.9 ±0.6)/point]were lower than before treatment[(8.6 ± 1.43,8.8 ± 1.382)/point]in two groups(t =2.301,2.335,all P<0.05) ;The scores of VAS after treatment 3d and 1 week in group A[(5.5 ±0.92) point,(2.7±1.14) point] were lower than group B[(8.2 ±0.59) point,(5.3 ±0.91) point] (t =2.405,2.435,all P<0.05);The hospital days(20.1 ±3.5)d in group A was significantly lower than group B(27.6 ±4.3)d( t = 2.314,P <0.05).Conclusion Western combined with traditional Chinese medicine had better clinical efficacy in teatrent of patients with acute lumbar disc hemiation.
7.Determination of 4′-O-?-D-glycosyl-5-O-methylvisamminol in Jingfuzhiyang Granules by RP HPLC

AIM: To establish a RP HPLC method for the determination of 4′ O ? D glycosyl 5 O methylvisamminol in Jingfuzhiyang Granules (Herba Schizonepetae, Fructus kochiae, Radix Saposhnikoviae, Fructus Crataegi, etc.). METHODS: The sample was separated on Turnner Kromasil C 18 column (4.6mm?200mm,5?m) with mobile phase of methanol 0.05mol?L -1 potassium dihydrogen phosphate acetic acid isopropyl alcohol (85∶170∶4∶4,v/v). The detection wavelength was set at 254nm and the flow rate was 1.0mL?min -1 . The column temperature was 30℃. RESULTS: The calibration curve was linear in the range of 0.0606~0.606?g (r=0.9994). The average recovery was 92.2%. CONCLUSION: This method proves to be relatively simple and accurate, and available for the quality control of Jingfuzhiyang Granules

10.Expression of p53, p21, PCNA and COX-2 and its relationship with recurrence in the early-stage laryngeal cancer with negative surgical margin.

OBJECTIVE:
To explore the relation between the expression of p53, p21, PCNA and COX-2 and local recurrence in resection margins of laryngeal carcinoma operation.
METHOD:
SElect 98 patients with laryngeal carcinoma, who came to our hospital from Nov, 2005 to Dec, 2010. Diagnosed with early laryngeal squamous cell carcinoma by a pathological examination, all these patients received CO2 laser surgery to cut the entire tumors. Keep the primary lesion and resection margin tissues after that and take 5 mm peritumoral tissue as a sample of operation resection margin. With the chemical method of SP, record in detail the local recurrence condition by clinical diagnosis every 3-6 months or telephone follow-up for 2 years.
RESULT:
(1)The positive rate of p53, p21, PCNA and COX-2 in the tumor tissues of the 98 patients with laryngeal carcinoma are 65. 3%, 52. 0%, 70. 4% and 69. 4%. The positive rate of p53, p21, PCNA and COX-2 in their resection margin tissues are 23. 5%, 39. 8%, 32. 7% and 30. 6%. So there is no relation between the expression of p53, p21, PCNA and COX-2 in tumor tissue and tumorous grading and TNM staging. (2)There appeared 17 cases of local recurrence in two years, with a recurrence rate of 17. 3%. The recurrence rate of patients with a positive expression of p53, p21, PCNA and COX-2 in resection margin tissue is higher than those with a negative expression(P<0. 05). Among positive cases, the recurrence rate of patients with double positive results is obviously higher than those with single positive results(P< 0. 05). (3)The expression of p21 in p53 positive resection margin tissue is greatly higher than that in negative tissue. The expression of PCNA in p53 positive resection margin tissue is greatly higher than that in negative tissue (P<0. 01); however, the PCNA positive expression in p53 positive cut edge organization rather than in the expression of negative cut edge groups(P>0. 05).
CONCLUSION
Through the discussion on the effect of p53, p21, PC- NA and COX-2 in the process of laryngeal carcinoma cell proliferation and local recurrence, the study proposes that biochemical metabolism and molecular structure level abnormal expression occur before the change of cell morphology apparent, and suggests that positive index should be examined regularly and effective foreseeability intervention can be applied to patients with positive expression.
