1.Expressions and clinical significances of IL-10 and IL-18 levels in patients with cervical intraepithelial neoplasias and cervical carcinoma

Objective To explore the expressions and.significancse of interleukin (IL)-10 and IL-18 in serum and tissues of patients with cervical intraepithelial neoplasias (CIN) and cervical cancer,and to analyze their relationships with clinical pathological characteristics and prognosis.Methods One hundred and eight patients' tissues and clinical blood samples (68 cases of CIN and 40 cases of squamous cell carcinoma) were enrolled in this study,including 25 with low-grade cervical intraepithelial neoplasias (CIN Ⅰ),43 with high-grade cervical intraepithelial neoplasias (CIN Ⅱ-Ⅲ),and 40 with invasive cervical squamous cell carcinoma (SCC);meanwhile,20 cases normal cervical tissues were enrolled.The expressions of IL-10 and IL-18 in cervical tissues and serum of the studied patients were measured by enzyme-linked immunosorbent assay and polymerase chain reaction.Meanwhile,second generation hybrid capture was used to detect human papilloma virus(HPV) DNA in cervical smears obtained from the studied patients with the different lesions.The relationships among IL-10 and IL-18 with HPV DNA and clinical prognosis were analyzed.Results The expressions of IL-10 and IL-18 in normal control serum were (212.75 ± 62.09),(187.84 ± 81.03)pg/ml respectively.The expressions of IL-10 in CIN Ⅰ,CIN Ⅱ-Ⅲ and SCC serums were (324.71 ±75.87),(397.43 ±68.56),(482.77 ± 104.05) pg/ml,with statistically significant difference (F =17.657,P =0.001).The expressions of IL-18 in CIN Ⅰ,CIN Ⅱ-Ⅲ and SCC serums were (305.53 ± 64.08),(392.74 ± 87.38),(499.86 ± 92.04) pg/ml,with statistically significant difference (F =13.309,P =0.003).The relative expressions of IL-10 in normal control,CIN Ⅰ,CIN Ⅱ-Ⅲ and SCC tissues were 0.99 ±0.01,3.24 ±0.68,7.32 ±0.99,13.24 ± 1.03,with statistically significant difference (F =21.694,P =0.000).The relative expressions of IL-18 in normal control,CIN Ⅰ,CIN Ⅱ-Ⅲ and SCC tissues were 0.98 ± 0.01,2.02 ± 0.84,7.01 ± 1.59,14.38 ± 2.10,with statistically significant difference (F =19.912,P =0.001).The expressions of IL-10 and IL-18 were positively associated with HPV infection (r =0.696,P =0.001;r =0.852,P =0.001).The median survival time of patients with high expressions of IL-10 was 9.74 months,which obviously shorter than those patients with low expressions of IL-10 (24.47 months),with statistically significant difference (x2 =21.363,P =0.001).The median survival time of patients with high expressions of IL-18 was 12.32 months,which obviously shorter than those patients with low expressions of IL-18 (22.88 months),with statistically significant difference (x2 =7.457,P =0.006).Conclusion High expressions of IL-10 and IL-18 are observed in patients with CIN and SCC,which can be used as biomarkers for predicting early cervical lesions.

4.Effects of glucose concentration on G6PD activity and respiratory burst of normal human′s neutrophils in vitro

Objective To determine the effects of glucose concentration on G 6PD activity and respiratory burst of normal hu-man′s neutrophils in vitro .Methods Normal human′s neutrophils were cultured in different glucose concentration for 8 hours ,as-sayed G6PD activity by spectrophotometric method and determining ROS content by fluorescent probe DCFH-DA .Results G6PD activity and ROS of 15 mmol/L group and 25 mmol/L group were significant lower than before ,when the 5 mmol/L group and L-GLU group didn′t have significant change with time goes by .And G6PD activity and ROS of 25 mmol/L group were the lowest in all groups(P<0 .01) .Conclusion High glucose may induce G6PD activity decreased and cause respiratory burst dysfunction as a stimulating factor .The stimulation intensity was increased with the increase of glucose concentration .It′s the probable mechanism on susceptibility to infections in patients with diabetes mediated by dysfunction of respiratory burst in leucocyte .
5.Determination of artesunate in artesunate and amodiaquine hydrochloride tablets by HPLC

Objective To establish an HPLC method for the determination of artesunate in artesunate and amodiaquine hydrochloride tablets. Methods WondaSil C18-WR column was used with mobile phase consisted of acetonitrile:phosphoric acid aqueous solution(adjust pH to 3,gradient elution);wavelength was 210 nm; flow rate was 1 mL/min and the column temperature was 30℃.Results The standard curve was linear in the range of 0.2~3.2 mg/mL(r=0.9997), average recoveries were 99.0%(RSD=1.35%, n=6).Conclusion The method is accurate and sensitive, and it can be used to control the quality of artesunate and amodiaquine hydrochloride tablets.

9.The diverse phenotypes of RAG mutation associated immunodeficiency

RAG mutations cause a spectrum of severe immunodeficiencies ranging from classical severe combined immunodeficiency( SCID) and Omenn syndrome to an increasing number of peculiar phenotypes. Based on the distinct levels of RAG expression in various patients, their immunophenotypes, histopathological findings and clinical manifestations are diverse. The subtypes of RAG-defect diseases have been described as classical SCID,SCID with maternal T cells,classical Omenn syndrome,atypical Omenn syndrome,granuloma-tous inflammation,predominance/expansion of γδ-T cells and maternal T-cell engraftment. The complete failure or partial blockage of lymphocyte development and differentiation can cause repeated infections with autoimmune reactions frequently,and may be lethal. Thorough assessment and interpretation of various phenotypes will guide accurate diagnosis,definitive treatment and the mechanism research.
10.Effect of valerian extract treatment on experimental pulmonary fibrosis in rats

Objective To evaluate the effects and mechanism of valerian extract treatment on bleomycin-induced pulmonary fibrosis of rats. Methods After healthy Wistar rats(irrespective of sex)were given diethyl ether inhalation anesthesia, giving a single intratracheal instillation of bleomycin at a dose of 5 mg/kg to make a pattern of pulmonary fibrosis. 65 survival rats were randomly divided into four groups: Valerian high-dose groups(n = 16), Valerian low-dose groups(n = 16), Colchicine group(n = 16)and Model group (n = 17).Each group was given a dose of 100 mg/kg valeian extract everyday, a dose of 20 mg/kg valeian extract Valeian extract , a dose of 100μg/kg colchicine and a dose of 10ml/kg after second day. And in each groups, 6 rats were killed on day 7, 10 rats were killed on day 28 after instillation respectively. Rats in control group (n = 8) were instilled with saline intractracheally and saline was given everyday with a dose of 10ml/kg. Control group was killed on day 28. After rats were killed, The right lower pulmonary lobes were harvested for HE-staining, Masson-staining was used to observe the transformation of pulmonary tissue and immunohistochemistry was used to examine transforming growth factor-β1 (TGF-β1) which was analyzed by image analysis system. Results No obvious transformation were found in control group. The alveolitis in valerian and colchicine groups were ameliorated, compared with model group on day 7. The expressions of TGF-β1 in control group were lower than that in model group, and the mean integrated optical densities of TGF-β1 in control group were lower than that in mod-el group(P<0.05). The pulmonary fibrosis in valerian and colchicine group were ameliorated, compared with model group on day 28. The expressions of TGF-β1 in valerian and colchicine group were lower than that in model group. There were no significant differences between the valerian group and the colchicine group. Conclusion Valerian extract could reduce the degree of alveolitis and fibrosis induced by bleo-mycin through suppressing of TGF-β1.
