1.A study of the time-dependent changes in myocardial ischemia/reperfusion injury

The time-dependent changes in myocardio membrane system during ischemia/reperfusion injury were studied with ligature of the left ventricular branch of coronary artery in rabbit. The permeability of cardiac membrane system being tested by using lanthanum tracer technique and stereological methods. Lanthanum granules were observed in the sarplacosma and sarccpiasmic tubes especially at ischemia for 20min. The pecmeability of membrane system for La~(+++) was more marked when hearts were reperfused for 20min. With the increase in time of ischemia, the changes became more and more serious. A lot of lanthanum granules were entered into mitochondria when the hearts were reperfused after 40min ischemia. There were neither intact mitichondria nor lanthanum granules in mitochondria observed when the hearts were reperfused after 60min ischemia. The structural changes were irreversible at this stage of reperfusion The results of our study indicated that the changes of permeability of cardiac cell for La~(+++) in ischcmia/reperfusion are time-dependent, 40min ischemia is the demarcation line between the reversible and irreversible injury during reperfusion. On the basis of our experiment, we like to propose a new conception of reversible and irreversible injury of reperfusion.
2.Serum anti-hepatitis E virus IgG antibody and risk factors among female residents of Xinmi in Henan Province.

OBJECTIVETo determine the distribution of anti-hepatitis E virus (HEV) IgG antibody and anti-human papilloma virus (HPV) IgG antibody among female residents of Xinmi and investigate the risk factors of HEV infection.
METHODSA questionnaire was used to collect data on the demographic characteristics and suspected risk factors of HEV infection, including behavioral habits. All questionnaire responders also provided peripheral blood samples for investigation by enzyme-linked immunosorbent assay to detect HEV-IgG. Demographic data were statistically evaluated by t-test and univariate analysis, and HEV infection risk factors were statistically evaluated by a binary logistic regression model.
RESULTSThe average age of the 952 questionnaire responders was 47.16 + 8.09 years. The demographic parameters of education level, income, experience of stillbirth, and age were associated with HEV-IgG positivity (all P less than 0.05). Age, occupation, and income were identified as independent risk factors for HEV-IgG positivity (all P less than 0.05). No statically association was found between sexual behavior and anti-HEV or anti-HPV levels, or HEV infection.
CONCLUSIONThe female population surveyed in Xinmi, Henan Province showed a higher HEV-IgG positive rate than generally reported in the literature, and this rate shows an increasing trend with age, Risk factors for HEV infection among this group are age, income and occupation.

3.Feasibility of HEV vaccine as control in HPV vaccine study and comparison of serological and risk factors between HEV and HPV infection among females.

OBJECTIVEInvestigating the distribution of anti-hepatitis E virus (HEV-IgG), anti-human papillomavirus (HPV L1-IgG) and risk factors among female residents in Xinmi County, to explore the influencing factors of HPV vaccine study using HEV vaccinated population as a control.
METHODSA screening study of cervical cancer in Xinmi County, Henan Province, was performed. The information of demographic characteristics and risk factors was collected using standard questionnaire. Nine ml blood was drawn from each woman for enzyme-linked immunosorbent assay to detect HEV-IgG and HPV L1-IgG antibody. Percentile, histogram and binary logistic regression model were used to describe the distribution of risk factors and their correlation to HPV and HEV infection.
RESULTSThe average age of the Xinmi female residents was 47.2 years, their positive rate of HPV L1 antibody was 26.8%, and that of HEV-IgG antibody was 31.0%, both of which were raised with age (P < 0.001). Single factor analysis showed that non-education, low-income and growing age were associated with HEV-IgG antibody positivity, and non-education, lowering ages of first sexual life and growing age were associated with HPV L1-IgG antibody positivity. Multivariable analysis showed that growing age, low-income and work as peasantry were independent risk factors for HEV-IgG antibody positivity, and lowering ages of first sexual life, non-education and growing age were independent risk factors for HPV L1-IgG antibody positivity.
CONCLUSIONSBoth the HEV-IgG and HPV L1-IgG antibodies positive rates increase with age. Age is the common risk factor of HEV-IgG and HPV L1-IgG antibodies in female residents in Xinmi County. The risk factors of HEV-IgG and HPV L1-IgG antibodies have no statistical association, neither cross reaction was found in the HEV-IgG and HPV L1-IgG detection.

4.Hyperlipidemia in hepatic MED1 deficient mice in response to fasting.

MED1 is a key transcription co-activator subunit of the Mediator complex that is essential for RNA polymerase II-dependent transcription. MED1 functions as a co-activator for PPARs and other nuclear receptors and transcription factors, and plays an important role in lipid metabolism. To examine how MED1 might affect plasma lipids, plasma triglyceride, cholesterol levels, and lipoprotein profiles, were measured in MED1(deltaLiv) mice fasted for 24, 48 and 72 hours. Histological changes in liver sections from MED1(deltaLiv) mice after 72 hours of fasting were also examined using H&E staining. There was no fat accumulation in livers of MED1(deltaLiv) mice compared to MED1(fl/fl) and PPARalpha -/- control mice after 72 hours of fasting. Compared with MEDl(fl/fl) mice, plasma triglycerides in MED1(deltaLiv) mice were significantly increased after 24, 48 and 72 hours of fasting, and plasma cholesterol was significantly increased after 48 and 72 hours of fasting. Lipoprotein profiles were similar in fed MED1(fl/fl) and MED1(deltaLiv) mice. However, very low density lipoprotein (VLDL) was significantly increased in MED1(deltaLiv) mice after 24 hours of fasting. We conclude that, hyperlipidemia in MED1(deltaLiv) mice in response to fasting is due to the accumulation of VLDL, which suggests that MED1 plays a pivotal role in the regulation of plasma triglyceride and cholesterol levels.

5.Electroacupuncture alleviates orofacial allo-dynia and anxiety-like behaviors by regulating synaptic plasticity of the CA1 hippocampal region in a mouse model of trigeminal neuralgia

OBJECTIVE To investigate whether electroacupuncture(EA)ameliorates abnormal trigeminal neuralgia(TN)orofacial pain and anxiety-like behavior by altering synaptic plasticity in the hippocampus CA1.METHODS A mouse infraorbital nerve transection model(pT-ION)of neuropathic pain was established,and EA or sham EA was used to treat ipsilateral acu-puncture points(GV20-Baihui and ST7-Xia-guan).Golgi-Cox staining and transmission elec-tron microscopy(TEM)were administrated to observe the changes of synaptic plasticity in the hippocampus CA1.RESULTS Stable and per-sistent orofacial allodynia and anxiety-like behav-iors induced by pT-ION were related to changes in hippocampal synaptic plasticity.Golgi stain-ings showed a decrease in the density of dendritic spines,especially mushroom-type dendritic spines,in hippocampal CA1 neurons of pT-ION mice.TEM results showed that the density of synapses,membrane thickness of the postsynap-tic density,and length of the synaptic active zone were decreased,whereas the width of the synap-tic cleft was increased in pTION mice.EA attenu-ated pT-ION-induced orofacial allodynia and anx-iety-like behaviors and effectively reversed the abnormal changes in dendritic spines and syn-apse of the hippocampal CA1 region.CONCLU-SION EA modulates synaptic plasticity of hippo-campal CA1 neurons,and reduces abnormal oro-facial pain and anxiety-like behavior,providing evidence for a TN treatment strategy.
6.Distribution of serum antibodies against human papillomavirus 16 and 18 among high-risk women to cervical cancer

Objective To explore the distribution of serum antibodies against human papillomavirus(HPV)16/18 among women at high-risk for cervical cancer. Methods All women when tested positive for anyone of the cervical cancer screening programs,from Xinmi county of Henan province in 2011,were recruited as the subjects of this study. Cervical exfoliated cells were collected,using cervical brush for HPV DNA testing,and 10 ml venous blood was drawn for HPV-16, 18 serum antibodies testing,by enzyme-linked immunosorbent assay. Results Among the 952 women unders study,230 cases(24.2%)showed HPV DNA positive,with positivity rates of HPV16 and 18 L1 virus-like particle(VLP)antibodies as 23.2%and 6.5%,respectively. The overall positivity rate of any type of HPV16,18 VLP antibodies was 26.8%. Geometric means of HPV16,18 VLP antibody titers were 79.1(Yangshengtang Unit,YU/ml)and 125.0(YU/ml). Positivity rate of HPV16 antibody was significantly associated with age,viral load of HPV DNA,and cervical lesion severity (P<0.05). Seropositvity of HPV18 was also increasing with the increase of viral load (P<0.01) with different cervical lesion significantly showing different titer of HPV18 antibody (P<0.01). Based on the results of HPV DNA detection among the two years of study,women with HPV persistent infection showed significant higher positive rate of HPV16/18 antibodies than women who did not have HPV infection or emerging infection (P<0.001). When comparing to those women without HPV infection,the ones with transient infection showed higher seropositivity rates on both HPV16 antibodies and titer of HPV16 antibody (P<0.001). Conclusion Seroprevalence rates on HPV16 and 18 among the unvaccinated high-risk women in Henan were high. Prevalence of both HPV16 and 18 antibodies were correlated with age,viral load,cervical lesion and history of infection. Women with high viral load,high grade cervical lesion or history of infection would more likely to be seropositive.
7.A qualitative study of user experience, knowledge and attitude of e-cigarette use in Shanghai

ObjectiveTo understand the experience, knowledge and attitudes of e-cigarette users in Shanghai, and to provide evidence for the development of intervention programs for e-cigarette use. MethodsThe Respondent-Driven Sampling (RDS) method was used to recruit 40 e-cigarette users for qualitative interviews from November 2020 to February 2021. NVivo (11.0) software was used to code interview records, and the thematic pattern and relationship analysis were conducted. ResultsIn terms of the reasons and user experience of e-cigarette use, most respondents used e-cigarettes for the first time after recommendation of friends; The reasons for the current use of e-cigarettes focus on the subjective feelings generated by the use of e-cigarettes (easy to use, replacing traditional tobacco, and having many flavors). Compared with traditional tobacco use, respondents said e-cigarettes tasted good and had no physical impact (they could be used all the time without making them uncomfortable). In terms of the awareness of e-cigarette use related knowledge, the respondents' opinions on whether e-cigarettes caused health risks were mostly based on their own experience of using them, and they said it was difficult to identify real information online. As for whether they were interested in relevant knowledge, most respondents said they wanted to learn about the professional knowledge related to e-cigarettes. Regarding the attitude related to the use of e-cigarettes, most respondents believed that e-cigarettes are addictive, e-cigarettes are not a safe alternative to traditional tobacco, ande-cigarettes couldn't help them quit smoking. Three out of ten respondents said they supported including e-cigarettes in the Regulation on Smoking Control in Public. ConclusionThere are certain characteristics of e-cigarette users' feelings, related knowledge and attitude towards e-cigarette use that should be targeted to carry out publicity, education and intervention.
8.Overexpression of PPARγ induces adipogenic steatosis in mouse primary hepatocytes.

OBJECTIVETo investigate the effects of PPARγ overexpression on steatosis in mouse primary hepatocytes.
METHODSPrimary hepatocytes isolated from C57BL/6J mice were infected with either Ad/LacZ or Ad/PPARγ for 48 h. Steatosis of the primary hepatocytes was checked by Oil Red O staining. The mRNA and protein expression of adipocyte-specific genes PPARγ, aP2 and CideA were analyzed by using RT Real-time PCR and Western Blot.
RESULTSPrimary hepatocytes were small and even. Hepatocyte nuclei were round with dispersed chromatin and prominent nucleoli. Accumulated lipid droplets were observed in Ad/PPARγ-infected hepatocytes, but in Ad/LacZ-infected hepatocytes. Moreover, compared with Ad/LacZ-infected hepatocytes, the mRNA expression of PPARγ, aP2, FGF21 and CideA in Ad/PPARγ-infected hepatocytes were significantly induced, the protein expression of PPARγ and its target aP2 strongly increased.
CONCLUSIONover expression of PPARγ induces adipogenic steatosis in mouse primary hepatocytes.

9.Effects of Low Frequency Transcutaneous Electric Acupoint Stimulation on Upper Limbs and Hands Function for Chronic Stroke Patients

Objective To explore the effects of transcutaneous electrical acupoint stimulation (TEAS) on upper limbs function in chronic stroke patients. Methods From March, 2016 to May, 2018, 53 patients were randomly divided into control group (n = 27) and research group (n = 26). All the patients received conventional rehabilitation, and the research group received additional intervention of TEAS, for six weeks. They were assessed with Manual Muscle Test (MMT) on upper limbs, modified Ashworth Scale (MAS), Fugl-Meyer Assessment-upper extremities (FMA-UE), Hand Motor Status Scale and modified Barthel Index (MBI) before, at the end of 6-week treatment and twelve weeks after treatment. Results There was no significant difference in all the scores between two groups at the end of 6-week treatment (t < 1.511, P > 0.05). The scores of MMT of wrist dorsal extension, FMA-UE and MBI were better in the research group than in the control group twelve weeks after end of treatment (t > 2.312, P < 0.05). Conclusion TEAS may promote the recovery of hands and upper limbs function in chronic stroke patients.
