The influence of cimetidine on the pharmacokinetics of ftorafur ( FT-207 ) in 9 rats after intragastric administration were investigated with cross-over design. The concentration of ftorafur in serum was determined by reversed-phase HPLC. Its curve of concentration vs time was fit to one compartment opened model. The serum concentrations and areas under the curve ( AUC ) vs time were increased when cimetidine orally administered daily for 5d or single dose. 2.Study on the pharmacokinetics of alprazolam at different dose in rats

The pharmacokinetics of alprazolam in 18 rats was studied by using reversed-phase HPLC. The pharmacokinetic characteristics were fit to one compartment opened model after intragastric administration of alprazolam at doses of 50,20 and 2 mg ? kg-1, respectively. 3.Changes of circulating ET, A Ⅱ, CGRP after PTCA in the patients associated with acute coronary events

Percutaneous transluminal coronary angioplasty (PTCA) was done successfully in 33 patients,6 of them were associated with 8 acute coronary events. Radioimmunoassay was used for the determination of plasmaET,AII and CGRP levels. The samples were drawn successively from femoral artery just before PTCA and 0min, 15min,1h,3h,24h after the final balloon inflation. The results showed that the levels of plasma AII were higher in the complication group than those of noncomplication group before PTCA and 0min, 24h after PTCA. The differences in plasma ET levels before and after PTCA between the complication group and the non-complication group were not significant. The level of plasma CGRP was decreased at 15min and returned to basal level at 1h after PTCA in the complication group.In the patients without complication,there were no significant differences in their plasma CGRP levels before and after PTCA. All + ET/CGRP in the complication group raised more rapidly,recovered later and the peak appeared earlier than that of non-complication group. For a better understanding pathophysiologic meaning of the above changes,further studies are needed.
4.STUDIES ON THE PHARMACDKINETICS AND ABSOLUTE BIO- AVAILABILITY OF 1-HEXYLCARBAMOYL-5-FLUOROURACIL TABLET IN DOGS

The pharmacokinetics and absolute bioavailability of 1-hexyl-carbamoyl-5-fluorouracil ( HCFU ) (10 mg/kg ) after oral and intravenous administration were studied in 5 dogs with cross-over design. The concentration of HCFU in serum was determined by reversed-phase high performanee liquid chromatography. After intravenous administration, the curve of serum HCFU concentration vs time was fit to a two-compartment opened model and the phar-macokinetic parameters were. T1/2?=1 .67 min, T1/2? = 34.55 min, Vc= 0.2525L/kg,C1 = 0.3205 L/kg?h~-1 & AUCiv =1.9375 mmol/min?L~-1. When HCFU tablets were tiken orally, the curve of concentration vs time was fit to an one-compartment opened model and its pharmacokinetie parameters were: T1/2ke=12.13 min, T1/2ke=38.51 min, Tmax=23.46 min,Cmax=8.140?10~-3mmol/L & AUCpo=1.5856 mmol/min?L~-1 . The absolute bioavailability calculated from AUCpo and AUCiv was 0.8214.

Objective To determine the content of luteolin in Callicarpa nudiflora Hook.et Arn..Methods HPLC was performed with chromatographic conditions as follows:Alltima C18 column(250 mm? 4.6 mm,5 ? m)with column temperature at 25 ℃,the mobile phase of methanol-water-phosphate(55:45:0.4),flow rate at 1 mL/min,and the detection wavelength at 348 nm.Results The linearity of luteolin was good in the range of 0.032 46~ 0.129 84 ? g(r=0.999 5).The average recovery was 99.52 % and RSD was 1.58 %.Conclusion The method is simple,feasible and reproducible,and can be used to control the quality of Callicarpa nudiflora Hook.et Arn..
7.Clinical application of selective renal artery embolization in treating kidney diseases

Objective To evaluate the clinical application of selective renal artery embolization (SRAE) for the treatment of kidney diseases. Methods Seventy-four cases of renal carcinomas, 11 cases of renal angiomyolipomas (RAML) and 72 cases of traumatic renal haemorrhages were first demonstrated by renal arteriography under Seldinger technique to ensure a site, range and neighbouring relation of lesions and then followed by percutaneous catheterized selective renal arterial embolization with embolic agents. Results The edema around the carcinomas and abscesses became obvious, and bleeding were reduced, which were convenient for operation after SRAE for the preoperative adjuvant treatment of renal carcinomas. The clinical symptoms were improved obviously in 9 eases with advanced renal carcinoma after palliative treatment. The tumour volumes of 11 RAML were decreased evidently by 15 %-65 % with the average of 42 %. The bleeding in 68 cases of traumatic renal haemorrhage were ceased completely in 1-4 days after embolism, 2 cases with serious renal fragmented injury and huge perirenal hematoma combined with shock received successful operation after SRAE, 2 cases bleeding again after SRAE were cured by the second SRAE. No severe complications occurred after embolization in all the patients. Conclusions As a minimal invasive technique,selective renal artery embolization is a safe, effective method with less complications and an adjuvant pre-op-erative therapy for renal carcinoma or a palliative treatment for advanced carcinoma and an effective treatment for RAML and traumatic renal haemorrhage.

Numerical simulation of the thyroxin producing process of a human body is presented to analyze the dynamic and static variation laws of the process. The relationship between the static value of thyroxin and the reaction coefficients of various physiologic chemical reactions upon the producing thyroxin is clarified. The simulation results provided in this paper can be utilized as some reference data for clinical pathogeny analysis of some thyroxin diseases.

9.Mathematical modeling and simulation study of the heart rate feedback regulation system.

The physiological feedback regulation mechanism of the heart rate variation due to the blood pressure variation is studied. The continuous closed-loop mathematical model of the heart rate feedback regulation system is constructed. Numerical simulation is completed based on the above model. The simulation results demonstrate that the regulation system has the ability to constrain the fluctuation of the blood pressure to certain extent. The results are helpful for the pathological studies of some hypertensive diseases.

10.Scanning and analysis of the KATP channel mutations in 12 cases of infancy onset type 1 diabetes mellitus

Objective To screen the mutation of KATP channel mutations in Chinese pedigrees with infantile onset type 1 diabetes mellitus (T1DM) and neonatal diabetes mellitus.Methods A cohort of 12 children of infant onset T1DM and neonatal diabetes mellitus admitted into Beijing Children's Hospital between March 2004 and June 2013 were selected.PCR amplification and direct sequencing were used to analyze the 39 exons of ABCC8 gene and one exon of KCNJ11.And the mutational sites of the parents of the probands was sequenced in order to identify the inheritance.Results Analysis revealed ABCC8 mutation in 25% (3/12 cases) of the patients,a case of transient neonatal diabetes (TNDM),a case of permanent neonatal diabetes mellitus (PNDM) and a case of infant onset T1DM.All positive patients showed a known heterozygosis mutation in the ABCC8 gene(R1182Q,c.3545G > A,D209E,c.627C > G,E208K c.622G > A).The residue R1182Q,which was located at a position involved in joining transmembrane domain 2 to nucleotide binding domain 2,the mutations E208K and D209E were located in the intracellular region that links the transmembrane domain with the gatekeeper module.All the three mutations were located throughout the cytoplasm part of SUR1 protein.The TNDM successfully transferred from insulin to oral sulfonylureas therapy.Conclusions There is a complex genetic pathogenesis in neonatal and infant-onset diabetes.The KATP channel activating mutations is one of the main causes of neonatal diabetes mellitus and may cause T1DM in infants in China.Oral Glibenclamide therapy seems highly effective for some patients with the KATP channel activating mutations.
