Objective To study pharmacological mechanism of Sanshanxiao granules in treating diabetes. Methods The rat model of diabetes was set up by injecting STZ intraabdominally. The modeled diabetic animals were divided into model group, Sanshanxiao group, and Xiaokewan control group. Normal rats were devised as blank-contrasted group. Objective To investigate the popularity and distribution of Paragonimus westermani in Nankun Mountain,Northern suburban of Guangzhou.Methods 2012 snails and 63 crabs were picked from creeks.Three feces samples of wild cats in epidemic area and two feces samples of domestic cats inoculated with P.westermani metacercariae were collected.The cercariae,metacercariae and eggs of P.westermani were detected.P.westermani-positive cats were identified by detection,in which adults of Paragonimus were checked.P.westermani eggs artificially inoculated were also anatomized.Results Infection ratio in snails was 0.15‰(3/2000),while infection ratio of P.westermani metacercariae in crab was 100%(59/59),with an intensity of infection of 2~516 metacercariae per crab,and 2~10 matacercariae per gram crab.Snails and crabs were identified to be Semisulcospira libertina type and Sinopotamon pinheense type,respectively.P.westermani eggs were also found in two feces samples from wild cats,with an infection ratio of 66.66%(2/3).Eleven adults of P.westermani were found in two domestic cats inoculated with P.westermani.Conclusion Nankun Mountain in northern suburban of Guangzhou as an ultra-high infectious focus of P.westermani(Grade I) is reported for the first time.In view of the fact that P.westermani is one of the major disease paragonimus and the infectious focus Nankun Mountain nature reserve is an 4 A scenic spot and popular summer resort,tourists can become infected by P.westermani by drinking water from streams,springs and ponds.More attention should be paid to prevent human infection with P.westermani in Nankun Mountain.

Objective To study the relationship between circumferential resection margin status and prognosis of patients with middle and lower rectal cancer.Methods Specimens from 49 patients with middle and lower rectal cancer undergoing total mesorectal excision were studied by the large slice pathologic technique.The local recurrence,metastasis and five-year survival rate were evaluated by Kaplan-Meier Survival analysis.The related clinicopathologic factors were also analyzed.Results The cancer involvement rate of the circumferential resection margins was 24%(12/49).The overall local recurrence rate was 12%(6/49),the distant metastasis and recurrence rate was 27%(13/49),and the five-year survival rate was 67%(33/49).For the 12 patients in which the eircumferential resection margin was tumor positive.the local recurrence rate was 33%compared with 5%in those with negative circumferential resection margin(X2=6.577,P=0.010),distant recurrence was 50%compared with 19%in those with tumor negative margin(X2=4.491,P=0.034).Kaplan-Meier survival analysis showed that patient's survival time was statistically correlative with the circumferential resection margin status(log-rank.P=0.009).Five-year survival rate was 33%in patients with positive circumferential resection margin,compared with 78%in those with negative margins.Tumor diameter(X2=4.451,P=0.035),T staging (X2=20.283,P=0.000),N staging(X2=7.773,P=0.018),the distance away from the anocutaneous line(X2=6.502,P=0.04),tumor location(X2=4.421,P=0.035)and operation type(X2=5.754,P=0.016)were significantly correlated with the circumferential resection margin status of the middle and lower rectal cancer.Conclusions The circumferential resection margin status was an important predictor of local and distant recurrence as well as survival of patients with middle and lower rectal carcinoma.and the status is significantly correlated with tumor diameter,T staging,N staging,the distance away from the anocutaneous line,tumor location and operation type.

OBJECTIVETo construct a lentivirus-mediated vector for RNA interference (RNAi) of Fas and establish a human umbilical cord-derived mesenchymal stem cells (UC-MSCs) line with stable Fas gene silencing.

METHODSFour short hairpin RNA sequences targeting the coding region of human Fas mRNA were designed. The synthesized oligonucleotides were ligated with the lentivirus vectors harvested from BamHI and EcoRI double digestion of LV3 recombinant vector. The recombinant lentivirus vectors were transfected into the packaging cells 293T, and the lentivirus titers were determined. Cultured UC-MSCs were infected with the lentivirus, and real-time PCR and Western blotting were used to detect the expressions of Fas mRNA and protein in the transfected cells.

RESULTSRestriction digestion and DNA sequencing showed that the lentiviral vectors were successfully constructed, and the titer of lentivirus reached 3 × 10⁸ TU/ml in the packaging cells. Real-time PCR and Western blot demonstrated significantly suppressed Fas gene expression in UC-MSCs after infection with the recombinant lentivirus.

CONCLUSIONLentivirus-mediated RNAi can effectively inhibit Fas gene expression in cultured UC-MSCs.

BACKGROUNDAllergic asthma is a chronic airway inflammatory disease partly characterised by high concentration of T help 2 (Th2) cytokines in bronchoalveolar lavage fluid (BALF). There is no report on the relation of peripherally circulating blood lymphocytes and asthma. We explored the balance of Th2/Th1 cytokines in asthmatic mice. Exogenous recombinant interleukin (IL) 33 acted on murine peripheral circulating blood lymphocytes, IL-5 cytokine was selected for assessing Th2 cytokines and interferon-gamma (IFN-γ) for Th1 cytokines.

METHODSFemale specific pathogen free BABL/c mice were sensitised by intraperitoneal injection of 20 µg of ovalbumin emulsified in 1 mg of aluminium hydroxide gel in a total volume of 200 µl, and challenged for 30 minutes in 7 consecutive days with an aerosol of 2 g ovalbumin in 100 ml of PBS. Then we collected BALF and isolated lymphocytes from the peripheral blood. The lymphocytes were divided into two groups: asthmatic group and normal group. Th1/Th2 cytokines was detected by enzyme-linked immunosorbent assay (ELISA) kits.

RESULTSIn the asthma group, we found numerous eosinophils and lymphocytes on the glass slides. We then confirmed that the optimal concentration of IL-33 was 10 ng/ml and time of IL-33 stimulating lymphocytes was 24 hours. In the asthma group, the production of IL-5 was significantly increased over normal group after stimulation with IL-33 (P < 0.05) and the production of IFNγ was supressed from IL-33 stimulated lymphocytes (P < 0.05).

CONCLUSIONIL-33 acts on lymphocytes of peripheral blood increasing secretion of Th2 cytokines and inhibiting secretion of Th1 cytokines.
