1.Progress in animal models of experimental osteoporsis

Animals for model of osteoporosis involve rat, mouse, rabbit, beagle dog, minipig, sheep, etc. The types of model include aged related model, ovariectomized model, orchietomied model, drug treated model, abolition degeneration model, and dietary bone loss. The rat with ovariectomized model is used widely. Biochemical determination, bone mineral density measurement, bone histomorphorphormetry and bone biomechanics are used to judge the formation of experimental osteoporosis.
2.Discussion on the Control Index of Higher Medical Education According to the Relation of Supply and Demand

Academic analysis was made on the relationship between the supply and demand of higher education on the condition of rapid economic development.The development target of the healthcare manpower up to 2020 in Zhejiang Province was discussed through a comparative study method.A simulated prediction of the demand for the healthcare manpower was made by the specific ratio method of healthcare manpower/population.This study shows that by 2020 the number of doctors per thousand will reach 2.4,which is the average figure of the developed countries in the 1990's,then can adapt to the effective demand in Zhejiang Province.
3.Early non-invasive positive pressure ventilation in treatment of serious hypoxemia following acute left heart failure

Objective To observe the effect of early non-invasive positive pressure ventilation(NIPPV)on the treatment of serious hypoxemia induced by acute left heart failure. Methods Forty patients with acute left heart failure( Grade Ⅳ heart function)were randomly divided into two groups. Patients in both groups accepted supportive treatment included cardiotonics, diuretics, vasodilators, in additional to these high concentrations of oxygen were given in conventional group, and non-invasive positive pressure ventilation were given in NIPPV group by biphasic positive airway pressure(BiPAP). Systolic blood pressure, heart rate, respiratory rate, blood-gas analysis( pH, PaO2, PaCO2, SaO2 )and clinical signs were observed at 2 hours after treatments. Results Compared to control,RR( [ 19.55 ± 1.88] vs [21.85 ±3.51 ] ) BPM] ,HR ( [96.40 ±2.80] vs[ 103.20 ±6.78 ] BPM), SBP ( [ 116.30 ± 8.95 ] mm Hg vs [ 122.50 ± 6.13 ] mm Hg), pH (7.404 ± 0.027 vs 7.358 ±0.05) ,SaO2 ( [93.57 ± 1.18]% vs [91.97 ± 1.85]% ) ,PaO2 ( [75.58 ±4.61 ]mm Hg vs [68.38 ±7.95]mm Hg), PaCO2 ( [ 38.69 ± 3.06 ] mm Hg vs [ 43.61 ± 2.65 ] mmHg) were significantly different in NIPPV group( t = 2.582,4.146,2.558,3.534,3.256,3.505,5.428, Ps < 0.05 ). We found no significant differences in the comparisons before treatments. Hypoxia improved in NIPPV group,and the total effective rate was 95% in NIPPV group and 70% in control group,which showed significant difference( x2 =4.329 ,P <0.05 ) Conclusion BiPAP non-invasive positive pressure ventilation combined with routine treatment in treating heart failure, could promptly correct hypoxia, improve heart function and shortening disease course.

5.The effect of silencing ATP1A1 gene expression by RNA interference on proliferation of human U 251 glioma stem cells

Objective To investigate the effects of ATP1A1 knockdown by RNA interference(RNAi) on proliferation of human U251 glioma stem cells .Methods The human U251 glioma stem cells were infected with lentivirus expressing ATP1A1-shRNA . The mRNA and protein expressions of ATP1A1 in U251 glioma stem cells were detected by RT-qPCR and Western blotting ,re-spectively .The cell cycle and apoptosis were evaluated by flow cytometry .The proliferation of U251 glioma stem cells was deter-mined by MTT assay .Results The expressions of ATP1A1 in U251 glioma stem cells transfected with ATP1A1-shRNA were in-hibited significantly at both mRNA and protein levels ,with an inhibitory rate of 84 .15% for ATP1A1 mRNA and of 55 .33% for ATP1A1 protein respectively .The proliferation of cells was inhibited ,the cell apoptotic rate was significantly increased and the cell cycle was arrested in G1 phase and S phase decreased significantly in ATP1A1-shRNA cells(P<0 .05) .Conclusion RNAi targe-ting ATP1A1 gene could down-regulates the ATP1A1 expression ,induces cell apoptosis ,regulates cell phase redistribution and in-hibits cell proliferation in U 251 glioma stem cells .
6.Prediction of schistosomiasis infection rates of population based on ARIMA-NARNN model

Objective To explore the effect of the autoregressive integrated moving average model?nonlinear auto?regressive neural network(ARIMA?NARNN)model on predicting schistosomiasis infection rates of population. Methods The ARIMA model,NARNN model and ARIMA?NARNN model were established based on monthly schistosomiasis infection rates from Janu?ary 2005 to February 2015 in Jiangsu Province,China. The fitting and prediction performances of the three models were com?pared. Results Compared to the ARIMA model and NARNN model,the mean square error(MSE),mean absolute error (MAE)and mean absolute percentage error(MAPE)of the ARIMA?NARNN model were the least with the values of 0.011 1, 0.090 0 and 0.282 4,respectively. Conclusion The ARIMA?NARNN model could effectively fit and predict schistosomiasis in?fection rates of population,which might have a great application value for the prevention and control of schistosomiasis.
7.Treatment of hypertensive intraventricular hemorrhage by urokinase

Objective To discuss the feasibility,efficiency and safety of urokinase application in patients with hypertensive intraventricular hemorrhage.Methods Sixty-nine patients were included,30 treated with ventricular drainage alone and 39 receiving adjunctive intraventricular urokinase.CT images and ADL scores were compared between the two groups.Results The intraventricular thrombolysis with urokinase significantly hastened the resolution of intraventricular blood clots as compared with ventricular drainage alone(P=0.030),with better outcome(P=0.029).Conclusion Urokinase application is a simple,effective,and safe in managing hypertensive intraventricular hemorrhage.
8.Transplantation of bone marrow mesenchymal stem cells preconditioned with hydrogen sulfide in the treatment of rat myocardial infarction

BACKGROUND:Bone marrow mesenchymal stem cells (BMSCs) transplantation can promote cardiac repair after myocardial infarction, but it has been limited by the low cellsurvival rate.
OBJECTIVE:To study the effect of hydrogen sulfide (H 2 S) on the BMSCs transplantation for treatment of myocardial infarction.
METHODS:BMSCs were separated and cultivated form Sprague-Dawley rats weighing (100±20) g. The 4th generation cells were used for later experiment, and marked by DAPI at 2 hours before use. Fifty male Sprague-Dawley rats, weighing (200±20) g had been divided into five groups:Sham group (n=10) and four transplantation groups:BMSCs (n=10), H 2 S-BMSCs (n=10), H 2 S (n=10), normal saline (n=10). The myocardial infarction model of four groups was established except of sham group (only thread without ligation). The cardiac function was measured by echocardiogram at 4 weeks after celltransplantation. The col agen in the infarction area was tested by Masson staining.
RESULTS AND CONCLUSION:Severe myocardial fibrosis was found in the normal saline group, with no myocardial regeneration in the infarct area. H 2 S-BMSCs group had less col agen and more cardiac muscle tissue than BMSCs or H 2 S groups. Left ventricular ejection fraction and left ventricular fractional shortening of the H 2 S-BMSCs group were significantly higher than those of the BMSCs or H 2 S groups (P<0.05). The cells survival rate and cardiac function of myocardial infarction rats can be promoted by H 2 S-preconditioned BMSCs transplantation, which is superior to BMSCs or H 2 S alone.
9.Application research of remifentanil in patients with mechanical ventilation in ICU

Objective To observe the efficacy and safety of remifentanil in patients with mechanical ventilation in Intensive Care Unit (ICU) .Methods Totally 90 cases were enrolled ,patients with mechanical ventilation were randomly divided into two groups (remifentanil vs .fentanyl) ,45 cases in each group .We used facial expression score (FPS) to monitoring pain ,richmond se‐dation agitation score (RASS) was the sedation tool for measuring quality and depth of sedation .Propofol wase used for sedation therapy .We recorded the data including FPS and RASS and vital signs during the treatment .The primary outcome was the time to reach the goal of analgesia sedation ,cases of using propofol and doses of propofol .The secondory outcome was mechanical ventila‐tion time ,ICU length of stay and adverse events .Results Fentanyl and remifentanil all could achieve the desired analgesic effect . Compared with fentanyl group ,FPS scores showed significantly decreased after 5 min application of remifentanil (P< 0 .05) ,but FPS scores showed significantly decreased after 10 min application of fentanyl .Analgesia goal time of the remifentanil and fentanyl were(5 .00 ± 1 .37)min and (30 .00 ± 4 .50)min ,respectively .Only 28 .9% of the patients should be combined use of propofol in remifentanil group ,but fentanyl group was 64 .4% (P<0 .01);the dosage of propofol of remifentanil group was lower than those of fentanyl group ;there was no significant difference in central venous pressure (CVP) and heart rate(HR) between the two groups at each time point(P>0 .05) .The mechanical ventilation time ,ICU length of stay in the patients with remifentanil were significantly shorter(P<0 .05) ,and adverse events occur less(all P<0 .05) .Conclusion Remifentanil analgesia in patients mechanical ventilated patients is fast onset of action ,analgesic effect significantly ,with less sedative drug dosage .It also can shorten mechanical ventila‐tion time ,ICU length of stay and has no obvious side effects .
10.Effect of apolipoprotein E polymorphisms on expression of p38MAPK in the early period of astrocytes injury

Objective To investigate the correlation between apolipoprotein E (APOE) polymorphisms and expression of p38MAPK of injured astrocytes in the early stage.Methods Scratch injury to astrocytes of three different alleles of APOE (ε2,ε3,andε4) was induced.RT-PCR and Western-blot were applied to detect dynamic changes of intracellular p38MAPK before injury and at 12,24,48,and 72 hours postinjury.Results Expression of p38MAPK in APOEε2,ε3,andε4 astrocytes increased gradually over time,whereas before and 12-hour after injury,the difference was insignificant in pair comparison (P > 0.05).p38MAPK in APOEε2,ε3,andε4 astrocytes revealed progressive up-regulation at 24,48,and 72 hours postinjury,but the expression in APOEε4 astrocytes was the highest (P < 0.05).Conclusion In the early period of injury,highly expressed p38MAPK in APOEε4 astrocytes indicates a more active p38MAPK-induced inflammatory response in APOEε4 carriers which may contribute to acute exacerbation and poor outcome.
