1.The basic research of primary lactase deficiency

Lactose is one of the important nutrients in milk.It is decomposed by the lactase enzyme and plays an important role in the life activities of cells in human health.Due to the lack of lactase,the symptoms of lactose intolerance occur highly in Chinese population,thus limiting the large part of the people to drink milk.Milk is a good source of high-quality protein,minerals and vitamins for the human,so the problem of lactase deficiency also seems particularly severe.
2.Analysis of 9260 cases of cervical cytology liquid-based thin-layer analysis

Objective To explore the role of thinprep cytology test(TCT)and HPV-DNA hybrid capture detection played in the screening of cervical lesions.Methods Do TCT examination on 9260 outpatients and do HPVDNA hybrid capture detection on 840 cases therein,836 positive cases continued to have biopsy by colposcope,conformed to the gold standard of histopathology and contrast with cytology in the meantime.Results 8374 cases were negative in the TCT examination;836 cases were cytological abnormal.Cytology-positive rate was 9.03%,ASC-US 398 were 4.29%,LSIL 239 were 2.58%,HSIL 179 are 1.93%,SGUS 9 are 0.097%,SCC 11 are 0.12%.The group of 30 ~40 ranked the highest in cytological positive,it's 43.66%,the second highest was the group of 40 to 50,it's 27.39%.The results of other groups were as follow:< 30 is 2.85%,50 ~ 60 is 5.14%,> 61 is 0.96% The difference of them was significant ( P < 0.01 ).HPV infection rate was 27.98% (235/840).In ASC-US,the detectable rate of CIN in the HPV-positive group was 81.58% (31/38),the HPV-negative group was 8.98% ( 15/167),there was significant differene between the two methods(P < 0.01 ).In LSIL,the detectable rate of CIN in the HPV-positive group was 95.45% (21/22).In HSIL,the detectable rate of CIN in the HPV-positive group was 100% (16/16).The infection rate in different age group is different,the difference was significant ( x2 =25.2 P < 0.01 ) ; 836positive cases had biopsy by colposcope,507 cases were diagnosed to be CIN,the detectable rate was 60.65% (507/836).To organize pathology as control,the diagnose accordance rate of cytology and pathology of LSIL and HSIL and SCC were 50.46%,82.12%,100% (P <0.01 ).Took their accordance rate into comparison,the differene was significant.Conclusion Cervical lesion was collected in sex activity period,the peak was 31-40 years.TCT is propitious to screen cervical cancer and precancerous lesion,combined with HPV-DNA hybrid capture detection,colposcopy and pathology,can improve the diagnostic accuracy and the detective rate of CIN and cervical cancer.

As the development of the microsurgical technique,cataract surgery rapidly got good result and visual acuity,and patients had been demanded for higher visual quality.Because of popular surgical equipments,shorter surgical time and better efficiency,some unpractical advertisements appeared.So the medical dispute associated with cataract surgery has been increasing.The factors associated with medical dispute and how to prevent them will be discussed.
4.Growth differentiation factor-15 in cardiovascular disease:from bench to bedside

Growth differentiation factor(GDF)-15,a distant member of the transforming growth factor-? cytokine superfamily,is a powerful cardioprotective factor.Many studies have demonstrated that serum GDF-15 level is a novel marker closely associated with the diagnosis,risk stratification,and prognosis of several cardiovascular diseases.This article mainly reviews the characteristics of GDF-15 and its multifunction in cardiovascular diseases.
5.Application of DNA microarray technology in immunological research and its inspiration to researches on traditional Chinese medicine

DNA microarray technology is a high throughout method that can analyze the expression of thousands of genes at the same time to form a complete cellular gene expression profile. It has been applied in immunological researches such as the development, maturation, activation and differentiation of immune cells, the regulation of immune responses, the molecular mechanism of allergy, the relation between phenotype and gene expression, and immunological pharmacology, etc. It has deepened our perception of the immune system. It will as well be helpful in the research of the regulative mechanism of traditional Chinese medicine (TCM) toward immune cells and immune responses, the therapeutic mechanisms of TCM toward allergy, the standardization of differentiation of syndrome and herbal pharmacology, etc. The data analysis of the results of DNA microarray experiments is aimed to compare the difference of gene expression levels of different samples and obtain diagnostic gene expression profiles. But there are controversies about the number of samples and choices of statistical methods. Researchers are striving in setting standards of DNA microarray experiments.
6.OBSERVATION ON ULTRASTRUCTURE OF NORMAL AND ISCHEMIC DOG MYOCARDIA BY FREEZEETCHING METHOD

The normal and ischemic dog myocardia were treated with freeze-etching method and observed by transmission electron microscope, with special reference to the ultrastructural changes of myocardial cell membrane.Following 6 hours of coronary occlusion, the myocardial samples were taken from central and borderline ischemic areas as well as from distant normal area. The samples were cut into slices of about 1.5?1.5?3 mm~3 in size and proceeded by the routine freeze etching procedure.The myocardial cell membrane were fractured into the P face and E face. In normal myocardial cell membrane, the number of intramembranous particles of the P face was exceedingly larger than that of the E face. There were numerous caveolae appearing as depressions on the P face and elevated structures on the E face. In central and borderline ischemic areas, intramembranous particles were significantly decreased and aggregated. Following coronary occlusion, the caveolae of the myocardial cell membrane were found to be decreased, deformed or absent. In the severe ischemic area, spherical elevations with "rosette" configuration were occasionally observed as elevation on P face and depressions on E face.
7.SCANNING ELECTRON MICROSCOPIC OBSERVATION ON THE RABBIT PANCREAS BY DMSO FREEZE CRACKING METHOD

The normal rabbit pancreas was treated with DMSO freeze-cracking method and observed by scanning electron microscope, with special reference to examine the ultrastructure of pancreatic exocrine portion.The pancreatic acinus was composed of five to six pyramidal acinar cells around the lumen. A few short microvilli were present at the apex of each acinar cell. The spherical nucleus was located in the basal region, with one to three nucleoli.Numerous rough surfaced endoplasmic reticula were found in the basal region of acinar cells arranged either in longitudinal arrays, or concentrically. Mitochondria were located between rough surfaced endoplasmic reticula. The rough surfaced endoplasmic reticulum may extend into supranuclear portion where well-developed Golgi apparatus and numerous spherical zymogen granules were situated.
8.Expression of endostatin in ischemic myocardium of myocardial infarction rats

AIM: To study the expression of endostatin in ischemic myocardium of myocardial infarction(MI) rats in various periods and the correlation with VEGF expression and microvascular density(MVD).METHODS: Thirty-two male Sprague-Dawley rats after myocardial infarction were randomly divided into 7,14,21 and 28 days group.The sham group was normal control group(eight rats in each group).The expression of endostatin,VEGF and MVD in ischemic myocardium were observed by immunohistochemistry.RESULTS: The expression of endostatin significantly increased in the ischemic myocardium after MI,peaked at 7 days,then gradually decreased at 14,21 and 28 days.The endostatin level at 28 days was the same as the shams.The changing trends of expression of endostatin in ischemic myocardium after MI were similar to that of VEGF and were significantly correlated with the MVD.CONCLUSION: The expression of endostatin increased in ischemic myocardium of myocardial infarction rats.The changing trends of endostatin were similar to that of VEGF and positively correlated with the MVD.These data suggest that endostatin may modulate ischemic myocardium angiogenesis after myocardial infarction.
10.Influence of Psoriatic T Lymphocytes on Epidermal c-myc,bcl-2 and p53 Protein Expression in vitro

Objective To investigate the mechanism of abnormal epidermal proliferation induced by psoriatic T lymphocytes.Methods Skin organ culture was established with psoriatic T cells mixed with epi-dermal cells.The expressions of c-myc,bcl-2and p53protein were examined with immunohistochemical method in epidermis before culture,on the3rd day and the6th day after culture.Results Significant up-regulation of c-myc and p53protein was found in psoriatic lesions,but bcl-2protein expression was rarely observed.The expressions of those proteins were normal in non-lesional psoriatic skin.The p53protein ex-pression was increased in normal skin and non-lesional psoriatic skin on the3rd day after culture with psori-atic T cells,and c-myc protein expression was enhanced while bcl-2was decreased on the6th day of co-culture.There was no significant difference of those proteins' expression between normal skin and non-lesion-al psoriatic skin stimulated by psoriatic T cells.Conclusions The abnormal expressions of c-myc,bcl-2and p53protein play an important role in abnormal epidermal proliferation and differentiation in psoriasis.Psoriatic T lymphocytes can influence c-myc,bcl-2and p53protein expression in normal skin and non-le-sional psoriatic skin.Pathogenic T cells rather than keratinocytes might be vital for initiation of psoriasis.
