1.Paying attention to the differential diagnosis of central serous chorioretinopathy

Central serous chorioretinopathy (CSC)is a common eye disease characterized by macular serous retinal detachment.However,sometimes there are some atypical features,such as diffuse retinal pigment epitheliopathy,choroidal neovascularization (CNV),acute bullous retinal detachment,subretinal fibrosis,etc.Atypical CSC often is misdiagnosed because of its similar manifestations to polypoidal choroidal vasculopathy (PCV),inflammatory diseases such as Vogt-Koyanagi-Harada syndrome (VKH),et al.Furthermore,there are some other ophthalmic and systematic diseases which can cause serous retinal detachment and other atypical features.Nevertheless,the therapeutic regimens of these diseases are in great variation.In view of this,oculist should pay more attention to the differential diagnosis of CSC by fully understanding its clinical findings and concerning the general history including gluococorticoid administration.In this way,a precise diagnosis and correct management can be done.
2.Clinical Study on the Treatment of Subluxation of the Small Joint of the Cervical Vertebrae by Acupuncture plus Tuina

To observe the clinical therapeutic effect of the subluxation of the small joints of the cervical vertebrae by acupuncture plus Tuina, 178 subjects were divided randomly into acupuncture group, Tuina group and acupuncture plus Tuina group to receive the corresponding treatment. The cure rate in acupuncture plus Tuina was 68.8%, which is higher than that in acupuncture group (27.8%) and that in Tuina group(28.6%), and the statistical management showed significant differences. Acupuncture combined with Tuina could enhance the therapeutic effect in the treatment of the subluxation of the small joints of the cervical vertebrae.
3.Influence of periodical health education on self-care capability and living quality of patients with connective tissue diseases complicated with interstitial lung disease

Objective To explore the influence of periodical health education on self-care capability and living quality of patients with connective tissue diseases complicated with interstitial lung disease. Methods A total of 92 cases of patients with connective tissue diseases complicated with interstitial lung disease who were received and treated from January 2013 to October 2015 were selected as objects of study. The patients were divided into the observation group and the control group with 46 cases in each group according to the admission time. Both of the two groups were given conventional oxygen uptake therapy, non-steroidal anti infection therapy, expansion of blood vessels therapy and immune regulation therapy. The control group adopted usual care, while the observation group adopted periodical health education. The clinical efficacy, self-management ability, living quality and other indicators of the two groups were compared. Results The effective rate of observation group was 93.48%(43/46), and the effective rate of control group was 78.26%(36/46), The difference between the 2 groups was statistically significant (χ2=4.389, P < 0.05). The observation group related disease knowledge, self-care behavior, attitude adjustment, oxygen inhalation and respiratory function exercise with 5 self-care scores were higher than control group, the difference between the 2 groups was statistically significant (t=3.468-4.348, P<0.05). The observation group related physiological function, physiological rate function, physical pain, overall health, vitality, social function, emotional function, mental health 8 quality of life scores were higher than control group, the difference between the 2 groups was statistically significant (t= 2.261-5.456, P < 0.05). Conclusions Periodical health education is conductive to improving the self-care capability and living quality of patients with connective tissue diseases complicated with interstitial lung disease.
4.Relationship between the expression of focal adhesion kinase and the sensitivity of HT29 cells to 5-fluorouracil

Objective RNA interference (RNAi) expression vector was constructed to inhibit the focal adhesion kinase(FAK)expression in colon carcinoma HT29 cells, and then the sensitivity of the cells to 5-fluorouracil (5-FU) was determined. Methods One specific pair of oligonucleotides with short hairpin and its negative control sequence were designed and synthesized based on FAK cDNA sequences, then they were inserted into pGenesil-l vector to generate the recombinant plasmids. After identification by the restriction endonuclease and DNA sequencing, the recombinant plasmids were transfected into HT29 cells by lipofectamine TM 2000. The stable transfected cells were selected in a medium containing geneticin G418. The change in FAK expression in HT29 cells before and after RNA interference was detected by reverse transcription and polymerase chain reaction (RT-PCR) analysis and SP immunocytochemistry technique. Sensitivity of HT29 cells to 5-FU was determined by MTT assay. Results The recombinant plasmids coincided completely with the designs in the restriction map and the sequence analysis. After FAK being targeted by RNA interference, immunocytochemistry showed the protein expression of FAK was reduced dramatically, and RT-PCR revealed FAK mRNA expression was down-regulated by 76.94% compared to that of untransfected cells. 5.Relationship between antitumor activity of Gefitinib and epidermal growth factor receptor expression in human colorectal cancer cell lines

Objective To investigate the antitumor activity of Gefitinib, a selected epidermal growth factor receptor-tyrosine kinase inhibitor, on human colorectal cancer cell lines in vitro, and to explore the relationship between the inhibitory effect of Gefitinib on cancer cells and the expression of epidermal growth factor receptor (EGFR). Methods The growth inhibitory effects of Gefitinib, which expressed as the half growth inhibition dose IC50, on colorectal cancer cells were assessed by MTT assay. EGFR mRNA expression was detected by reverse transcriptional PCR (RT-PCR). Western blot was used to determine the expression of EGFR protein as well as its phosphorylated forms (p-EGFR). Results Gefitinib inhibited growth of all the six colorectal cancer cell lines in vitro with an IC50 range from 6.5 to 172.7?mol/L. Lovo cell line, with an IC50 value less than 10?mol/L, was the most sensitive one to Gefitinib, HT29 and SW480 were moderate sensitive to 10?mol/L
6.Effects of the crude extracts of Abrotani herba on the activity of NF-?B in colon carcinoma HT-29 and Lovo cells

Objective To study the NF-?B activity in colon carcinoma HT-29 and Lovo cells treated with different crude extracts of Abrotani herba obtained by column chromatography with macroporous resin.Methods The decoction of Abrotani herba absorbed by macroporous resin AB-8 was mounted into the column and then eluted with distilled water and 30%,50%,75% and 95% alcohol.To select appropriate concentrations for treatment,HT-29 cells were pretreated for 24 hours with each elution phase of the rude extracts in different concentrations(0,50,100,200,400 and 600?g/ml),and then their activity was detected by MTT.The HT-29 and Lovo cells were thereafter cultured in DMEM complete media respectively containing distilled water extract and 30%,50%,75% and 95% alcohol extracts in the concentrations as obtained by MTT assay,and the cells in control group were cultured in DMEM only.The cells were then harvested and the nucleic proteins were extracted for measurement with electrophoretic mobility shift assay(EMSA).Changes in NF-?B activity in HT-29 and Lovo cells treated with different concentrations of crude extracts were observed by EMSA.Results Viability of HT-29 cells treated with 400 and 600?g/ml crude extracts were significantly lower than those treated with 0-200?g/ml crude extracts(P0.05).Conclusion Crude extracts of Abrotani herba,extracted by column chromatography with macroporous resin and eluted with 30% alcohol,can inhibit NF-?B activity in colon carcinoma HT-29 and Lovo cells.

8.Effect of water-immersion restraint stress on nitric oxide production pathway in rat platelets

Objective To investigate the effect of water-immersion restraint stress(WRS) on nitric oxide(NO) production and its mechanism in rat platelets.Methods Male SD rats underwent WIR stress for 2,4 and 8 hours.Ulcer index(UI) of stomach was chedsed by Guth method,nitrite production was measured by Greiss assay,nitric oxide synthase(NOS) activities and L-arginine transport rate were determined by isotope tracer method.ResultsAfter 2 h WRS,the NO production,the NOS activity and L-arginine transport rate in rat platelets obviously increased and then gradually decreased.After 8 h of WRS,these parameters were decreased as compared with the control group.The gastric ulcer appeared and got more serious after 4 h WIR stress.Conclusion Short-term WRS up-regulates L-arginine /NO pathway and increases nitric oxide production in the platelets but long-term WIR stress has the reversed effect.
9.Relationship between PTEN expressions and inhibitory effects of Gefitinib on the growth of colon cancer cells

Objective To observe the inhibitory effect of Gefitinib,a selective oral epidermal growth factor receptor tyrosine kinase inhibitor,on the growth of human colon cancer cells,and investigate the relationship between the sensitivity of colon cancer cells to Gefitinib and PIEN expressions.Methods The inhibitory effects of Gefitinib on 6 kinds of colon cancer cells(Lovo,HCT116,HT29,Lsl74T,SW480 and SW620),the levels of PTEN mRNA in different colon cancer cells.and PTEN protein expressions in colon cancer cells were detected by MTT assay,RT-PCR and Westem blot,respectively.Results The inhibitory effects of Gefitinib on the 6 colon cancer cells in vitro varied a lot.When the concentration of Gefitinib was 1 μmol/L,LoVO cells had the most sensitivity to Gefitinib with an IC50<10μmol/L;HT29 and SW480 had moderate sensitivity,and the IC50 ranged from 10 μnol/L to 100 μmol/L;HCT116,LS174T and SW620 were insensitive to Gefitinib,and their IC50>100 μmol/L.All the colon cancer cell lines exhibited PTEN mRNA and protein expressions.Conclusions PTEN mRNA andprotein expressions might not be associated with the inhibitory effects of Gefitinib on the growth of colon cancer cells.The expression of PTEN can not be taken as the indication of the sensitivity of colon cancer cells to Gefitinib.
10.Therapeutic Efficacy of Buzhong Yiqi Decoction Containing Different Doses of Radix Astragali for Stage Ⅲ Internal Hemorrhoids Patients with Spleen Deficiency and Sinking of Qi and Its Effect of on Fibulin-3 Expression in Hemorrhoid Tissues

Objective To compare the effect of Buzhong Yiqi Decoction (BYD) containing different doses of Radix Astragali on fibulin-3 expression in the hemorrhoid tissues of stage Ⅲ internal hemorrhoids patients with spleen deficiency and sinking of qi, and to evaluate its therapeutic efficacy and possible mechanism. Methods Fifty-five qualified patients were randomly divided into control group(N = 15), Chinese medicine group 1(N =20), and Chinese medicine group 2(N=20). All of the 3 groups were treated with operation, and additionally, Chinese medicine group 1 was given BYD containing Radix Astragali 20 g, and Chinese medicine group 2 was given BYD containing Radix Astragali 50 g orally after operation. The scores of anal pendant expansion and anal prolapse were evaluated, and the expression level of fibulin-3 in the hemorrhoid tissues was detected by Western blot method. Results After treatment, the symptoms of anal pendant expansion and anal prolapse were improved in the 2 Chinese medicine groups (P < 0.05) , and the improvement of anal pendant expansion symptom in Chinese medicine group 2 was superior to that of Chinese medicine group 1(P<0.05). Compared with the control group, the expression level of fibulin-3 in the hemorrhoid tissues of the two Chinese medicine groups was increased (P < 0.05); the expression level of Chinese medicine group 2 showed an increasing trend as compared with that of Chinese medicine group 1, but the difference was statistically insignificant (P > 0.05). Conclusion BYD with large dose of Radix Astragali exerts stronger therapeutic efficacy for the treatment of prolapsed hemorrhoids than BYD with small dose of Radix Astragali, and its therapeutic mechanism has no obvious relation with promoting the increase of fibulin-3 expression.
