1.Long-term follow up of polypoid malignant colorectal tumor after endoscopic polypectomy or resection by surgery

Objective To compare the therapeutic result of endoscopic po lypectomy with colectomy for polypoid malignant colorectal tumor. Methods A tot al of 61 cases of polypoid malignant colorectal tumor including thirty- nine pa tients treated by endoscopic polypectomy and surveillance,twenty- two patients treated by colectomy.Follow- up ranged from 28 to 168(mean 64) months. Results During the follow- up period,one patient died relating to this disease,two rec urred at the original site,and one metastasized to the liver.The distribution of the adverse outcome had no significant statistical difference between the group of endoscopic polypectomy and colectomy;significant difference existed between the sessile and pedunculated malignant polyps (P=0.006),as well as between favou rable(carcinoma invading the muscularis mucosa or above,limited to the head of t he pedunculated polyp,polypectomy margin negative,and well and moderately differ entiated)and unfavourable(without those criteria) histologic characteristics(P=0 .002).Conclusion The polypoid malignant colorectal tumors with favourable his tologic characteristics could be treated by endoscopic polypectomy alone,tumors with unfavourable histologic characteristics should be treated by colectomy.
2.Proliferation, survival and differentiation of nerve stem cell in rat hippocampal after chronic stress

ObjectiveTo investigate the effect of chronic stress on hippocampal neurogenesis.MethodsChronic unpredictable mild stress(CUMS) was used as animal model to provoke a decrease of neurogenesis in hippocampus.Simultaneously,hippocampal neurogenesis was monitored by assessing cell proliferation,survival,and differentiation.All rats were divided into control group,14 d CUMS group,28 d CUMS group.ResultsThe cell proliferation in 14d CUMS group(2254.17 ± 164.41 ) or 28d CUMS( 1900.33 ± 104.10) group were decreased contrast to the control (2919.50 ± 188.80) (P<0.01).The cell survival in 28 d CUMS group ( 1845.33± 126.88 ) were decreased contrast to the control (2404.50 ± 148.77 ) (P < 0.01 ).The cell differentiation had no difference between control group and CUMS group( NeuN/BrdU(71.63 ± 10.21 ) vs (69.11 ± 11.30),GFAP/BrdU ( 14.34 ± 2.03 ) vs ( 17.27 ± 2.93 ) ) (P > 0.05 ).ConclusionThese results suggest that chronic stress decreases cell proliferation and survival,but does not affect the phenotype of newborn cells in the hippocampus.
3.Incidences of malnutrition and abdominal obesity in elderly type 2 diabetes

Objective To investigate the incidences of malnutrition (including undernutrition, overweight, and obesity) and abdominal obesity in elderly type 2 diabetes. Methods Totally, 133 elderly type 2 diabetes patients [study group, aged (66.9 5±.4) years] and 133 age-matched healthy subjects [control group, aged (66. 3 ±5.8) years] who met entry criteria and obtained informed consent were randomly enrolled into this study. Body weight, total body fat (TBF), abdominal fat, visceral fat, visceral fat area, and waist-to-hip ratio (WHR) were measured by multi-frequency bioelectric impedance analysis. The incidences of undernutrition, overweight, obesity, and abdominal obesity judged by BMI and WHR respectively were compared between the two groups. Results Com- pared to control group, BMI [(25.7 3.8) vs. (24.2 2.2) kg/m2, P = 0.001 ], TBF [ (20.1±6.9) vs. (17.4 5.0) kg, P = 0.001], WHR (0. 92±0.10 vs. 0.87±0.06, P =0.001), abdominal fat [(10.2 3.4) vs. (8.6 2.5)kg, P= 0.001], visceral fat [(2.7±0.9) w. (2.3 0.7)kg, P =0.001 ], and visceral fat area [ (89.1±28. 8) vs. (75. 7±21. 6) cm2, P =0.001 ] significantly increased in study group. The incidences of undernutrition (BMI<18.5, 3.8% vs. 0, P=0.024) and obesity (BMI^28.0, 26.3% vs. 6.0%, P = 0.001) judged by BMI Chinese standard in study group were significantly higher than those in control group. The incidences of "over-TBF" (52. 1% vs. 34. 1% , P =0.023) and abdominal obesity in males of study group (83.6% vs. 67.1% , P =0. 017) were significantly higher than those in control group. Conclusion Measurement and control of TBF, waist circumference, and abdominal and visceral fats should be emphasized in elderly patients with type 2 diabetes.
4.Mitochondrial apoptotic pathway is involved in apoptosis in hippocampus of post-traumatic stess disorder rats

ObjectiveTo observe the expressions of Caspase-9,Caspase-3, cytochrome C and their relationships, and investigate apoptotic mechanisms in hippocampus of post-traumatic stress disorder(PTSD) rats. Methods Sixty male Wistar rats were used in the present study. The single-prolonged stress (SPS)-method was used to set up the rat PTSD models. There were six groups:after SPS 1 day,4 days,7 days,14 days,28 days groups and control group. The expressions of caspase-9, Caspase-3 and Cytochrome C proteins were detected with immunohistochemistry, immunofluorescence and Western blotting. Results Immunohistochemical and immunofluorescent results showed that the expression of cytochrome C peaked at 4 days and maintained higher level at 7days after SPS. Expressions of Caspase-9 and Caspase-3 were increased and peaked at 7days after SPS. Western blotting results showed that compared with control group, cytochrome C protein was up-regulated and peaked at 4 days after SPS in the cytosol. Compared with the control group, cytochrome C tended to decrease in mitochondrial fractions. The activated form of Caspase-9 and caspase-3 were not detected in control group. They were present in model groups. Cleaved- caspase-9 and cleaved-caspase-3 peaked at 7 days after SPS, and then gradually decreased at 14 days. Conclusion The neuronal apoptosis in hippocampus of PTSD rats may be one of the causes inducing hippocampus atrophy. Mitochondrial apoptotic pathway is involved in apoptosis in the hippocampus of PTSD rats.
5.Changes of nuclear receptors-glucocorticoid receptors in locus ceruleus neurons of posttraumatic stress disorder-like rat

Objective To investigate expression changes of nuclear receptors-glucocorticoid receptors (GR) in the locus ceruleus neurons of posttraumatic stress disorder(PTSD)like rats. Methods Sixty adult healthy male Wistar rats were enrolled in the experiment. PTSD-like rat model was established by single prolonged stress (SPS). There was no SPS stimulation in the control group and the rats of the other five groups were undisturbedly raised for 24 hours, 4 days,7 days,14 days and 28 days respectively after SPS stimulation.The expressions of glucocorticoid receptors in the locus ceruleus nucleus of all groups were detected with Nissl staining, immunohistochemistry,Western blotting and PCR, and image analysis and statistical analysis were performed. Results GR was distributed in the nucleus of coeruleus neurons, GR expression was showed after 24 hours, 4 days, 7 days treatment and gradually increased, restorative downregulation was seen after 14 days and 28 days, but still high(P<0.05). Conclusion After SPS, the GR locus ceruleus temporarily increased and then lowered, suggesting that PTSD rat locus coeruleus neurons in nuclear receptor-GR expression changes may directly involve in the continuing spirit of PTSD symptoms occurred in the development process.
6.Expression and significance of cytochrome C and Bax in hippocampus of posttraumatic stress disorder rat

Objective To observe the expression of cytochrome C (Cyt C)and apoptosis-related gene Bax in hippocampus of posttraumatic stress disorder(PTSD)rats in order to investigate their effects and relationships. Methods Sixty male Wistar rats were used in the present study. The single-prolonged stress(SPS)-method was carried out to set up the rat PTSD models. There were six groups:after SPS 1,4,7,14,28 days groups and control group. The expression of Cyt C and Bax proteins were detected with immunohistochemistry, double immunofluorescence, confocal laser scanning microscopy and Western blotting. Results The expression of Cyt C peaked at 4 days and maintained higher level at 7 days after SPS. At 7 days after SPS, strong immunoreactivity of Bax was noticed,At 14 days after SPS, the expression of Bax was down-regulated and then gradually decreased. Conclusion Bax is upregulated and promotes the releasing of Cyt C. And the releasing of Cyt C may be one of mechanisms of inducing apoptosis in the hippocampus of PTSD rats.
7.Application value of human papillomavirus DNA detection and ThinPrep liquid-based cytology testing in cervical lesions

Objective To evaluate the application value of high-risk human papillomavirus (HR-HPV)DNA de-tection and ThinPrep liquid-based cytology testing(TCT)in cervical disease screening.Methods Cervical specimens of women with cervical lesions in a hospital between October 2012 and December 2013 were taken and performed hu-man papillomavirus DNA genotyping (HPV DNA)and ThinPrep liquid-based cytology testing(TCT).Positive pa-tients were performed colposcopy pathological detection.HPV DNA positive rates among different TCT groups, and different cervical lesion groups were compared,the sensitivity and specificity of TCT and HPV DNA detection, as well as differences between separate and joint detection were also compared.Results The positive rate of HPV DNA was 28.07% (1 045/3 723),most were HR-HPV (21 .57%,n=803),the major HR-HPV genotypes were HPV 16,58,52,and 18 type.HR-HPV positive rates were statistically different among different age groups(χ2 =31 .74,P <0.001 ),positive rate was highest in 20 -30 year old age group .Positive rate of TCT was 13.46%(n=501),a total of 971 patients were performed pathological detection,293 were positive.Patients were divided according to different TCT and different lesion type,χ2 testing of HR-HPV DNA positive rate showed that positive rate of HR-HPV had a increasing tendency with the increase in severity of diseases(all P <0.01).Pathological de-
tection was as a gold standard ,the sensitivity of HR-HPV DNA and TCT was 90.44%(265/293)and 85.32%(250/293)respectively,the sensitivity of joint HR-HPV DNA detection and TCT was 95.90%.In positive patho-logical group,the detection rate of TCT and HR-HPV DNA was 85.32% and 90.44%,respectively,joint detec-tion rate was 95.90%,the difference among three groups was significant (χ2 =18.185,P <0.001).Joint detection rate was higher than separate detection rate of TCT or HPV DNA.Conclusion HPV DNA detection is a useful supplement for cervical cancer screening,HPV DNA detection combined with TCT can reduce the misdiagnosis rate.
8.THE MORPHOLOGICAL ANALYSIS OF APOPTOSIS IN HIPPOCAMPUS OF POSTTRAUMATIC STRESS DISORDER RATS AND THE EXPRESSION OF APOPTOSIS-RELATED GENES

Objective To investigate relationships between the expression of Bcl-2,Bax protein and neuronal apoptosis in hippocampus of posttraumatic stress disorder(PTSD) rats.Methods The SPS-method was used to set up the rat PTSD models.There were five groups:after SPS 1d,4d,7d,14d groups and control group.Apoptotic cells were detected by electron microscopy and TUNEL method.The expressions of Bcl-2 and Bax proteins were detected with immunohistochemistry,double fluorescent,confocal laser scanning microscopy and Western blotting.Results Apoptotic cells were present in hippocampus of PTSD rats.The number of apoptotic cells increased with the development of PTSD and peaked on the 7th day after SPS,then decreased gradually.The expression of Bcl-2 protein peaked on the 4th day after SPS and Bax protein peaked on the 7th day after SPS,then decreased gradually.The ratio of Bcl-2/Bax increased at the beginning and then gradually decreased.Conclusion The neuronal apoptosis in hippocampus of PTSD rats may be one of reasons inducing hippocampus atrophy.The increase of Bcl-2 and Bax protein and the change of Bcl-2/Bax ratio would play an important role in regulating the hippocampal neuronal survival or death during posttraumatic stress disorder.
9.Studies on the relationship of apoptosis and multidrug resistance in 5-fluorouracil resistant human hepatocellular carcinoma cell line Bel7402/5-FU

AIM To elucidate whether apoptosis is involved in the development of multidrug resistance in 5-fluorouracil resistant cells Bel7402/5-FU. METHODS Multidrug resistance of Bel7402/5-FU cells was determined by SRB assay, apoptosis was measured by Annexin V-FITC/PI double labeled assay, cell cycle phases were analysed by flow cytometry, and expression of genes related to apoptosis bcl-2, bcl-xl, bcl-xs, bax, fas, p53, and cpp32 was detected by RT-PCR assay. RESULTS Bel7402/5-FU cells were resistant not only to 5-FU but also to multiple anticancer drugs. When treated with the same concentration of 5-FU (30 ?mol?L -1 or 150 ?mol?L -1 ), the proportion of apoptosis was significantly increased and G_0/G_1 phase was increased and S and G_2/M phases were reduced in Bel7402 cells, but the proportion of apoptosis and cell cycle was not changed in Bel7402/5-FU cells. The expression of bcl-xl, bcl-xs, and bax mRNA were significantly increased and the expression of p53 and cpp32 mRNA were significantly decreased in resistant Bel7402/5-FU cells, as compared with Bel7402 cells. CONCLUSION Decrease of apoptosis may be related to multidrug resistance of human hepatocellular carcinoma cells Bel7402/5-FU. The up-regulation of anti-apoptotic genes and down-regulation of pro-apoptotic genes are involved in the development of multidrug resistance in Bel7402/5-FU cells.
