OBJECTIVE:To determine aminophenazone,phenazone and barbital in antodine injection.METHODS:Taking 0.1mol/L NaOH as solvent,the detection data was caculated with partial least square method.RESULTS:The average recoveries were 99.7% for aminophenazone(RSD=0.67%),99.1% for phenazone(RSD=0.76%) and 99.5% for barbital(RSD=0.72%).CONCLUSION:The method is simple,rapid and good in repeatability.

γ-Secretase is aⅠtransmembrane protease associated with Alzheimer disease(AD), and including four subunits:presenilin,presenilin enhancer-2,anterior-pharynx-defective1 and nicastrin. In recent studies,the ultra-high-resolution cryo-electron microscope has been used for the first time, revealing the humanγ-secretase″horseshoe″,the three-dimensional structure and the arrangement of the subunits. This technique has shed light on the regulation of the enzyme pathway and mechanism. In addition,the γ-secretase modulators,including non-steroidal anti-inflammatory drugs,have been shown in vitro to inhibit γ-secretase activity and selectively reduce the level of Aβ42 against AD. They have provided an effective approach,with broad prospects for development. Studies in this area have become a hot topic in recent years. Here we summarized the γ-secretase subunits regulatory pathways, humanγ-secretase fine three-dimensional structure andγ-secretase modulators .
3.Research on the correlation between subacromial impingement syndrome and the relative position of acromion and humeral head

[Objective]To evluate the influence of subacromial space caused by the relative position of acromion and humeral head,and to provide theoretical guidance for diagnosis,prevention and treatment of subacromial impingement syndrome.[Method]From October 2006 to March 2007,31 shoulders(13 men and 18 women with average age of 54.3)with subacromial impingement syndrome,from our department,were selected to form the patient group,An age and gender-matched of 40 asymptomatic shoulders(18 men and 22 women with average age of 52.8)formed the control group.The acromion index was measured on the Grashey view.The distance from the glenoid plane to the lateral border of the acromion was divided by the distance from the glenoid plane to the lateral aspect of the humeral head to calculate the acromion index.[Result]The average acromion index(and standard deviation)was(0.72?0.05),in patient group,(0.61?0.04)in the asymptomatic,normal shoulders.The difference between the index in the shoulders with subacromial impingement syndrome and the index in normal shoulder was highly significantly(P
4.Dexmedetomidine relieves oxidative stress and inflammatory damage after tourniquet-induced ischemia/reperfusion injury

BACKGROUND: Dexmedetomidine has been shown to fight against ischemia/reperfusion injury induced by tourniquets. OBJECTIVE: To study the effects of dexmedetomidine on the oxidative stress and inflammatory damage caused by tourniquet-induced ischemia/reperfusion injury. METHODS: Seventy-six patients scheduled for lower limb operation were randomized into two groups: patients in dexmedetomidine group were given the intravenous injection of 1 μg/kg dexmedetomidine for 10 minutes, followed by 0.5 μg/kg?h until the end of operation; while the controls were subjected to 0.9% saline injection at an equivalent velocity and volume. The levels of serum propanediol, lactic dehydrogenase, superoxyde dismutase, tumor necrosis factor-α, interleukin-6 and -8 were detected before tourniquet inflation, 10, 60 and 120 minutes after tourniquet release. RESULTS AND CONCLUSION: In both two groups, the serum levels of propanediol, lactic dehydrogenase, tumor necrosis factor-α, interleukin-6 and -8 after tourniquet release were significantly higher and the serum superoxide dismutase level was significantly lower than those before tourniquet inflation (P < 0.05). Compared with the control group, dexmedetomidine significantly reduced the serum levels of propanediol, lactic dehydrogenase, tumor necrosis factor-α, interleukin-6 and -8, and increased the serum superoxyde dismutase level after tourniquet release (P < 0.05). These results suggest that dexmedetomidine can attenuate the oxidative stress and inflammatory damage resulting from tourniquet-induced ischemia/reperfusion injury probably by up-regulating the serum superoxyde dismutase level, and down-regulating the serum levels of propanediol, lactic dehydrogenase, tumor necrosis factor-α, interleukin-6 and -8.
5.Clinical application of metal-on-metal hip joint prosthesis with large heads(ASRTM XL)

Objective To explore clinical characteristic of metal-on-metal hip joint prosthesis with large heads(ASRTMXL).Methods Total hip arthroplasty(THA)with ASRTMXL was performed in 14 patients including two with ankylosing spondylitis combined with hip amalgamation,three with avascular necrosis,two with secondary osteoarthritis and seven with femoral neck fractures.Based on shortterm follow-up results,we analyzed its design characteristic and clinical properties.Results All patients walked with crotches one week after operation and began weight bearing without support six weeks later.An improvement was seen on the average range of motion of the hip including anteflexion,retroextension,internal rotation,external rotation,adduction and abduction.The mean Harris score was increased from precperative 30 points(13-50 points)to postoperative 90 points(75-100 points),with statistical difference.Postoperative X-ray showed right positions of the prostheses,normal abduction angle and anteversion angle,without complications cccurred.Condusion Short-term follow-up results show that metal-on-metal hip joint prosthesis with large heads has advantages of lower wearing,large range of motion,few dislocation and small deformation of acetabular cup.
6.Viper system and pedicle screw fixation for treating thoracolumbar compression fractures

BACKGROUND:Thoracolumbar spine as highly concentrated stress, often prone to vertebral fractures. With the further development of the biomechanics and anatomical structure of the spine, posterior open reduction and internal fixation with pedicle screw has been widely accepted by clinicians. OBJECTIVE:To explore the clinical results and safety of percutaneous pedicle screw fixation system (Viper system) used in thoracolumbar compression fractures. METHODS:We retrospectively analyzed 40 patients with thoracolumbar compression fractures from Department of Orthopedics, Changzhou Traditional Chinese Medicine Hospital and Nanjing General Hospital of Nanjing Military Area Command of Chinese PLA from January 2013 to February 2014. According to the type of graft, patients were randomly divided into experiment group and control group, with 20 patients in each group. They were respectively subjected to Viper percutaneous pedicle screw fixation system and open reduction and pedicle screw fixation. RESULTS AND CONCLUSION:Al vertebra got bone unions. Operative time and time to bone union were shorter in the experiment group than in the control group. Moreover, intraoperative blood loss was less in the experiment group than in the control group. Cobb’s angle, height percentage of leading edge and wedge angle were similar between the two groups. However, at 12 months after internal fixation, height percentage of leading edge was lower in the experiment group than in the control group. Visual Analogue Scale scores and Oswestry Disability Index were noticeably improved after fixation in both groups. Visual Analogue Scale scores and Oswestry Disability Index were lower in the experiment group than in the control group immediately after fixation. These results suggest that compared with open reduction and pedicle screw fixation, Viper percutaneous pedicle screw fixation system for thoracolumbar compression fractures can stably restore the structure and function of spine, and does not increase perioperative complications.

Recent metabolomics research revealed a new ginseng ginsenoside IH901 that is synthesized by intestinal microbial transformation in oral administration of ginseng. IH901 shows various biological activities, including anti-tumor, anti-inflammatory, anti-diabetic, and anti-aging. In recent years, great effort has been made to prepare IH901 by microbial and enzymatic transformation in a large scale. In this paper, we reviewed the biotransformation pathways both in vivo and in vitro and bioactive properties of rare ginsenoside IH901.

8.Study on Medication Regularity of Chinese Medicine for Polycystic Ovary Syndrome Based on Data Mining and Syndrome Dis-tribution

[Objective]To explore the medication rule of traditional Chinese medicine(TCM)in the treatment of polycystic ovary syndrome(PCOS)through data mining based on the distribution of syndromes.[Methods]Randomized controlled trials of TCM syndrome differentiation in the treatment of PCOS from CNKI,Wanfang Data Platform,VIP full-text database,Web of Science and PubMed were systematically searched.Exce12019,SPSS Modeler 18.0,SPSS Statistics 26.0 and other softwares were used for data analysis to explore the drug characteristics and rules of TCM treatment of PCOS based on syndrome distribution.[Results]A total of 281 literatures were included,including 3 427 frequent Chinese drugs,among which spleen deficiency phlegm dampness was the most involved syndrome type,and Angelicae Sinensis Radix,Poria cocos,Cuscuta chinensis Lam and so on were the most frequently used Chinese medicines.The amount of prescription drugs was mostly between 10~20 g,deficiency tonic drugs were the most common efficacy type in prescription,and warm and plain were the most common drug properties.Sweet and pungent were the most common drug flavors,while liver,kidney and spleen channels were the most common drug normalization channels.Drug types were also different under different syndrome types.Association rule analysis and cluster analysis were conducted on the top 3 syndrome types,and complex network diagram and tree diagram were drawn to explore the characteristics and rules of TCM treatment of PCOS under different syndrome types.[Conclusion]The clinical manifestations of PCOS are highly heterogeneous,and the treatment mainly focuses on tonifying the spleen and kidney,regulating Qi,resolving phlegm and removing blood stasis.The treatment methods of patients with different syndrome types are also different.TCM syndrome differentiation has unique advantages in the treatment of PCOS with significant effects,and the clinical treatment should be based on reasonable syndrome differentiation.
9.Diagnositic value of urine heparin-binding protein, interleukin-6 and white blood cell in bacterial urinary tract infection

Objective To evaluate the diagnostic values of urinary heparin-binding protein (HBP), interleukin-6 (IL-6) and white blood cell (WBC) levels in bacterial urinary tract infection (UTI). Methods A case-control method was used. Urine of 157 cases of bacterial UTI, 61 cases of non-infection, and 40 cases of normal controls were collected in the Third Xiangya Hospital of Central South University from September 2017 to March 2018. U-HBP levels were measured in duplicate using a commercial HBP ELISA, U-IL-6 concentrations were analyzed with an up-conversion luminescence. The method of quantitative culture of bacteria was used to identify pathogenic species. Rapid dipstick tests and urinary sediment analyses were detected by FUS-2000 Urinalysis Hybrid. For continuous variables with skewed distributions, comparisons among the three groups were performed using the nonparametric Kruskal-Wallis test, and Mann-Whitney U test was used to further evaluate the difference between two groups. The Chi-square test was applied to analyze dichotomous. Receiver operating characteristic curve (ROC curve) was constructed to analyze the clinical diagnostic values of U-HBP, U-IL-6 and U-WBC for bacterial UTI. Results The levels of U-HBP in UTI group, non-UTI group and control group were 513.43 (50.45-644.40) ng/ml, 55.65 (20.43-314.55) ng/ml and 4.83 (3.28-12.63) ng/ml. The scores of U-IL-6 were 5.72 (3.84-9.02) pg/ml, 5.31 (4.31-6.39) pg/ml and 5.06 (4.56-6.18) pg/ml. The scores of U-WBC were 205 (24-754) cells/μl, 34 (13-117) cells/μl and 0 (0-0) cell/μl. There were statistically significant differences of U-HBP and U-WBC among the three groups (HU-HBP=83.192, HU-WBC=100.416, P<0.05), but no significant difference for U-IL-6 (HU-IL-6=2.585, P>0.05). The best Youden indexes of U-HBP and U-WBC diagnosing bacterial UTI were 0.475 and 0.441, respectively. The best cut-off level of U-HBP and U-WBC was 64.35 ng/ml and 119.25 cells/μl, respectively. Conclusions Testing the level of U-HBP was important for auxiliary diagnosing bacterial UTI, but testing U-IL-6 wasn't.
10.Inhibitory effect of siRNA-Pax6 on biological behavior and epithelial-mesenchymal transition of human lens epithelial cells

Objective:To explore the effect of knockdown of the homeobox gene paired-box 6 ( Pax6) on the biological behavior and epithelial-mesenchymal transition (EMT) of human lens epithelial cells (LECs). Methods:The SRA01/04 human LECs were divided into small interfering RNA-Pax6 (siRNA-Pax6) group transfected with siRNA-Pax6 and siRNA negative control (siRNA-NC) group transfected with disordered siRNA.Cell survival rate was detected by cell counting kit-8 method at 24, 48 and 72 hours after transfection.Cell cycle distribution and apoptosis were analyzed by flow cytometry at 48 hours after transfection.Migratory capability of cells was examined by cell scratch test at 24 hours after transfection.The mRNA relative expression levels of Pax6, α-crystallin A (CRYAA), α-crystallin B (CRYAB), Sox2, α-smooth muscle actin (α-SMA) and E-cadherin were detected by quantitative real-time PCR at 48 hours after transfection.The relative expression of Pax6 protein was detected by Western blot at 48 hours after transfection.Results:There was a significant difference in cell survival rates at different time points between the two groups ( Fgroup=4.776, P<0.05; Ftime=13.535, P<0.05). The cell survival rate of siRNA-Pax6 group was obviously lower than that of siRNA-NC group at 48 and 72 hours after transfection, and the differences were statistically significant (both at P<0.05). Compared with siRNA-NC group, the proportion of cells in G 0/G 1 phase was significantly increased and the proportion of cells in S phase was significantly reduced in siRNA-Pax6 group ( t=9.971, -5.063; both at P<0.05). The cell migration rate of siRNA-Pax6 group was (19.73±6.07)%, which was lower than (70.56±2.97)% of siRNA-NC group, showing a statistically significant difference ( t=-7.245, P<0.05). The relative expressions of Sox2 mRNA and α-SMA mRNA were lower, and the relative expression of E-cadherin mRNA was higher in siRNA-Pax6 group than siRNA-NC group, with statistically significant differences between them ( t=-23.254, -5.294, 6.062; all at P<0.01). The relative expression of CRYAA mRNA and CRYAB mRNA was significantly higher in siRNA-Pax6 group than siRNA-NC group, and the differences were statistically significant ( t=5.521, 8.270; both at P<0.01). The relative expressions of Pax6 mRNA and protein in siRNA-Pax6 group were 0.27±0.01 and 0.24±0.05, respectively, which were both lower than 1.00±0.05 and 1.14±0.10 in siRNA-NC group, showing statistically significant differences ( t=-14.456, -4.458; both at P<0.001). Conclusions:Silence of Pax6 can suppress the proliferation and EMT of human LECs and enhance the expression of crystallin.
