Objective To observe the effect of gastric motility and gastric electric activity on sleep deprivation and the changes of calcitonin gene-related peptide(CGRP) and nitro oxygen(NO). Methods The 192 SD rats were divided into three groups randomly. Gastric emptying was analyzed with radioimmunoassay, using nuclide 99m Tc and gastric emptying of liquids and EGG with the serosa method .By using the immuno-chemistry and chemistry analysis methods, we observed the changes of some connective factors, for example, CGRP and NO. Results The acute deprivation can cause the change of gastric emptying and which is decreased with the time of deprivation, in the meantime, we found the gastric electric activity would be decreased before food, otherwise, the gastric electric activity is irregulary and the gut hormone, for example, CGRP and NO have been changed in the plastic of rats after acute deprivation. In the study we found the gastric emptying have a change obviously in the short time. But with the time of deprivation the gastric empting no have change. And the gastric similar to this, the gut hormone no any change. Conclusions The acute sleep deprivation caused the change of gastric emptying and the change of gastric electric activity obviously. In plastic, the level of CGRP and NO have a change after sleep deprivation. The chronic sleep deprivation caused the change of gastric emptying and the change of gastric electric activity, and caused the function of gastric motility disturbing, and the effect is prolong after chronic sleep deprivation.

Objective To study the expression of Fos gene and GFAP in medullary visceral zone (MVZ) of rats under stress with their gastric mucosa protected by acupuncture. Methods 24 male SD rats were randomly divided into 3 groups: control, stress and acupuncture group, with 8 for each group. Immunohistochemical methods were employed to detect the expression of Fos and GFAP in the nucleus of solitary tract (NTS) and dorsal motor nucleus of vagus (DMV) of medullary visceral zone. The extent of gastric mucosal damage was evaluated by ulcer index (UI). Results Fos-positive neurons and GFAP-positive stellar microgliocytes were found mainly in NTS and DMV of the medullary visceral zone. 7.A study on detecting chlorophyll-related genes of plankton in the diagnosis of death by drowning

Objective To evaluate the value of detecting chlorophyll related genes of plankton in the diagnosis of drowning. MethodsEighteen rabbits were divided randomly into three groups: death by drowning (n=10), postmortem submersion (n=6) and control (n=2). The heart blood, lung, liver, kidney and brain tissues were taken from every rabbit. After isolated plankton from tissues with percoll and extracted their DNA, the chlorophyll-related genes, including EG (EG1 and EG2) and SK (SK1 and SK2), were detected using PCR technique. Meanwhile, diatom test was also performed from lung and liver tissues by nitric acid digestion method. ResultsFor the drowning group, the specific amplification products for EG1 were detected from 9 samples in heart blood, 10 samples in lung, 9 samples in liver, 7 samples in kidney and 8 samples in brain. The products for EG2 were detected from 8 samples, 10 samples, 7 samples, 5 samples and 7 samples accordingly. There were a small number of positives in heart blood, lung and kidney with SK1 and SK2 (≤2). For the postmortem submersion group, only one case was positive from heart blood and lung tissue respectively for EG1. No amplified product was detected for EG1 and EG2 in various tissues in control group, and also no product was detected for SK1and SK2 in other groups. In addition, diatoms were detected from 9 lung and 3 liver tissues in drowning group with the nitric acid digestion, and only one sample of lung was positive in the postmortem submersion group. ConclusionThe detection rate of the chlorophyll-related gene EG with PCR method was higher than that of diatom with nitric acid digestion method in drowning victims, and it can be used as a potentially useful tool for diagnosing drowning.

Objective The chemical components of lyophilized powder of Sinisan were analyzed by the method of RP-HPLC.Methods HPLC Method was applied for quality assessment on lyophilized powder of Sinisan.Column: Kromasil C18 column(250 mm ? 4.6 mm,5 ?m);wavelength: 210 and 240 nm;flow-rate: 1.0 mL/min;column temperature: 28 ℃.Mobile phase: acetonitrile(A) and H2O(acidified to pH=3 with phosphoric acid)(B).Concentration of lyophilized powder of Sinisan was resolved into different parts and analyzed by the same method,the variation of the chemical components was analyzed.Results At 240 nm,21 peaks were detected and two of them were confirmed to be from Bupleurum chinense,three from Paeonla lactiflora,ten from Citrus aurantium,and five from Glycyrrhiza uralensis.At 210 nm,24 peaks were detected and one of them was confirmed to be from B.chinense,four from P.lactiflora,fourteen from C.aurantium,and five form G.uralensis.Conclusion This method can be used to establish the fingerprint chromatograph of lyophilized powder of Sinisan and elucidate the variation of chemical components completely.
