1.Immunohistochemical Detection of ras P_(21) Expression in Pancreatic Adenocarcinoma and Its Clinicopathological Significance

By using antr-ras P21 mouse monoclonal (MoAb), SCI-Oncogema 1, the authors examined immunohistochemica! staining in pancreatc adeocarcinoma. The percentage of positive staining was 24/43 (558%). Furthermore it was indicated that the positive staining rate of antr-ras P21 MoAb was related to either histopathological grade or clinical stage. Upon statistical analysis of the correlation between the staining of anti-ras P21 and patient prognosis with Kaplan-Meier curve and Log-rant test, the positive staining cases showed comparatively better prognosis than the negative ones. Our study suggests that ras P21 expression may be important in the early stage of pancreatic carcinoma.
2.Apoptosis in gastric cancer cells induced by trichosanthin is related with down-expression of bcl-2

Objective To investigate the apoptosis in gastric cancer induced by trichosanthin, and the relationship between this apoptosis and expression of bcl2. Methods In in vitro experiments, morphologic test and TUNEL staining method were used to quantitatively and qualitively detect the apoptosis status of gastric adenocarcinoma cell line SGC7901 before and after the trichosanthin treatment. Immunohistochemical staining method and Northern Blot hybridization were used for detecting expression status of apoptosisrelated genes bcl2, before and after trichosanthin treatment. Results When SGC7901 cells were treated with trichosanthin (0.1 ?g/ml, 36 h), they presented some typical apoptotic morphologic changes observed by fluorescent staining. These morphologic changes include nuclear condensation, nucleosomal fragments forming a lunate body under nuclear membrane, etc. When SGC7901 cells were treated with trichosanthin at the concentration 0.1 ?g/ml for 36 h,42 h and 48 h, respectively, TUNEL staining showed a significant increase of apoptotic index (AI), from 3.78%?1.11%, 3.98%?1.12%,3.85%?1.08%, respectively, to 11.30% ? 2.33%, 10.22% ?2.00%,11.18%?1.85%(P
3.Isolation and identification of macrophages from liver of Microtus fortis

Objective To separate and purify intrahepatic macrophages from Microtus fortis Mf and identify its phagocy?tosis. Methods The intrahepatic macrophages from Mf were separated and purified by perfusion collagenase digestion and density gradient centrifugation. The function of the cells was identified by FACS analysis and ink phagocytosis activity. Results The macrophage cells from the liver of Mf were obtained. These cells were bright and circular and grew adhering to the wall. The proportion of the living cells was 95%. The binding rate of these cells from Mf with anti?mouse CD14 antibody Clone Sa2?8 was about 50%of the rate of macrophage from C57BL/6 mice with this monoclonal antibody. The result of ink?phagocytosis ex?periment of macrophage cells from the liver of Mf was positive. Conclusion The method above mentioned is useful to separate and purify macrophage from the liver of Mf. The study builds the foundation for further research on macrophages of Mf against Schistosoma japonicum.
4.Clinical value of procalcitonin detection in neonatal necrotizing enterocolitis

Objective To investigate the clinical value of of procalcitonin detection in neonatal necrotizing enterocolitis (NEC).Methods Sixty cases eligible NEC infants were randomly divided into observation group and control group(n=30 each group).All the infants were divided into Ⅰ grade group(19cases),Ⅱ-Ⅲ grade group(41 cases) according to NEC classification.All infants received PCT detection after NEC diagnosis.The observation group received antibiotic treatment base on the serum PCT levels.The control group received antibiotic treatment according to treatment routine and physician's clinical experience.Results PCT increased in 28 (46.7%) of the 60 NEC infants.In the Ⅰ grade group and Ⅱ-m grade group,the proportion of PCT increased cases were 26.3%,61.3% respectively,and the PCTmax in PCT positive cases were (0.81±0.25) μg/L,(1.76±0.89) μg/L respectively,the differences were signifiacnt(x2 =4.627,t=-2.608,P<0.05).The time of antibiotic usage in observation group was significantly less than that in control group ((4.9±3.0) d vs.(8.6±2.5) d,t=-3.645,P<0.01).The fasting time and the cure time of the two groups had no significant difference (P> 0.05).Conclusion PCT detection can help to understand the cause and severity of NEC.Using antibiotics according to PCT levels can reduce the time of antibiotic usage without affecting the effect of the NEC treatment.
5.A retrospective study on the outcomes of the oncology, fertility and pregnancy in patients with early-stage cervical cancer after undergoing the fertility-sparing treatments

Objective To explore the outcomes of the oncology, the fertility and pregnancy on patients with early-stage cervical cancer after undergoing fertility-sparing treatments, and to investigate its value on clinical treatment. Methods A total of 29 patients with cervical cancer (stage Ⅰa2-Ⅰb1) who had undergone the fertility-sparing treatments in Renji Hospital, School of Medicine, Shanghai Jiaotong University from February 2010 to September 2015 were analyzed retrospectively. Surgical procedures included transvaginal cervical cold knife conization (CKC)+laparoscopic pelvic lymphadenectomy (PLD) or total laparoscopic uterine cervix extensive resection (RT; contains PLD); 48 patients with early cervical cancer under going laparoscopic uterine extensive resection (RH; contains PLD) by the same group of physicians in the same period were chosen as the control group. The perioperative related indicators, postoperative specimen examination, pregnancies after surgery and postoperative tumor follow-up results were retrospectively analyzed and compared between the two groups. Results (1) Among these 29 patients in the study group, 26 cases were underwent the fertility-sparing surgery quick frozen pathological examinations were positive in 3 cases, which underwent total laparoscopic RT eventually. (2) The perioperative related indicators: compared with the study group and the control group, the operation time [(182 ± 21), (147 ± 24) minutes, respectively] has significant difference (t=6.563, P<0.01). There were no significant difference (P>0.05) in intra-operative blood loss [(102±26), (99±34) ml], postoperative aerofluxus time [(1.3±0.4), (1.1±0.9) days], the average length of hospital stay [(11.2 ± 2.1), (10.6 ± 3.5) days], rate of urine retention [10%(3/29), 10%(5/48)] and rate of postoperative infection [3%(1/29), 2%(1/48)]. (3)Postoperative specimen examination:compared with the study group and the control group, there were no significant difference (P>0.05) innumber of removed lymphatic nodes (23.4 ± 4.1, 22.8 ± 3.9), length of cardinal ligament [(2.9 ± 0.5), (3.0 ± 0.6) cm], lengthof uterosacral ligament [(2.6±0.7), (2.8±0.4) cm], length of removed vaginal [(3.4±0.3), (3.5±0.3) cm]. (4) Pregnancies after surgery and postoperative tumor follow-up results:in the study group, only 14 patients had fertility requirement after treatments. Pregnancies occurred in 5 women (5/14), which included1 case of full-term labor, 1 case of preterm labor, and 3 cases of spontaneous abortion. The Average follow-up time in postoperative patients of the study group and control group was 29.4, and 30.2 months respectively. In follow-up period, compare with study group and the control group, there was no significant difference (χ2=0.004, P>0.05) in the recurrence rate [4%(1/26), 4%(2/48)]. Conclusion Fertility-sparing surgery of early-stage cervical cancer is safe but the outcome of the fertility and pregnancy is still need toimprove.
6.Normal development of refractive state and ocular dimensional components in guinea pigs

Objective To study the emmetropization in guinea pig eyes during the normal development from birth in order to provide theoretical basis to use guinea pigs as model for research on near-sightedness.Methods Sixty-four guinea pigs were assigned to 8 groups (n=8,4 male/4 female).Each group underwent a series of ocular examinations at one of the 8 time-points (0,1,2,3,5,7,9 and 11 weeks),including refraction(R),radius of corneal curvature (RCC),depth of anterior segment(AS),thickness of crystalline lens (CL),length of vitreous chamber (VC)and axial length (AL).Pooled results from both eyes of the same animals with mixed sexes can be used for further assessment of the emmetropization with the associated biometrical changes of the eye.Correlations were made between RCC,AS,CL,VC,AL and R. Results There were no significant differences between the right eye and left eye or between male and female in results of all the examinations. The refraction at birth was (+5.25?0.22 )D in guinea pigs.This value rapidly decreased during the first 3 weeks,approaching(+1.34?0.61) D by 11 weeks.There was no significant difference in refraction between 9 and 11 weeks (P= 0.215). The RCC was (3.23?0.01) mm at birth,AS was (1.20?0.00) mm at birth,CL was (2.72?0.02) mm at birth and VC was (3.27?0.01) mm at birth,they increased within the first 3 weeks despite a transient decrease in the RCC within the first week.Such an increase continued except the AS which became constant after 3 weeks.The VC was more correlated to the emmetropization (r=-0.818,P
7.Construction and identification of a recombinant lentivirus harboring expressing Mus musculus STAT3 gene

Objective To construct recombinant lentivirus with the gene STAT3 of the Mus musculus,measure the expres-sion of STAT3,and conduct lentivirus packing and identification.Methods The mRNA of mouse myoblast was extracted and transformed into STAT3 cDNA by the special primer.then,STAT3 cDNA was amplified and reclaimed and inseted into pLVX-IRES-ZsGreen1 vector.Cleavage map and sequencing analysis were used for identification of the recombinant lentivirus vector (pLVX-IRES-ZsGreen1-STAT3).293 T cells were transfected with main vector pLVX-IRES-ZsGreen1-STAT3.and 48 h later, Western blott detected the expression of STAT3 protein.Lentiviral vectors were packaged and the titer was determined.Results The lentiviral vector plasmid pLVX-IRES-ZsGreen1-STAT3 was identified correctly by cleavage map and Co-transfection of 293 T cells with 48 h,the expression of STAT3 was significantly enhanced by western blot.And DNA sequencing analysis confirmed that STAT3 gene sequencing was exactly the same with that reported by genbank.Conclusion Lentiviral vector carrying STAT3 was successfnlly constructed and could express STAT3 with high efficiency,and can be used in further study.
8.Role of thoracic electrical bioimpedance monitoring in the evaluation of cardiac function in peritoneal dialysis patients

Objective To evaluate the effectiveness of thoracic electrical bioimpedance(TEB)in monitoring the cardiac function of peritoneal dialysis patients.Methods One hundred and one patients with continuous ambulatory peritoneal dialysis (CAPD) and 30 healthy persons (control group)were included in the study.Thoracic electrical bioimpedance (TEB) noninvasive hcmodynamic monitoring and echocardiography were taken to analyze the correlation between indexes.Results Echocardiography showed that left atrial diameter (LAD),left ventricular end diastolic diameter (LVDd),left ventricular end systolic diameter (LVDs),interventricular septal thickness (IVST),interventricular septal thickness (PAP),left ventricle weight index (LVMI) of CAPD group were higher than that of the control group (all P < 0.05),early and late wave of mitral valve flow (E/A) of CAPD group was lower than that of control group (P < 0.05).TEB monitoring showed that cardiac output (CO),stroke volume (SV),acceleration index (ACI),ejection fraction (EF),velocity index (Ⅵ) of CAPD group were significantly lower than that of control group (all P < 0.01),systolic time ratio (STR),SVR,TFC of CAPD group were significantly higher than that of control group (P < 0.01).Correlation analysis show that left ventricular ejection fraction (LVEF) was negatively correlated with BNP (r =-0.467,P < 0.01),LVMI was positively correlated with BNP (r=0.416,P < 0.01),PEP,STR and TFC were positively correlated with BNP (r =0.404,P < 0.01; r =0.572,P < 0.01; r=0.471,P < 0.01),EF was negatively correlated with BNP (r =-0.664,P < 0.01).Correlation analysis between echocardiogaphy and TEB monitoring index showed there was significant correlation between EF and LVEF (r =0.451,P < 0.01),SVR and TFC were positively correlated with LVMI (r =0.232,P < 0.05; r =0.284,P < 0.05),SV was positively correlated with E/A (r =0.285,P < 0.05),pre-ejection period (PEP) and STR were negatively correlated with LVEF (r =-0.389,P < 0.01; r =-0.446,P < 0.01),TFC was positively correlated with LAD (r=0.279,P < 0.05).Conclusion TEB monitoring can accurately evaluate the cardiac function with the advantage of dynamic monitoring and simple operation.It can partly replace the echocardiography test.
9.Development of a Software for Automatically Generated Contours in Eclipse TPS.

OBJECTIVEThe automatic generation of planning targets and auxiliary contours have achieved in Eclipse TPS 11.0.
METHODSThe scripting language autohotkey was used to develop a software for automatically generated contours in Eclipse TPS. This software is named Contour Auto Margin (CAM), which is composed of operational functions of contours, script generated visualization and script file operations. RESULTS Ten cases in different cancers have separately selected, in Eclipse TPS 11.0 scripts generated by the software could not only automatically generate contours but also do contour post-processing. For different cancers, there was no difference between automatically generated contours and manually created contours.
CONCLUSIONThe CAM is a user-friendly and powerful software, and can automatically generated contours fast in Eclipse TPS 11.0. With the help of CAM, it greatly save plan preparation time and improve working efficiency of radiation therapy physicists.

Pharmacological studies have shown that Bruceae Fructus has unique antitumous effects, which has broad market prospects. In recent years, the demand of Bruceae Fructus has increased year by year. Production and marketing thrive, and prices continue to rise. Therefore, it has become one of the Chinese medicinal herbs with antitumour effects that obtained people’s attention. This author summarized the dynamic condition of production and marketing of Bruceae Fructus during the last 10 years, and analyzed the major reasons for price changes and forecast development tendency so as to provide references for the reasonable layout of production and marketing of Bruceae Fructus.
