1.Expression of collagen type Ⅲ, laminin and fibronectin in Mooren's ulcer

Objective To compare the locations and levels of collagen type Ⅲ, laminin, and fibronectin in Mooren's ulcer and normal human corneas. Methods An indirect immunofluorescent technique and immunohistochemical staining were used to determine the distribution of collagen type Ⅲ, laminin, and fibronectin. The positive results were quantitatively analyzed with an image-processing system. Results Collagen type Ⅲ was not detected in the normal cornea. However, the staining could be seen in the corneal stroma close to Mooren's ulcer focus. Laminin was expressed faintly in the basement membrane of the normal cornea and the positive expression increased in the basement membrane and Bowman's membrane of Mooren's ulcer. In the normal cornea basement membrane, fibronectin was located continuously. In Mooren's ulcer basement membrane, however, fibronectin could not be found in all sections in which one expressed in epithelium and the others in stoma close to the ulcer focus. Conclusion Epithelial basement membrane may play a potential role in the pathogenesis of Mooren's ulcer.
2.A research on the corrosion resistance of Ni-Cr alloy with different coatings

Objective: To evaluate the corrosion resistance of Ni-Cr alloy with different coatings in neutral and acid saliva by electrochemistry method. Methods: Measured the self-corrosion potential (Ecorr) and polarization resistance(Rp) curves of Ni-Cr alloy models with different coatings in neutral and acid artificial salivas. Results: The Ecorr of the Ni-Cr alloys all showed positive values, the electrochemistry corrosion and galvanic corrosion were not detected. Conclusion: Ni-Cr alloy with titanium nitride coating, gold electroplating coating, gold-paste layer and gold-porcelain blended layer have both anti-corrosion and esthetical function without changing its own property.
3.Effect of a novel oncolytic herpes simplex virus type Ⅱ on lung adenocarcinoma

Objective To establish the subcutaneous transplantation tumor models with Lewis lung adenocarcinoma in C57BL/6 mice, and to observe the influence of oHSV2, DDP and drug combination on tumor volume, median survival time and weight of tumor-burdened mice.Methods Subcutaneous transplantation tumor models were established with Lewis lung adenocarcinoma in tumor-burdened mice.Tumor-burdened mice were randomly divided into the control group, oHSV2 group, DDP group, oHSV2/DDP sequential group, DDP/oHSV2 sequential group and oHSV2+DDP combination group with 12 rats in each group using the random number table method.The tumor size and weight of mice were measured every 3 days.Results On the 21st day, the tumor size of tumor-burdened mice in every group was as follows: control group (1.82±0.06)cm3, oHSV2 group (0.63±0.05)cm3, DDP group (0.58±0.03)cm3, oHSV2/DDP sequential group (0.49±0.05)cm3, DDP/oHSV2 sequential group (0.42±0.04)cm3, and the difference was statistically significant (F=1 359.01, P=0.000).The data in oHSV2+DDP group were put away because of premature death in mice.The differences were statistically significant between control group and oHSV2 group (P=0.000), control group and DDP group (P=0.000), control group and oHSV2/DDP sequential group (P=0.000), control group and DDP/oHSV2 sequential group (P=0.000), oHSV2 group and DDP group (P=0.017), DDP group and DDP/oHSV2 sequential group (P=0.000), oHSV2/DDP sequential group and DDP/oHSV2 sequential group (P=0.001).The weight of tumor-burdened mice in every group was listed as follows: control group (21.64±0.40)g, oHSV2 group (21.34±0.37)g, DDP group (15.96±0.43)g, oHSV2/DDP sequential group (19.04±0.31)g, DDP/oHSV2 sequential group (16.34±0.30)g, and the difference was statistically significant (F=588.67, P=0.000).The difference was not statistically significant between control group and oHSV2 group (P=0.076).However, the differences were statistically significant between control group and DDP group (P=0.000), control group and oHSV2/DDP sequential group (P=0.000), control group and DDP/oHSV2 sequential group (P=0.000), oHSV2 group and DDP group (P=0.000), oHSV2 group and oHSV2/DDP sequential group (P=0.000), DDP group and DDP/oHSV2 group (P=0.013), oHSV2/DDP sequential group and DDP/oHSV2 sequential group (P=0.000).The median survival time of tumor-burdened mice in every group was displayed as follows: control group 23 d , oHSV2 group 32 d, DDP group 30 d, oHSV2/DDP sequential group 37 d, DDP/oHSV2 sequential group 39 d, oHSV2+DDP combination group 16 d, and the difference was statistically significant (χ2=120.81, P=0.000).The differences were statistically significant between control group and oHSV2 group (χ2=10.88, P=0.001), control group and DDP group (χ2=10.69, P=0.001), oHSV2 group and DDP/oHSV2 sequential group (χ2=10.09, P=0.001), DDP group and DDP/oHSV2 sequential group (χ2=9.67, P=0.002).However, the differences were not statistically significant between oHSV2 group and DDP group (χ2=0.00, P=0.996), oHSV2/DDP sequential group and DDP/oHSV2 sequential group (χ2=2.70, P=0.100).Conclusion On the premise of that the weight of mice is no affected, oHSV2 can inhibit the tumor size and prolong the median survival time of tumor-burdened mice effectively, and the effect of DDP/oHSV2 sequential group is the most significant.This article provides an experimental basis for exploring therapeutic methods of lung adenocarcinoma.
4.Screening of human anti-ricin ScFv from large phage library

Objective:To clone human anti-ricin antibodies from large phage antibody library.Methods:Panning for a large phage library against ricin toxin was conducted to select specific antibodies against ricin. The binding activities and specificities were tested by ELISA method. Soluble ScFvs were prepared through infecting E coli. HB2151 with the selected phage antibodies and induction with IPTG. Results:Forty positive clones were obtained after 5 rounds of panning, and 12 clones had specific binding ability to ricin toxin. DNA fingerprinting showed 7 different band patterns indicating 7 different positive clones. DNA sequencing showed that variable regions of these ScFvs belonged to different subgroups.Conclusion:Human anti-ricin antibodies were successfully obtained from large phage antibody library.
5.Research advances in determination of hepatitis B core antibody level

With the development and application of the double antigen sandwich method for quantification of hepatitis B core antibody (HBcAb) in recent years, there is increasing knowledge of the ability of HBcAb to reflect the body′s anti-viral capability. This article introduces the commonly used measurement methods for HBcAb and the new trends in HBcAb measurement and summarizes the association of serum HBcAb level with viral antigen and the body′s immune response, as well as research advances in effective prediction of antiviral effect with baseline HBcAb measurement before antiviral therapy. It is also pointed out that the clinical application of HBcAb needs further investigation.
6.Increased ostial pulmonary vein diameter in congestive heart failure: a multi-slice computed tomography angiography evaluation

Background and Objectives The relationship between left atrial (LA) size and congestive heart failure (CHF) is well recognized;however, there is little information on the association of pulmonary vein (PV) diameter and CHF.The purpose of this study was to investigate the changes of PV and LA sizes in CHF patients by multislice computed tomography (MSCT) angiography using a new 64-slice scanner. Methods and Results We assessed diameters of PVs ostium and LA by 64-slice MSCT with three-dimensional reconstruction in 25 CHF patients and in 26 age- and sex-matched non-CHF controls. Compared with controls, CHF patients showed significant greater diameters of left superior pulmonary vein (LSPV) and right inferior pulmonary vein (RIPV) in both anteriorposterior(AP) and superior-inferior (SI) directions (P<0.01), significant dilation of right superior pulmonary vein (RSPV) in AP direction (P<0.05), as well as significant increase of LA transverse, AP, and SI diameters (P<0.01). Conclusion Significant dilation of PVs with simultaneous LA enlargement was demonstrated in CHF patients. This anatomic and geometric changes may participate in the perpetuation of AF.
7.Dual-energy CT lung perfusion imaging and perfusion scintigraphy in rabbit models of acute peripheral pulmonary embolism

Objective To explore the clinical value of dual-energy CT lung perfusion imaging and perfusion scintigraphy in acute pulmonary embolism (PE) rabbit models. Methods Acute PE models were established in 20 rabbits with femoral vein injection of sponge as embolus materials, and 4 rabbits were injected saline as control group. Two hours after embolization, dual-energy CT and perfusion scintigraphy imaging were performed respectively. Taking the pathologicall findings as the reference standards, the sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV) of both imaging methods were compared. CT values and enhancement degree (overlay value) of the embolic areas and normal pulmonary parenchyma were measured in DEPI image after embolization. Results Normal lung was color-coded as homogeneously yellowish red, perfusion scintigraphy displayed rarefaction or absence. The sensitivity, specificity, PPV, NPV of DEPI was 100%, 97.50%, 95.24%, 100%, and of perfusion scintigraphy was 67.50%, 81.25%, 64.29%, 83.33%, respectively. Increased CT values and overlay values of embolism areas in DEPI images were lower than those of normal pulmonary parenchyma (P<0.05). Conclusion DEPI has higher sensitivity and specificity than perfusion scintigraphy for detection of pulmonary emboli in an acute PE rabbit models.
8.Computer simulation study on physiological feedback parameters during chest compression.

To have a thorough understanding of the CPR quality based on patients' various physiological states, the doctors must do something to simulate the chest compression physiological feedback parameters (CCPFP). The CCPFP simulation plays an important role in raising efficiency of CPR training and improving chest compression quality. In this study, the CCPFP, including cardiac output (CO), coronary perfusion pressure (CPP), partial pressure of End-tidal CO2 (PETCO2) and mean arterial relaxation pressure (MARP), was simulated using Charles F. Babbs' Model. Simulation results showed that the effect of compression depth upon CCPFP was important in the range of 2-6 cm, whereas compression rate had little effect on the CCPFP higher than 100/min; the thoracic factor is inversely proportional to the CCPFP with fixed compression depth and compression rate. The CCPFP simulation can be implemented at the various physiological statuses, and verified well with the animal experimental results and the clinical results.

9.Laparoscopic Diagnosis and Treatment of Small Bowel Tumors in 24 Patients

Objective To evaluate the clinical value of laparoscopy in the diagnosis and surgical treatment of small intestinal tumors. Methods After the establishment of pneumoperitoneum with umbilicus puncture, a laparoscope was inserted.Two 5-mm trocars were placed through lateral right and left rectus abdominis to explore the location of the small intestine tumors.A 3 -4 cm incision was made along the abdominal midline or lower abdomen median.With the use of incision protector,benign tumors were given local excision of small intestine,or malignant tumors were given radical resection.End to end or side to side anastomosis of bowel was carried out,and then the mesenteric hole was closed and the puncture hole sutured. Results All the patients were diagnosed and treated by laparoscopic procedure successfully, including 23 cases of laparoscopic-assisted partial intestinal resections and 1 case of right hemicolectomy.The mean operative time was(70.3 ±21.7)min,the mean blood loss was(18.5 ±3.1)ml,the mean length of incision was(3.5 ±1.1)cm, and the mean time for passage of flatus was(2.5 ±0.5)d.Postoperative pathological examinations showed benign tumors in 8 cases,most of which were leiomyoma(5 cases), and malignant tumors in 16 cases, most of which were stromal tumors(6 cases).The postoperative hospital stay was(6.5 ±2.5)d.Follow-ups were conducted for 0.5-4.5 years(mean, 2.6 years)in 19 cases,including 6 cases of benign tumor and 13 cases of malignant tumor.No recurrent tumor was found except one fatal case of advanced intestinal carcinoma. Conclusion Laparoscopic exploration can diagnose small intestinal tumors accurately, and laparoscopic-assisted surgical treatment is safe.
10.Self-efficacy and relevant influencing factors for cardiac disease patients when discharged

Objective To investigate the status of self-efficacy in cardiac disease patients when discharged, and to explore the influencing factors .Methods A total of 403 patients were selected from Cardiology and Cardiothoracic Surgery Department in a three-A hospital during March to August in 2013 .The chronic disease self-efficacy scale and the basic information questionnaire of cardiac disease patients ( self-designed) were administered to these patients when discharged .Differences of self-efficacy between medical and surgical patients were compared , and the influencing factors were analyzed .Results The average score of self-efficacy for these patients was (6.63 ±1.90), which belonged to the medium level.There was a significant difference between medical and surgical patients in self-efficacy (7.27 ±1.83 vs 6.01 ±1.75;t=-7.080,P<0.01).Univariate analysis showed that the influencing factors of self-efficacy in patients including age, education background , heart function level , and the number of hospital admissions and length of hospital stay . Conclusions The self-efficacy in cardiac disease patients is in the medium level , and the self-efficacy of surgical patient is lower than that in the medical patients .
