1.Changes of serum PCⅢ,Ⅳ-C,LN,TPO and EPO in patients with Leukemia

Objective To detect the levels of serum procollagen type Ⅲ (PCⅢ), Ⅳ collagen(Ⅳ-C) ,laminin ( LN ) , erythropoietin ( EPO ) and thrombopoietin ( TPO) in patients with acute and chronic leukemia. Methods PCⅢ , Ⅳ-Cand LN levels were determined by radioimmunoassay and TPO,EPO levels were determined by enzyme-linked immunosorbent assay,for 26 acute leukemia patients,16 chronic leukemia patients,and 16 normal controls. Results PC Ⅲ[(164.02 ± 118.53) μg/L], Ⅳ-C[(66.54 ± 19.48) μg/L] ,LN[(122.85 ±25.10)μg/L], EPO[(52.90±34.87)mU/ml]and TPO[(642.54±318.49)ng/L]levels of patients with acute and chronic leukemia were higher than that of the healthy control group before chemotherapy (P < 0. 05). EPO [( 72. 21 ± 52. 15 )mU/ml]and TPO[(642.54 ±318.49)ng/L]levels of acute leukemia patients after treatment were significantly lower than before treatment(P<0.05),while PC Ⅲ ,Ⅳ-C,LN levels had no significant difference(P >0.05). There was no significant difference before and after chemotherapy for PC Ⅲ, Ⅳ-C,LN,TPO and EPO levels of chronic leukemia patients (P> 0.05). Conclusion PC Ⅲ,Ⅳ-C,LN,TPO and EPO of patients with acute and chronic leukemia were abnormal, and dynamic monitoring of EPO and TPO levels could reflect the effect of AL chemotherapy, but conld not contribute to the prediction of CML blast crisis.
2.Studies on the Apoptotic Cell Death of Interleukin-2 Gene Modified Mouse Hepatoma Cells

Retroviral vector was used to introduce the interleukin - 2 (IL -2) gene into H22 cells, a BALB/c mouse hepatoma cell line. The IL-2 gene and marker gene (NeoR) were assessed by using PCR and RT-PCR methods. FACS analysis demonstrated that the cells with low DNA content in IL-2 gene modified H22 cells were more than these in control H22 cells. Electron microscopic morphological structure showed that some cells in IL-2 gene modified H22 went into apoptotic cell death. The study demonstrated that apoptotic cell death of H22 - IL-2 cells was induced by the IL-2 gene modification. It may be one of mechanisms of decreased tumorigenicity of IL - 2 gene modified tumor cells.
3.Study on Anti-Tumor Effect of Vaccination with Irradiated Tumor Cells Engineered to Secrete TNF-?

We compared the ability of tumorigenicity of TNF-? gene-modified and unmodified H22 tumor cells, and therapeutic functions of the irradiated cells. Results indicated that H22-TNF-? cells were less tumorigenic as compared to H22 and H22-LXSN cells. In case of treatment groups, injected with irradiated tumor vaccine in 3 or 6 days after inoculation of H22 cells, the tumor growth was suppressed.
4.Current status of standards for laboratory diagnosis of pertussis and clinical relevance of serological diagnosis

Despite high coverage of vaccinations, incidence of pertussis (whooping cough) has been increasing throughout the world and large outbreaks were reported in several countries.Adolescents and adults with atypical pertussis symptoms have become the main sources of infants′ pertussis.Since clinicians are lack of experience in making diagnosis based on atypical symptoms, laboratory methods are needed.The currently recommended laboratory methods include bacterial culture, PCR and serology ELISA.It is well known that sensitivity and specificity of the above-mentioned methods may vary depending on many factors such as status of vaccination, timing of specimen collection and onset of symptoms.Serology ELISA to measure specific anti-pertussis toxin IgG antibodies in serum has been proven to be one suitable method for the diagnosis of pertussis infection in adolescents and adults.In this review, we summarize the current status of methods used in China and other countries for the laboratory diagnosis of pertussis and discuss the clinical relevance of serology ELISA for pertussis diagnosis.
5.Construction of a vector suitable for making large phage antibody library

Objective:To construct a vector that suits the construction of large phage antibody libraries.Methods:Hie phage antibody vector p3MH was modified by insertion of synthesized oligos and PCR mediated site-specific mutations. Hie resultant vector was checked by expression of phage antibody, ELBA and in vivo recombination. Results: Vector pAL was obtained by following modifications of p3MH: insertion of Ioxp511 and loxp sequences, substitution of Lac promoter by Ara promoter, and modification of the cloning site for antibody genes. pAL was proved capable of expressing functional Fab phage antibodies under tighter control. In ere+ bacteria, pAL exhibited loxp-cre mediated recombination. Conclusion: pAL is useful for construction of large phage antibody libraries.
6.Activation of endogenous retinal stem cells in RCS rats during onset and development of retinal degeneration

Objective To investigate whether endogenous retinal stem cells can be activated in the retina of Royal College of Surgeons (RCS) rats during the onset and development of retinitis pigmentosa. Methods The RCS-p+ rats with inherited retinal dystrophy were divided into 3 groups: the initial stage group (15-day RCS rats), the mid-stage group (30-day RCS rats) and the advanced stage group (90-day RCS rats) according to the severity of degeneration (n=4 in each group). RCS-rdy+p+ rats without retinal degeneration served as controls, and divide into three groups (15-day control, 30-day control, 90-day control) matched with RCS-p+ rats. A transcription factor (Chx10) expressed by embryonic retinal progenitors was detected using immunofluorescence and Western blotting. Results All of the retinal layers in the three control groups and in the 15-day RCS rats did not express Chx10, while the positive expression was observed in the photoreceptor layers of the 30-day and 90-day RCS rats. 7.Comparison of electrophysiological and morphological properties of retinal ganglion cells in Long Evans and Wistar rats

Objective To compare the differences of the electrophysiological and morphological properties of retinal ganglion cells (RGCs) in Long Evans and Wistar rats. Methods Whole cell patch clamp recordings were made from RGCs in the acute retinal slices of Long Evans and Wistar rats aged postnatal 0-31 d. RGCs were stained with Lucifer Yellow diffused into cells during whole-cell recordings for the purpose of morphological study. The sections of rat retinas by HE staining were examined by light microscopy. Results The differences of electrophysiological properties in Long Evans and Wistar rats were not significant. There were pigment granules in retinal pigment epithelial cells of Long Evans but not in those of Wistar rats. Conclusion Long Evans rats can be the good animal model for visual and neural science research in place of Wistar rats.
8.Ultrasound-guided injection of sclerosant for the treatment of ovarian endometriosis cysts:therapeutic comparison between different sclerosants in 86 cases

Objective To compare the curative effects of CT-guided ethanol injection and lauromacrogol injection into the sac cavity in treating ovarian endometriosis cysts. Methods A total of 86 patients with ovarian endometriosis cyst were enrolled in this study. The patients were divided into ethanol group (n=44) and lauromacrogol group (n=42). Under CT guidance, injections of ethanol or lauromacrogol into the sac cavity of ovarian endometriosis cysts were respectively performed for the patients of both groups. The patients were followed up for six months, and the curative effects and the complications were analyzed. Results Six months after the treatment, the cure rates of ethanol group and lauromacrogol group were 95.50%and 92.86%respectively, and no statistically significant difference in cure rate existed between the two groups (P>0.05). The preoperative serum CA125 levels of the ethanol group and lauromacrogol group were (48.42±23.68)μg/L and(49.21±22.83) μg/L respectively, and the post operative ones were (23.56±5.89) μg/L and (25.49± 6.10) μg/L respectively; the differences between the preoperative data and the postoperative data were statistically significant in both groups (P<0.05), although the differences in serum CA125 levels between the two groups were not significant (P>0.05). The incidence of postoperative complications in the lauromacrogol group was obviously lower than that in the ethanol group (P<0.05). The cure time in the ethanol group was shorter than that in the lauromacrogol group, although the difference was not significant after six months. Conclusion For the treatment of ovarian endometriosis cysts, CT-guided lauromacrogol injection into the sac cavity has reliable curative effect. Compared to ethanol injection, injection of lauromacrogol is safer and has fewer adverse reactions. Therefore, this technique should be recommended in clinical practice. Serum CA125 can be used as an indicator for the evaluation of curative effect.
9.A study on pathway of apoptosis in murine thymocytesinduced by candida albicans

Objective:To investigate the pathway of C.albicans-induced murine thymocyte apoptosis.Methods:The chages of TNF-? level in murine plasma and caspase-3 activity were measured by ELISA and flurorescence spectrophoto meter respectively;FCM measurement was adopted for the expression levels of related genes.Results:The level of TNF rise,the caspase-3 activity serengthen significantly and bax,p53 gene expression are upregulated.Conclusion:It is possible that C.albicans elicits apoptosis in murine thymocytes by upregulating bax and p53 gene expression as well as raising the serum TNF-? level to strenghen the activity of caspase-3.
10.Investigating the killing effect of photodynamic therapy induced by different photosensitizers on human leukemia cell in vitro

Objective To investigate and compare the killing effect of photodynamic therapy (PDT)induced by hematoporphyrin derivative (HpD),hematoporphyrin monomethyl ether (HMME) and photocarcinorin (PsD007) on human leukemia cells K562 in vitro.Methods Human leukemia cells were cultured with serial concentrations of photosensitizers followed by irradiation of different dosage of laser light,then MTT colorimetric assay was applied to measure the relative survival rate of PDT for the cells.Results Significant difference in the inhibitory between the PDT group and control group was observed (P<0.05).The survival rate of PDT for the cells elevated along with the increase in the concentration of sensitizer and dose of laser light.When the photosensitizer concentration was bigger (25 μg/ml) or the energy density was bigger (7.2 J/cm2),the effect of PsD007 was better than HMME,and they were significantly better than HpD (P<0.05).Conclusion PDT has significant killing effect on human leukemia cells K562,and its relative inhibitory rate appears to be correlated with the dose of sensitizer and laser light irritation.The effect of PDT is related to the photosensitizers.The effect of HpD-PDT is not as effective as PsD007 and HMME.On the conditions of higher energy density and larger photosensitizer concentration,the effect of PsD007-PDT is better than HMME-PDT.
