1.Effect of acupuncture combined with acupoint sticking therapy on intractable hiccups of elderly patients with cerebral apoplexy

Objective To study the effect of acupuncture combined with acupoint sticking therapy on intractable hiccups of elderly patients with cerebral apoplexy. Methods Sixty patients with intractable hiccups after stroke were randomly divided into observation group and control group with 30 cases in each group. The control group was treated by routine nursing, while the observation group was treated by acupuncture on the accupoints of Neiguan, Zusanli, Zanzhu, Zhongwan, Fengchi, 2 times a day and meantime, the accupoint of Yongquan point was externally addressed with evodiarutaecarpa once before going to bed. The two groups were compared in terms of healing time and effectiveness time. Result The total effectiveness rate of the observation group was higher than that of the control group (P < 0.05). Conclusion Acupuncture combined with acupuncture addressing is effective in treating of intractable hiccups.
2.Acarophenax tribalii found in stored grains

Objective To observe and understand the morphology and structure of Acarophenax tribalii in order to create a new record of this mite in China. Methods The stored grain samples(rice and brown rice)were collected from the storage of lo?cal dwellers,and mites breeding in the products were isolated. Then mite samples were prepared and identified as the previous protocols,and the species was classified in accordance with Hughes. Results A. tribalii was detected in the grain samples. The light microscopic examination exhibited ovally shaped idiosoma of this species,and the abdomen of the male was more inflated than that of the female. By general view,the propodosomatal plate appeared triangle?like,and its anterior margin was covered by the gnathosoma. Leg I consisted of four segments,and its tibia and tarsus healing were fused into a stubby tibiotarsus. A tarsulus occurred at the end of legⅡ,ⅢandⅣ,with a claw born at each leg. Conclusion The morphology and structure of A. tribalii are characterized. This may lay a basis for following study on this species.
3.A new device and method for tail vein injection in mice

Objective To provide a device and an effective method for tail vein injection in mice. Methods Doing the tail vein injection in mice with the self-designed device which is consisted of constant temperature part, lighting part and holding part.The difficulty and time of injection with and without the device were compared.Results It was faster and more accurate to perform the tail vein injection in mice with this self-designed device.Conclusion Using this self-designed device can significantly improve the efficiency and save the injection time.
4.Efficacy of oxycodone for painless gastroscopy in pediatric patients when combined with propofol

Objective To evaluate the efficacy of oxycodone for painless gastroscopy in pediatric patients when combined with propofol.Methods Sixty pediatric patients of either sex,aged 2-7 yr,weighing 10-28 kg,of American Society of Anesthesiologists physical status Ⅰ or Ⅱ,scheduled for elective painless gastroscopy,were divided into 2 groups (n=30 each) using a random number table:oxycodone group (O group) and sufentanil group (F group).The pediatric patients held by their parents entered the gastroscopy room,and propofol 1.5-2.0 mg/kg and penehyclidine hydrochloride 0.3 mg were intravenously injected.Oxycodone 0.1 mg/kg and sufentanil 0.1 μg/kg were intravenously injected in O and F groups,respectively.When body movement occurred during operation,additional propofol 1.0 mg/kg was intravenously injected.The occurrence of adverse reactions such as intraoperative body movement,laryngospasm and respiratory depression was recorded.The occurrence of postoperative vomiting and pain (FLACC pain scale scores >3) was also recorded.Results The incidence of intraoperative body movement,laryngospasm and respiratory depression and postoperative pain was significantly lower in O group than in F group (P<0.05).Conclusion When combined with propofol,oxycodone produces better efficacy than sufentanil for painless gastroscopy in pediatric patients.
5.Design,preparation and screening of siRNA of proliferating cell nuclear antigen

AIM: To design,prepare and screen out functional small interfering RNA(siRNA) for specifically silencing proliferating cell nuclear antigen(PCNA) gene expression and effectively inhibiting cell proliferation on human hepatoma cell line SMMC-7721,human gastric carcinoma cell line SGC-7901 and human colorectal carcinoma cell line Caco2.METHODS: PCNA siRNA was designed based on previous studies about design guidelines and synthesized in vitro by T7 Mega short script reaction kit according to the manufacturer's instructions.After purification,the integrities of siRNA were identified through 19% denaturing polyacrylamide gel electrophoresis,and the concentrations of the generated siRNA were measured by testing the absorbance at 260 nm using a spectrophotometer.Four synthesized double-strand siRNA were transfected into three types of carcinoma cell lines by LipofectamineTM2000 reagent,respectively.WST-8 assay was employed to examine the proliferative inhibitions of these three cell lines.The subsequent alterations on PCNA mRNA and protein levels were determined by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) and immunocytochemistry,respectively.RESULTS: 3 sequences,No.2,No.3 and No.4 PCNA siRNA showed effective inhibitions on tumor cells among the 4 candidate siRNA,and a single dose of 50 nmol/L of No.2,No.4 PCNA siRNA showed the most effective inhibitory rates as more than 62% at 48 h after transfection.50 nmol/L of No.2 and No.4 PCNA siRNA transfection caused 72% decrease of PCNA mRNA level and almost completely loss of the protein in human Caco2 cells.CONCLUSION: No.2 and No.4 PCNA siRNA have been screened out in this study,which show the capability to effectively down-regulated PCNA expression and significantly inhibit the proliferation of carcinoma cells.The optimal concentration is 50 nmol/L and satisfactory effects are achieved 48 h after transfection in vitro.

Objective:To evaluate the effect of presurgical nasal mode(PNM)combined with nasal diorthosis in the treatment of na-sal deformity and incomplete unilateral cleft lip(IUCL)in infants.Methods:35 infants with IUCL were treated by PNM followed by nasal diorthosis and cheiloplasty.The nasal asymmetry was analysed by measurments of nostrils height,nostrils width and nasal colu-mella angle skewness on the photographs at the initial visit(T0),pre-operation(T1),1 week after operation(T2),1 month after opera-tion(T3)and a year after operation(T4).The other 35 infants with IUCL without PNM treatment were served as the controls.Re-sults:Compared with the controls,the symmetry of nostrils height,nostrils width,nasal columella angle skewness in PNM treated children were significantly improved at T0-T1 and T1-T2(P <0.05).there was no significant difference at T2-T3 and T3-T4(P >0. 05).Conclusion:Nasal asymmetry can be improved by presurgical nasal mode treatment followed by preliminary nasal deformity di-orthosis and cheiloplasty.
7.Immunogenicity of Poria cocos polysaccharides PCP-Ⅰand PCP-Ⅱas vaccine adjuvants

OBJECTIVE To investigate the immunogenicities of Poria cocos polysaccharides, PCP-Ⅰand PCP-Ⅱ, as a vaccine adjuvant. METHODS ①Keyhole limpet hemocyanin (KLH) was linked to PCP-Ⅰor PCP-Ⅱrespectively to prepare immuno-antigen KLH-PCP-Ⅰor KLH-PCP-Ⅱ. Bovine serum albumin (BSA) was also linked to PCP-Ⅰor PCP-Ⅱrespectively to prepare screening-antigen. Rabbits were immunized with KLH-PCP-Ⅰor KLH-PCP-Ⅱplus Freund adjuvant by intradermal injection twice, and serum specific antibody titers were determined by ELISA. ②BALB/c mice were immunized with PCP-Ⅰ or PCP-Ⅱ alone intramuscularly twice, and serum polysaccharide antibody titers were determined by ELISA.③BALB/c mice were co-immunized intramuscularly or subcutaneously with PCP-Ⅰor PCP-Ⅱplus hepatitis B surface antigen (HBsAg) or porcine reproductive and respiratory syndrome virus inactivated vaccine (PRRSV) twice, and serum polysaccharide-antibody titers were determined by ELISA. RESULTS ①Serum anti-KLH and anti-polysaccharides (PCP-Ⅰor PCP-Ⅱ) antibodies were pro?duced after rabbits were immunized with KLH-PCP-Ⅰor KLH-PCP-Ⅱplus Freund adjuvant twice.②Serum anti-PCP-Ⅰor anti-PCP-Ⅱantibodies were not found after mice were immunized with PCP-Ⅰand PCP-Ⅱalone twice.③After mice were immunized with HBsAg or PRRSV plus PCP-Ⅰor PCP-Ⅱtwice, serum anti-PCP-Ⅰor anti-PCP-Ⅱantibodies were not found. CONCLUSION PCP-Ⅰand PCP-Ⅱshow weak immunogenicity, which may be quite safe as a vaccine adjuvant.
8.Different expressions of p53 gene family members and their clinical significance in human non-small cell lung cancer.

BACKGROUNDTo investigate the different expressions of p53 gene family members p53, p63 and p73, and their clinical significance in non small cell lung cancer (NSCLC).
METHODSp53, p63 and p73 protein expressions were detected in 60 NSCLC tissues and 7 normal lung tissues by immunohistochemistry.
RESULTSIn NSCLC, positive rate of p53, p63 and p73 protein was 61.67%(37/60), 80.00%(48/60), 73.33% (44/60) respectively. There were significant differences in positive rate of three proteins as compared to normal lung tissue ( P < 0.05). p53 protein expression was closely associated with tumor cell differentiation degree ( P =0.023), but was not associated with histological classification, lymph node metastasis and clinical stages ( P > 0.05). Expression of p63 protein was closely related to lymph node metastasis ( P =0.028) and histological classification ( P =0.001), but not to cell differentiation degree and clinical stages ( P > 0.05). There was no significant relationship between p73 protein expression and clinical characteristics of NSCLC ( P > 0.05). A positive correlation was present between p63 and p73 protein expressions ( P =0.000 1). No statistical correlation was found between p53 and p73 ( P > 0.05).
CONCLUSIONSp53 gene family may be related to the oncogenesis and development of NSCLC. p63 and p73 proteins may have different biological function from p53 protein, and both might play oncogenic roles.
9.Therapeutic Effects of rhIL-11 on Carboplatin-Induced Thrombocytopenia in Monkeys

A model of myelosuppression with thrombocytopenia was produced in monkey by i.v. administration of carboplatin to the evaluate effects of rhIL-11 treatment in monkeys. Following myelosuppression, rhIL-11 was subcutaneously injected for 19 consecutive days at the dose of 50 or 100 micro g/kg. In myelosuppressed monkeys treated with rhIL-11, peripheral blood platelet started to drop at the day 8 after the administration of carboplatin, and reaching the nadir between the day 12 - 14, the decrease in blood platelet was less severe compared with untreated monkeys; peripheral blood platelet began to recovery on day 11 - 13 (D14 - D16) after rhIL-11 treatment, and reached or surpassed the baseline value before carboplatin administration after 13 - 15 days rhIL-11 treatment. Blood platelet counts remained high level after discontinuation of rhIL-11 administration and returned to baseline after 4 days. The results demonstrated that rhIL-11 has a significant thrombopoietic activity, it can reduce the severity of thrombocytopenia as well as shorten the duration of thrombocytopenia caused by myeloablastive agents, and is likely to become an effective agent against thrombocytopenia induced by chemotherapy.
10.Promotion of Thrombopoiesis by rhIL-11 in Normal and Myelosuppressed Mice

Hematological effects of rhIL-11 on normal and myelosuppressed male BALB/c mice were observed. Mice were subcutaneously injected with rhIL-11 for 7 consecutive days, at the dose of 200 or 400 micro g/kg per day, peripheral blood platelet counts were moderately elevated on 5 days after administration and returned to base level within 4 days after discontinuation of injection. In myelosuppressed mice, treatment with rhIL-11 significantly ameliorated the degree of thrombocytopenia, the recovery of thrombocytopenia was also significantly accelerated at the dose range of 100 - 400 micro g/kg per day, and blood platelet counts reached pre-irradiated level after 13 - 15 days of treatment. The magnitudes of platelet count elevation were similar among groups of 100, 200 and 400 micro g/kg per day, although recovery appeared earlier in group of 400 micro g/kg per day. Significant increases in CFU-Meg were observed both in normal and myelosuppressed mice. Our results suggest that rhIL-11 promotes the increase of peripheral blood platelets both in normal and myelosuppressed mice, and can be used as a potential therapeutic agent for thrombocytopenia induced by chemotherapy.
