1.Effect of remote limb ischemic postconditioning combined with electro-acupuncture postconditioning on focal cerebral ischemia-reperfusion injury in rats

Objective To evaluate the effect of remote limb ischemic postconditioning (RLIP) combined with electro-acupuncture postconditioning (EAP) on focal cerebral ischemia-reperfusion (I/R) injury in rats.Methods Seventy male Sprague-lDawley rats,weighing 220-250 g,were randomly assigned into 5 groups (n =14 each) using a random number table:sham operation group (group S),group I/R,RLIP group,EAP group and RLIP combined with EAP group (group RLIP +EAP).Focal cerebral I/R was induced by middle cerebral artery occlusion (MCAO).MCAO was maintained for 120 min.The animals were subjected to 3 cycles of 15 min ischemia of bilateral hind limbs followed by 30 s reperfusion starting from onset of reperfusion.Dazhui and Baihui acupoints were stimulated with electric stimulator (frequency 15 Hz/2 Hz,intensity 1 mA) for 30 min starting from onset of reperfusion.Neurological deficit scores (NDSs) were evaluated at 24 h of reperfusion.The animals were then sacrificed and brains were removed to measure the infarct size and for microscopic examination of pathological changes in hippocampal CA1 region.The number of survival neurons was counted.The apoptosis in neurons was determined by TUNEL.Apoptosis index was calculated.Results Compared with group S,the NDSs,percentage of cerebral infarct size and AI were significantly increased,and the number of survival neurons was decreased in the other four groups.Compared with group I/R,the NDSs,percentage of cerebral infarct size and AI were significantly decreased,and the number of survival neurons was increased in EAP,RLIP and RLIP+EAP groups.Compared with RLIP and EAP groups,the NDSs,percentage of cerebral infarct size and AI were significantly decreased,and the number of survival neurons was increased in group RLIP+ EAP.Conclusion The combination of RLIP and EAP provides better efficacy than either alone in attenuating focal cerebral I/R injury in rats.

Objective To solve completely the problem which the software in No.1 Military Medical Project do not be updated automatically in C/S mode.Methods By directly connecting log in the database server,the query table whether there are new procedures and downloaded to a local client,and then run the client grogram.Results Not only the program was updated automatically but also the program can be started automatically and the many attached programs may be updated in the same time.Conclusion With the programs updated automatically,the workload of engineers and errors are reduced,and the work efficiency is also improved greatly.
3.The effect of interferon ?-2a on the radiosensitivity for nasopharyngeal carcinoma cell line CNE-I

Objective To discuss the effect of interferon ?-2a on the radiosensitivity and cell cycle for nasopharyngeal carcinoma cell line CNE-I. Methods Interferon ?-2a was given by different concentration. Then the cells were radiated with X-ray (6?MV) and the cell survival rate was calculated. The change of cell cycle dynamics was measured with flow cytometry. Results The cell survival fraction was 0.62, 0.43 and 0.20 respectively after the interferon ?-2a was given by different concentration(0.5?106, 1.0?106 and 1.5?106?IU/L). The radiosensitization ratio was 1.16, 1.57 and 1.93 respectively. Compared with the control group, increasing cell percentage in G_1 and G_2+M phage and decreasing cell percentage in S phage were observed on 24 h after the interferon ?-2a(1?106?IU/L) was given(P
4.The reliability and validity of the Chinese version of Infant Feeding & Nutrition Checklist For Congenital Heart Disease

Objective To translate the English version of Infant Feeding & Nutrition Checklist For Congenital Heart Disease (IFNC:CHD) into Chinese,and to test the reliability and validity of the Chinese version of IFNC:CHD.Methods After obtaining authorization from the developer,the IFNC:CHD was translated and culturally adapted into Chinese version.The reliability and validity of the Chinese version of IFNC:CHD were tested in 168 patients.Results The Cronbach's α coefficient was 0.804.Two nurses used the scale to evaluate the consistency of the results,and the Kappa coefficient was 0.812.The content validity index of item ranged from 0.80 to 1.00,and the average index was 0.90 for the total scale.The correlations coefficients between each dimension score and the total score ranged from 0.339 to 0.474 (P<0.01),and the correlations coefficients between each dimension score ranged from 0.198 to 0.861(P<0.01,P<0.05),Three factors were extracted from exploratory factor analysis,and the cumulative variance contribution rate was 72.66%.Conclusion The Chinese version of IFNC:CHD has been proved to have good reliability and validity.It can be used to assess infant feeding and nutritional risk of congenital heart disease in the Chinese settings.
5.Study on the construction of job evaluation index system for clinical nurse specialist in comprehensive children′s hospital

Objective To formulate job evaluation index system for clinical nurse specialist in comprehensive children′s hospital. Methods A total of 19 experts were consulted by two-round Delphi method to ensure the item of index system, the order and weight of each index was determined by hierarchy analysis. Results The evaluation index system included 5 level-1 dimensions, 13 level-2 dimensions and 30 level-3 items. The authority of the expert advisory coefficient was 0.84, determine coefficient was 0.91, familiarity cofficient was 0.77. The coordination coefficient of one, two, three indexes were 0.44, 0.52, 0.51, with statistical significance (P < 0. 05). Conclusions The job evaluation index system is reliable and valid, which can provide basis on job management.

BACKGROUND: CpG oligodeoxynucleotide (ODN) is a type of highly effective immune adjuvant with low toxicity, which has an extensive application in gene therapy for many diseases. However, the specificity for species and cells leading to low uptake by cells and degradation by nuclease blocks its clinical application. OBJECTIVE: To explore the specific delivery and its immunologic efficacy of CpG ODN targeting B lymphocytes of umbilical cord blood by CD40 ligand-receptor-mediated carrier system. DESIGN, TIME AND SETTING: An observation and control experiment was performed at the Department of Hematology, and Department of Pediatric, the First Affiliated Hospital of Jinan University from April 2004 to October 2007. MATERIALS: Fresh umbilical cord blood with heparin was obtained from healthy, natal infant. Informed consent was obtained from his parents, and the experiment was approved by the hospital Ethics Committee. METHODS: CD40 ligand (CD40L)-EDC-PLL-CpG ODN conjugated complex was prepared. Mononuclear cells (MNCs) from umbilical cord blood were co-cultured with conjugated complexes. Uptake rate, mean fluorescence intensity of FAM marked CpG ODN, expressions of MNCs, proliferations of lymphocytes and the IgG levels of culture supematants were detected by flow cytometry, fluorescence techniques, MTT assay and ELISA, respectively. MAIN OUTCOME MEASURES: The uptake rate, the mean fluorescence intensity of CpG ODN by MNCs, subgroups and proliferations of lymphocytes, and IgG levels of culture supematants. RESULTS: Compared to the pure CpG ODN group, the uptake rate of the conjugated complexes group was higher (98%), the peak level of up-taking occurred earlier, and intracellular fluorescence intensity maintained much more stable. Expressions of CD19+, CD22+, and CD20+ was increased, A value and IgG levels in supematants were all higher than that of the control group. CONCLUSION: CD40 tigand-receptor-mediated carrier system is helpful for CpG ODN delivery targeting to B lymphocyte, enhancing its immunological efficiency.
8.Brain injury induced by ischemia-reperfusion of rat hindlimbs and its mechanisms

AIM:To investigate the pathologic changes in the brain and its underlying mechansims during ischemia-reperfusion of rat hindlimbs.METHODS:SD rats were divided into the normal(N), sham(S), 4 h ischemia without reperfusion(I), and 4 h ischemia-2, 6,12,18 or 24 h reperfusion (I-R) groups at random. Ischemia and ischemia-reperfusion were established with the occlusion or/and re-opening of the terminal of abdominal aorta, respectively. The pathologic changes in the brain tissue were morphologically observed. The expression of inducible nitric oxide synthase (iNOS) mRNA, and iNOS protein and the nitrotyrosine, a marker of peroxynitrite (ONOO-),in the brain tissue were detected with RT-PCR and immunohistochemical technique, respectively. The brain superoxide dismutase (SOD) activity and malondialdehyde (MDA) contents were spectraphotometrically measured.RESULTS:Hydropic degeneration and severe injury to neurons were only showed in I-R group. Expressions of iNOS mRNA and protein were demonstrated in I-R, I and S groups, which were maximal in I-R 6 h group. iNOS positive neurons and microglias were more spread in I-R 6 h group than those in S and I groups. NT positive neurons were localized in the cerebral cortex and hippcampus of I-R 6 h group. The contents of MDA markedly increased, while the activity of SOD significantly decreased in I-R 6 h group compared to the N, S and I groups. There were no significant changes in MDA and SOD in N, S and I groups.CONCLUSION:Severe ischemia-reperfusion of rat hindlimbs could induce brain injury, and its mechanisms might be related to enhanced expression of iNOS-NO-ONOO- in the brain.

10.Changes of HO-1 gene expression in brain following ischemia and reperfusion of limbs

AIM: To detect the changes of heme oxygenase-1(HO-1) expression in brain following ischemia-reperfusion of hindlimbs in rats and to elucidate their significance. METHODS: Ischemia-reperfusion was established using the occlusion for 4 h and re-opening for 2-24 h of the femoral arteries. The expression of HO-1 mRNA in brain was detected with reverse transcription-polymerase chain reaction (RT-PCR). The expression and location of HO-1 protein were detected with immunohistochemical technique. The observation of pathologic changes of brain was made following the inhibition of HO-1 by zinc protoporphyrin (ZnPP). RESULTS: The relative expression level of HO-1 mRNA significantly increased in I-R group, compared to those in control group.
