1.Evaluation of the life quality in the floating population tuberculosis patients

Objective To evaluate the life quality in front and at the back of the cure of the floating population tuberculosis patients.Methods The SF-36 scale was applied to the new smear-positive pulmonary tuberculosis patient in floating population using face to face interview in Shenzhen 6 districts.The contrast was tuberculosis patient in the population of the household register,the people stayed temporarily in the same term.Results The score of general health(43.36 ± 15.67),vitality(57.96 ± 16.54) and mental health(61.46 ± 14.26) in the floating population was lower than the appraisal result with the population of the household register(49.28 ± 17.85,66.38 ± 17.03 and 66.70 ± 15.12),the people stayed temporarily(45.00 ± 17.22,60.99 ± 16.59 and 63.13 ±13.71 ) (P < 0.05 ).It was still lower than the other two groups after 2 (3) months and 6 (7) months (P < 0.05 ).But the score of social functioning in floating population ( 88.92 ± 14.88 ) was higher than the other two groups (82.62 ± 14.44,68.02 ± 27.30).The change rate of general health ( (76.05 ± 132.82) %,(69.32 ± 111.98 ) %and (48.36 ± 101.31 ) % ) and social functioning scores( (46.49 ± 85.18 ) %,( 26.33 ± 50.25 ) % and ( 51.09±73.44)% ) were significant difference in the three groups(x2 =12.088,P=0.002;x2 =11.611,P=0.003).The change rate of the general health score in floating population was higher than the household register population and people stayed temporarily.Conclusion It is necessary to implement the management in whole course to the floating population tuberculosis patients.
2.Non-contact Coculture Reveals a Comprehensive Response of Chondrocytes Induced by Mesenchymal Stem Cells Through Trophic Secretion

Coculture between mesenchymal stem cells (MSCs) and chondrocytes has significant implications in cartilage regeneration. However, a conclusive understanding remains elusive. Previously, we reported that rabbit bone marrow-derived MSCs (rbBMSCs) could downregulate the differentiated phenotype of rabbit articular chondrocytes (rbACs) in a non-contact coculture system for the first time. In the present study, a systemic investigation was performed to understand the biological characteristics of chondrocytes in coculture with MSCs. Firstly, cells (MSCs and chondrocytes) from different origins were cocultured in transwell system. Different chondrocytes, when cocultured with different MSCs respectively, consistently demonstrated stimulated proliferation, transformed morphology and declined glycosaminoglycan secretion of chondrocytes. Next, cell surface molecules and the global gene expression of rbACs were characterized. It was found that cocultured rbACs showed a distinct surface molecule profile and global gene expression compared to both dedifferentiated rbACs and rbBMSCs. In the end, cocultured rbACs were passaged and induced to undergo the chondrogenic redifferentiation. Better growth and chondrogenesis ability were confirmed compared with control cells without coculture. Together, chondrocytes display comprehensive changes in coculture with MSCs and the cocultured rbACs are beneficial for cartilage repair.

3.Design of Experiment Platform for Noninvasive Ventilator Based on LabVIEW.

OBJECTIVE:
The study presents an experiment platform to perform ventilation and develop the control strategy for ventilator.
METHODS:
The software designed by LabVEIW displays and saves data in ventilation support through connecting with data acquisition device and signal processing circuit, which is improved by control strategy in order to achieve the more scientific ventilation.
RESULTS:
The experiment platform realizes the ventilation support and is close to the data outputted from ventilator.
CONCLUSIONS
The platform provides values for ventilation control strategy in development phase.

The rehabilitation compliance of stroke patients is generally low.Evaluating the rehabilitation motivation of patients is helpful to promote the rehabilitation management of patients,enhance the rehabilitation enthusiasm and compliance of patients,and improve the rehabilitation outcome.This paper reviews the existing stroke patients rehabilitation motivation assessment tools,and expounds the main contents,application status,characteristics and limitations of stroke patients rehabilitation motivation assessment tools,in order to provide references for the appropriate selection of clinical assessment tools,the rehabilitation management of stroke patients and the development of domestic localized stroke rehabilitation motivation assessment tools.
5.Study on the drying processing methods of Peucedanum praeruptorum in producing area

OBJECTIVE To compare the effects of different drying processing methods in the producing area on the quality of Peucedanum praeruptorum, and screen the optimal drying processing in the producing area. METHODS P. praeruptorum was processed by traditional drying in the sun, drying in the shade, and modern drying processing methods such as electric heating and blast drying, vacuum drying, infrared drying, and vacuum freeze drying. Technique for order preference by similarity to ideal solution (TOPSIS) method was used to perform the comprehensive quality evaluation using drying rate, water content, extract content, color chroma value, and the contents of 4 coumarins as indexes. RESULTS Under different drying processing methods, the drying rates of 12 samples of P. praeruptorum were 31.56%-40.10%; the moisture contents were 6.96%-8.58%, and the extract contents were 27.56%-43.10%; the color chroma values red green degree, yellow blue degree and brightness were 1.80-7.50, 19.90- 30.20, 46.90-59.90, respectively. The contents of umbelliferolactone, praeruptorin A, praeruptorin B and praeruptorin E were ≤ 0.05, 3.70-14.05, 0.72-2.37, 0.81-3.90 mg/g, respectively; and the total contents were 5.47-19.65 mg/g. The comprehensive score of P. praeruptorum was the highest under the condition of drying at 40 ℃ by electric heating and blast . CONCLUSIONS The drying method at 40 ℃ by electric heating and blast is the first choice for P. praeruptorum in the producing area.
6.Mental health of front-line staff in prevention of coronavirus disease 2019.

OBJECTIVES:
The coronavirus disease 2019 (COVID-19) pandemic is a global public health crisis, which elicits psychological problems in different population This study is to investigate the impact of COVID-19 on mental health in the front-line staff.
METHODS:
Patient Health Questionnare-9 (PHQ-9), Self-Rating Anxiety Scale (SAS), and Fatigue Self-assessment Scale (FSAS) were used to assess the depression, anxiety, and fatigue in front-line staff.
RESULTS:
The detection rates of depression, anxiety, and fatigue were 49.1%, 21.8%, and 76.0% among the front-line staff. The rates of depression, anxiety, and fatigue in community workers were higher than those in medical workers and other occupational staff (<0.01). The PHQ-9 of front-line staffs was negatively correlated with age, family income, family members' support, satisfaction of service objects, and sleep quality (all <0.01), while positively correlated with education level, fatigue, fear of pneumonia, and the duration of daily attention to the COVID-19 (all <0.01). SAS was negatively correlated with age, family income, family support, satisfaction of objects service, and sleep quality (all <0.01), while positively correlated with gender, fatigue, fear of pneumonia, and duration of daily attention to the COVID-19 (all <0.01).
CONCLUSIONS
The front-line workers should manage work and rest time reasonably to adjust their negative mood and fatigue. The government and the society should pay more attention to the psychological state of the front-line staff, particularly for the staff working in the community or villages and towns in preventing the COVID-19 pandemic. Thus, front-line staff can be obtained mental intervention or be taken a rest from the high-intensive work.

7.CRISPR-assisted transcription activation by phase-separation proteins.

The clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 system has been widely used for genome engineering and transcriptional regulation in many different organisms. Current CRISPR-activation (CRISPRa) platforms often require multiple components because of inefficient transcriptional activation. Here, we fused different phase-separation proteins to dCas9-VPR (dCas9-VP64-P65-RTA) and observed robust increases in transcriptional activation efficiency. Notably, human NUP98 (nucleoporin 98) and FUS (fused in sarcoma) IDR domains were best at enhancing dCas9-VPR activity, with dCas9-VPR-FUS IDR (VPRF) outperforming the other CRISPRa systems tested in this study in both activation efficiency and system simplicity. dCas9-VPRF overcomes the target strand bias and widens gRNA designing windows without affecting the off-target effect of dCas9-VPR. These findings demonstrate the feasibility of using phase-separation proteins to assist in the regulation of gene expression and support the broad appeal of the dCas9-VPRF system in basic and clinical applications.
