1.Clinical Observation on New Adjuvant Chemotherapy with THP-containing Regimen for the Treatment of Locally Advanced Breast Cancer

Objective To observe the THP for locally advanced breast cancer in recent efficacy and toxicity. Methods 32 patients with locally advanced breast cancer received chemotherapy: THP 50mg/m~2 , intravenous injection; day 1: CTX600mg/m 5 - FU500mg/m~2, ivgtt on day 1 and day 8,for a period of 21 days. Results 73% had efficiency,with small heart poisonous side effects. Conclusion New adjuvant chemotherapy with THP - containing regimen for the treatment of locally advanced breast cancer has a higher efficacy, and adverse reactions can be tolerated.
2.Construction and expression of human interleukin-13 and its mutant and subcellular location

Objective:To construct a eukaryotic expression vector for wild-type human Interleukin-13(whIL-13)and its mutant(mhIL-13) gene fused with enhanced green fluorescent protein(EGFP) and to analyze the expression and subcellular location of the fusion protein in COS-7 cells.Methods:The whole whIL-13 and mhIL-13 coding gene was amplified by RT-PCR. And then the expression vector for EGFP-whIL-13, EGFP-mhIL-13 fusion protein were constructed.The recombinant fusion gene were transfected into COS-7 cells.The expression and subcellular location of the fusion protein were detected by confocal microscopy.Results:The construction of expression vector were all corrected.In the positive cells, green fluorescent was found to be localized in cytoplasm,while empty in nucleus.Conclusion:The expression vectors for EGFP-whIL-13, EGFP-mhIL-13 fusion protein were constructed successfully and expressed in COS-7 cells. Fusion proteins were distributed in cytoplasm and no significant difference was observed between them.
3.Improvement of determination of related substances in piracetam and its injection by HPLC

Objective To improve and establish an RP-HPLC method for the determination of related substances of piracetam and its injection. Methods Using thermo syncronis C18(4.6 mm ×250 mm,5μm)chromatographic column, mobile phase was acetonitrile-phosphate buffer (adjust pH of 1.0 g/L potassium hydrogen phosphate solution to 6.0 with phosphoric acid), gradient elution with initial mobile phase 5% acetonitrile.The flow rate was 1 mL/min, detected at 205 nm.Five calibration factors of the known impurities were separately measured.the related substances were determined using main component self-compare with calibration factor.Results Piracetam and their related substances were well separated.The calibration curves of five known impurity A~E were linear within the concentration range measured.The average recoveries were 99.46%,98.21%,97.22%,100.23% and 97.58%, respectively(n=9), RSDs of the average recoveries were lower than 2.0%.The calibration factors of five known impurity A ~E were 1.1, 1.4,1.5,0.8, 1.4, respectively.Conclusion The method can be used to determine the related substances in piracetam and its injection.The related substances can be determined accurately using main component self-compare with calibration factor.

Saponin and sapogenin are main components in chinese medicine Anemarrhena asphodeloides Bge.Modern research has shown that Anemarrhena asphodeloides Bge and its effective components could markedly enhance the ability of learning and memory in dementia animals,as well as improve the descent of brain function.The effect was related with many factors such as inhancing the cholinergic receptor(M,N),improving the activity of ChAT and inhibiting the activity of AChE,regulating the balance of ? receptor-cAMP and M receptor-cGMP,improving the metabolite of free radicals in model animal brains and so on.
5.Effects of valsartan on nuclear factor-?B protein expression in the kidney of spontaneously hypertensive rats

AIM: To observe the local expression of nuclear factor-?B(NF-?B) in spontaneously hypertensive rat(SHR) kidney and effects of AT1 receptor contagonist valsartan.METHODS: 16 SHRs were randomly divided into two groups: SHR control group and valsartan group.Another 8 WKY rats act as normal control group.Systolic blood pressure(SBP) of SHR was measured at the beginning and the end of 2,4,6 and 8 weeks of intervention treatment.Radioimmunoassay was used to determine the activities of rennin and angiotensin II(AngII).The renal tissue NF-?B protein expression was detected by immunobiochemistry.RESULTS: SBP of SHR was remarkably decreased after valsartan intervention.However,the rennin activities and AngII level in plasma increased in valsartan group.In the renal tissue of SHR,there was remarkably increased in expression of NF-?B protein.Valsartan could significantly reduced NF-?B expression.CONCLUSION: Valsartan can depress NF-?B renal expression in protein level and might benefit hypotension renal function.
6.Research on the present situation of Hangzhou medical service scientific competitiveness

Objective We try to find the current situation of Hangzhou health research.Methods Using established evaluation index system to evaluate 8 medical service.Results Scientific research development level of 8 medical service is unbalanced,but input-output is balance.Conclusions According to the appraisal results,we suggest reduce the gap among the scientific level of medical service,train high-level personnel,strengthen the construction of key discipline and specialties,create the atmosphere of project application,and establish scientific research incentive system.

8.Synergetic Inhibition Effects of Methioninase and Cisplatin against the Growth of Human Lung Adenocarcinoma Cell Line GLC

Objective To purify recombinant methioninase and investigate the synergetic inhibitory effect of methioninase and cisplatin on the proliferation of human lung adenocarcinoma cell line GLC. Methods Recombinant methioninase was purified with GST-column from supernatant after ultrasonic disruption of cultured Escherichia coli in the prokaryotic expression system pGEX-4T-1-Met/Dh5a. MTT assay was used to determine the inhibition rate of methioninase in combined with cisplatin on cell proliferation,and their synergistic effect was evaluated by using the q value judge method. Results The concentration of recombinant methionine cleaving enzyme was 0.22 mg/mL, the purity was 95%, and the activity was 0.568 IU/mg. After 72 h culturing, the inhibition rate of cisplatin methionine was 24.80%and 27.49%respectively,while the inhibition rate of the combined drugs was 66.80% ( =1.47>1.15) which showed a significant synergistic effect. Conclusion Both methioninase and cisplatin have the inhibition effect,and the combined drugs display a significant synergistic effect on the proliferation of GLC.
9.Effects of pervanadate on radiation-induced G2/M arrest and apoptosis in BET-2 cells

Objective To study the effects of Pervanadate(Per) on BET-2 cells cell cycle arrest and apoptosis induced by ionizing radiation. Methods The BET-2 cells were radiated with 0, 1, 3, 5, 7, 10 Gy and divided into PTP(protein tyrosine phosphatases)-specific inhibitor Per-treated group and non-pervanadate group(served as a control) and then incubated after ?-ray irradiation. Cell cycle and apoptosis were measured by FCM and Hematoxylin-Eosin staining at different time phases. Results G2/M phase arrest and apoptosis were induced in a pattern of dose-effect and time-course after irradiation in the BET-2 cells. In Per-treated group, G2/M phase arrest increased more notably, and apoptosis decreased markedly. Conclusions These studies suggested that Pervanadate potentialy enhance the ratio and prolong the term of G2/M phase arrest, and facilitate the damaged hematopoietic cells to be repaired, remarkably prevent radiated-cells from apoptosis.
10.Comparison of scientific research papers from 2000 to 2009 of 15 general hospital in subprovincial city in China

To reveal the scientific research situation of general hospitals in recent years in china,the authors retrieved papers included in CBM、CCD and ISI Web of Knowledge,and compared the production of papers from the general hospital in 15 sub-provincial cities during the period of 2000 -2009.Also the citation frequency of all papers and the average citation frequency were compared.
