1.A Study on the Effects of Interferon-alpha on Hepatitis B virus in Vitro

Objective To evaluate the in vitro inhibiting activities of interferon-alpha (INF-?) against hepatitis B virus (HBV). Methods HepG2 2 2 15 cells were treated with INF-?. At 3rd, 6th and 9th days after treatment, the supernatant was collected for HBsAg quantitative assay, and total RNA of the cells was isolated for RT-PCR and realtime-PCR assay of HBV mRNA. Results There were no significant differences in HBsAg titer and virus mRNA level at 3rd and 6th days between the experimental and control groups. At 9th day, HBsAg titer and virus mRNA level in INF-? group were significantly lower than those in control group. And 500IU/ml INF-? could get maximal inhibiting effect against HBV. Conclusion INF-? can inhibit transcription and expression of HBV gene in HepG2 2 2 15 cells, which indicated that INF-? can inhibit HBV replication directly.
2.Chemical constituents of whole herb of Dicliptera chinensis

Objective To study the chemical constituents in the whole herb of Dicliptera chinensis.Methods Separation and purification were conducted by silica gel column chromatographic and preparative TLC methods,the structures were identified by IR,NMR,and MS.Results Thirteen compounds were isolated from petroleum ester extract fraction.Their structures were identified as hexatriacontanol(Ⅰ),stearic acid(Ⅱ),lupenone(Ⅲ),lupeol(Ⅳ),4-sitost-4-en-3-one(Ⅴ),stigmast-5-en-7-oxo-3?-yl palmitate(Ⅵ),?-sitosterol(Ⅶ),oleanolic acid(Ⅷ),3?,6?-stigmast-4-en-3,6-diol(Ⅸ),6?-hydroxy-stigmast-4-en-3-one(Ⅹ),3?-hydroxy-stigmast-5-en-7-one(Ⅺ),dehydrovomifoliol(ⅩⅡ),and vomifoliol(ⅩⅢ).Conclusion All the compounds except Ⅱ and Ⅶ are obtained from the plants of Dicliptera Juss.for the first time.
3.Development and Application of a New Clinic Drug Dispensing Information System for Automated Dispensing and Rational Drug Use

OBJECTIVE: To promote automated dispensing and rational drug use in the inpatient dispensary by applying information technology.METHODS: Powerbuilder9.0 development software and bar code technology were used to build a new drug unit dose automated dispensing system based on hospital information system.RESULTS: Two sub-system programs including clinical prescription screening and transmitting function were developed,and the rational drug use examination and reporting system was build up,which enhanced the efficiency of clinical dispensing,facilitated the accurate management of the drug stock amount in the medicine box of the machine.CONCLUSIONS: To develop software by taking consideration of the developmental trend of pharmacy can better meet the demand and adapt to the changing work flow in drug dispensing.
4.In vitro antiviral efficacy of mycophenolic acid against hepatitis B virus

Objective To evaluate the in vitro inhibitive activities of mycophenolic acid against hepatitis B virus in vitro. Methods The different concentrations of the test drugs in DMEM were added to the well with the monolayer growth of HepG2.2.15 cells. The culture medium was refreshed with DMEM containing the appropriate drugs every 3 days. At 3rd, 6th and 9th days, the supernatant was collected for HBsAg quantitative assay, total RNA was isolated from cells for RT-PCR and realtime-PCR assays. Results It was found no significant anti-HBV effect of mycophenolic acid on concentration of 2~250 ?g/ml, and mini inhibitory efficacy of HBV replication on concentration of 1250 ?g/ml. There was significant anti-HBV effect of lamivudine on concentration of 5 ?g/ml. When combined with lamivudine mycophenolic acid can significantly potentiate anti-HBV activities of lamivudine, and it can get maximal inhibition effect on concentration of 10 ?g/ml. Conclusions Mycophenolic acid could inhibit HBV replication and potentiate anti-HBV activities of lamivudine.
5.Construction of Rat Extracellular Signal-regulated Kinase 1 Gene 3' Untranslated Regions Dual-luciferase Reporter Plasmids and Effect of rno-miR-15b-5p on Its Activitiy

Objective To construct dual-luciferase reporter plasmids containing the wild type and mutant rat extracellular signal-regulat-ed kinase 1 (ERK1) gene 3' untranslated regions (UTR) which were used to detect rno-miR-15b-5p's putative target gene. Methods The rat ERK1 gene 3' UTR fragment was amplified by polymerase chain reaction (PCR) from PC12 cell cDNA and cloned into pmiR-RB-ReportTM vector. The mutant rat ERK1 gene 3' UTR fragment was obtained by overlap PCR and inserted into pmiR-RB-ReportTM vector. Successful wild type and mutant recombinant plasmids were confirmed by DNA sequencing. PC12 cells were co-transfected with rno-miR-15b-5p mim-ic and pmiR-ERK13' UTR or pmiR-ERK1-mut 3' UTR and then analyzed by dual-luciferase reporter assay system. The achieved mutation sequence of the target site TGCTGCT was mutated to CGAACGT and GTACACG, respectively. Results The wild-type reporter vector pmiR-ERK13' UTR and the mutant reporter vector pmiR-ERK1-mut 3' UTR were successfully constructed. The rno-miR-15b-5p mimic de-creased the activity of pmiR-ERK13' UTR plasmid (P<0.001) but did not decrease the activity of pmiR-ERK1-mut 3' UTR plasmid. Conclu-sion The recombinant pmiR-ERK13' UTR and pmiR-ERK1-mut 3' UTR plasmids were constructed successfully, and luciferase activities demonstrated that the 3' UTR of ERK1 gene might be a potential target of rno-miR-15b-5p.
6.Target genes of the hsa-miR-106b-25 cluster:Prediction and bioinformatic analysis

Objective The hsa-miR-106b-25 gene cluster is involved in various biological processes of carcinoma .This study aims at a prediction and function analysis of the hsa-miR-106b-25 gene cluster, miR-106b, miR-93, and miR-25, so as to provide some evidence for further studies on the functions of the three miRNAs and the mechanisms of their interaction . Methods We obtained the sequences of miR-106b, miR-93, and miR-25 from the miRBase, predicted their target genes with TargetScan , PicTar, and miRanda, and used 3 or more experimentally verified target genes from the miRTarbase as the gene set for further bioinformatic analysis .We predic-ted the biological processes of the target genes by GeneOntology analysis and enriched KEGG ( Kyoto Encyclopedia of Genes and Genomes) by pathway analysis, produced protein-protein interaction ( PPI) networks with STRING . Results The target genes of the miR-106b-25 gene cluster were significantly enriched in such biological processes as the regulation of macromolecule metabolism , regulation of metabolic process , and cell cycle process , while the KEGG pathway mainly in glioma, melanoma, prostate cancer , and gallbladder carcino-ma.The proteins encoded by the targeted genes of showed complicated interactions , and those encoded by the KAT2B, PTEN, TP53, CDH1, MDM2, E2F1, RB1, and SMAD7 plaid a core role in the interac-tion network. Conc lusoi n MiRNAs of the miR-106b-25 gene cluster regulate the downstream target proteins involved in tumorigenesis by participating in the cell cycle and cancer signaling pathway .
7.Comparative Study of LISS and the Condylar Support Plates in the Treatment of AO Type C Distal Femoral Fractures in Adults

Objective To summarize complications and early clinical effect of less invasive stabilization system and the femoral condylar support plates in treatment of AO type C distal femoral fractures. Methods We reviewed 46 patients who had internal fixation of AO type C distal femoral fractures. Of all 46 patients, 25 were with less invasive stabilization sys-tem and 21 were with femoral condylar support plates fixation. Comparative analysis was performed using intraoperative in-dex, postoperative complications and the Evanich score at follow-up. Results All 46 patients were followed up with a mean time of 19.6 months after surgery. The difference in incision length, blood loss, fracture healing time was significant between the 2 groups(P<0.05)but not in the duration of operations and hospital stays(P>0.05). The statistical signifi-cance was also found in the total incidence of postoperative complications and the Evanich score at the last follow-up(P<0.05). Conclusion Patients with less invasive stabilization system fixation had the characteristics of less trauma, shorter fracture healing time, less postoperative complications and better functional recovery compared with femoral condylar sup-port plates. Less invasive stabilization system had became an ideal internal fixation in treatment of AO type C distal femoral fractures.
8.Meta-analysis and associated mechanism on prognostic role of miR-31 in carcinomas

Objective To synthetically evaluate the relationship between miR-31 and the prognosis of carcinoma and to investi-gate its related mechanism.Methods The correlative literatures of tumor prognosis were retrieved from the electronic databases PubMed,EMBASE and ISI Web of Science.The pooled hazard ratios (HRs)and 95% confidence interval(95%CI )were extracted. The prognostic data were performed the synthesis analysis.Results A total of 7 trials conformed to the inclusion criteria including accumulated 2 012 cases of carcinoma.Meta-analysis revealed that the decrease of miR-31 expression in the tumor patients had the poor prognosis (HR=0.784,95%CI :0.630-0.974);in the subgroup analysis,the synthesis results adopting the multivariable a-nalysis and China subjects were 3.512 (95%CI :1.797-6.865)and 1.574 (95%CI :1.062-2.333),which indicating that the in-crease of miR-31 expression predicted the poor prognosis;miR-31 had no statistical significance in the digestive system (P >0.05). Conclusion The prognostic role of miR-31 may possess the histological and regional specificity and has the potential as a novel marker.
9.Simultaneous determination of calycosin and fermononetin in Radix Astragali and its fried product with honey by HPLC

AIM: To compare the contents of calycosin and fermononetin between Radix Astragali and its fried product with honey. METHODS: The chromatographic column was a Waters SunfireTM C18 column(250 mm ? 4. 6 mm,5 ?m). The mobile phase was the mixture of methanol and water with a linear gradient elution at a flow rate of 1 mL/min. The ultraviolet wavelength was set up 254 nm,and the column temperature was maintained at 35 ℃. RESULTS: The contents of calycosin and fermononetin in Radix Astragali were 0. 247 9 mg/g and 0. 125 2 mg/g respectively,the contents of calycosin and fermononetin in Radix Astragali fried with honey were 0. 225 6 mg/g and 0. 108 2 mg/g,respectively. CONCLUSION: The contents of calycosin and fermononetin in Radix Astragali fried with honey decreased.
