1.Salmonella typhimurium Flagellin as adjuvants can enhance immune response of DnaJ protein

To make sure the role of flagellin as mucosal adjuvants and the protective effect of streptococcus pneumonia infection when combined with DnaJ protein.Methods:Recombinant plasmid pET-28 ( a)/flagella was transferred to E.coli BL21(DE3).Over-expression of flagella was induced by IPTG and purification for animal study.All the C57BL/6 mice were randomly divided into three groups ,then respectively intranasally given the mixture of flagellin and DnaJ protein ( experimental group ) , DnaJ protein( control group 1 ) and the mixture of GST and DnaJ protein ( control group 2 ).The serum IgG and its subtype , cytokines secreted by mice spleen cells were detected by ELISA.At last all the C57BL/6 mice were intranasally immunized with Streptococcus pneumoniaD39.The protective effect by survival times were evaluated.Results: The mice of experimental group could secrete high level of serum IgG and cytokines IFN-γ,IL-4 and IL-17A.What more,the survival rate of mice in experimental group was 60%,a significant statistical difference with the control group.Conclusion:The flagellin as an adjuvant can reinforce the immune response of DnaJ protein and have better protection of resistance D 39 infection.We suggest that flagellin can be used as protein vaccine adjuvants.
2.Application and prospect of artificial intelligence in diagnosis, treatment and prognosis of prostate cancer

Prostate cancer is one of the most common malignant tumors in the world.Without typical early manifestations in the early stage, it is often too late when found. Therefore, early diagnosis, treatment, and prognosis are critical to improving the survival rate of patients with prostate cancer. Over the past few years, artificial intelligence(AI) has developed rapidly in the field of prostate cancer. In terms of diagnosis, AI is used as a tool to screen the images to reduce the error caused by the professionalism and subjectivity of the technician and to improve the repeatability of the results; In the prediction of prognosis, the algorithm calculates and evaluates disease-related parameters such as recurrence rate, lymph node metastasis rate and mortality rate, so as to assistant clinicians in decision-making and treatment improvements. This reviews aims to introduce the application of artificial intelligence in the diagnosis, treatment and prognosis of prostate cancer in recent years, as well as the prospect and challenges faced by artificial intelligence in the medical field.
3.Artificial intelligence in pathological diagnosis and molecular typing of prostate cancer:research progress

Artificial intelligence (AI) has important significance and great promise in the pathological diagnosis,imaging diagnosis,prognosis prediction,and molecular subtyping of prostate cancer (PCa). This review focuses on the progress of AI for the diagnosis and molecular classification of PCa,and briefly introduces the application of AI in the pathological diagnosis of needle biopsy and Gleason grading,pathological diagnosis and grading after prostatectomy,and prognosis prediction of PCa patients based on pathological sections. For the pathological diagnosis of needle biopsy and Gleason grading,AI has already comparable to general pathologists;for the pathological diagnosis and grading after prostatectomy,AI can accurately grade and classify tumors;and for the prognosis prediction of PCa patients,AI can directly extract relevant prognostic information from pathological tissue sections for prognosis prediction. In addition,AI can also predict gene mutations in PCa patients and suggest the probability of gene mutation by analyzing the pathological sections.
4.ADAMDEC1 regulates growth and migration of pancreatic carcinoma cells through Wnt/β-catenin signaling pathway

AIM:To investigate the effect of a disintegrin and metalloproteinase(ADAM)domain-like decy-sin 1(ADAMDEC1)knockdown on the proliferation,migration and invasion of pancreatic carcinoma cells.METHODS:Expression levels of ADAMDEC1 in pancreatic carcinoma tissues were analyzed using the GEPIA and UALCAN online da-tabases.Western blot analysis was employed to detect the protein expression levels of ADAMDEC1 in pancreatic carcino-ma cell lines(MIA PaCa-2 and PANC-1)and pancreatic ductal cell line(hTERT-HPNE).The effects of ADAMDEC1 knockdown on cell proliferation,migration and invasion were evaluated using CCK-8,colony formation,wound-healing and Transwell assays.Additionally,Western blot analysis was used to detect the effects of ADAMDEC1 knockdown on the expression levels of migration and invasion markers,as well as Wnt/β-catenin signaling pathway-related proteins in pancre-atic carcinoma cells.Furthermore,a recovery experiment was conducted to assess the role of Wnt/β-catenin signaling path-way agonist CHIR-99021 in ADAMDEC1 knockdown-induced inhibition of pancreatic carcinoma cell growth and migra-tion.RESULTS:(1)ADAMDEC1 was highly expressed in pancreatic carcinoma cells.(2)Knockdown of ADAMDEC1 led to a significant reduction in the proliferation,migration and invasion of pancreatic carcinoma cells.(3)Knockdown of ADAMDEC1 resulted in increased E-cadherin protein expression and decreased levels of matrix metalloproteinase 9,N-cadherin and vimentin proteins,alongside a reduction in the expression of Wnt/β-catenin signaling pathway-related pro-teins.(4)Co-treatment of pancreatic carcinoma cells with CHIR-99021 and ADAMDEC1 small interfering RNA reversed the inhibitory effects of ADAMDEC1 knockdown on cell proliferation,migration,and invasion.CONCLUSION:ADAMDEC1 is highly expressed in pancreatic carcinoma.Targeted silencing of ADAMDEC1 has the potential to inhibit the prolifera-tion,migration and invasion of pancreatic carcinoma cells by regulating the Wnt/β-catenin signaling pathway.
5.Free trajectory cone beam computed tomography reconstruction method for synchronous scanning of geometric calibration phantom and imaging object.

In order to suppress the geometrical artifacts caused by random jitter in ray source scanning, and to achieve flexible ray source scanning trajectory and meet the requirements of task-driven scanning imaging, a method of free trajectory cone-beam computed tomography (CBCT) reconstruction is proposed in this paper. This method proposed a geometric calibration method of two-dimensional plane. Based on this method, the geometric calibration phantom and the imaging object could be simultaneously imaged. Then, the geometric parameters could be obtained by online calibration method, and then combined with the geometric parameters, the alternating direction multiplier method (ADMM) was used for image iterative reconstruction. Experimental results showed that this method obtained high quality reconstruction image with high contrast and clear feature edge. The root mean square errors (RMSE) of the simulation results were rather small, and the structural similarity (SSIM) values were all above 0.99. The experimental results showed that it had lower image information entropy (IE) and higher contrast noise ratio (CNR). This method provides some practical value for CBCT to realize trajectory freedom and obtain high quality reconstructed image.

6.Effects of Dual-antiplatelet Medication Time on Efficacy and Safety of Postoperative Complications after Transcatheter Aortic Valve Implantation :A Meta-analysis

OBJECTIVE: To systematically evaluate the effects of dual-antiplatelet medication time on efficacy and safety of postoperative complications after transcatheter aortic valve implantation (TAVI), and to provide evidence-based reference for the formulation of antiplatelet therapy after TAVI. METHODS: Retrieved from Cochrane clinical controlled trial registration center, PubMed, Embase, Web of Science, Wanfang database and CJFD, during database establishment to Feb. 2019, RCTs and observational study about efficacy (all-cause mortality and incidence of stroke) and safety (the incidence of major bleeding events) the effects of dual-antiplatelet therapy for postoperative complications after TAVI at different time points were collected. After data extraction of clinical studies met inclusion criteria, quality evaluation with Cochrane bias risk evaluation tool 5.1.0 (for RCT) or Newcastle- Ottawa Scale (for observational study), Meta-analysis was conducted by using Rev Man 5.3 and Stata 14.0 statistical software. Meta-regression analysis was also conducted for outcome and different treatment duration. RESULTS: A total of 3 RCTs and 10 observational studies were included, involving 2 868 patients. The results of Meta-analysis showed that the incidence of all-cause mortality one month and 6 months after medication were 0.05 [95%CI (0.03, 0.07), P<0.001] and 0.07 [95%CI (0.05, 0.08), P<0.001]. The incidence of major bleeding events 1, 3 and 6 months after medication were 0.14 [95%CI (0.08,0.19), P<0.001], 0.11 [95%CI (0.03, 0.19), P=0.007] and 0.13 [95%CI (0.05, 0.22), P=0.002]. The incidence of stroke after one month after medication was 0.04 [95%CI (0.03, 0.05), P<0.001]. Results of Meta-regression analysis showed that the all-caused mortality [regression coefficient=0.005 7, 95%CI (-0.001 6, 0.013 0), P=0.116], major bleeding [regression coefficient=-0.000 5,95%CI(-0.022 4,0.021 4), P=0.959] or the incidence of stroke [regression coefficient=0.001 4, 95%CI (-0.003 8, 0.006 5), P=0.570] were not related to medication duration of dual-antiplatelet therapy. CONCLUSIONS: The prolongation of the medication time of the dual-antiplatelet therapy has no significant effect on the efficacy and safety of TAVI.

Oral diseases concern almost every individual and are a serious health risk to the popula-tion.The restorative treatment of tooth and jaw defects is an important means to achieve oral function and support the appearance of the contour.Based on the principle of"learning from the nature",Deng Xu-liang's group of Peking University School and Hospital of Stomatology has proposed a new concept of"microstructural biomimetic design and tissue adaptation of tooth/jaw materials"to address the worldwide problems of difficulty in treating dentine hypersensitivity,poor prognosis of restoration of tooth defects,and vertical bone augmentation of alveolar bone after tooth loss.The group has broken through the bottle-neck of multi-stage biomimetic technology from the design of microscopic features to the enhancement of macroscopic effects,and invented key technologies such as crystalline/amorphous multi-level assembly,ion-transportation blocking,and multi-physical properties of the micro-environment reconstruction,etc.The group also pioneered the cationic-hydrogel desensitizer,digital stump and core integrated restora-tions,and developed new crown and bridge restorative materials,gradient functionalisation guided tissue regeneration membrane,and electrically responsive alveolar bone augmentation restorative membranes,etc.These products have established new clinical strategies for tooth/jaw defect repair and achieved inno-vative results.In conclusion,the research results of our group have strongly supported the theoretical im-provement of stomatology,developed the technical system of oral hard tissue restoration,innovated the clinical treatment strategy,and led the progress of the stomatology industry.
8. Analysis of mental state of allergic rhinitis patients in Chengdu city by symptom check list 90 (SCL-90) scale

Objective:
To analyse the mental state of patients with allergic rhinitis (AR) in Chengdu.
Methods:
One thousand five hundred and thirty-six AR patients from Sichuan Provincial Integrated Traditional Chinese and Western Medicine Hospital, West China Hospital of Sichuan University, Affiliated Hospital of Chengdu University of Traditional Chinese Medicine, Sichuan People′s Hospital, Sichuan Second Hospital of Traditional Chinese Medicine were selected from July 2013 to January 2018. Eight hundred and twenty-seven patients were screened into study group by inclusion and exclusion standards. The symptom check list 90 (SCL-90) was used to group and score the mental state of these patients according to nine classification criteria: gender, BMI, age, marital status, monthly salary, disease duration, living environment, education level and working environment. Then, the scores were compared within groups. Inter-group comparison was made between the study group and the Chinese norm, and the positive factors for psychological disorders were extracted. Four symptoms in the study group, i.e. nasal itching, sneezing, clear discharge and nasal congestion, were scored on the visual analogue scale (VAS). SPSS 19.0 software was used to carry out statistical analysis. Partial correlation analysis was performed between the positive factors and the symptom scores by multiple regression statistical method.
Results:
The total score of SCL-90 in the study group was 2.64±0.25, which was accorded with mild to moderate mental health impairment. There were 124 (15.0%) without mental health damage, 176 (21.3%) with mild damage, 474 (57.3%) with mild to moderate damage, 41 (5.0%) with moderate to severe damage and 12 (1.4%) with severe damage. The in-group comparison showed that the top three categories of different items were the living environment, gender and working environment. The scores of somatization, obsessive-compulsive symptoms, interpersonal sensitivity, depression, anxiety, psychosis, other (sleep, diet) and total average score of urban residents were higher than that of country residents (3.29±0.61
9.Meta-analysis of Efficacy and Safety of Single or Dual Antiplatelet Therapy for Short-term Complications after Transcatheter Aortic Valve Implantation

OBJECTIVE: To systematically evaluate effectiveness and safety of single antiplatelet therapy (SAPT) versus dual antiplatelet therapy (DAPT) on short-term complications after transcatheter aortic valve implantation (TAVI), and to provide evidence-based reference for clinical treatment. METHODS: Retrieved from PubMed, Cochrane clinical controlled trials registry, Web of Science, CNKI, Wanfang database, CBM and Chinese Clinical Trial Registry, RCTs and observational studies about effectiveness (all-cause mortality, incidence of stroke and incidence of myocardial infarction 30 days after operation) and safety (the incidence of bleeding events at 30 days after operation) of SAPT versus DAPT on short-term complications of TAVI were collected during the date of database establishment to Jan. 2019. After data extraction of included studies and quality evaluation with Cochrane system evaluator manual 5.1.0 (for RCT) and the Newcastle-Ottawa Scale (NOS) (for observational studies), Meta-analysis was conducted by using Rev Man 5.3 statistical software. RESULTS: Totally 3 RCTs and 7 cohort studies were included, involving 3 188 patients. Results of Meta-analysis showed that the incidence of all-cause mortality 30 days after operation [OR=0.48, 95% CI (0.32, 0.73), P<0.001] and the incidence of bleeding events 30 days after operation [OR=0.43, 95%CI (0.30, 0.59), P<0.001] in SAPT group were significantly lower than DAPT group, with statistical significance. There was no statistical significance in the incidence of stroke 30 days after operation [OR=0.63, 95%CI (0.38, 1.06) , P=0.08] or the incidence of myocardial infarction 30 days after operation [OR=1.09, 95%CI (0.46, 2.59), P=0.85] between 2 groups. CONCLUSIONS: Compared with DAPT, SAPT can decrease the incidence of all-cause mortality 30 days after TAVI and the incidence of bleeding events 30 days after TAVI.
