1.Inhibitory effects of LaCl_3 on hepatocellular carcinoma

Objective To observe the inhibitory effect of LaCl_3 on the growth of human hepatocellular carcinoma cell lines SMMC-7721 and its mechanism.Methods The cells were treated with different doses of LaCl_3 and the growth curve,colony inhibitory rates,the levels of AFP of cultured cells were detected at 1,3,5 and 7 days after they were dealed with LaCl_3.The changes of cell cycle were detected by flow cytometry and the expressions of cell cycle protein CyclinD_1,PCNA,CDK_4 and P16 were observed by immunocytochemistry staining.Results MTT test showed that SMMC-7721 cell proliterating activity was obviously inhibited by LaCl_3 in a time and dose related manner at concentrations of 0.50 and 1.00 mmol?L~(-1).The colony forming inhibitory rate of 0.10,0.50 and(1.00 mmol?L~(-1))LaCl_3 were 51%, 67% and 97% (P
2.Analysis of clinical effectiveness and complications of different internal fixation treatment for intertrochanteric femoral fractures

Objective To investigate the clinical effectiveness of dynamic hip screw (DHS) internal fixation and proximal femoral nail (PFN) internal fixation for intertrochanteric femoral fractures.Methods One hundred and sixty-five patients with intertrochanteric femoral fractures were randomly divided into 2 groups,85 patients were in DHS group and 80 patients were in PFN group.The clinical effectiveness,index in operative procedure and complications were compared between two groups.Results The fine rate of DHS group and PFN group was 75.3%(64/85 ) and 83.8%( 67/80 ) (P > 0.05 ).The indexes of function recover time,fracture healing time,amount of bleeding in operation,operation time in PFN group were significant lower than those in DHS group [(54.3 ± 11.7 ) d vs.(92.6 ± 10.5 ) d,(63.8 ± 12.2) d vs.(71.3 ± 10.6) d,(453.3 ± 50.7) ml vs.(627.5 ± 46.8) ml,(77.9 ± 25.2) min vs.( 115.7 ± 32.8) min](P<0.05 ).The rate of short-term complications was 16.2% (13/80) in PFN group,and 12.9 % ( 11/85 ) in DHS group(P > 0.05 ).The rate of long-term complications was 1.2%( 1/80 ) in PEN group and 4.7%(4/85 ) in plts group (P < 0.05).Conclusions As the treatment of intertrochanteric femoral fractures,PFN internal fixation has less bleeding and better efficacy,and can improve function recovery.PFN internal fixation is a better method for unstable femoral fracture.
3.Analysis of amplification and bioinformatics on mycobacterium tuberculosis protein higA

Objective To amplify the higA gene from the Mycobacterium tuberculosis,and to analyze the structure and function of their encoded proteins by using bioinformatics.Methods Total DNA was extracted from Mycobacterium tuberculosis.PCR of higA was performed and the products were sequenced.The biological features of the higA protein including,its physical and chemical properties,signal peptide,spatial structure and epitopes were analyzed by using software online.Results The PCR products of higA were 450 bp in length,which were consistent with the expected size.The higA protein consisted of 149 amino acids and had the following characteristics:a theoretical isoelectric point of 7.93,a fat-soluble factor of 94.30,and instability coefficient of 36.57.The higA protein had no signal peptide,containing 10 phosphorylation sites and multiple potential epitopes.Conclusion Mycobacterium tuberculosis higA gene can be amplified by PCR and the characteristics of higA protein is identified.
4.Virulence of VEG strain Toxoplasma gondii oocysts against Kunming mice

To explore the pathogenicity of VEG strain Toxoplasma gondii oocysts on China Kunming mice,T.gondii oocysts of < 1 and 102-108 were chosen to feed the mice orally.And the modified agglutination test (MAT),H&E and IHC were used to check the infection of mice.The infection rate,the survival rate of mice,and number of cysts in brain were analyzed.Results showed that 100% of the mice fed with ≥102 oocysts were infected,the minimum lethal dose was 102 oocysts and the 100% lethal dose was 108 oocysts.The time of death in acute infection was 7 DPI to 14 DPI.T.gondii cysts formation rate was 32.14% (9/28),and the number of cysts was 9 to 857 per mouse.The survival rate of infected mice was 67.44% (29/43),and the longest survival time was more than 390 DPI.Accordingly,the virulence of T.gondii VEG strain is medium,and has a higher cysts formation rate.

7.TESTIN tumor suppressor gene in endometrial cancer tissues

ObjectiveTo investigate the expression of TESTIN gene in endometrial carcinoma and explore the functions of this gene in tumor development and progression.MethodsqRT-PCR and immunochemical staining assay were used to determine the mRNA and protein level of TESTIN in the tumor tissues,and the relationship between TESTIN expression and clinical pathology characteristics was analyzed.Results Compared to normal tissue,76.5% (52/68) tumor tissues showed TESTIN reduced ( P < 0.01 ),furthermore,this reduction in the subgroup of endometrioid adenocarcinoma was significant,but it was rarely observed in the subgroup of serous papillary adenocarcinoma.ConclusionsTESTIN was obviously down regulated in endometrail carcinoma,especially in endometrioid adenocarcinoma,which indicated TESTIN played an important role in tumorigenesis of uterine.
8.Hepatic artery reconstruction in operations for hilar cholangiocarcinaoma

Objective To investigate the feasibility and outcome of resection and reconstruction of hepatic artery in hilar cholangiocacinoma (HCC).Methods The data of 29 patients with HCC with hepatic artery reconstruction carried out from March 2009 to August 2013 in our center were retrospectively analyzed.23 right hepatic arteries and 6 common hepatic arteries were involved.In-situ anastomosis was carried out in 20 patients and a double anastomosis using gastrodoudenal artery grafts was carried out in 9 patients.Results There were no arterial thrombosis or other related complications on prolonged follow-up.Conclusion Hepatic artery resection and reconstruction should be carried out if the artery was invaded by a hilar cholangiocarcinoma to produce a high resection rate and a better outcome.
9.Study on mechanisms of IL-1βpromoted lung cancer cells proliferation

Objective:To investigate the mechanisms of IL-1β promoted lung cancer cells proliferation. Methods: The“Transwell? Inserts” system was used to coculture lung cancer cells A549,NCI-H520 with macrophages. BrdU ELISA used to measure the effect of macrophages promoted lung cancer cells proliferation. Expression of mRNA of IL-1β in A549 and NCI-H520 cells were analysed by Real-time PCR analysis. IL-1β was responsible for macrophage-promoted lung cancer cells growth, IL-1β neutralizing antibody was added. The autophagy marker Beclin1 protein was detected by Western blot. Results:The BrdU ELISA assay showed that after coincubation with macrophages in the proportion of 1:0. 5,the OD value of A549 increased from(0. 41±0. 06)to(1. 13±0. 10). There was statistical significance(P<0. 05). It also showed that the growth of the A549 cell was dependent on the macrophage number (P<0. 05). The OD value variability of NCI-H520 cells was as same as A549 cell upon cocultured with macrophages. Real-time PCR results showed that the expression of IL-1β mRNA in macrophages was remarkably enhanced in a time dependent manner upon coincubated with lung cancer cell,and the expression level was higher than lung cancer cells. Addition of IL-1β neutralizing antibody markedly inhibited macrophage-promoted lung cancer cells proliferation. The OD value of these two cells were decreased from ( 3. 63 ± 0. 33) to (1. 46±0. 18),from (2. 94±0. 38) to (1. 53±0. 20),respectively (P<0. 05). After treatment with IL-1β,the expression of Beclin1 was significantly inhibited in tumor cells. Conclusion:Over-expression of IL-1βfrom macrophages and lung cancer cells is re-sponsible for proliferation of tumor cells in coculture condition. Inhibition of autophagy in tumor cells may be the important mechanisms of IL-1β promotes lung cancer cells proliferation.
10.Ex-vivo liver resection combined liver autotransplantation for the treatment of hepatic alveolar echinococcosis

Ex-vivo liver resection and liver autotransplantation offers a chance to cure extensive regional liver diseases which are otherwise deemed untreatable in selected cases. A 33-year-old male patient with advanced alveolar echinococcosis (AE) which invaded the right lobes and adjacent retrohepatic inferior vena cava, while the left lobes were free from lesion and had proliferated to over 1200 ml in size except for a 1 cm × 1 cm solid lesion in the segment Ⅳ. No extrahepatic metastases were found in this patient. A lesion with a size of 4300 ml and other small lesions in the right lobes were removed extracorporeally,and the retrohepatic vena cava was repaired, then the remaining AE-free left lobes (more than 700 ml in size) and the retrohepatic inferior vena cava were re-implanted in situ. A temporary end-to-end cavo-caval shunt with interposition of a blood vessel prosthesis and end-to-side portocaval shunt were established to keep the blood flow during the four-hour anhepatic phase. The patient was followed up for six months, and he had no signs of residual liver disease with good hepatic function.
