Objective To investigate the expression level of miRNA-146a (miR-146a) in retinal pigment epithelial (RPE) cells aging and age-related macular degeneration (AMD),and discuss its regulation mechanism of AMD by repressing VEGF-A.Methods The expressions of miR-146 and VEGF-A were examined by qRT-PCR in RPE cells in mice aged 2 months,8 months,12 months,18 months or 24 months, and in RPE cells from 75 years old AMD patients. The protein level of VEGF-A was also detected by Western Blotting. Finally, the effects of overexpression of miR-146a in APRE-19 cell line on expression of VEGF-A was checked. Results MiR-146 was up-regulated to 8 times or 24 times at 18 months or 24 months aged mice, and the expression of VEGFA was down-regulated in aging RPE from 1.5 times to 0.8 times. However, the expression of miR-146 decreased to 14.5 times and VEGF-A increased in RPE cells of AMD.In cultured cells, overexpression of miR-146a inhibited the expression of VEGF-A.Conclusion Up-regulation of miR-146a in aging RPE cells and its down-regulation in AMD suggest a potential of miR-146a as molecular marker.MiR-146a overexpression inhibits the expression of VEGF-A,supporting a potential clinical treatment of miR-146 in AMD.

Objective To observe the effect of serum containing Dahuang Zhechong Pills on vascular endothelial growth factor ( VEGF) expression in K562 cells of chronic myelogenous leukemia ( CML) and to develop new antiangiogenic drugs for CML treatment. Methods Enzyme-linked immunosorbent assay ( ELISA) was used for the detection of serum VEGF content in 38 CML patients, 11 acute myelogenous leukemia patients and 9 healthy volunteers. SD rats were used for the preparation of serum containing Dahuang Zhechong Pills, and then ELISA was applied for the detection of VEGF content in K562 cells pretreated with serum containing Dahuang Zhechong Pills. Results Compared with the healthy volunteers, myelogenous leukemia patients at chronic phase and acute phase had high serum VEGF content ( P<0.05 or P<0.01) . Compared with blank serum control group, the secretion of VEGF in K562 cells pretreated with serum containing Dahuang Zhechong Pills was decreased obviously ( P<0.01) , and VEGF expression decreased with the increase of drug concentration. Conclusion VEGF content is increased in myelogenous leukemia patients at chronic phase and acute phase, which may be related to the pathogenesis, progress and prognosis of chronic myelogenous leukemia. Serum containing Dahuang Zhechong Pills can inhibit the expression of VEGF in chronic myelogenous leukemia K562 cells and decrease VEGF protein expression in concentration-dependent manner.

Objective To provide data evidence for early diagnosis of bone and joint damage derived from brucellosis by analyzing its clinical and imaging characteristics. Methods Patients with brucellosis accepted in Ji'nan Infectious Disease Hospital form December 2013 to December 2014 were selected.Patients with bone and joint damage confirmed through imaging were further studied, their epidemiological and clinical characters,CT and MRI characteristics, treatment and outcome were summarized. Results Total of 97.8% (45/46) patients had a clear contact history, most of them worked in poultry farming, taking up to 67.4% (31/46).July to October was its peak time for attacking, taking up to 58.7% (27/46).The clinical manifestations of patients were joint pain, swelling and activities obstacles. In the CT images, there was obvious bony destruction, characterized by multiple round or large areas of low density insect damage sample spots. There was osteosclerotic bone lesions or osteophyte formatted in edge. The paravertebral soft tissue was swelling, and the vertebral body deformation was not obvious, and there was vertebra small joint damage occasionally. In the MRI images, there was vertebral ligaments damage,or soft tissue damage,or osteoproliferation at the edge of the vertebral bodies. There was abnormal signal T1W1 or T2W1 signal, and FS-T1W1 showed high signal, while FS-T2W1 showed slightly high signal. The diagnosis rate of CT,MRI and CT combined MRI for bone and joint damage derived from brucellosis was 76.1% (35/46),84.8% (39/46) and 91.3% (42/46),respectively.The main therapeutic drugs were doxycycline and rifampicin, and the clinical effective rate was 91.3% (42/46).Conclusion The epidemiological characters,CT and MRI characteristics of bone and joint damage derived from brucellosis have certain representativeness, and the comprehensive investigation is beneficial for clinical diagnosis.

Objective To study the mechanism of ferulic acid(FA)on hepatic fibrosis(HF)based on network pharmacology, and establish an in vitro model of rat hepatic stellate Cell-T6(HSC-T6)according to the results. Methods The potential targets of FA were screened through PubChem, swisstargetprediction and pharmmapper, and overlapped with the FA targets screened in disgenet, genecards and OMIM.Then, protein protein interaction(PPI)was analyzed by using string platform.Gene ontology(go)and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis were carried out for key targets by using R64 4.0.3,and the"component target disease"network diagram was constructed by Cytoscape 3.7.2 software.Based on this, the proliferation of HSC-T6 was detected by cell counting kit-8(CCK-8)method,and the grouping was determined:blank group and low-dose group(100 μg·mL-1 FA),medium dose group(200 μg·mL-1 FA),high dose group(400 μg·mL-1 FA)and positive control group(200 μg·mL-1 colchicine),the migration ability of HSC-T6 was detected by scratch test, and the content of HSC-T6 was detected by enzyme linked immunosorbent assay(ELISA)α-Alpha smooth muscle actin,α-Flow cytometry was used to detect the changes of HSC-T6 cycle, quantitative real time polymerase chain reaction(qRT-PCR)was used to detect the relative expression of JAK2 and STAT3 mRNA,and Western Blot was used to detect the molecular expression of JAK2 and STAT3 protein. Results 254 intersection targets of FA and HF were obtained. The core targets were signal transducer and activvator of Transcription(STAT3),albumin(ALB),protein kinase B(AKT1),tumor suppressor protein p53(TP53),epidermal growth factor receptor(EGFR)and caspase-3(CASP3).KEGG analysis showed that the action pathway of FA on HF mainly involved phosphatidylinositol 3 kinase protein kinase B(PI3K-Akt),vascular endothelial growth factor(VEGF),Janus kinase/signal transducer and activator of transcription(JAK/STAT)Tumor necrosis factor(TNF)and other pathways. The experimental results showed that in CCK-8 experiment, scratch experiment and ELISA experiment, compared with the blank group, the cell proliferation rate, migration ability and the expression of α-SMA protein decreased significantly(P<0.05).Compared with the blank group, the cycle arrest rate of low, medium and high dose groups and positive control group increased significantly(P<0.05).Compared with the blank group, the molecular weight and mRNA expression of JAK2 and STAT3 protein in low, medium and high dose groups and positive control group decreased gradually(P<0.05).Conclusion FA has the characteristics of multi-channel and multi-target.FA may inhibit the apoptosis of hepatic stellate cell(HSC)by down regulating JAK2 and STAT3 targets.
