1.The research of myocardial fibers character in isolated rats heart based on 7.0 T diffusion tensor ;imaging

Objective Using 7.0 T diffusion tensor imaging(DTI) to study myocardial fibers character in isolated rats heart in aspect of water molecular diffusion, myocardial mechanics and microstructure. Methods Fixed male rats heart (n=10) underwent DTI and the myocardial structures were quantitatively measured by Diffusion Toolkit and Matlab. The characteristic of myocardial fibers arrangement was observed and FA values and ADC values of myocardial fibers, myocardial fibers density and length, mean helix angle of left ventricular were calculated as well. Mean helix angle of anterior wall, ventricular septum, lateral wall and posterior wall of epicardium and endocardium were compared by using one?way ANOVA. Results The SD rats' myocardial fibers appeared in compact and inerratic distribution from epicardium to endocardium. Compared with HE staining, regular direction and arrangement in fibers were showed in myocardial fibers tracer map and tensor map in papillary muscle plane. Quantitative analysis showed that ADC value of SD rats' myocardial fibers was (9.6 ± 3.6) × 10-4 mm2/s, FA value was 0.80 ± 0.04, myocardial fibers density was (981±24) tracks/mm3 and myocardial fibers length was (6.18±1.71)mm. Mean helix angle of left ventricular epicardium to endocardium ranged from-81.37° to 0° and then gradually change to 82.83° . There were no significant differences in mean helix angle of anterior wall, ventricular septum, lateral wall and posterior wall of epicardium and endocardium, respectively (F=2.25, 0.40, P>0.05). Conclusions This study shows that DTI is an effective method to quantitatively measure the characteristics of myocardial fibers. This study provides useful information for further study of myocardial fibers in heart disease models.
2.Determination of Peoniflorin in Shenshaogankang Oral Liquid by HPLC

OBJECTIVE:To establish HPLC method for the determination of the content of paeoniflorin in Shenshao?gankang oral liquid.METHODS:Paeoniflorin was separated on Kromasil C 18 ;methanol-0.05mol/L KH 2 PO 4 solution-avantin(60∶173∶4,which was adjusted to pH=4.0with acetic acid)was taken as the mobile phase with a flow rate at1.0ml/min and detection wavelength at230nm.RESULTS:Good linear relationship was achieved when the detection concentration range of peoniflorin was0.027~0.230mg/ml(r=0.9999);The average recovery of peoniflorin was100.97%(RSD=1.28%,n=5). CONCLUSION:The method was simple,rapid,precise and reproducible,which can be used as the quality control of Shenshaogankang oral liquid.
3.Expressions of OPN, COX-2 and CyclinD1 in breast infiltrating carcinoma and their relationship with clinicopathological features

Objective To observe the expressions of OPN, COX-2 and CyclinD1 in breast infiltrating carcinoma and evaluate their relationships with clinic pathological features. Methods Expression of the above three indexes were detected from 70 breast cancinoma patients by immunohistochemistry. The relationships among them and clinicopathological features were analyzed. Results The positive expression rates of OPN were 78.8% in cases (≤45 years old) and 73.0% in cases (> 45 years old); the positive expression rates were 79.3%(tumor diameter ≤ 3 cm) and 73.2% (tumor diameter > 3 cm); the positive expression rates were 77.8%, 73.8% and 78.9% in cases ofⅠgrade, Ⅱgrade and Ⅲ respectively, the positive rates had no statistical significances(P > 0.05). The expression rates of OPN in cases of breast infiltrating carcinoma without and with axillary node metastasis were 62.5% and 93.3%, in cases at stage Ⅰ~Ⅱ and Ⅲ ~Ⅳ were, 68.0% and 95.0% respectively, the positive rates had statistical significances(P < 0.05). The expression of OPN was negatively correlated with ER and PR while positively correlated with CerbB2, COX-2 and CyclinD1. Conclusions OPN plays an important role in the invasion and metastasis of breast carcinoma coordinated with COX-2 and CyclinD1.
4.Clinical characteristics of 42 patients with cardiac amyloidosis

Objective To characterize the clinical features of patients with cardiac amyloidosis (CA).Methods Totally 42 patients with CA admitted to Guangdong General Hospital since 2008 were included and retrospectively analyzed in the present study.CA was confirmed by abdomen and endocardium biopsy examination.Clinical manifestations,electrocardiogram and echocardiography were collected for the evaluation.Results Several clinic features are common in CA.In the present study,37 cases (88.1%) presented with chest tightness,dyspnea,20 cases(47.6%) with chest pain,27 cases(64.3%) with right heart failure,27 cases (64.3%) with fatigue,and 30 cases (71.4%) with renal insufficiency and proteinuria.Electrocardiogram (ECG) showed that 32 of the patients (76.2%) were with low voltage in limb leads,29 cases (69%) of them were with poor R wave progression in precordial leads,17 cases (40.5%) with ST-T change,28 cases(66.7%) with pseudo-necrotic Q wave and 36 cases (85.7%) with various kinds of arrhythmia.Echocardiography indicated that all of the subjects (100%) were with different degrees of left ventricular posterior wall or ventricular septal thickness,and left atrial hypertrophy with different degree of myocardial grain appearance or ground-glass opacity.Thirty-six cases (85.7%) were with pericardial effusion,and 27 cases (64.3%) were with abnormal left ventricular eject function.Conclusion For those who were with unexplained clinical cardiac insufficiency,renal insufficiency,myocardial hypertrophy,but normal of ventricular size in echocardiography and low voltage on ECG limb leads,a tissue biopsy from abdomen,labial glands or endocardium should be considered in the diagnosis of CA.
5.Gab2-Akt-ARK5 signaling pathway is associated with the invasion of glioma cells

Objective:This study aimed to investigate the effect and significance of a binding protein-2 (Gab2)-Akt-ARK5 signaling pathway on the invasion of glioma cells. Methods:Immunohistochemical methods were used to detect the expressions of Gab2 and ARK5 in 45 cases of glioma tissue. siRNA plasmid was used to transfect LN-229 cells, and western blot was performed to analyze the protein expressions of Gab2 and ARK5. In vitro Matrigel invasion assay was conducted to detect variations in the invasiveness of transfected cells. Western blot was also conducted to analyze the protein phosphorylation of Akt and ARK5 in the cells transfected with Gab2 plasmid. Results:Immunohistochemical assay revealed that the expressions of ARK5 and Gab2 in glioma cells were positively correlated, and both expressions were higher in high-grade glioma (WHO gradeⅢ,Ⅳ) than in low-grade glioma (WHO gradeⅠ,Ⅱ). LN-229 cells transfected with ARK5 plasmid, Gab2 plasmid, ARK5 and Gab2 plasmid, and control plasmid were named siARK5/LN-229, siGab2/LN-229, siARK5 and siGab2/LN-229, and SCR/LN-229, respectively. After transfection was performed, the protein expressions of ARK5 and Gab2 were respectively decreased in siARK5/LN-229 and siGab2/LN-229. The protein expressions of ARK5 and Gab2 in siARK5 and siGab2/LN-229 were also respectively decreased. After ARK5 or Gab2 was downregulated, the number of glioma cells, which invaded and penetrated Matrigel, was decreased (P<0.01). The number of glioma cells also decreased significantly after ARK5 and Gab2 were downregulated. The phosphorylation of Akt and ARK5 in siGab2/LN-229 cells was decreased after these cells were stimulated by insulin-like growth factor-1. Conclusion:The silencing of ARK5 or Gab2 impaired glioma cell invasiveness. The decreased protein expression of Gab2 inhibited the phosphorylation of Akt and ARK5. These results suggested that the Gab2-Akt-ARK5 signaling pathway could be relevantly involved in glioma cell invasion.
6.The efficacy of different regimens in the salvage therapy of elderly patients with refractory diffuse large B-cell lymphoma

Objective To evaluate the efficacy and toxicities of gemcitabine plus oxaliplatin with R-GemOx or without (GemOx) rituximab regimen in the treatment of relapsed or refractory diffuse large B-cell lymphoma in elderly patients.Methods A total of 39 patients with relapsed or refractory diffuse large B-cell lymphoma received R-GemOx or GemOx chemotherapy.There were 16 patients in R-GemOx and 23 patients in GemOx group.Patients in both groups received gemcitabine 1000 mg/m2,d1,at land 8 day and oxaliplatin 130 mg/m2,d1 at lday.Patients in R-GemOx additionally received rituximab 375 mg/m2.Every 21-28 days was 1 cycle.The toxicities were evaluated after 1 cycle of chemotherapy.The efficacy was evaluated after 2 cycles of chemotherapy.Results In R-GemOx group,the total response rate was 62.5%,and the clinical benefit rate was 87.5%.In GemOx group,the total response rate was 47.8%,and the clinical benefit rate was 73.9% There was no significant differences between the two groups.There was a significant difference in the median time-to-progression (TTP) between R-GemOx group (6.4 months) and GemOx group (5.0 months) (P < 0.05).The major toxicities were marrow suppression and gastrointestinal reaction,which had no significant differences between the two groups.Conclusions R-GemOx and GemOx regimen are effective and safe for the elderly patients with relapsed or refractory diffuse large B-cell lymphoma(DLBCL).But the patients with relapsed/refractory DLBCL treated with R-GemOx had a longer median time-to-progression than with GemOx regimen.
7.Prognostic factors of surgical therapy for abdominal aortic aneurysm in the area of Tibet plateau

Objective:To analyze the feasibility of surgical therapy and the risk factors affecting prognosis for patients with abdominal aortic aneurysms in Tibetan plateau.Methods:Data of 48 abdominal aortic aneurysm cases were retrospectively analyzed at our hospital from Jun 2013 to Dec 2017.Results:Five cases received conservative treatment for suspected rupture of abdominal aortic aneurysm and all died. Eight cases left hospital without treatment, among them 5 cases died for rupture of abdominal aortic aneurysm.Thirty-five patients had abdominal aortic aneurysm resection and artificial vascular reconstruction. Successful rate was 100%. Perioperative complications developed in 5.7% patients; blood transfusion rate was 11.4%, there were non-perioperative death. Single-factor regression analysis showed that chronic altitude erythrocytosis as an independent risk factor with poor surgical prognosis ( P<0.05). Conclusion:Even in Tibet plateau, traditional surgery is a safe and effective method for treating abdominal aortic aneurysms. Chronic altitude erythrocytosis is an independent risk factor for poor surgical prognosis.
8.Role of Hes1 in malignant transformation of human bronchial epithelial cells induced by tobacco

AIM: To investigate the role of transcription factor hairy and enhancer of split 1 (Hes1) in the malignant transformation of human bronchial epithelial cell line BEP2D induced by tobacco.METHODS: The BEP2D cells were chronically exposed to cigarette smoke condensate (CSC) at 1 cigarette per L until the 70th generation.The phenotype of malignant transformation of the cells induced by CSC was detected by soft agar clony formation assay.RT-PCR and Western blot were used to determined the expression of Hes1 at mRNA and protein levels in each generation of the cells.The proliferation and apoptosis of the BEP2D cells exposed to CSC were analyzed with the methods of MTT assay, flow cytometry and cell colony formation assay after treatment with Notch pathway bloker DAPT or liposome transfection with Hes1-siRNA.The expression of Hes1 in the peripheral small airway tissues of the smoking rats was evaluated by immunohistochemical staining.The expression of Hes1 in non-small-cell lung cancer and normal airway tissues was also detected by the methods of immunohistochemistry and RT-PCR.RESULTS: The BEP2D cells in the 70th generation had a malignant transformation phenotype.The expression of Hes1 in the BEP2D cells exposed to CSC for different time showed an increa-sing trend.DAPT and liposome transfection with Hes1-siRNA down-regulated the expression of Hes1, inhibited the cell proliferation and induced cell apoptosis.The expression of Hes1 in the airway mucosa of the rats exposed to cigarette smoke for 1 month and 6 months was significantly higher than that in control group.Cigarette smoking induced the expression of Hes1 in lung cancer and normal airway tissues.CONCLUSION: Hes1 may be involved in smoking-induced lung cancer by promoting the imbalance between apoptosis and proliferation.

Astragalus membranaceus is one of the frequently used qi tonifying herbal medicines.It can regulate blood pressure.This paper reviewed the studies on the two-way regulation of blood pressure of Astragalus membranaceus in recent years,even thought the Astragalus membranaceus has been regarded to cure hyperhension due to qi deficiency.It can lower the pressure with large dosage or in liquid-fried form.Astragalus membranaceus,Astragalus membranaceus as the sovereign drug of the formulas or Astragalus membranaceus combined with western medicine can regulate blood pressure.It was found that the effect of reducing blood pressure was mainly related to the decrease of heart rate,dilation of blood vessels,inhibition of oxidative stress,reduction of calcium ion level,diuretics,change the content of growth factor and receptor,lowering uric acid,reduction of cysteine levels,and increase insulin sensitivity.Itelevated the blood pressure,mainly related to the increase of cardiac output and vasoconstriction.
