1.EXPRESSION AND REGULATORY FUNCTION OF INTER RELATED POLYPEPTIDES DURING DEVELOPMENT OF RENAL TUBULES IN CHICK EMBRYO

Objective To observe the expression of EGFR,TGF-?,AQP-2 and Bax during development of renal tubules in chick embryo,and explore the role and regulating significance of them. Methods The expression of EGFR,TGF-?,AQP-2 and Bax were observed by immunohistochemistry combined with stereological methods in 36 chicks kidney at 10th-46th incubation stages.The expression intensity of them was analyzed by IPP software. Results The epithelial cells of proximal tubules in chick embryo appeared EGFR immunoreactivity at 26th-46th stages;At 26th-37th stages the epithelial cells of proximal tubules and distal tubules of mesonephros appeared TGF-? immunoreactivity,the epithelial cells of proximal tubules and distal tubules of metanephros appeared TGF-? immunoreactivity at 33rd-40th stages,at 40th-46th stages the epithelial cells of proximal tubules of metanephros appeared TGF-? immunoreactivity but the epithelial cells of distal tubules did not appeare TGF-? immunoreactivity;The epithelial cells of collecting tubules in chick embryo appeared AQP-2 immunoreactivity at 26th-46th stages;The epithelial cells of proximal tubules and distal tubules of mesonephros appeared Bax immunoreactivity at 26th-37th stages,the epithelial cells of proximal tubules of metanephros appeared Bax immunoreactivity at 33rd-46th stages.Images showed the expressions of EGFR,TGF-?(?_1,?_2,?_3)and Bax first displayed up then down,but expression of AQP-2 always displayed up during development of chick embryo.Conclusion Mesonephros of chick embryo has the function of excreting.Degeneration of renal tubules of mesonephros might be related with the excessive expression of TGF-? and Bax.Development,differentiation and maturation of renal tubules and collecting tubules of metanephros might be due to the synergetic expression of EGFR,TGF-?,AQP-2 and Bax.AQP-2 relats to the balance of water during development and maturation of renal tubules in chick embryo.
2.Mechanism progress on enteric nervous system of acupuncture for slow transit constipation.

In recent years, according to the etiology and pathology researches of slow transit constipation (STC) STC is considered as a kind of "enteric neuropathy", indicating it is a kind of disease caused by abnormity of the enteric nervous system (ENS). Through reviewing the mechanism of acupuncture to regulate STC, it is found out that there is a close relationship between acupuncture regulating STC and ENS. Through various channels including ganglion cells, nerve plexus, neurotransmitter and TRPV1 (the primary sensory neurons receptor of the ENS) of the ENS, acupuncture is likely to make comprehensive adjustment on STC.

3.LUNG HISTOGENESIS AND EXPRESSION OF TGF-? IN BUFO RADDEI STRAUCH

Objective To analyze the tissue development rule of the Bufo raddei Strauch lung,to observe the expression of transforming growth factor-?(?_1,?_2,?_3)in developing lung,and to discuss their functions and relationships. Methods The tissue development of the(218) Bufo raddei Strauch lungs was studied by biological microscope technology,and the expression of TGF-? was measured in lung development by immunohistochemistry and images were analyzed by IPP,providing a basis for study of the zoology and developmental biology. Results(1) Lung bud occured in larval 22nd stage from pharynx caudal part.In 26th stage the minutenss locule began to appear.The lung bud gave rise to obvious lung cavity in 31st stage.In 33rd stage originality alveolus began to appear.In 36th stage obvious septum and alveolus appeared.There were many capillary vessel in the wall of lung.In 39th stage the lung of tadpole was like as one of young toad.The respiratory epithelium differentiated from columnar into squamous.The entire developmental process was divided into four phases,lung bud Ⅰphase,lung bud II phase,originality alveolus phase and alveolus phase.(2)In the early developmental phase,TGF-? expressed in the lung wall.In the late developmental phase,TGF-? expressed in mesenchymal at high levels.Conclusion The expression of TGF-? displayed intermittent changes during the period of development.The functions of TGF-? might be respectively different in different development stages.They not only stimulate the process of epithelium of the reticular septa,but also modulate epithelium of alveolar sac and differention.
4.Influencing factors of caries among Tibetan primary school studentsin Maqu County

Objective:
To investigate the prevalence and influencing factors of caries among Tibetan primary school students in Maqu County, Gansu Province, so as to provide insights into caries control among children living in ethnic minority areas.
Methods:
Tibetan primary school students at grades three to six were sampled from two primary schools in Maqu County using a multi-stage cluster sampling method. Primary school students' demographics, dietary habits and oral health habits were collected using questionnaire surveys, and the caries was tested. Factors affecting the development of caries were identified using a multivariable Poisson regression model.
Results:
A total of 378 Tibetan primary school students were tested, and there were 334 students with caries, with a caries prevalence rate of 86.30%, and a mean caries number of (4.72±2.69) teeth. Multivariable Poisson regression analysis showed that grade (grade 5, IRR=0.791, 95%CI: 0.706-0.886; grade 6, IRR=0.698, 95%CI: 0.593-0.822), frequency of vegetable intake (less than once daily, IRR=0.872, 95%CI: 0.778-0.978; once daily, IRR=0.769, 95%CI: 0.677-0.873; twice and more daily, IRR=0.571, 95%CI: 0.469-0.696), frequency of tooth brushing (once daily, IRR=0.814, 95%CI: 0.734-0.904; twice and more daily, IRR=0.606, 95%CI: 0.521-0.703) and frequency of oral examinations (IRR=0.736, 95%CI: 0.594-0.911) were influencing factors of caries.
Conclusion
The prevalence of caries is 86.30% among Tibetan primary school students at grades 3 to 6 in Maqu County, and age, dietary habit and oral health habit are factors affecting the development of caries.
5.Construction and identification of the double auxotrophic Candida parapolymorpha

Objective To construct a genetically-stable double auxotrophic,in which the uracil and leucine were mutated,using the Candida parapolymorpha ATCC26012 as materials.Methods Based on the physical and genetic engineering methods,the chromosome of the C.parapolymorpha strain was modified,where the ura3 and leu2 genes were directly mutated,to obtain the uracil and leucine double auxotrophic strain.Then the constructed strain was identified by the analysis of its biological properties,such as genetic stability,the change of the genes,and the physiologic and biochemical characteristics.Results The uracil and leucine double auxotrophic strain is obtained by screening.The biological identification results show that the obtained strain is genetically stable and the targeted genes are directly altered.In addition,the physiologic and biochemical analyses indicate that the auxotrophic can utilize various kinds of carbon and nitrogen nutrient sources,and its growth is good.Conclusion The successful construction of double auxotrophic mutant strains facilitated the genetic studies on C.parapolymorpha to meet various investigational purposes.Moreover,the constructed auxotrophic strains can be applied as advantageous host cells to express multiple proteins/antigens simultaneously,which is of great significance in the development of vaccines.

7.Comparison of Therapeutic Efficacy of Desflurane and Sevoflurane for Anesthesia of Elderly Patients

OBJECTIVE:To compare therapeutic efficacy and safety of desflurane and sevoflurane for anesthesia of elderly pa-tients. METHODS:A randomized,parallel-controlled method was used,120 elderly patients undergoing anesthesia were random-ized into trial group and control group,with 60 cases in each group. Based on routine medication and disposal,control group was additionally given sevoflurane for anesthesia,and trial group given desflurane for anesthesia. The blood pressure and heart rate were recorded before anesthesia,2 min and 10 min after intubation,2 min and 30 min after the start of the operation;the extubation time,eye opening time,consciousness recovery time and postoperative complications were also recorded. RESULTS:10 min after intubation,the heart rate of trial group was significantly faster then that of control group,with statistical significance(P<0.05);the blood pressure and heart rate of 2 groups had no statistically significant difference between 2 groups at other time points(P>0.05). The extubation time and consciousness recovery time of trial group were significantly shorter than those of control group, with statistical significance(P<0.05). The postoperative complications mainly were hypopiesia,hypertension,bronchospasm,nau-sea and vomiting,and dysphoria;the incidence of postoperative complications in trial group and control group were 25.0% and 40.0%,with statistical significance (P<0.05). CONCLUSIONS:Desflurane is effective with less postoperative complications for the anesthesia of elderly patients.
8.Clinical Observation of Low-dose Ropivacaine Combined with Sufentanil in Upper Limb Anesthesia

OBJECTIVE:To observe the efficacy and safety of low-dose ropivacaine combined with sufentanil in the upper limb anesthesia. METHODS:100 patients with upper limb anesthesia were randomly divided into control group and test group. Control group was intravenously injected 20 ml 0.5% ropivacaine;test group was intravenously injected 20 ml 0.375% ropivacaine +10 μg sufentanil. The anesthesia effect,disappearance time of pain,duration of analgesia,and pain visual analogue (VAS) score before anesthesia(T0),5 min(T1),15 min(T2)and 30 min(T3)after anesthetic block,and 4 h(T4)and 8 h(T5)after surgery,and in-cidence of adverse reactions in 2 groups were observed. RESULTS:VAS scores at T1-T5 in test group and T2-T4 in control group were significantly lower than T0,test group was lower than control group at T1,T2 and T5,the differences were statistically signifi-cant(P<0.05). The total effective rate of anesthesia in test group was significantly higher than control group,disappearance time of pain was significantly shorter than control group,duration of analgesia was significantly longer than control group,incidence of adverse reactions was significantly lower than control group,the differences were statistically significant(P<0.05). CONCLU-SIONS:Both the efficacy and safety of low-dose ropivacaine combined with sufentanil in the upper limb anesthesia are good.
9.Neuroprotective effects of sufentanil preconditioning on spinal cord injury in mouse models

BACKGROUND:Sufentanil exerts protective effects on tissues, but its roles in the repair of nervous system injury and the underlying mechanism are stil unknown.
OBJECTIVE:To explore the protective effect of sufentanil preconditioning in the repair of spinal cord injuries and the underlying mechanism.
METHODS:Mouse models of spinal cord injuries were prepared through clipping spinal cord fol owed by intraperitoneal injection of 3 and 6μg/kg sufentanil, respectively.
RESULTS AND CONCLUISON:(1) Western blotting, ELISA and TUNEL assays showed that 6μg/kg sufentanil significantly down-regulated the protein expression levels of TLR4, nuclear factor-κBp65, cleaved-caspase 3, tumor necrosis factor-αand interlenkin-1βin the spinal cord of mice (P<0.05);at the same time, the number of apoptotic neurons was significantly decreased (P<0.05). (2) Furthermore, high-dose sufentanil preconditioning significantly ameliorated the recovery of limb function at 14 days after injury (P<0.05). (3) These results administrate that the neuroprotection provided by 6μg/kg sufentanil preconditioning for spinal cord injuries in mice maybe related to the TLR4/nuclear factor-κB signaling pathway inactivation.
10.Analysis of the Use of Analgesics in 100 Postoperative Patients

Objective:To analyze the analgesic method and analgesic use for postoperative patients in our hospital. Method:100 cases with postoperative pain in the general surgery department,orthopedic department and gynecological department were chosen from March to June in 2009.Their analgesic method,analgesic use,visual analog scale(VAS) scores and adverse drug reaction were analyzed.Result:The rate of patient-controlled analgesia(PCA) use was 25.0%. The cases who used opioid and non-steroidal anti-inflammatory drugs(NSAIDs) in 3 days after operations were 20 and 44, respectively.The rate of the drug combination was low.Pethidineand diclofenac potassium were major drugs to relieve their postoperative pain.VAS score in 1 day post-operation was 3.43.Conclusion:The use of analgesics was positive in our hospital, but the insufficient analgesia and little drug combination existed,and drug selection is also limited.
