1.Effects of PB-19 on ischemia-reperfusion arrhythmia in rats

Aim To investigate the effects of PB-19 on action potential (AP) of ventricular cells in vitro and ischemia and reperfusion induced arrhythmias in vivo in rats. Methods ① We established the model of isolated rat right ventricular wall and perfused the models with normal Tyrode’s solution and PB-19.Microelectrode was inserted into the cells to record the AP. ② We ligated the left anterior descending coronary artery of rats to study the effects of PB-19 on ischemia and reperfusion induced arrhythmias. 50 rats were divided into 5 groups randomly and were given iv. injection with normal saline and PB-19 of different concentrations respectively. 2.Effects of bradykinin preconditioning on cerebral ischemia

Aim To investigate the effects of bradykinin preconditioning on the damage produced by focal cerebral ischemia-reperfusion in rats.Methods Rats were scheduled to undergo middle cerebral artery ischemia-reperfusion by intraluminal suture.Prior to ischemia,bradykinin was pumped into the brain via extra carotid artery and control group was given the same amount of normal saline.The infarct volume、brain water content、permeability of blood brain barrier and histological neuronal changes were evaluated after 2 h ischemia and 24 h reperfusion.Results Compared with other groups, bradykinin preconditioning 15min before ischemia reduced infarct volume、brain edema、permeability of blood brain barrier and histological neuronal damage.Conclusion Bradykinin preconditioning may provide protective effects against cerebral ischemic injury.This protection may be due to the protection of cerebral vasculature and the decrease of infarct volume.
3.Nursing care after heart valve replacement operation for patients with cardiomegaly

Objectives:To discuss the characteristics of nursing care of patients with cardiomegaly after heart valve replacement operation. Methods:From October 1997 to January 2000, 65 patients with cardiomegaly received heart valve replacement operations. According to the degree of cardiomegaly, these patients were divided into 3 types: left atrium, left ventricle and whole heart cardiomegaly. Different nursings were applied depending on the features of each type. Results:Complications occurred in 28 patients(12 in left atrium,11 in left ventricle and 5 in whole heart cardiomegaly),all of whom were cured;2 patients died, the mortality rate was 3.08%. Conclusions: To lower the morbidity and mortality, different kinds of nursing care should be applied to different cardiomegaly after heart valve replacement operations.
4.Effect of femoral intramedullary guides on prosthesis arrangement in total knee arthroplasty

BACKGROUND: The femoral intramedul ary guides in total knee arthroplasty require high precision, complex operation, it is very important for prosthesis and joint function to choose more precise positioning method and determine the correct needle point. OBJECTIVE: To study the effect of needle point position on prosthesis arrangement when applying different femoral intramedul ary guides methods in total knee arthroplasty. METHODS: Total y 80 patients who received the treatment of total knee arthroplasty in Changshu No.1 People’s Hospital from January 2012 to July 2015 were selected and divided into test and control groups according to random number table (n=40/group). The patients in the test group accepted CT scan for femoral intramedul ary guides. The theoretical position of femoral intramedul ary guides entry point was marked using radiographic parameters. In the control group, the traditional total knee arthroplasty technology was used to mark the entry point of femoral intramedul ary guides. The needle point position when applying different femoral intramedul ary guide methods was observed. The effect of femoral intramedul ary guides on prosthesis arrangement in total knee arthroplasty was discussed. RESULTS AND CONCLUSION: Compared with the control group, the distance from entry point to femoral anatomic line on positive and lateral X-ray film in the test group was shorter, femoral prosthesis lateral angle and physiological valgus angle were closer to the theoretical value, distance from intersection of femur axis and femoral condyle to block center was shorter; the differences were statistical y significant (P < 0.05). These results demonstrate that compared with the traditional two-dimensional intramedul ary positioning, the needle point position of three-dimensional CT scan stimulative positioning is more accurate. The location more concentrates on within intercondylar fossa 2-5 mm, more front of intercondylar fossa 3-10 mm, three-dimensional CT scan stimulative positioning is a reliable choice for femoral intramedul ary guides.
5.Total knee arthroplasty with Gemini MKII rotating-platform prosthesis

BACKGROUND:Total knee arthroplasty mainly used fixed platform, but more and more movable platform was selected year by year in the clinic. Rotary platform theoretical y can apparently improve the curative effects, but it has not been verified in the clinic.
OBJECTIVE:To summarize the design characteristics and clinical results of Gemini MKII (Link, Germany) rotary platform.
METHODS:From January 2004 to January 2009, 108 patients (119 knees) including 31 males and 77 females were treated with total knee arthroplasty by using Gemini MKII system. There were 97 unilateral cases and 11 bilateral cases at the first stage of replacement. Preoperative diagnosis:1 case of hemophilic arthritis, 1 case of venereal Charcot’s disease, 8 cases of post-traumatic arthritis, 11 cases of rheumatoid arthritis, and 87 cases of degenerative osteoarthritis. The prosthesis was fixed with bone cement. None received patel ar replacement. RESULTS AND CONCLUSION:None experienced infection, femoral condyle fracture or nerve and vascular damage. Except bilateral replacement, the average operation time of unilateral replacement was (56±13) minutes. Average postoperative hemoglobin decreased (25±5) g/L. After replacement, hemoglobin maintained over 100 g/L in 83 patients (86%), without blood transfusion. Knee joint score and function score recovered to (92.0±4.7) points and (90.3±6.8) points at half years after replacement, and became (94.4±3.2) points and (91.6±5.9) points during final fol ow-up, which was significantly improved as compared with that pre-replacement (P<0.01). No revision surgery should be done for lining prolapse or spin out, osteolysis and prosthesis loosening. These results indicated that the early and midterm results of Gemini MKII rotating-platform prosthesis were satisfactory, but the long-term results remain to be confirmed.

This article was aimed to study the acute toxicities on intragastric administration of differentRadix Aconiti Lateralis Praeparataprocessed products to Beagle dogs. A total of 16 healthy and qualified Beagle dogs were randomly divided into the blank group,Pao-Fu-Pian(PFP) group,Pao-Tian-Xiong(PTX) group andHei-Shun-Pian(HSP) group according to the body weight. The intragastric administration of 4 g crude herb per kg was given. Before medication, 1 h, 24 h, and 3, 7, 14 days after medication, the body weight, food consumption, rectal temperature, electrocardiogram, blood routine and blood biochemistry were measured. The results showed that after medication, all dogs in three experimental groups were depressed. And there were significant differences in the electrolytes of blood. Among them, the HSP group was the most obvious one. The red blood cells, blood sugar and triglycerides of dogs in the PFP group had significant difference. The lymphocytes and blood sugar had significant difference of dogs in the PTX group. However, after the medication of HSP, the lymphocytes of the dogs were decreased significantly. It was concluded that the toxicity of three processed products followed the order of HSP > PFP > PTX.
7.Construction, identification and immunoreactivity of Mycobacterium tuberculosis DNA vaccine pVAX1/ESAT-6 plasmid.

OBJECTIVETo construct a Mycobacterium tuberculosis DNA vaccine pVAX1/ESAT-6 plasmid and investigate its immunoreactivity.
METHODSThe ESAT-6 gene fragment amplified from Mycobacterium tuberculosis genome was inserted into pVAX1 vector to construct the recombinant plasmid pVAX1/ESAT-6, which was identified by restriction enzyme digestion and sequencing. The recombinant plasmid was transformed into Hela cells using Sofast® Transfection reagent, and the cellular expressions of ESAT-6 mRNA and protein were analyzed by RT-PCR and immunofluorescence assay, respectively. The recombinant plasmid pVAX1/ESAT-6 was also transfected into mouse by electronic pulse method, and the mouse serum IFN-γ level and anti-ESAT-6 IgG antibody level were detected by ELISA, mouse lymphocyte proliferation assessed with flow cytometry, and IFN-γ-secreting lymphocytes counted using ELISPOT.
RESULTSDouble restriction-enzyme digestion and sequencing showed that the inserted fragment in the recombinant plasmid pVAX1/ESAT-6 was identical to ESAT-6 gene with an inframe insertion. RT-PCR yielded the target band as expected on agarose gel, and immunofluorescence assay of the transfected cells showed specific green fluorescence signals. The mice transfected with the recombinant plasmid showed significantly elevated serum level of anti-ESAT-6 IgG antibody and increased serum IFN-γ level, spleen cell proliferation, and number of IFN-γ-secreting lymphocytes.
CONCLUSIONThe Mycobacterium tuberculosis DNA vaccine pVAX1/ESAT-6 plasmid we constructed can induce high levels of cellular and humoral immunoreactivity in mice.

8.IL-22 and immunity in female reproductive tract

IL-22 is a member of IL-10 cytokine family. In recent years, increasing evidence has shown that IL-22 is closely related to the immunity in female reproductive tract, and its role in disease development is two-sided. It can not only maintain the balance of microbiota, enhance the resistance to pathogens and reduce the tissue damage caused by infection, but also promote the development and progression of malignant diseases via various signaling pathways. More studies on the biological characteristics and functions of IL-22 are needed for clarifying the pathogenic mechanism and providing new insight into the diagnosis and treatment of female reproductive tract diseases.
9.Progress of adipokines in cancer treatment

Obesity is a well-known high-risk factor for many kinds of neoplasms. Adipokines secreted by adipose tissue play an important role in the process of obesity promoting tumorigenesis and development, and different adipokines play a role in promoting or suppressing cancer via different signaling pathways. Currently, lifestyle modification to control weight and targeted therapy of adipokines and their receptors are major research directions of cancer treatment, but most of the studies are still in the stage of basic and pre-clinical research. Therefore, the molecular mechanisms of adipokines in promoting or suppressing cancer should be further explored, and the dual inhibitors as well as combined therapy are the key research strategies for adipokines in cancer treatment in the future.
10.Qualitative researchon the psychological feelings during radiological examination of parents of children fracture of ulna and radius

Objective:To investigate the psychological feelings of parents of children with fracture of ulna and radius during radiological examination, and to provide the basis for taking scientific and effective intervention program.Methods:Using the purposive sampling methods, a total of 30 parents of children who were treated in the pediatric orthopedics department of the emergency department of Beijing Jishuitan Hospital affiliated to Capital Medical University from January 2022 to June 2023 were selected as the research objects. A descriptive research method was adopted to conduct semi-structured interviews with parents of children, and Colaizzi′s phenomenological analysis was applied to analyze, refine, and summarize the interview information.Results:Four themes were summarized: negative emotions, information needs, seeking multi-party support and post-traumatic growth.Conclusions:The medical staff should pay close attention to the psychological status of the parents, meet their needs for information during the radiological examination, and provide targeted support measures to promote the prognosis of the children, improve the psychological adjustment and quality of life of the children and their parents.
